| 1 | β-elemene enhances the radiosensitivity of gastric cancer cells by inhibiting Pak1 activation显示文摘AIM:To explore the potential of β-elemene as a radiosensitizer for gastric cancer cells and the underlying mechanisms.METHODS:SGC7901,MKN45,MKN28,N87,and AGS human gastric cancer cell lines were used to screen for radioresistant gastric cancer cell lines. A 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium(MTT) assay was used to determine the effects of β-elemene and IPA-3 on cell viability in MKN45 and SGC7901 gastric cancer cell lines. A clonogenic survival assay and annexin V-FITC/PI apoptosis detection assay were used to evaluate cellular radiosensitivity and radiation-induced cell death,respectively. A proteomic method,isobaric tags for relative and absolute quantitation(i TRAQ),was employed to screen the proteins regulated by β-elemene pretreatment prior to ionizing radiation(IR) in SGC7901 gastric cancer cell line. IPA-3 was used as a specific small molecule inhibitor of p21-activated protein kinase 1(Pak1) to target Pak1 signaling. Protein levels of PAK1IP1(p21-activated protein kinase-interacting protein 1),total Pak1(t-Pak1),phospho-Pak1(T423),phospho-ERK1/2( Thr202/Tyr204),and cleaved caspase-3(17 k Da) were assessed by western blotting.RESULTS:MKN45 and SGC7901 gastric cancer cell lines were relatively more resistant to IR. β-elemene pretreatment decreased clonogenic survival following IR in MKN45 and SGC7901 gastric cancer cell lines. Additionally,β-elemene pretreatment prior to IR increased radiation-induced cell death compared with IR alone in MKN45(10.4% ± 0.9% vs 34.8% ± 2.8%,P < 0.05) and SGC7901(11.6% ± 0.9% vs 46.7% ± 5.2%,P < 0.05) human gastric cancer cell lines,respectively,consistent with the level of cleaved caspase-3(17 k Da). Through i TRAQ analysis and western blot validation,we found that β-elemene upregulated PAK1IP1 and downregulated phospho-Pak1(T423) and phosphoERK1/2 in SGC7901 gastric cancer cells. IR increased the level of phospho-Pak1(T423). Pretreatment with β-elemene decreased radiation-induced Pak1 and ERK1/2 phosphorylation. Inhibition of Pak1 using IPA-3 decreased clonogenic survival following IR. In addition,IPA-3 increased radiation-induced cell death in MKN45(13.4% ± 0.3% vs 26.6% ± 1.0%,P < 0.05) and SGC7901(16.0% ± 0.6% vs 37.3% ± 1.7%,P < 0.05) gastric cancer cell lines,respectively,consistent with the level of cleaved caspase-3(17 k Da). | 3 | The Effect of Hydrolysis with Neutrase on Molecular Weight,Functional Properties,and Antioxidant Activities of Alaska Pollock Protein Isolate显示文摘In this study, the Alaska pollock protein isolate(APPI) was hydrolyzed by Neutrase for 20, 40, 80, 120, 160, 200, and 240 min. Hydrolysates with different molecular weights were produced and they were named as H1–H7. Furthermore, the effects of hydrolysis on the average molecular weights, functional properties(solubility, oil-holding capacities, foaming activities, and emulsifying properties), and antioxidant activities(1, 1-diphenyl-2-picrylhydrazyl, superoxide, and hydroxyl free radical-scavenging activities) were determined. It was found that when the degree of hydrolysis(DH) increased, the average molecular weights of the hydrolysates decreased significantly. The functional properties of APPI were also significantly improved. The hydrolysates of APPI exhibited better solubility, emulsifying activities, and foaming activities. Hydrolysates with low molecular weights(<1 kDa) had better solubility, oil-holding capacities, and emulsifying activities, while hydrolysates with higher molecular weights(>1 kDa) had better foaming activities. In addition, the hydrolysates exhibited excellent antioxidant properties, while the inhibition values of 1, 1-diphenyl-2-picryl hydroxyl(DPPH), superoxide, and hydroxyl free radical-scavenging activities, were 85.22%, 53.56%, and 75.00% respectively, when the concentration of the hydrolysates was 5.0 mg mL^(-1). The lower the average molecular weight was, the higher was the antioxidant activity. These results indicated that hydrolysis with Neutrase is an effective method for improving the functional and antioxidant properties of APPI. | 4 | Preparation of zeolite NaA for CO_2 capture from nickel laterite residue显示文摘Zeolite NaA was successfully prepared from nickel laterite residue for the first time via a fusion–hydrothermal procedure. The structure and morphology of the as-synthesized zeolite NaA were characterized with a range of experimental techniques, such as X-ray diffraction, scanning electronic microscopy, and infrared spectroscopy. It was revealed that the structures of the produced zeolites were dependent on the molar ratios of the reactants and hydrothermal reaction conditions, so the synthesis conditions were optimized to obtain pure zeolite NaA. Adsorption of nitrogen and carbon dioxide on the prepared zeolite NaA was also measured and analyzed. | 14 | Experimental study on influence of operating parameters on tar components from corn straw gasification in fluidized bed显示文摘Gasification is a promising approach for converting solid fuel sources, including renewable ones like biomass, for use. The main problem in biomass gasification is the formation of condensable tars, including polycyclic aromatic hydrocarbons (PAHs). This paper investigated the conversion of tar components during corn straw gasification. It analyzed collected tar components using a gas chromatograph-mass spectrograph (GC-MS). Experimental results indicate that, with increasing temperature from 700°C to 900°C, the concentrations of benzene, indene, phenanthrene, naphthalene, acenaphthylene, fluorene, and pyrene increased whereas those of toluene, phenol, 1-methylnaphthalene, and 2-methylnaphthalene decreased. As the equivalence ratio (ER) increased from 0.21 to 0.34, the concentrations of indene and phenanthrene increased from 0.148% and 0.087% to 0.232% and 0.223%, respectively. Further, the phenol content increased as ER increased from 0.21 to 0.26 and then decreased as the ER increased further to 0.34. Other parameters like the steam/biomass (S/B) ratio and catalyst also played a critical role in tar reduction. | 15 | Bacterial diversity in Arctic marine sediment determined by culture-dependent and-independent approaches显示文摘Bacterial diversity in surface sediment from the Arctic Ocean was investigated by culture-dependent and-independent approaches. Conventional culture-dependent techniques revealed 11 strains based on their distinct morphological characteristics on marine Zobell 2216E agar plates. Phylogenetic analysis showed that these isolates belonged to three major lineages of the Bacteria, γ-proteobacteria, Bacteroidetes and Actinobacteria, and that they included 10 genera. Most isolates were psychrotrophic, and NaCl was not necessary for their growth. Furthermore, they exhibited activity of at least one extracellular hydrolytic enzyme at 4°C and had various abilities to assimilate carbon sources. A total of 67 phylotypes were detected among 142 clones based on the 16S rRNA library of the total community DNA and grouped into nine major lineages of bacteria. Phylotypes affiliated with γ-, δ- and ε-proteobacteria accounted for 36.7%, 21.8% and 16.9% of the total clones, respectively. | 17 | Discrepancy in photosynthetic responses of the red alga Pyropia yezoensis to dehydration stresses under exposure to desiccation,high salinity,and high mannitol concentration显示文摘Macroalgae that inhabit intertidal zones are exposed to the air for several hours during low tide and must endure desiccation and high variations in temperature, light intensity, and salinity. Pyropia yezoensis (Rhodophyta, Bangiales), a typical intertidal red macroalga that is commercially cultivated in the northwestern Pacific Ocean, was investigated under different dehydration stresses of desiccation, high salinity, and high mannitol concentration. Using chlorophyll fluorescence imaging, photosynthetic activities of P. yezoensis thalli were analyzed using six parameters derived from quenching curves and rapid light curves. A distinct discrepancy was revealed in photosynthetic responses to different dehydration stresses. Dehydration caused by exposure to air resulted in rapid decreases in photosynthetic activities, which were always lower than two other stresses at the same water loss (WL) level. High salinity only reduced photosynthesis significantly at its maximum WL of 40% but maintained a relatively stable maximum quantum yield of photosystem II (PSII) (Fv/Fm). High mannitol concentration induced maximum WL of 20% for a longer time (60 min) than the other two treatments and caused no adverse influences on the six parameters at different WL except for a significant decrease in non-photochemical quenching (NPQ) at 20% WL. Illustrated by chlorophyll fluorescence images, severe spatial heterogeneities were induced by desiccation with lower values in the upper parts than the middle or basal parts of the thalli. The NPQ and rETRmax (maximum relative electron transport rate) demonstrated clear distinctions for evaluating photosynthetic responses, indicating their sensitivity and applicability. | 20 | Coupling Effects of A Deep-Water Drilling Riser and the Platform and the Discharging Fluid Column in An Emergency Disconnect Scenario显示文摘As drilling operations move into remote locations and extreme water depths, recoil analysis requires more careful considerations and the incidence of emergency disconnect is increased inevitably. To accurately capture the recoil dynamics of a deep-water riser in an emergency disconnect scenario, researchers typically focus on modelling the influential subsystems (e.g., the tensioner, the mud discharge and seawater refilling process) which can be solved in the preprocessing, and then the determined parameters are transmitted into an existing global riser analysis software. Distinctively, the current study devotes efforts into the coupling effects resulting from that the suspended riser reacts the platform heave motion via the tensioner system in the course of recoil and the discharging fluid column follows the oscillation of the riser in the mud discharge process. Four simulation models are established based on lumped mass method employing different formulas for the top boundary condition of the riser and the discharging flow acceleration. It demonstrates that the coupling effects discussed above can significantly affect the recoil behavior during the transition phase from initial disconnect to the final hang-off state.
