2A 2-kb Mycovirus Converts a Pathogenic Fungus into a Beneficial Endophyte for Brassica Protection and Yield Enhancement显示文摘Mycoviruses are viruses that infect fungi,and hypovirulence-associated mycoviruses have the potential to control fungal diseases.However,it is unclear how mycovirus-mediated hypovirulent strains live and survive in the field,and no mycovirus has been applied for field crop protection.In this study,we found that a previously identified small DNA mycovirus(SsHADV-1)can convert its host,Sclerotinia sclerotiorum,from a typical necrotrophic pathogen to a beneficial endophytic fungus.SsHADV-1 downregulates the expression of key pathogenicity factor genes in S.sclerotiorum during infection.When growing in rapeseed,the SsHADV-1-infected strain DT-8 significantly regulates the expression of rapeseed genes involved in defense,hormone signaling,and circadian rhythm pathways.As a result,plant growth is promoted and disease resistance is enhanced.Field experiments showed that spraying DT-8 at the early flowering stage can reduce the disease severity of rapeseed stem rot by 67.6%and improve yield by 14.9%.Moreover,we discovered that SsHADV-1 could also infect other S.sclerotiorum strains on DT-8-inoculated plants and that DT-8 could be recovered from dead plants.These findings suggest that the mycoviruses may have the ability to shape the origin of endophytism.Our discoveries suggest that mycoviruses may influence the origin of endophytism and may also offer a novel strategy for disease control in which mycovirus-infected strains are used to improve crop health and release mycoviruses into the field.Hongxiang Zhang Jiatao Xie Yanping Fu Jiasen Cheng Zheng Qu Zhenzhen Zhao Shufen Cheng Tao Chen Bo Li Qianqian Wang Xinqiang Liu Binnian Tian David B.Collinge Daohong Jiang 2020Molecular Plant2020,13,10:9
3Characterization of glucose-6-phosphate dehydrogenase deficiency and identification of a novel haplotype 487G>A/IVS5-612(G>C) in the Achang population of southwestern China显示文摘The prevalence of glucose-6-phosphate dehydrogenase (G6PD) deficiency and its gene mutations were studied in the Achang population from Lianghe County in Southwestern China. We found that 7.31% (19 of 260) males and 4.35% (10 of 230) females had G6PD deficiency. The molecular analysis of G6PD gene exons 2―13 was performed by a PCR-DHPLC-Sequencing or PCR-Sequencing. Sixteen inde-pendent subjects with G6PD Mahidol (487G>A) and the new polymorphism IVS5-612 (G>C), which combined into a novel haplotype, were identified accounting for 84.2% (16/19). And 100% Achang G6PD Mahidol were linked to the IVS5-612 C. The percentage of G6PD Mahidol in the Achang group is close to that in the Myanmar population (91.3% 73/80), which implies that there are some gene flows between Achang and Myanmar populations. Interestingly, G6PD Canton (1376G>T) and G6PD Kaiping (1388G>A), which were the most common G6PD variants from other ethnic groups in China, were not found in this Achang group, suggesting that there are different G6PD mutation profiles in the Achang group and other ethnic groups in China. 4Fractality of grain composition of debris flows显示文摘Debris flows in essence are the process of mass transportation controlled by the constitution featured by a wide-ranged distribution of grain size. Debris-flow samples of different densities collected from different regions and gullies reveal that cumulative curve of grain composition, in particular for debris flows of high density, ps >2 g/cm3, can be fitted well by exponential function with exponents varying with regions and gullies. Debris flows fall into a narrow-valued domain of the exponent, as evidenced by Jiangjiagou Gully (JJG) with high occurrence frequency of debris flows. Furthermore, fractality of grain composition and porosity have been derived from cumulative curves in a certain size range, a range that determines the upper limit of grains constituting the matrix of debris flows. 5Neonatal Maternal Deprivation Followed by Adult Stress Enhances Adrenergic Signaling to Advance Visceral Hypersensitivity显示文摘The pathophysiology of visceral pain in patients with irritable bowel syndrome remains largely unknown.Our previous study showed that neonatal maternal deprivation(NMD) does not induce visceral hypersensitivity at the age of 6 weeks in rats. The aim of this study was to determine whether NMD followed by adult stress at the age of 6 weeks induces visceral pain in rats and to investigate the roles of adrenergic signaling in visceral pain. Here we showed that NMD rats exhibited visceral hypersensitivity 6 h and 24 h after the termination of adult multiple stressors(AMSs). The plasma level of norepinephrine was significantly increased in NMD rats after AMSs. Whole-cell patch-clamp recording showed that the excitability of dorsal root ganglion(DRG) neurons from NMD rats with AMSs was remarkably increased. The expression of β_2 adrenergic receptors at the protein and mRNA levels was markedly higher in NMD rats with AMSs than in rats with NMD alone. Inhibition of β_2 adrenergic receptors with propranolol or butoxamine enhanced the colorectal distention threshold and application of butoxamine also reversed the enhanced hypersensitivity of DRG neurons. 6Study on correlation between humic acid and selenium in Kaschin-Beck disease areas显示文摘The relationship between humic acid and selenium content was studied in this research. The results of calculation showed that there is more humic acid and less selenium in Kaschin-Beck disease areas in China. Fulvic acid in soil could increase selenium level in plants. The chemical bonding between humic acid and selenium was observed with an I.R. spectroscope. 9SARS-CoV-2 triggers inflammatory responses and cell death through caspase-8 activation显示文摘Severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)infection can lead to respiratory illness and multi-organ failure in critically ill patients.Although the virus-induced lung damage and inflammatory cytokine storm are believed to be directly associated with coronavirus disease 2019(COVID-19)clinical manifestations,the underlying mechanisms of virus-triggered inflammatory responses are currently unknown.Here we report that SARS-CoV-2 infection activates caspase-8 to trigger cell apoptosis and inflammatory cytokine processing in the lung epithelial cells.The processed inflammatory cytokines are released through the virus-induced necroptosis pathway.Virus-induced apoptosis,necroptosis,and inflammation activation were also observed in the lung sections of SARS-CoV-2-infected HFH4-hACE2 transgenic mouse model,a valid model for studying SARS-CoV-2 pathogenesis.Furthermore,analysis of the postmortem lung sections of fatal COVID-19 patients revealed not only apoptosis and necroptosis but also massive inflammatory cell infiltration,necrotic cell debris,and pulmonary interstitial fibrosis,typical of immune pathogenesis in the lung.The SARS-CoV-2 infection triggered a dual mode of cell death pathways and caspase-8-dependent inflammatory responses may lead to the lung damage in the COVID-19 patients.These discoveries might assist the development of therapeutic strategies to treat COVID-19.Shufen Li Yulan Zhang Zhenqiong Guan Huiling Li Meidi Ye Xi Chen Jun Shen Yiwu Zhou Zheng-Li Shi Peng Zhou Ke Peng 2020Signal Transduction and Targeted Therapy2020,5,1:3
10Overexpressed long noncoding RNA CRNDE with distinct alternatively spliced isoforms in multiple cancers显示文摘Alternative splicing is a tightly regulated process that contributes to cancer development.CRNDE is a long noncoding RNA with alternative splicing and is implicated in the pathogenesis of several cancers.However,whether deregulated expression of CRNDE is common and which isoforms are mainly involved in cancers remain unclear.In this study,we report that CRNDE is aberrantly expressed in the majority of solid and hematopoietic malignancies.The investigation of CRNDE expression in normal samples revealed that CRNDE was expressed in a tissue- and cell-specific manner.Further comparison of CRNDE expression in 2938 patient samples from 15 solid and hematopoietic tumors showed that CRNDE was significantly overexpressed in 11 malignancies,including 3 reported and 8 unreported,and also implicated that the overexpressed isoforms differed in various cancer types.Furthermore,anti-cancer drugs could efficiently repress CRNDE overexpression in cancer cell lines and primary samples,and even had different impacts on the expression of CRNDE isoforms.Finally,experimental profiles of 12 alternatively spliced isoforms demonstrated that the spliced variant CRNDE-g was the most highly expressed isoform in multiple cancer types.Collectively,our results emphasize the cancer-associated feature of CRNDE and its spliced isoforms,and may provide promising targets for cancer diagnosis and therapy.Xuefei Ma Wei Zhang Rong Zhang Jingming Li Shufen Li Yunlin Ma Wen Jin Kankan Wang 2019Frontiers of Medicine2019,13,3:3
11Pan-cancer analysis of long non-coding RNA NEAT1 in various cancers显示文摘Changes in the abundance and activity of long non-coding RNAs(lncRNAs)have an important impact on the development of cancer.The nuclear paraspeckle assembly transcript 1(NEAT1)has been reported to be overexpressed in many types of cancer since its discovery.However,inconsistencies exist as NEAT1 can also function as a tumor suppressor in certain types of cancer,such as acute promyelocytic leukemia.Here we systematically describe our current understanding of NEAT1 in tumor initiation and progression.First,we analyzed the expression patterns of NEAT1 in various normal tissues and malignant cancers using data from public data portals,the Genotype-Tissue Expression Project(GTEx)and the Cancer Genome Atlas(TCGA),together with recent progress in the study of NEAT1 in various types of cancer.Second,we discussed the functions and mechanisms of NEAT1 in modulating tumor activity.Then,the upstream transcription factors and downstream microRNA targets of NEAT1 in the transcription cascade of cancers were also summarized.These data highlight the emerging role of NEAT1 in tumorigenesis,and present promising targetable pathways and clinical opportunities for tumor prevention and classifications.Shufen Li Jingming Li Chen Chen Rongsheng Zhang Kankan Wang 2018Genes & Diseases2018,5,1:2
12The model study of water mass and energy exchange between the inland water body and atmosphere显示文摘Based on a one-dimensional eddy diffusion model,a model to study the water mass and energy exchange between the water body(such as lake and wetland) and the atmosphere is developed,which takes the phase change process due to the seasonal melting and freezing of water and the convection mixing process of energy caused by temperature stratification into consideration. The model uses enthalpy instead of temperature as predictive variable,which will help to deal with the phase change process and to design an efficient numerical scheme for obtaining the solution more easily. The performance of the model and the rationality of taking convection mixing into the consideration are validated by using observed data of Kinneret Lake in Israel and Lower Two Medicine Lake in Montana State in America. The comparison of model results with observed data indicates that the model presented here is capable of describing the physical process of water mass and energy between the water body(lake and wetland) and atmosphere.
