1Pluronic L-81 ameliorates diabetic symptoms in db/db mice through transcriptional regulation of microsomal triglyceride transfer protein显示文摘AIM: To test whether oral L-81 treatment could im-prove the condition of mice with diabetes and to investigate how L-81 regulates microsomal triglyceride transfer protein (MTP) activity in the liver. METHODS: Genetically diabetic (db/db) mice were fed on chow supplemented with or without L-81 for 4 wk. The body weight, plasma glucose level, plasma lipid profile, and adipocyte volume of the db/db mice were assessed after treatment. Toxicity of L-81 was also evaluated. To understand the molecular mecha-nism, HepG2 cells were treated with L-81 and the effects on apolipoprotein B (apoB) secretion and mRNA level of the MTP gene were assessed.RESULTS: Treatment of db/db mice with L-81 sig-nificantly reduced and nearly normalized their body weight, hyperphagia and polydipsia. L-81 also markedly decreased the fasting plasma glucose level, improved glucose tolerance, and attenuated the elevated levels of plasma cholesterol and triglyceride. At the effective dosage, little toxicity was observed. Treatment of HepG2 cells with L-81 not only inhibited apoB secretion, but also signif icantly decreased the mRNA level of the MTP gene. Similar to the action of insulin, L-81 exerted its effect on the MTP promoter. CONCLUSION: L-81 represents a promising candidate in the development of a selective insulin-mimetic mol-ecule and an anti-diabetic agent.Wo-Shing Au Li-Wei Lu Sidney Tam Otis King Hung Ko Billy KC Chow Ming-Liang He Samuel S Ng Chung-Man Yeung Ching-Chiu Liu Hsiang-Fu Kung Marie C Lin 2009World Journal of Gastroenterology2009,15,24:1

8Fish-on-Chips:unveiling neural processing of chemicals in small animals through precise fluidic control显示文摘Precise chemical cue presentation alongside advanced brainwide imaging techniques is important to the study of chemosensory processing in animals.Nevertheless, the dynamic nature of chemical-carrying media, such as water or air, poses a significant challenge for delivering highly-controlled chemical flow to an animal subject.Moreover,contact-based cue manipulation and delivery easily shift the position of the animal subject, which is often undesirable for high-quality brain imaging.Additionally, more advanced interfacing tools that align with the diverse range of body part sizes of an animal, ranging from micrometer-scale neurons to meter-long limbs, are much needed. This is particularly crucial when dealing with dimensions that are beyond the reach of conventional experimental tools.Samuel K.H.Sy Ho Ko 2024Neural Regeneration Research2024,19,11:0
9Antibiofilm and immunomodulatory resorbable nanofibrous filing for dental pulp regenerative procedures显示文摘Multifunctional scaffolds with host defense peptides designed for regenerative endodontics are desirable nanobiotechnological tools for dentistry. Here, different scaffolds were tested for use during the pulp revascularization process, including poly(vinyl alcohol)-PVA hydrogels or resins, collagen hydrogels and poly(vinyl alcohol)PVA/Chitosan(PVA/CS)nanofibers.Based on time to degradation(21 days),nanofibers were chosen to be incorporated with ciprofloxacin and IDR-1002(each at 50 mg/g).Nanofibers containing ciprofloxacin and IDR-1002 had anti-biofilm activity against Enterococcus faecalis,Staphylococcus aureus and a multispecies oral biofilm, besides anti-inflammatory activities. The in vivo subcutaneous tissue response to tooth fragments filled with nanofibers demonstrated a pulp-like tissue formation, when compared to empty teeth fragments. Thus,we designed a strong antimicrobial, immunomodulatory and regenerative candidate for pulp revascularization and regeneration procedures.Mauricio Gonçalves da Costa Sousa Gabriela Conceiçao de Almeida Danilo Cesar Martins Mota Rosiane Andrade da Costa Simoni Campos Dias Samuel Nunes Limberger Frank Ko Li Ting Lin Evan FHaney Hashem Etayash Beverlie Baquir Michael J.Trimble Ya Shen Zheng Su Markus Haapasalo Daniel Pletzer Letícia Chaves de Souza Glaucia Schuindt Teixeira Renato M.Silva Robert E.W.Hancock Octavio Luiz Franco Taia Maria Berto Rezende 2022Bioactive Materials2022,7,10:0
