| 4 | Induction of apoptosis by arsenic trioxide and hydroxycamptothecin in gastric cancer cells in vitro显示文摘AIM To study the effects of arsenic trioxide andHCPT on different degrees of differentiated gastriccancer cells(SGC-7901,MKN-45,MKN-28)withrespect to both cytotoxicity and induction ofapoptosis in vitro.METHODS The cytotoxicity of As2O3 and HCPTon gastric cancer cells was determined by MTTassay.Morphologic changes of apoptosis ofgastric cancer cells were observed by lightmicroscopy and transmission electron microscopy.Apoptosis and cell cycle changes of gastric cancercells induced by HCPT and As2O3 were investigatedby TUNEL method and flow cytometry.RESULTS As2O3 and HCPT had remarkablecytotoxic effects on different degrees ofdifferentiated gastric cancer cells.The IC50ofAs2O3 on well differentiated gastric cancer cellMKN-28,moderately differentiated gastric cancercell SGC-7901,and poorly differentiated gastriccancer cell MKN-28 were 8.91 μmol/L,10.57μmol/L,and 11.65 μmol/L,respectively.The IC50of HCPT on MKN-28,SGC-7901,and MKN-45 were9.35 mg/L,10.21 mg/L,and 12.63 mg/Lrespectively after 48 h treatment.After 12 h ofexposure to both drugs,gastric cancer cellsexhibited morphologic features of apoptosis,including cell shrinkage,nuclear condensation, and formation of apoptotic bodies.A typicalsubdiploid peak before G0/G1 phase was observedby flow cytometry.The apoptotic rates of SGC-7901,MKN-45,and MKN-28 were 13.84%,22.52%,and 9.68%,respectively after 48 hexposure to 10 μmol/L As2O3.The apoptotic ratesof SGC-7901,MKN-45,and MKN-28 were 21.88%,12.35%,and 30.26%,respectively after 48 hexposure to 10 mg/L HCPT.The apoptotic indicewere 7%-15% as assessed by TUNEL method.The effect of As2O3 on SGC-7901 showedremarkable cell cycle specificity,which inducedcell death in G1 phase,and blocked G2/M phase.HCPT also showed a remarkable cell cyclespecificity,by inducing cell death and apoptosis inG1 phase and arrest of proliferation at S phase.CONCLUSION As2O3 and HCPT exhibitsignificant cytotoxicity on gastric cancer cells byinduction of apoptosis.As2O3 and HCPT mighthave a promising prospect in the treatment ofgastric cancer,which needs to be further studied. | Tu SP Zhong J Tan JH Jiang XH Qiao MM Wu YX Jiang SH | 2000 | World Journal of Gastroenterology2000，6，4： | 43 |

| 8 | Ileoscopy in patients with ileocolonic tuberculosis显示文摘AIM: To study the role of retrograde terminal ileoscopy in patients suspected to have ileocolonic tuberculosis. METHODS: A retrospective analysis was performed for patients undergoing colonoscopy for suspected ileocolonic tuberculosis between January 2000 and June 2004, in whom retrograde ileoscopy had been performed. Only patients diagnosed with tuberculosis on the basis of histological findings of either a caseating granuloma or those having non-caseating granuloma or a collection of epithelioid cells at a minimum of one endoscopic lesion (either in the colon or the terminal ileum) on histology with good responses to conventional anti-tuberculous drugs were enrolled for the study. RESULTS: Fifty-three patients were included. The terminal ileum was involved in only 11 patients. Eight of these patients had involvement of the cecum too. Two patients had ileal lesions without cecal involvement; however, lesions were noted in the ascending colon. In one patient the whole colon was normal and only the terminal ileum showed nodularity and ulceration. Histological examination of the ileal biopsies obtained from the ileal lesions showed noncaseating granulomas in five, collection of epithelioid cells in four and nonspecific histology in two patients. Mucosal biopsies obtained from the lesion in the terminal ileum, in the patient in whom mucosal abnormality was observed to be confined to the ileum, showed noncaseating granulomas. In two other patients the ileal biopsies alone showed histological evidence of tuberculosis with biopsies from the colonic lesions showing non-specific inflammatory changes only. CONCLUSION: Retrograde ileoscopy should be performed in all patients undergoing colonoscopy for suspected ileocolonic tuberculosis. In some patients only the terminal ileum may be involved and histological examination may reveal evidence of tuberculosis only from the lesions in the terminal ileum. | 12 | Isolation of the mucilages from Hibiscus rosasinensis linn.and Okra(Abelmoschus esculentus linn.) and studies of the binding effects of the mucilages显示文摘Objective:To isolate and evaluate comparatively the binding efficacy of the mucilages obtained from the plants of Hibiscus rosasinensis and Okra(Abelmoschus esculentus).Methods:Extraction of mucilages from the leaves of Hibiscus and pods of Okra(Ladies finger) was carried out by a cold maceration process.The extracted mucilages were subjected to various physicochemical properties for its suitability as an excipient in the formulation of tablet dosage form.Different concentrations(10,8,5,2 and 1%w/v) of binder solutions of Hibiscus and Okra were used for the formulation of tablets and the formulated tablets were evaluated by studying the standard parameters like diameter,thickness,weight variation,hardness,friability,disintegration and in vitro dissolution.Stability studies of the formulated tablets were conducted for four weeks. Results:The formulated tablets prepared using the mucilages of both Hibiscus and Okra had good appearance.The in vitro drug release profile of the tablets prepared using Okra mucilage had an optimum of 90%at a mucilage concentration of 1%w/v concentration mucilage itself within 4 h.Conclusions:According to the observations,the lower concentration levels of Okra can be used as an alternative binder to starch.The higher concentration levels of Okra mucilage show a slow and sustained release,and can be considered as an alternative natural excipient in the modified drug delivery systems.At the same time,the above natural excipient of Hibiscus mucilage could be used as a platform for prolonged release if its binder concentrations are increased. | Ameena K Dilip C Saraswathi R Krishnan PN Sankar C Simi SP | 2010 | Asian Pacific Journal of Tropical Medicine2010，3，7： | 8 |

Appropriate DNA markers must be selected for the phylogenetic analyses of Cyprinidae. In present study, the 1st intron of the S7 ribosomal protein (r-protein) gene is first used to examine the relationships among cyprinid fishes. The length of the 1st intron obtained by PCR amplification ranges from 655 to 859 bp in the 16 cyprinid species investigated, and is 602 bp in Myxocyprinus asiaticus. Out of the alignment of 925 nucleotide sites obtained, the parsimony informative sites are 499 and occupy 54% of the total sites. The results indicate that the 1st intron sequences of the S7 r-protein gene in cyprinids are rich in informative sites and vary remarkably in sequence divergence from 2.3% between close species to 66.6% between distant species.
