| 1 | Euphorbia factor L2 induces apoptosis in A549 cells through the mitochondrial pathway显示文摘Euphorbia factor L2, a lathyrane diterpenoid isolated from caper euphorbia seed(the seeds of Euphorbia lathyris L.), has been traditionally applied to treat cancer. This article focuses on the cytotoxic activity of Euphorbia factor L2 against lung carcinoma A549 cells and the mechanism by which apoptosis is induced. We analyzed the cytotoxicity and related mechanism of Euphorbia factor L2 with an MTT assay, an annexin V-FITC/PI test, a colorimetric assay, and immunoblotting. Euphorbia factor L2 showed potent cytotoxicity to A549 cells. Euphorbia factor L2 led to an increase in reactive oxygen species(ROS) generation,a loss of mitochondrial electrochemical potential, release of cytochrome c, activation of caspase-9 and caspase-3, and cleavage of poly(ADP-ribose) polymerase, suggesting that Euphorbia factor L2 induced apoptosis through a mitochondrial pathway. | 4 | Remineralization of enamel caries by an amelogenin-derived peptide and fluoride in vitro显示文摘Dental caries is one of the most common oral diseases in the world.This study was tantamount to investigate the combinatory effects of an amelogenin-derived peptide(called QP5)and fluoride on the remineralization of artificial enamel caries.The peptide QP5 was synthesized and characterized,and the binding capability of the peptide on hydroxyapatite(HA)and demineralized tooth enamel surface was analysed.Then,the mineralization function of the peptide and fluoride was studied through the spontaneous mineralization testing and remineralization on enamel caries in vitro.First,the novel peptide QP5 could bind on the hydroxyapatite and demineralized tooth enamel surfaces.Second,QP5 can transitorily stabilize the formation of amorphous calcium phosphate and direct the transformation into hydroxyapatite crystals alone and in combination with fluoride.In addition,compared to blocks treated by peptide QP5 alone or fluoride,the sample blocks showed significantly higher surface microhardness,lower mineral loss and shallower lesion depth after treatment with a combination of QP5 and fluoride at high or low concentrations.The peptide QP5 could control the crystallization of hydroxyapatite,and combinatory application of peptide QP5 and fluoride had a potential synergistic effect on the remineralization of enamel caries. | Longjiang Ding Sili Han Kun Wang Sainan Zheng Wenyue Zheng Xiu Peng Yumei Niu Wei Li Linglin Zhang | 2020 | Regenerative Biomaterials2020,7,3: | 6 |
| 5 | Design and syntheses of functionalized copper-based MOFs and its adsorption behavior for Pb(Ⅱ)显示文摘A novel copper-based MOFs adsorbent(Cu-BTC-Th) was prepared using an one-step method by introducing a new organic ligand of 4-thioureidobenzoicacid(Th) with active groups for selectively adsorbing Pb(Ⅱ) from aqueous solutions. The chemical composition and structure of the prepared MOFs materials were characterized by scanning electron microscope(SEM), X-ray diffraction(XRD), fourier transform infrared spectroscopy(FTIR), X-ray photoelectron spectroscopy(XPS), Brunner-Emmet-Teller(BET) analysis, and zeta potential measurements. The adsorption capability of the prepared Cu-MOFs was significantly enhanced by introducing the new organic ligand of Th in the materials. The maximum adsorption capacity of the Cu-BTC-Th for Pb(Ⅱ) attains 732.86 mg/g under the optimal conditions. In addition,the adsorption kinetics and adsorption isotherm analysis showed that the adsorption process followed the pseudo-second-order kinetic model and Langmuir adsorption model, indicating that the adsorption of Pb(Ⅱ) by Cu-BTC-Th was a monolayer chemisorption. The adsorption mechanism of Cu-BTC-Th for Pb(Ⅱ)was discussed and revealed. On one hand, the adsorption of Pb(Ⅱ) is mainly through ion exchange with the Cu(Ⅱ). | 7 | Endothelial METRNL determines circulating METRNL level and maintains endothelial function against atherosclerosis显示文摘METRNL is a recently identified secreted protein with emerging functions.This study is to find major cellular source of circulating METRNL and to determine METRNL novel function.Here,we show METRNL is abundant in human and mouse vascular endothelium and released by endothelial cells using endoplasmic reticulum-Golgi apparatus pathway.By creating endothelial cell-specific Metrnl knockout mice,combined with bone marrow transplantation to produce bone marrow-specific deletion of Metrnl,we demonstrate that most of circulating METRNL(approximately 75%)originates from the endothelial cells.Both endothelial and circulating METRNL decrease in atherosclerosis mice and patients.By generating endothelial cell-specific Metrnl knockout in apolipoprotein E-deficient mice,combined with bone marrow-specific deletion of Metrnl in apolipoprotein E-deficient mice,we further demonstrate that endothelial METRNL deficiency accelerates atherosclerosis.Mechanically,endothelial METRNL deficiency causes vascular endothelial dysfunction including vasodilation impairment via reducing eNOS phosphorylation at Ser1177 and inflammation activation via enhancing NFκB pathway,which promotes the susceptibility of atherosclerosis.Exogenous METRNL rescues METRNL deficiency induced endothelial dysfunction.These findings reveal that METRNL is a new endothelial substance not only determining the circulating METRNL level but also regulating endothelial function for vascular health and disease.METRNL is a therapeutic target against endothelial dysfunction and atherosclerosis. | Sili Zheng Zhiyong Li Jie Song Pin Wang Jian Xu Wenjun Hu Yi Shi Qi Qi Zhuwei Miao Yunfeng Guan Chaoyu Miao | 2023 | Acta Pharmaceutica Sinica B2023,13,4: | 2 |
