| 1 | α-淀粉酶抑制剂AAI的核磁共振氢谱谱峰归属及结构分析显示文摘α -淀粉酶抑制剂AAI(α -AmylaseInhibitor)是从墨西哥苋属植物Amaranthushypochondriacus种子中提取的由32个氨基酸残基组成的多肽。它能强烈地抑制几种昆虫幼虫的α -淀粉酶活性 ,而不抑制人及哺乳类动物的α -淀粉酶活性。使用核磁共振方法收集了AAI的二维氢谱谱图 ,完成了全部主链和绝大部分侧链质子共振峰的序列归属 ,并利用X -PLOR软件计算出AAI的三维结构。AAI由三股反平行 β-折叠片与三对二硫键组成抑制剂胱氨酸结模体(inhibitorcystine -knotmotif)结构,与来自虎纹捕鸟蛛的神经毒素HWTX -I和凝集素SHL -I及其它来源的多种小分子活性多肽的结构类似 ,但它们的生物活力各不相同 ,表明该模体适用于结构功能关系的比较研究和作为蛋白质工程活力改造的结构骨架 (scaffold)。AAI对昆虫α -淀粉酶具有专一活性 ,可用于通过转基因技术来赋予植物抗虫活性。 | 邓鹏翅 吕善运 罗静初 Sandor Pongor 刘修才 曲红 梁宋平 韩汝珊 顾孝诚 | 2000 | 生物物理学报2000,16,3: | 7 |

| 6 | Evolution of disease phenotype in adult and pediatric onset Crohn's disease in a population-based cohort显示文摘AIM:To investigate the evolution of disease phenotypein adult and pediatric onset Crohn's disease(CD) populations,diagnosed between 1977 and 2008.METHODS:Data of 506 incident CD patients were analyzed(age at diagnosis:28.5 years,interquartile range:22-38 years).Both in-and outpatient records were collected prospectively with a complete clinical follow-up and comprehensively reviewed in the population-based Veszprem province database,which included incident patients diagnosed between January 1,1977 and December 31,2008 in adult and pediatric onset CD populations.Disease phenotype according to the Montreal classification and long-term disease course was analysed according to the age at onset in time-dependent univariate and multivariate analysis.RESULTS:Among this population-based cohort,seventy-four(12.8%) pediatric-onset CD patients were identified(diagnosed ≤ 17 years of age).There was no significant difference in the distribution of disease behavior between pediatric(B1:62%,B2:15%,B3:23%) and adult-onset CD patients(B1:56%,B2:21%,B3:23%) at diagnosis,or during follow-up.Overall,the probability of developing complicated disease behaviour was 49.7% and 61.3% in the pediatric and 55.1% and 62.4% in the adult onset patients after 5-and 10-years of follow-up.Similarly,time to change in disease behaviour from non stricturing,non penetrating(B1) to complicated,stricturing or penetrating(B2/B3) disease was not significantly different between pediatric and adult onset CD in a Kaplan-Meier analysis.Calendar year of diagnosis(P = 0.04),ileal location(P < 0.001),perianal disease(P < 0.001),smoking(P = 0.038) and need for steroids(P < 0.001) were associated with presence of,or progression to,complicated disease behavior at diagnosis and during follow-up.A change in disease location was observed in 8.9% of patients and it was associated with smoking status(P = 0.01),but not with age at diagnosis.CONCLUSION:Long-term evolution of disease behavior was not different in pediatric-and adult-onset CD patients in this population-based cohort but was associated to location,perianal disease and smoking status. | Barbara Dorottya Lovasz Laszlo Lakatos Agnes Horvath Istvan Szita Tunde Pandur Michael Mandel Zsuzsanna Vegh Petra Anna Golovics Gabor Mester Mihaly Balogh Csaba Molnar Erzsebet Komaromi Lajos Sandor Kiss Peter Laszlo Lakatos | 2013 | World Journal of Gastroenterology2013,19,14: | 2 |

| 8 | Mechanism of Methylene Blue adsorption on hybrid laponite-multi-walled carbon nanotube particles显示文摘The kinetics of adsorption and parameters of equilibrium adsorption of Methylene Blue(MB)on hybrid laponite-multi-walled carbon nanotube(NT)particles in aqueous suspensions were determined.The laponite platelets were used in order to facilitate disaggregation of NTs in aqueous suspensions and enhance the adsorption capacity of hybrid particles for MB.Experiments were performed at room temperature(298 K),and the laponite/NT ratio(X_l)was varied in the range of 0–0.5.For elucidation of the mechanism of MB adsorption on hybrid particles,the electrical conductivity of the system as well as the electrokinetic potential of laponite-NT hybrid particles were measured.Three different stages in the kinetics of adsorption of MB on the surface of NTs or hybrid laponite-NT particles were discovered to be a fast initial stage Ⅰ(adsorption time t=0–10 min),a slower intermediate stage Ⅱ(up to t=120 min)and a long-lasting final stage Ⅲ(up to t=24 hr).The presence of these stages was explained accounting for different types of interactions between MB and adsorbent particles,as well as for the changes in the structure of aggregates of NT particles and the long-range processes of restructuring of laponite platelets on the surface of NTs.The analysis of experimental data on specific surface area versus the value of X_l evidenced in favor of the model with linear contacts between rigid laponite platelets and NTs.It was also concluded that electrostatic interactions control the first stage of adsorption at low MB concentrations. | Maryna Manilo Nikolai Lebovka Sandor Barany | 2016 | Journal of Environmental Sciences2016,28,4: | 2 |
| 9 | mRNA expression, functional profiling and multivariate classification of colon biopsy specimen by cDNA overall glass microarray显示文摘AIM: To understand the local pathophysiological altera- tions and gene ontology-based functional classification of colonic biopsies into inflammatory and neoplastic dis- eases. METHODS: Total RNA was extracted from frozen biop- sies and amplified by T7-method. Expression profile was evaluated by Atlas Glass 1K microarrays. After microar- ray quality control, applicable data were available from 10 adenomas, 6 colorectal adenocarcinomas (CRCs), and 6 inflammatory bowel diseases (IBDs). Multivariate statistical and cell functional analyses were performed. Real-time RT-PCR and immunohistochemistry were used for validation. RESULTS: Discriminant analysis of selected genes, could correctly reclassify all 22 samples using 4 parameters (heat shock transcription factor-1, bystin-like, calgranu- lin-A, TRAIL receptor 3). IBD samples were characterized by overregulated chemokine (C-X-C motif) ligand 13, replication protein A1, E74-like factor 2 and downregu- lated TNF receptor-associated factor 6, BCL2-interacting killer genes. In adenomas upregulation of TNF receptor- associated factor 6, replication protein A1, E74-like factor 2 and underexpression of BCL2-associated X protein, cal- granulin-A genes were found. CRC cases had significantly increased epidermal growth factor receptor, topoisomer- ase-1, v-jun, TNF receptor-associated factor 6 and TRAIL receptor 3, and decreased RAD51 and RAD52 DNA repair gene, protein phosphatase-2A and BCL2-interacting killer mRNA levels. Epidermal growth factor receptor RT-PCR and immunohistochemistry, topoisomerase-1 RT-PCRconfirmed the chip results. CONCLUSION: Different histological alterations can be reclassified by functional, multivariate analysis using cDNA microarrays. | 20 | 3D printed fiber-optic nanomechanical bioprobe显示文摘Ultrasensitive nanomechanical instruments,e.g.atomic force microscopy(AFM),can be used to perform delicate biomechanical measurements and reveal the complex mechanical environment of biological processes.However,these instruments are limited because of their size and complex feedback system.In this study,we demonstrate a miniature fiber optical nanomechanical probe(FONP)that can be used to detect the mechanical properties of single cells and in vivo tissue measurements.A FONP that can operate in air and in liquids was developed by programming a microcantilever probe on the end face of a single-mode fiber using femtosecond laser two-photon polymerization nanolithography.To realize stiffness matching of the FONP and sample,a strategy of customizing the microcantilever’s spring constant according to the sample was proposed based on structure-correlated mechanics.As a proof-of concept,three FONPs with spring constants varying from 0.421 N m^(−1)to 52.6 N m^(−1)by more than two orders of magnitude were prepared.The highest microforce sensitivity was 54.5 nmμN^(−1)and the detection limit was 2.1 nN.The Young’s modulus of heterogeneous soft materials,such as polydimethylsiloxane,muscle tissue of living mice,onion cells,and MCF-7 cells,were successfully measured,which validating the broad applicability of this method.Our strategy provides a universal protocol for directly programming fiber-optic AFMs.Moreover,this method has no special requirements for the size and shape of living biological samples,which is infeasible when using commercial AFMs.FONP has made substantial progress in realizing basic biological discoveries,which may create new biomedical applications that cannot be realized by current AFMs. | Mengqiang Zou Changrui Liao Yanping Chen Lei Xu Shuo Tang Gaixia Xu Ke Ma Jiangtao Zhou Zhihao Cai Bozhe Li Cong Zhao Zhourui Xu Yuanyuan Shen Shen Liu Ying Wang Zongsong Gan Hao Wang Xuming Zhang Sandor Kasas Yiping Wang | 2023 | International Journal of Extreme Manufacturing2023,5,1: | 1 |
