1SARS-CoV-2 spike protein receptor-binding domain perturbates intracellular calcium homeostasis and impairs pulmonary vascular endothelial cells显示文摘Exposure to the spike protein or receptor-binding domain(S-RBD)of SARS-CoV-2 significantly influences endothelial cells and induces pulmonary vascular endotheliopathy.In this study,angiotensin-converting enzyme 2 humanized inbred(hACE2 Tg)mice and cultured pulmonary vascular endothelial cells were used to investigate how spike protein/S-RBD impacts pulmonary vascular endothelium.Results show that S-RBD leads to acute-to-prolonged induction of the intracellular free calcium concentration([Ca^(2+)]i)via acute activation of TRPV4,and prolonged upregulation of mechanosensitive channel Piezo1 and store-operated calcium channel(SOCC)key component Orai1 in cultured human pulmonary arterial endothelial cells(PAECs).In mechanism,S-RBD interacts with ACE2 to induce formation of clusters involving Orai1,Piezo1 and TRPC1,facilitate the channel activation of Piezo1 and SOCC,and lead to elevated apoptosis.These effects are blocked by Kobophenol A,which inhibits the binding between S-RBD and ACE2,or intracellular calcium chelator,BAPTA-AM.Blockade of Piezo1 and SOCC by GsMTx4 effectively protects the S-RBDinduced pulmonary microvascular endothelial damage in hACE2 Tg mice via normalizing the elevated[Ca^(2+)]i.Comparing to prototypic strain,Omicron variants(BA.5.2 and XBB)of S-RBD induces significantly less severe cell apoptosis.Transcriptomic analysis indicates that prototypic S-RBD confers more severe acute impacts than Delta or Lambda S-RBD.In summary,this study provides compelling evidence that S-RBD could induce persistent pulmonary vascular endothelial damage by binding to ACE2 and triggering[Ca^(2+)]i through upregulation of Piezo1 and Orai1.Targeted inhibition of ACE2-Piezo1/SOCC-[Ca^(2+)]i axis proves a powerful strategy to treat S-RBD-induced pulmonary vascular diseases.Kai Yang Shiyun Liu Han Yan Wenju Lu Xiaoqian Shan Haixia Chen Changlei Bao Huazhuo Feng Jing Liao Shuxin Liang Lei Xu Haiyang Tang Jason X-J.Yuan Nanshan Zhong Jian Wang 2023Signal Transduction and Targeted Therapy2023,8,8:2
2Experiment and analysis of the formation,expansion and dissipation of gasbag in fine sediments based on pore water pressure survey显示文摘Deep-seated gas in seabed sediments migrates upwards from effect of external factors,which easily accumulates to form gasbags at interface of shallow coarse-fine sediments.Real-time monitoring of this process is important to predict disaster.However,there is still a lack of effective monitoring methods,so we attempt to apply multi-points pore water pressure monitoring technology when simulating forming and dissipation of gasbags in sediments through laboratory experiment.This study focuses on discussion of sensitivity of pore water pressure monitoring data,as well as typical changing characteristics and mechanisms of excess pore water pressure corresponding to crack generation,gasbag formation and gas release.It was found that the value of excess pore water pressure in sediments is negatively correlated with vertical distance between sensors and gas source,and the evolution of gasbag forming and dissipation has a good corresponding relationship with the change of excess pore water pressure.Gasbag formation process is divided into three stages:transverse crack development,longitudinal cavity expansion,and oblique crack development.Formation of gasbag begins with the transverse crack at the interface of coarse-fine sediments while excess pore water pressure attenuates rapidly and then drops,pressure remains almost unchanged when cavity expanses longitudinally,oblique crack appeared in final stage of gasbag evolution while excess pore water pressure accumulated and dissipated again.The variation curve of excess pore water pressure in gas release stage has saw-tooth fluctuation characteristics,and the value and time of pressure accumulation are also fluctuating,indicating the uncertainty and non-uniqueness of gas migration channels in sediments.Shiyun Lei Xiujun Guo Haoru Tang 2022Acta Oceanologica Sinica2022,41,4:0
3Transition-metal-catalyzed switchable divergent cycloaddition of para-quinone methides and vinylethylene carbonates:Access to different sized medium-sized heterocycles显示文摘Divergent synthesis of medium-sized rings with controllable ring sizes represents a longstanding challenge in organic synthesis.Herein,we developed a transition-metal-catalyzed switchable divergent cycloaddition of para-quinone methides and vinylethylene carbonates by controlling the steric hindrance of substituent.Different from reported alkoxide-triggered annulations,this process undergoes a regiodivergent allylation of para-quinone methides followed by 1,6-addition reaction,providing a new route to selectively synthesize seven-to ten-membered nitrogen-containing heterocycles in high yields with excellent regioselectivities.This protocol features a broad substrate scope,wide functional group tolerance as well as operational simplicity.The reaction mechanism was investigated by conducting a series of control experiments as well as DFT calculations and the origins of the regioselectivities of the cycloaddition process were rationalized.Junwei Wang Lei Zhao Chen Zhu Ben Ma Xiaolong Xie Jian Liu Shiyun He Magnus Rueping Kun Zhao Lihong Hu 2022Chinese Chemical Letters2022,33,10:0
