| 1 | NAT2*6A, a haplotype of the N-acetyltransferase 2 gene, is an important biomarker for risk of anti-tuberculosis drug-induced hepatotoxicity in Japanese patients with tuberculosis显示文摘AIM: To investigate an association between N -acetyltransferase 2 (NAT2 )-haplotypes/diplotypes and adverse effects in Japanese pulmonary tuberculosis patients. METHODS: We studied 100 patients with pulmonary TB treated with anti-TB drugs including INH. The frequencies and distributions of single nucleotide polymorphisms, haplotypes, and diplotypes of NAT2 were determined by the PCR-restriction fragment length polymorphism method, and the results were compared between TB patients with and without adverse effect, using multivariate logistic regression analysis.RESULTS: Statistical analysis revealed that the frequency of a variant haplotype, NAT2*6A , was signifi cantly increased in TB patients with hepatotoxicity, compared with those without hepatotoxicity [P = 0.001, odds ratio (OR) = 3.535]. By contrast, the frequency of a wild-type (major) haplotype, 'NAT2*4', was signif icantly lower in TB patients with hepatotoxicity than those without hepatotoxicity (P < 0.001, OR = 0.265). There was no association between NAT2-haplotypes and skin rash or eosinophilia. CONCLUSION: The present study shows that NAT2 is one of the determinants of anti-TB drug-induced hepatotoxicity. | 2 | Association of polymorphic alleles of CTLA4 with inflammatory bowel disease in the Japanese显示文摘AIM: To examine an association between the cytotoxic T-lymphocyte antigen 4 (CTLA4) gene that plays a role in downregulation of T-cell activation and inflammatory bowel disease consisting of ulcerative colitis (UC) and Crohn's disease (CD) in the Japanese.METHODS: We studied 108 patients with UC, 79 patients with CD, and 200 sex-matched healthy controls, with respect to three single nucleotide polymorphisms (SNPs)in CTLA4, such as C-318T in the promoter region, A+49G in exon 1 and G+6230A in the 3' untranslated region (3'-UTR) by a PCR-restriction fragment length polymorphism method, and to an (AT)n repeat polymorphism in 3'-UTR by fragment analysis with fluorescence-labeling on denaturing sequence gels. Frequency of alleles and genotypes and their distribution were compared statistically between patients and controls and among subgroups of patients, using χ2 and Fisher exact tests.RESULTS: The frequency of 'A/A' genotype at the G+6230A SNP site was statistically lower in UC patients than in controls (3.7% vs 11.0%, P= 0.047, odds ratio (OR) = 0.311). | 3 | Crohn's disease in Japanese is associated with a SNP-haplotype of N-acetyltransferase 2 gene显示文摘AIM: To investigate the frequency and distribution of N-acetyltransferase 2 (NAT2) and uridine 5′-diphosphate (UDP)-glucuronosyltransferase 1A7 (UGT1A7) genes in patients with ulcerative colitis (UC) and Crohn's disease (CD).METHODS: Frequencies and distributions of NAT2 and UGT1A7SNPs as well as their haplotypes were investigated in 95 patients with UC, 60 patients with CD, and 200gender-matched, unrelated, healthy, control volunteers by PCR-restriction fragment length polymorphism (RFLP),PCR-denaturing high-performance liquid chromatography (DHPLC), and direct DNA sequencing.RESULTS: Multiple logistic regression analysis revealed that the frequency of haplotype, NAT2*7B, significantly increased in CD patients, compared to that in controls (P= 0.0130, OR = 2.802, 95%CI = 1.243-6.316). However,there was no association between NAT2 haplotypes and UC, or between any UGT1A7haplotypes and inflammatory bowel disease (IBD).CONCLUSION: It is likely that the NAT2 gene is one of the determinants for CD in Japanese. | 4 | A novel suturing technique for filtering glaucoma surgery:the accordion suture显示文摘AIM:To present a novel scleral flap suturing technique for filtering glaucoma surgery in order to control high postoperative intraocular pressure(IOP).METHODS:Description of ‘the accordion suture' technique for mitomycin C augmented trabeculectomy.In cases of postoperative high IOP,pulling the loop of the suture helps to lift up the scleral flap by an even pressure on both edges.By means of this technique,the scleral flap opens up in an ‘‘accordion'' manner,thus preventing flap obstruction and providing adequate aqueous flow.RESULTS:Our study group consisted of 8 eyes of 8 patients with neovascular glaucoma.Mean age of the subjects was 67.42±8.21 y and female/male ratio was 4/4.Mean preoperative IOP was 37±7.48 mm Hg.Mitomycin C augmented trabeculectomy was carried out on the subjects without any complications.The scleral flap closure is performed with three separate sutures;initially,our accordion suture through the center of the flap,and two releasable sutures on both corners.All the patients received removal of two side releasable sutures concomitant with pulling the accordion suture,without any complications.The average traction time was 3.5±0 wk postoperatively.The mean postoperative IOP was 11.37±2.72 mm Hg.No suture related complications were observed.CONCLUSION:This technique can be the suture of choice for filtering glaucoma surgery in experienced hands by its easy learning curve for precisely indicated patients. | Mehmet Baykara Basak Can Ermerak Huri Sabur Selim Gent | 2017 | International Journal of Ophthalmology(English edition)2017,10,12: | 2 |

| 15 | Spatial distribution and risk assessments of mercury in topsoils of Central Asia显示文摘Central Asia is one of the largest arid areas on earth,yet little is known about the concentration levels and risks of mercury(Hg)in the soils of this region.In this study,extensive sampling of topsoils(0-10 cm)from representative landscapes was carried out over Central Asia(i.e.,Tajikistan,Uzbekistan,and Kyrgyzstan).The total mercury(THg)concentrations in topsoils varied widely from 1.6 to 908.0 ng/g,with high values observed in samples collected in the capital cities and urban areas.Topsoil THg concentrations among different landscapes showed a decreasing order of urban(79.8±184.0 ng/g)>woodland(27.3±28.9 ng/g)>grassland(20.6±15.9 ng/g)>farmland(18.3±9.5 ng/g)>desert(12.3±8.0 ng/g).High THg concentrations were found in the capital cities/urban clusters,followed by a gradual decrease towards the peripheries.THg concentrations were found to be negatively correlated with the distance from the sampling sites to their nearest cities,indicating that anthropogenic emissions significantly influenced the spatial distribution of topsoil Hg.A significant correlation between THg concentrations and topsoil total organic carbon(TOC)contents was also observed,suggesting that TOC played an essential role in the spatial distribution of topsoil Hg.The assessments of pollution and potential ecological risk suggested that topsoils in highly densely-populated areas were contaminated by Hg and had higher degrees of potential ecological risks.The health risk assessment results showed that the exposure risk of topsoil Hg to children was higher than that to adults.Fortunately,there was no unacceptable human health risk of topsoil Hg.This study clarified the spatial distribution and risks of Hg in the Central Asian topsoils,offering new insight into the risk prevention and control of soil Hg. | Zhengzheng Yang Junming Guo Shiwei Sun Dingming Ni Pengfei Chen Dipesh Rupakheti Huhu Kang Sabur F Abdullaev Salamat Abdyzhapar uulu Shichang Kang | 2023 | Geoscience Frontiers2023,14,4: | 0 |
| 16 | Modifications in aerosol physical, optical and radiative properties during heavy aerosol events over Dushanbe, Central Asia显示文摘The location of Central Asia,almost at the center of the global dust belt region,makes it susceptible for dust events.The studies on atmospheric impact of dust over the region are very limited despite the large area occupied by the region and its proximity to the mountain regions (Tianshan,Hindu Kush-KarakoramHimalayas,and Tibetan Plateau).In this study,we analyse and explain the modification in aerosols’physical,optical and radiative properties during various levels of aerosol loading observed over Central Asia utilizing the data collected during 2010–2018 at the AERONET station in Dushanbe,Tajikistan.Aerosol episodes were classified as strong anthropogenic,strong dust and extreme dust.The mean aerosol optical depth (AOD) during these three types of events was observed a factor of ~3,3.5 and 6.6,respectively,higher than the mean AOD for the period 2010–2018.The corresponding mean fine-mode fraction was 0.94,0.20 and 0.16,respectively,clearly indicating the dominance of fine-mode anthropogenic aerosol during the first type of events,whereas coarse-mode dust aerosol dominated during the other two types of events.This was corroborated by the relationships among various aerosol parameters (AOD vs.AE,and EAE vs.AAE,SSA and RRI).The mean aerosol radiative forcing (ARF) at the top of the atmosphere (ARF_(TOA)),the bottom of the atmosphere (ARF_(BOA)),and in the atmosphere (ARFATM) were -35±7,-73±16,and38±17 Wm^(-2)during strong anthropogenic events,-48±12,-85±24,and 37±15 Wm^(-2)during strong dust event,and -68±19,-117±38,and 49±21 Wm-2during extreme dust events.Increase in aerosol loading enhanced the aerosol-induced atmospheric heating rate to 0.5–1.6 K day^(-1)(strong anthropogenic events),0.4–1.9 K day^(-1)(strong dust events) and 0.8–2.7 K day^(-1)(extreme dust events).The source regions of air masses to Dushanbe during the onset of such events are also identified.Our study contributes to the understanding of dust and anthropogenic aerosols,in particular the extreme events and their disproportionally high radiative impacts over Central Asia. | Dipesh Rupakheti Maheswar Rupakheti Xiufeng Yin Julian Hofer Mukesh Rai Yuling Hu Sabur F.Abdullaev Shichang Kang | 2021 | Geoscience Frontiers2021,12,6: | 0 |
| 17 | 补肾解毒方抗核辐射损伤作用机制研究显示文摘目的:探讨补肾解毒方辐射防护作用的靶点及其作用机制,为补肾解毒方作为防治辐射损伤药物使用提供实验基础。方法:将138只TLR4_(+/+)小鼠随机分为TLR4_(+/+)空白对照组18只、TLR4_(+/+)辐射模型组60只、TLR4_(+/+)补肾解毒方组60只;将138只TLR4_(-/-)小鼠随机分为TLR4_(-/-)空白对照组18只、TLR4_(-/-)辐射模型组60只、TLR4_(-/-)补肾解毒方组60只。空白对照组、辐射模型组给予生理盐水灌胃,补肾解毒方组予中药灌胃,并于第11天对辐射模型组与补肾解毒方组进行6Gy全身照射。检测并比较各组CD_(14)、TLR4受体、MD_2和CD_4~+CD_(25)~+Treg细胞水平。结果:CD_(14)检测结果显示辐射后第14天,TLR4_(+/+)辐射模型组与补肾解毒方组均高于空白对照组,且补肾解毒方组高于辐射模型组(P<0.01)。辐射后第30天,两组CD_(14)水平下降,TLR4_(+/+)补肾解毒方组高于辐射模型组和空白对照组(P<0.01)。CD_4~+CD_(25)~+Treg检测显示辐射后第14天,TLR4_(+/+)补肾解毒方组高于辐射模型组和空白对照组(P<0.01);辐射后第30天,TLR4_(+/+)补肾解毒方组高于空白对照组(P<0.01)。上述指标同时间点的TLR4_(-/-)补肾解毒方组与辐射模型组比较,差异无统计学意义(P>0.05)。结论:补肾解毒方可通过TLR4信号通路发挥辐射损伤防护作用。 | 杨云霜 赵雪 杜磊 李延晖 石鹏展 张蓉 | 2021 | 陕西中医2021,42,3: | 0 |
| 18 | 丝裂霉素C联合粘小管成形术治疗开角型青光眼的疗效(英文)显示文摘目的:评估丝裂霉素C(MMC)联合粘小管成形术治疗开角型青光眼的效果。方法:回顾性研究。收集2007-12/2014-03接受粘小管成形术患者104例122眼。记录术前术后眼内压(IOP),青光眼用药数量,视力,并发症,辅助治疗(激光前房角穿刺或青光眼用药)和成功率。IOP≤21 mmHg无额外用药为完全成功,IOP≤21 mmHg有或无青光眼用药为部分成功。结果:术前平均IOP为27.5±9.2 mmHg,术后平均IOP为14.5±6.6 mmHg(P<0.001)。术前平均视力为0.42±0.4,术后为0.32±0.4(P=0.726)。部分成功106眼(86.9%),完全成功62眼(50.8%)。激光前房角穿刺术43眼(35.2%),青光眼药物使用率为49.1%。术后平均随访27.29±16.78(1~79)mo。结论:粘小管成形术并发症发生率低且术后视力稳定,是一种安全的选择,术后补充进行激光前房角穿刺术可以提高手术成功率。丝裂霉素C对于粘小管成形术的作用还需进行深入的比较研究。 | Sabur Huri Baykara Mehmet | 2017 | 国际眼科杂志2017,17,9: | 0 |
