| 5 | The protection effect of β-CD on DNA damage induced by ultrafine TiO_2显示文摘Under the photocatalysis of 365 nm ultraviolet radiation,ultrafine TiO2 caused the oxidative damage of Teasy plasmid DNA. The damage was determined by gel-electrophoresis. Then,a different dose of β-CD was added to the reaction,and the damage was restrained. The rate of damage restraining reached 97% when the mass of β-CD was 4 times as that of TiO2. Through UV scan and IR spectroscopy,it was found that the Ti-O of ultrafine TiO2 was bound with -OH of β-CD cavum and the -OH on the surface of ultrafine TiO2 disappeared,so the formation of ?OH was controlled. The ultrafine TiO2 has been widely used,but it was determined to be carcinogenic by some research. | 6 | In-situ synthesis of PHEMA magnetic nanogels via photochemical method显示文摘One-pot synthesis of magnetic nanogels via photochemical method is reported in this paper. Poly(2-hydroxyethyl methacrylate)(PHEMA) magnetic nanogels are synthesized by in-situ polymeriza-tion of 2-hydroxyethyl methacrylate(HEMA) and N,N'-methylene-bis-(acrylamide)(MBA) in Fe3O4 aqueous suspension under UV irradiation. The structure and compositions of magnetic nanogels are characterized by FTIR,TGA,SEM,TEM and PCS. TGA measurement indicates that magnetic nanogels contain 90% magnetite. Both naked Fe3O4 and magnetic nanogels are superparamagnatic at room temperature according to magnetization curves. The swollen capability of the hydrogel shell is proved by contrasting the particles sizes obtained by SEM,TEM and PCS. Particle diameters can be manipu-lated by changing monomer concentration and irradiation time. | 7 | Orbital control on cyclical organic matter accumulation in Early Silurian Longmaxi Formation shales显示文摘High resolution(939 samples)total organic carbon content(TOC)analyses were conducted on the Shuanghe Section of^152.6 m in the Changning area,Sichuan Basin.The sampling section was divided into two units considering the distinct-different deposit environment and sediments accumulation rate.The lower part(Unit 1)and the peer part(Unit 2)with high resolution sample spacing(0.08–0.4 m)enables the identification of the precession cycle in two sedimentary sequences with distinct different sedimentary accumulation rate.MTM Power spectral analyses on untuned TOC series reveals significant peaks exceeding above the 95%confidence level and shows that both Unit 1 and Unit 2 have recorded Milankovitch cycles of 405 kyr long eccentricity,short eccentricity,obliquity and precession.The floating astronomical time scale(ATS)was constructed on the Shuanghe Section in the Early Silurian(~439.673–444.681 Ma),and which was calibrated by 405 kyr long eccentricity cycles.The total duration of the Wufeng and Longmaxi shales is 5.01 Myr.The floating ATS used for estimating the duration of the graptolite zones and each stage in the study interval.Finally,we postulated two models that could verify the linkage between orbital cycle and organic accumulation.To make sure whether productivity or preservation is the main factor that under long eccentricity control,the phase correlation between the obtained filtered signal and the theoretical orbital solution should be made clear in the further research. | Siding Jin Hucheng Deng Xing Zhu Yan Liu Sibing Liu Meiyan Fu | 2020 | Geoscience Frontiers2020,11,2: | 4 |

| 11 | Impact and mechanism of TiO_2 nanoparticles on DNA synthesis in vitro显示文摘The impact of TiO2 nanoparticles on DNA synthesis in vitro in the dark and the molecular mechanism of such impact were studied. The impact of TiO2 nanoparticles on DNA synthesis was investigated by adding TiO2 nanoparticles in different sizes and at various concentrations into the polymerase chain reaction (PCR) system. TiO2 nanoparticles were premixed with the DNA polymerase, the primer or the template, respectively and then the supernatant and the precipitation of each mixture were added into the PCR system separately to observe the impact on DNA synthesis. Sequentially the interaction be- tween TiO2 nanoparticles and the DNA polymerase, the primer or the template was further analyzed by using UV-visible spectroscopy and polyacrylamide gel electrophoresis (PAGE). The results suggest that TiO2 nanoparticles inhibit DNA synthesis in the PCR system in the dark more severely than mi- croscale TiO2 particles at the equivalent concentration and the inhibition effect of TiO2 nanoparticles is concentration dependent. The molecular mechanism of such inhibition is that in the dark, TiO2 nanoparticles interact with the DNA polymerase through physical adsorption while TiO2 nanoparticles do with the primer or the template in a chemical adsorption manner. | 16 | Construction and expression of nucleic acid vaccine pVAX1-h-alpha S_(1-140) coding human alpha-synuclein显示文摘BACKGROUND: The deposition of α-synuclein (α-syn) aggregates is a neuropathological feature of Parkinson’s disease. It remains impossible to involve α-syn aggregation in the treatment of Parkinson’s disease. A nucleic acid vaccine will provide a new pathway to immunotherapy for Parkinson’s disease. OBJECTIVE: To construct a recombinant eukaryotic expression vector pVAX1 coding human α-syn and to observe its expression level in COS-7 cells. DESIGN AND SETTING: The present bioengineering and molecular biology experiment was performed at Department of Neurology, First Affiliated Hospital of Chongqing Medical University & Chongqing Key Laboratory of Neurology. MATERIALS: The eukaryotic expression plasmid pVAX1, human embryonic brain tissue, healthy human blood cells, and COS-7 cells were purchased from Promega Company, USA. METHODS: The full-length CDS sequence of the human α-syn gene was amplified by RT-PCR, which contained restriction sites for the enzymes KpnⅠ, XbaⅠand Kozak consensus sequence. Then the PCR products and eukaryotic expression vector pVAX1 were digested with KpnⅠand XbaⅠsimultaneously, and were extracted and ligated by T4 ligase. The recombinant constructs pVAX1-hα-S1-140 were transformed into competent E. coli TOP 10 cells and the positive clones were screened and selected using PCR analysis, restriction digestion analysis, and DNA sequencing. The constructs were then tested for protein expression in COS-7 cells by RT-PCR and Western blotting. MAIN OUTCOME MEASURES: Identification of an eukaryotic expression vector containing the human α-syn gene, pVAX1-hα-S1-140, and detection of the expression in mammalian cell COS-7. RESULTS: The pVAX1 vector was successfully cloned with humanα-syn in the correct orientation and in-frame. The DNA vaccine constructs pVAX1-hα-S1-140 with the humanα-syn gene were shown to be expressed in COS-7 cells. Humanα-syn was successfully expressed in the mammalian cell line and was detected by RT-PCR and western blotting. | 17 | Poly(PEGMA) magnetic nanogels:Preparation via photochemical method,characterization and application as drug carrier显示文摘One-pot synthesis of magnetic nanogels with excellent biocompatibility via the photochemical method is reported in this paper.Poly(PEGMA) modified superparamagnetic nanogels(poly(PEGMA) magnetic nanogels) were synthesized by in-situ polymerization using poly(ethylene glycol) methacrylate(PEGMA) as the monomer and N,N'-methylene-bis-(acrylamide)(MBA) as the cross-linking agent in magnetite aqueous suspension under UV irradiation.The surface functional groups and components of magnetic nanogels were analyzed by Fourier transform infrared spectroscopy(FTIR) and a thermogra-vimetric analyzer(TGA).The results indicated that the poly(PEGMA) magnetic nanogels were synthe-sized successfully by coating poly(PEGMA) on the Fe3O4 nanoparticles under UV irradiation,and the Fe3O4 nanoparticles content in this nanogels was above 50 wt%.The morphology,size,zeta-potential and magnetic property were also characterized.The magnetic nanogels had a nearly spherical shape and core-shell structure,the average size in aqueous system measured by photon correlation spec-troscopy(PCS) was 68.4 nm,which was much bigger than that in the dry state,the nanogels behaved superparamagnetically with saturated magnetization of 58.6 emu/g,and the zeta-potential was-16.3-_17.3 mV at physiological pH(6.8-7.4) which could help to maintain stability in blood.The preliminary application as drug carrier was made and the doxorubicin-loaded magnetic nanogels had an excellent property in slow-release.The experiment indicated that the magnetic nanogel was an ideal candidate carrier in target drug delivery systems and other biomedical application. | SUN HanWen ZHANG LianYing ZHU XinJun KONG ChunYan ZHANG CunLan YAO SiDe | 2009 | Science China Chemistry2009,52,1: | 2 |
