| 1 | Characterization and Evaluation of OsLCT1 and OsN ramp5 Mutants Generated Through CRISPR/Cas9-Mediated Mutagenesis for Breeding Low Cd Rice显示文摘To explore how rice(Oryza sativa L.) can be safely produced in Cd-polluted soil, OsLCT1 and OsNramp5 mutant lines were generated by CRISPR/Cas9-mediated mutagenesis. One of OsLCT1 mutant(lct1×1) and two of OsNramp5 mutants(nramp5×7 and nramp5×9) were evaluated for grain Cd accumulation and agronomic performances. In paddy field soil containing approximately 0.9 mg/kg Cd, lct1×1 grains contained approximately 40%(0.17 mg/kg) of the Cd concentration of the wild type parental line, less than the China National Food Safety Standard(0.20 mg/kg). Both OsNramp5 mutants showed low grain Cd accumulation(< 0.06 mg/kg) in the paddy(approximately 0.9 mg/kg Cd) or in pots in soil spiked with 2 mg/kg Cd. However, only nramp5×7 showed normal growth and yield, whereas the growth of nramp5×9 was severely impaired. | 3 | Spreading Social Influence with both Positive and Negative Opinions in Online Networks显示文摘Social networks are important media for spreading information, ideas, and influence among individuals.Most existing research focuses on understanding the characteristics of social networks, investigating how information is spread through the 'word-of-mouth' effect of social networks, or exploring social influences among individuals and groups. However, most studies ignore negative influences among individuals and groups. Motivated by the goal of alleviating social problems, such as drinking, smoking, and gambling, and influence-spreading problems, such as promoting new products, we consider positive and negative influences, and propose a new optimization problem called the Minimum-sized Positive Influential Node Set(MPINS) selection problem to identify the minimum set of influential nodes such that every node in the network can be positively influenced by these selected nodes with no less than a threshold of ?. Our contributions are threefold. First, we prove that, under the independent cascade model considering positive and negative influences, MPINS is APX-hard. Subsequently, we present a greedy approximation algorithm to address the MPINS selection problem. | 4 | Reliable and Energy Efficient Target Coverage for Wireless Sensor Networks显示文摘A critical aspect of applications with Wireless Sensor Networks (WSNs) is network lifetime.Power-constrained WSNs are usable as long as they can communicate sense data to a processing node.Poor communication links and hazardous environments make the WSNs unreliable.Existing schemes assume that the state of a sensor covering targets is binary:success (covers the targets) or failure (cannot cover the targets).However,in real WSNs,a sensor covers targets with a certain probability.To improve WSNs' reliability,we should consider that a sensor covers targets with users' satisfied probability.To solve this problem,this paper first introduces a failure probability into the target coverage problem to improve and control the system reliability.Furthermore,we model the solution as the α-Reliable Maximum Sensor Covers (α-RMSC) problem and design a heuristic greedy algorithm that efficiently computes the maximal number of α-Reliable sensor covers.To efficiently extend the WSNs lifetime with users' pre-defined failure probability requirements,only the sensors from the current active sensor cover are responsible for monitoring all targets,while all other sensors are in a low-energy sleep mode.Simulation results validate the performance of this algorithm,in which users can precisely control the system reliability without sacrificing much energy consumption. | Jing He Shouling Ji Yi Pan Yingshu Li | 2011 | Tsinghua Science and Technology2011,16,5: | 4 |
| 5 | Identification of a major quantitative trait locus and its candidate underlying genetic variation for rice stigma exsertion rate显示文摘Stigma exsertion in male sterile lines of hybrid rice is important for seed yield.In the present study, ZS616 [Oryza sativa subsp.Xian(indica)], a male sterile line with a stigma exsertion rate(SER) as high as 94.5%, was crossed to DS552, a japonica line with almost no exserted stigmas.F3 plants with extremely low and high SER were sequenced to identify SER-associated quantitative trait loci(QTL).A major QTL for SER, qSER-3.1, was identified along with other QTL on chromosome 3 in a 3.9 Mb region.A total of 307 nonsynonymous single-nucleotide polymorphisms(SNPs) and 27 frame-shift insertion/deletions(InDels)differentiating ZS616 and DS552 were identified in the region containing qSER-3.1.Most SNPs(294) and InDels(25) were excluded after further analysis because they were shared by ZS616 and low(<2.0%) SER accessions in the Huazhong Agricultural University(HAU) core rice collection.Association analysis using the full HAU collection identified a 17-bp InDel in OS03 G0689400 as the most likely causal genetic variant underlying qSER-3.1.ZS616-type accessions(n = 54, with the 17-bp insertion) in the HAU collection had minimum(16.5%)and mean(39.6%) SERs significantly greater than those(n = 424) without the insertion(with minimum and mean SERs of 0.2% and 20.6%, respectively).Thus, this study identified a major QTL for stigma exsertion and revealed the mutation in a candidate gene for the QTL. | Shouling Xu Yunchao Zheng Yang Liu Xiaohao Guo Yuanyuan Tan Qiuping Qian Qingyao Shu Jianzhong Huang | 2019 | The Crop Journal2019,7,3: | 2 |

| 17 | Spatiotemporal evidence of tree-growth resilience to climate variations for Yezo spruce(Picea jezoensis var. komarovii) on Changbai Mountain, Northeast China显示文摘Global warming-induced changes in tree-growth resilience to climate variations have been widely reported for mid-and high-latitude regions around the world. Most studies have focused on the spatial variability of trees in radial growth-climate relationships on Changbai Mountain in Northeast China, but little is known about temporal changes in tree growth in response to climate. We explored the stability of effect of climate variables on radial growth of Yezo spruce [Picea jezoensis Carr. var. komarovii(V.Vassil.) Cheng et L.K.Fu] at 1200, 1400, and 1600 m above sea level, representing low, middle, and upper ranges of the spruce-fir mixed forest on Changbai Mountain. The results showed that the relation between tree growth and climate did not vary with altitude, but the stability of the tree-growth-climate relationship did vary with altitude as the climate changed. Radial growth of Yezo spruce at allthree elevations was influenced primarily by maximum temperature during May(Tmax5) and mean minimum temperature from January to March(Tmin1-3). More specifically, the relationship strengthened significantly at lower elevations, but weakened significantly at higher elevation, and fluctuated at mid elevations since 1980.Increase in Tmin1-3 and decrease in Tmax5 were the main reasons for the decrease in the radial growth at three altitudes. | 18 | Towards Understanding the Security of Modern Image Captchas and Underground Captcha-Solving Services显示文摘Image captchas have recently become very popular and are widely deployed across the Internet to defend against abusive programs. However, the ever-advancing capabilities of computer vision have gradually diminished the security of image captchas and made them vulnerable to attack. In this paper, we first classify the currently popular image captchas into three categories: selection-based captchas, slide-based captchas, and click-based captchas. Second, we propose simple yet powerful attack frameworks against each of these categories of image captchas. Third, we systematically evaluate our attack frameworks against 10 popular real-world image captchas,including captchas from tencent.com, google.com, and 12306.cn. Fourth, we compare our attacks against nine online image recognition services and against human labors from eight underground captcha-solving services. Our evaluation results show that(1) each of the popular image captchas that we study is vulnerable to our attacks;(2) our attacks yield the highest captcha-breaking success rate compared with state-of-the-art methods in almost all scenarios; and(3) our attacks achieve almost as high a success rate as human labor while being much faster.Based on our evaluation, we identify some design flaws in these popular schemes, along with some best practices and design principles for more secure captchas. We also examine the underground market for captcha-solving services, identifying 152 such services. We then seek to measure this underground market with data from these services. | 19 | Cumulative burden of lipid profiles predict future incidence of ischaemic stroke and residual risk显示文摘Objectives Incident ischaemic stroke(IS)risk may increase not only with lipids concentration but also with longer duration of exposure.This study aimed to investigate the impact of cumulative burden of lipid profiles on risk of incident IS.Methods A total of 43836 participants were enrolled who participated in four surveys during 2006-2013.Individual cumulative lipid burden was calculated as number of years(2006-2013)multiplied by the levels of low-density lipoprotein cholesterol(LDL-C),total cholesterol(TC),high-density lipoprotein cholesterol(HDL-C),non-HDL C and triglyceride(TG),respectively.The primary outcome was defined as the incident IS during 2012-2017.Results During 4.67 years(±0.70 years)follow-up on average,we identified 1023(2.33%)incident IS.Compared with respective reference groups,the HRs(95%CIs)of the upper tertile in cumulative TG burden,cumulative LDL-C burden,cumulative TC burden and cumulative non-HDL C burden were 1.26 mmol/L(1.02-1.55 mmol/L),1.47 mmol/L(1.25-1.73 mmol/L),1.33 mmol/L(1.12-1.57 mmol/L)and 1.51 mmol/L(1.28-1.80 mmol/L)for incidence of IS,respectively.However,this association was not significant in cumulative HDL-C burden and IS(HR:1.09;95%CI:0.79 to 1.52),after adjustment for confounding variables.Among 16600 participants with low cumulative LDL-C burden,HRs(95%CI)for TC,TG,non-HDL C and HDL-C with IS were 1.63 mmol/L(1.03-2.57 mmol/L),1.65 mmol/L(1.19-2.31 mmol/L),1.57 mmol/L(1.06-2.32 mmol/L)and 0.98 mmol/L(0.56-1.72 mmol/L),respectively.Conclusions We observed the correlation between cumulative burden of lipid profiles,except for cumulative burden of HDL-C,with the risk of incident IS.Cumulative burden of TC,TG and non-HDL C may still predict IS in patients with low cumulative LDL-C burden.Trial registration number ChiCTR-TNRC 11001489. | Liye Dai Jie Xu Yijun Zhang Anxin Wang Zimo Chen Jinglin Mo Hao Li Xia Meng Shouling Wu Yongjun Wang | 2021 | Stroke & Vascular Neurology2021,6,4: | 0 |
| 20 | Composition and Functional Capacity of Gut Microbes are Associated with Arterial Stiffness:A Prospective Study显示文摘Objective:Arterial stiffness is an important predictor of cardiovascular disease.Microbial diversity in the gut has been shown to be associated inversely with arterial stiffness in Caucasian populations.However,due to the different profiles of the gut microbiota among ethnicities,the relationship between gut-microbiota dysbiosis and the progression of arterial stiffness merits further investigation.This study aimed to investigate the association between the composition and functional capacity of the gut microbiota and the progression of arterial stiffness.Methods:“Shotgun”metagenomics sequencing were undertaken in 96 individuals from a hypertension-associated gut-microbiota study in the Kailuan cohort,who measured brachial-ankle pulse wave velocity(baPWV)and provided fecal samples between September 2014 and February 2015 at Kailuan General Hospital and 11 affiliated hospitals.The different composition and functional capacity of the gut microbiota were compared between individuals without arterial stiffness(normal arterial stiffness group,baPWV<1,400 cm/s,n=27)and participants with arterial stiffness(increased arterial stiffness group,baPWV≥1,400 cm/s,n=69)at baseline.These participants were followed up prospectively for a mean duration of 2.6 years,and 50 underwent a repeat baPWV measurement.Associations between the gut microbiota and severity and progression of arterial stiffness were assessed using MaAsLin2 software after adjustment for age,sex,and mean arterial blood pressure and correction for multiple testing.Gene“catalogs”were aligned to the Kyoto Encyclopedia of Genes and Genomes(KEGG)database to obtain information for potential functional capacities of the gut microbiota.Results:In this study,14 genera and 50 species of bacteria were identified to be abundant in participants with normal arterial stiffness compared with those with increased arterial stiffness.Of 14 genera,the prevalence of beneficial bacteria of the genera Leadbetterella and Cytophaga was correlated inversely with baPWV(P<0.05).Analyses of functional capacity revealed gut-microbial dysfunctions in the synthetic processes of“threonine dehydratase”“hypothetical protein”“mannosyl transferase”and“type-IV secretion-system proteins”in individuals suffering from arterial stiffness.During follow-up,bacteria of the proinflammatory genera Escherichia,Shigella,and Ruegeria were enriched in individuals with increased baPWV.Functional analyses showed that 26 KEGG orthologs of gut microbes were associated with an increase in baPWV and involved in“carbohydrate metabolism”“amino acid metabolism”and“protein families related to genetic information processing.”Conclusions:The composition and functional capacity of the microbial community in the gut of people suffering from arterial stiffness differed from those in individuals not suffering from arterial stiffness.Our data provide a new direction for the causality of the host-gut microbiota in arterial stiffness. | Jing Li Yixuan Zhong Jingjing Bai Shuohua Chen Jun Cai Shouling Wu Weili Zhang | 2023 | Cardiology Discovery2023,3,2: | 0 |
