1Review on Zinc Oxide Nanoparticles: Antibacterial Activity and Toxicity Mechanism显示文摘Antibacterial activity of zinc oxide nanoparticles(Zn O-NPs) has received significant interest worldwide particularly by the implementation of nanotechnology to synthesize particles in the nanometer region. Many microorganisms exist in the range from hundreds of nanometers to tens of micrometers. Zn O-NPs exhibit attractive antibacterial properties due to increased specific surface area as the reduced particle size leading to enhanced particle surface reactivity. Zn O is a bio-safe material that possesses photo-oxidizing and photocatalysis impacts on chemical and biological species. This review covered Zn O-NPs antibacterial activity including testing methods, impact of UV illumination, Zn O particle properties(size, concentration, morphology, and defects), particle surface modification, and minimum inhibitory concentration. Particular emphasize was given to bactericidal and bacteriostatic mechanisms with focus on generation of reactive oxygen species(ROS) including hydrogen peroxide(H2O2), OH-(hydroxyl radicals), and O2-2(peroxide). ROS has been a major factor for several mechanisms including cell wall damage due to Zn O-localized interaction, enhanced membrane permeability, internalization of NPs due to loss of proton motive force and uptake of toxic dissolved zinc ions.These have led to mitochondria weakness, intracellular outflow, and release in gene expression of oxidative stress which caused eventual cell growth inhibition and cell death. In some cases, enhanced antibacterial activity can be attributed to surface defects on Zn O abrasive surface texture. One functional application of the Zn O antibacterial bioactivity was discussed in food packaging industry where Zn O-NPs are used as an antibacterial agent toward foodborne diseases. 2Relations of Budd-Chiari syndrome to prothrombin gene mutation显示文摘BACKGROUND: Budd-Chiari syndrome (BCS) is a type of disease characterized by portal hypertension and/or hy- pertension of the inferior vena cava (IVC) due to the ob- struction of the hepatic veins (HV) and/or intrahepatic IVC outlet. Being etiologically complicated and obscure, BCS can be acquired or idiopathic and several gene muta- tions may be contributable. This study was to explore whether prothrombin gene mutation (F G20210A) takes part in the pathogenesis of BCS and to investigate their cor- relativity. METHODS: In 38 proven BCS patients and 70 controls, polymerase chain reaction-restriction fragment length poly- morphism (PCR-RFLP) was used to find F G20210A mutation. To detect whether there are any mutations, four steps were taken: purification of genome DNA from whole blood, amplification of special fragment by polymerase chain reaction, digestion of the fragment via restriction en- donuclease, and analysis of results by polyacrylamide gel electrophoresis. RESULTS: F G20210A mutation was not detected in all patients and controls. CONCLUSIONS: No F G20210A mutation exists in Chi- nese patients with BCS, nor correlativity between the oc- currence of BCS and F G20210A mutation. 3南海4种珊瑚附生可培养真菌多样性分析显示文摘本研究利用从南海采集的4种珊瑚分离附生真菌,并通过菌株的分类鉴定及系统发育分析获得珊瑚附生可培养真菌的多样性信息.选用酵母麦芽糖琼脂培养基(YM)、马铃薯葡萄糖琼脂培养基(PDA)、马丁氏培养基(Martin)、察氏琼脂培养基(CDA)和孟加拉红培养基(RBM)等5种培养基,采用平板涂布法培养分离附生真菌,基于ITS-rRNA基因序列对真菌菌株进行鉴定和系统发育分析.从4种珊瑚上共分离获得10个属的120株真菌,分别是:青霉属(Penicillum)56株、曲霉属(Aspergillus)25株、枝顶孢属(Acremonium)2株、枝孢属(Cladosporium)5株、毕赤酵母属(Meyerozyma)14株、隔孢伏革属(Peniophora)2株、拟茎点霉属(Phomopsis)1株、裂褶菌属(Schizophyllum)1株、梗孢酵母属(Sterigmatomyces)13株、篮状菌属(Talaromyces)1株.通过对不同珊瑚品种分离获得的附生真菌的比较,发现小叶鹿角珊瑚(Acropora microphthalma)和花鹿角珊瑚(Acropora floride)上分离到的菌株多样性最高,包含6属10种.进一步的分析发现不同珊瑚样品上真菌种类分布有较大差异,隔孢伏革属和裂褶菌属真菌均分离自小叶鹿角珊瑚,梗孢酵母属和篮状菌属均分离自角孔珊瑚(Goniopora sp.),扁脑珊瑚(Platygyra sp.)上则分离到拟茎点霉属.同时还发现不同培养基分离真菌的能力不同,YM培养基分离的菌株最多,CDA培养基分离真菌的多样性最多.以上的研究结果表明,南海珊瑚上存在大量且丰富的真菌资源,可以为后续生物活性物质的开发利用提供良好的菌种资源.郭双双 徐炜 Siti Aisyah Alias 何高阳 骆祝华 2018应用海洋学学报2018,37,1:7
4Antibiotic sensitivity pattern of bacterial pathogens in the intensive care unit of Fatmawati Hospital,Indonesia显示文摘Objective:To evaluate the sensitivity pattern of bacterial pathogens in the intensive care unit(ICU) of a tertiary care of Falmawati Hospital Jakarta Indonesia.Methods:A cross sectional retrospective study of bacterial pathogen was carried out on a total of 722 patients that were admitted to the ICU of Fatmawati Hospital Jakarta Indonesia during January 2009 to March 2010. All bacteria were identified by standard microbiologic methods,and(heir antibiotic susceptibility testing was performed using disk diffusion method.Results:Specimens were collected from 385 patients who were given antimicrobial treatment,of which 249(64.68%) were cultured positive and 136(35.32%) were negative.The most predominant isolate was Pseudomonas aeruginosa(P.aeruginosa)(26.5%) followed by Klebsiella pneumoniae(K.pneumoniae)(15.3%) and Staphylococcus epidermidis(14.9%).P.aeruginosa isolates showed high rate of resistance to cephalexin(95.3%),cefotaxime(64.1%),and ceftriaxone(60.9%).Amikacin was the most effective(84.4%) antibiotic against P.aeruginosa followed by imipenem(81.2%),and meropenem(75.0%).K.pneumoniae showed resistance to cephalexin(86.5%),ceftriaxone(75.7%),ceftazidime(73.0%),cefpirome(73.0%) and cefotaxime(67.9%),respectively.Conclusions:Most bacteria isolated from ICU of Fatmawati Hospital Jakarta Indonesia were resistant to the third generation of cephalosporins,and quinolone antibiotics.Regular surveillance of antibiotic susceptibility pallerns is very important for setting orders to guide the clinician in choosing empirical or directed therapy of infected patients.Maksum Radji Siti Fauziah Nurgani Aribinuko 2011Asian Pacific Journal of Tropical Biomedicine2011,1,1:7
5Survival analysis of cholangiocarcinoma:A 10-year experience in Malaysia显示文摘AIM:To investigate the clinical features and survival of patients treated for cholangiocarcinoma in our institution and to analyze the factors affecting their survival.METHODS:This retrospective cohort study assessed patients diagnosed with cholangiocarcinoma between January 1997 and December 2007 at the University Malaya Medical Centre in Malaysia.The clinical data and associated outcomes were collected using a structured proforma.RESULTS:Of the 69 patients diagnosed with cholangiocarcinoma,38 (55%) were male;mean patient age was 61 years.Twelve patients (17%) had intrahepatic,38 (55%) had perihilar and 19 (28%) had distal tumors.Only 12 patients underwent curative surgery,including seven R0 resections.Only one patient died within 30 d after surgery.The overall median survival was 4 mo,whereas the median survival of R0 resected patients was 16 mo.The overall 1-,2-and 3-year cumulative survival rates were 67%,17% and 17%,respectively.Survival rates were significantly associated with curative resection (P=0.002),intrahepatic tumor (P=0.003),negative margin status (P=0.013),early tumor stage (P=0.016),higher tumor differentiation (P=0.032) and absence of jaundice (P=0.038).Multivariate analysis showed that tumor location was a significant independent predictor of patient survival.CONCLUSION:Curative,margin-negative resection of early stage,well-differentiated intrahepatic tumors is associated with improved patient survival.Ahmad Ramzi Yusoff Mohd Muzammil Abdul Razak Yoong Boon Koon R Vijeyasingam Siti Zuraidah Mahmud 2012World Journal of Gastroenterology2012,18,5:7
6Dual-specificity phosphatase 6(DUSP6): a review of its molecular characteristics and clinical relevance in cancer显示文摘Mitogen-activated protein kinases(MAPKs) are the main regulators of cellular proliferation, growth, and survival in physiological or pathological conditions. Aberrant MAPK signaling plays a pivotal role in carcinogenesis, which leads to development and progression of human cancer. Dual-specificity phosphatase 6(DUSP6), a member of the MAPK phosphatase family, interacts with specifically targeted extracellular signal-regulated kinase 1/2 via negative feedback regulation in the MAPK pathway of mammalian cells. This phosphatase functions in a dual manner, pro-oncogenic or tumor-suppressive, depending on the type of cancer. To date, the tumor-suppressive role of DUSP6 has been demonstrated in pancreatic cancer, non-small cell lung cancer, esophageal squamous cell and nasopharyngeal carcinoma, and ovarian cancer. Its pro-oncogenic role has been observed in human glioblastoma, thyroid carcinoma, breast cancer, and acute myeloid carcinoma. Both roles of DUSP6 have been documented in malignant melanoma depending on the histological subtype of the cancer. Loss-or gain-of-function effects of DUSP6 in these cancers highlights the significance of this phosphatase in carcinogenesis. Development of methods that use the DUSP6 gene as a therapeutic target for cancer treatment or as a prognostic factor for diagnosis and evaluation of cancer treatment outcome has great potential. 7Risk modification of colorectal cancer susceptibility by interleukin-8-251T>A polymorphism in Malaysians显示文摘AIM: To investigate the allele and genotype frequencies and associated risk of interleukin (IL)-8-251T>A polymorphism on colorectal cancer (CRC) susceptibility risk. METHODS: Peripheral blood samples of 255 normal controls and 255 clinically and histopathologically confirmed CRC patients were genotyped for IL-8-251T>A polymorphism employing allele-specific polymerase chain reaction. The relative association of variant allele and genotypes with CRC susceptibility risk was determined by calculating the odds ratios (ORs). Corresponding χ 2 tests on the CRC patients and controls were carried out and 95% confidence intervals (CIs) were determined using Fisher's exact test. The allele frequencies and its risk association were calculated using FAMHAP, haplotype association analysis software. RESULTS: On comparing the frequencies of genotypesof patients and controls, the homozygous variant AA was significantly higher in CRC patients (P = 0.002) compared to controls. Investigation on the association of the polymorphic genotypes with CRC susceptibility risk, showed that the homozygous variant IL-8 -251AA had a significantly increased risk with OR 3.600 (95% CI: 1.550-8.481, P = 0.001). In the case of allele frequencies, variant allele A of IL-8 -251 showed a significantly increased risk of CRC predisposition with OR 1.32 (95% CI: 1.03-1.69,P = 0.003). 9Mechanical and degradation behaviour of biodegradable magnesium-zinc/hydroxyapatite composite with different powder mixing显示文摘Magnesium-based biomaterials have recently gained great attention as promising candidates for the new generation of biodegradable implants.This study investigated the mechanical performance and biodegradation behaviour of magnesium-zinc/hydroxyapatite(Mg-Zn/HA)composites fabricated by different powder mixing techniques.A single step mixing process involved mechanical alloying or mechanical milling techniques,while double step processing involved a combination of both mechanical alloying and mechanical milling.Optimum mechanical properties of the composite were observed when the powders were prepared using single step processing via mechanical alloying technique.However,Mg-Zn/HA composite fabricated through single step processing via mechanical milling technique was found to have the most desirable low degradation rate coupled with highest bioactivity.The composite achieved the lowest degradation rate of 0.039×10^−3 mm/year as measured by immersion test and 0.0230 mm/year as measured by electrochemical polarization.Ca:P ratio of the composite also slightly more than enough to aid the initial bone mineralization,that is 1:1.76,as the required Ca:P ratio for initial bone mineralization is between 1:1 and 1:1.67.Siti Nur Hazwani Mohamad Rodzi Hussain Zuhailawati B.K.Dhindaw 2019Journal of Magnesium and Alloys2019,7,4:5
10Experimental Investigation of Immiscible Supercritical Carbon Dioxide Foam Rheology for Improved Oil Recovery显示文摘This paper presents the rheological behaviour of supercritical CO_2(s CO_2) foam at reservoir conditions of 1 500 psi and 80 °C. Different commercial surfactants were screened and utilized in order to generate a fairly stable CO_2 foam. Mixed surfactant system was also introduced to generate strong foam. Foam rheology was studied for some specific foam qualities using a high pressure high temperature(HPHT) foam loop rheometer. A typical shear thinning behaviour of the foam was observed and a significant increase in the foam viscosity was noticed with the increase of foam quality until 85%. A desired high apparent viscosity with coarse texture was found at 85% foam quality. Foam visualization above 85% showed an unstable foam due to extremely thin lamella which collapsed and totally disappeared in the loop rheometer. Below 5_2%, a non-homogenous and unstable foam was found having low viscosity with some liquid accumulation at the bottom of the circulation loop. 11Antimicrobial activity and essential oils of Curcuma aeruginosa,Curcuma mangga,and Zingiber cassumunar from Malaysia显示文摘Objective:To analyze the chemical composition of the essential oils of Curcuma aeruginosa (C.aeruginosa),Curcuma mangga(C.mangga),and Zingiber cassumunar(Z.cassumunar). and study their antimicrobial activity.Methods:Essential oils obtained by steam distillation were analyzed by gas chromatography-mass speclrometry(GC-MS).The antimicrobial activity of the essential oils was evaluated against four bacteria:Bacillus cereus(H.cereus).Staphylococcus aureus(S.aureus).Escherichia coli(E.coli).and Pseudomonas aeruginosa(P.aeruginosa);and two fungi:Candida albicans(C.albicans) and Cyptococcus neoformans(C.neoformans),using disc-diffusion and broth microdilution methods.Results:Cycloisolongifolene,8.9-dehydro-9- formyl(35.29%) and dihydrocoslunolide(22.51%) were the major compounds in C.aeruginosa oil; whereas caryophyllene oxide(18.71%) and caryophyllene(12.69%) were the major compounds in C.mangga oil:and 2,6.9,9-tetramethyl-2.6.10-cycloundecatrien-1-one(60.77%) andα-caryophyllene(23.92%) were abundant in Z.cassumunar oil.The essential oils displayed varying degrees of antimicrobial activity against all lested microorganisms.C.mangga oil had the highest and most broad-spectrum activity by inhibiting all microorganisms tested,with C.neoformans being the most sensitive microorganism by having the lowest minimum inhibitory concentration(MIC) and minimum fungicidal concentration(MFC) values of 0.1μL/mL. 13A NEW DESCRIPA NEW DESCRIPTOR OF AMINO ACIDS BASED ON THE THREEDIMENSIONAL VECTOR OF ATOMIC INTERACTION FIELD显示文摘A noval molecular structural expression method, three-dimensional vector of atomic interac- tion field (3D-VAIF), has been newly developed based on electrostatic and steric interaction between different types of atoms. Feature descriptors of single amino acid, i.e. principal component scores of struc- tural information for amino acids (SSIA), are obtained through calculation of structural information of 20 coded amino acids using principal component analy- sis (PCA) method, and the strict tests are performed on the property of SSIA by three quantitative struc- ture-activity relationships (QSARs)/quantitative se- quence-activity models (QSAMs) models of 58 ngio- tensin-converting enzymes (ACE), 48 bitter tasting thresholds (BTT) and 31 bradykinin potentiating pentapeptides (BPP). Cumulative multiple correlation coefficients (Rc2um) are 0.789, 0.856 and 0.838; and corresponding cross-validated correlation coefficients (QL2OO) are 0.773, 0.837 and 0.815, respectively. 14Distribution of Chironomidae (Insecta: Diptera) in polluted rivers of the Juru River Basin, Penang, Malaysia显示文摘The influence of physical and chemical parameters on the abundance and diversity of chironomids was studied in six rivers with moderate to highly polluted water in the Juru River Basin. The rivers: Ceruk Tok Kun (CTKR) as reference site, and polluted rivers of Pasir (PR), Juru (JR), Permatang Rawa (PRR), Ara (AR) and Kilang Ubi (KUR) were sampled over a period of five months (November 2007-March 2008). Nine chirnomid species: Chironimus kiiensis, C. javanus, Polypedilum trigonus, Microchironomus sp., Dicrotendipes sp., Tanytarsus formosanus, Clinotanypus sp., Tanypus punctipennis and Fittkauimyia sp. were identified. Assessment of their relationships with several environmental parameters was performed using the canonical correspondence analysis (CCA). Tanytarsus formosanus was the most dominant in the relatively clean CTKR and moderately polluted JR with mean densities of 19.66 and 25.32 m 2 , respectively while C. kiiensis was abundant in more polluted rivers. Tanytarsus formosanus, Dicrotendipes sp. and Microchironomus sp. were grouped under moderate to high water temperature, total organic matter (TOM), total suspended solids (TSS), velocity, pH, phosphates and sulphates. However, Tanypus punctipennis, Fittkauimyia sp., and Clinotanypus sp. were associated with high contents of river sediment such as TOM, Zn and Mn and water ammonium-N and nitrate-N and they were associated with higher dissolved oxygen (DO) content in the water. Chironomus kiiensis, C. javanus and P. trigonus showed positive relationships with TOM, ammonium-N and nitrate-N as well as trace metals of Zn, Cu and Mn. These three species could be considered as tolerant species since they have the ability to survive in extreme environmental conditions with low DO and high concentrations of pollutants. Based on the water parameter scores in all rivers, the highest diversity of chironomid larvae was reported in CTKR. With higher concentrations of organic and/or inorganic pollutants as reported in PPR, KUR and AR, the chironomid larval diversity decreased, and the abundance of tolerant species, mainly Chironomus spp., increased.Salman A. 15A comprehensive review of edible bird nests and swiftlet farming显示文摘Edible bird's nest(EBN) is currently widely consumed by the Chinese community as tonic food and functional food,which is believed to have many medicinal benefits.Some studies have reported the biochemical compositions of EBN,graded on the basis of colour,nitrate and nitrite contents.Other studies have shown significant biological effects,while ongoing research is in progress to explore potential pharmacological applications.The high demand for EBNs in the global market has forced the local regulatory bodies to monitor swiftlet farming activities,including the EBN cleaning process.Furthermore,numerous techniques have been developed to authenticate EBN;proteomics is likely to be the most promising of these methods.However,there are limited numbers of relevant protein sequences deposited at the database.More research is needed at the molecular level to explore the mechanisms behind the biological functions,such as bone strength improvement,skin rejuvenation,epidermal growth factor activity and cell proliferation.The current and future prospects of EBN and swiftlet farming are critically reviewed in this article.Lee Suan Chua Siti Najihah Zukefli 2016Journal of Integrative Medicine2016,14,6:4
16Genetic barriers in transplantation medicine显示文摘The successful of transplantation is determined by the shared human leukocyte antigens(HLAs) and ABO blood group antigens between donor and recipient. In recent years, killer cell receptor [i.e., killer cell immunoglobulinlike receptor(KIR)] and major histocompatibility complex(MHC) class I chain-related gene molecule(i.e., MICA) were also reported as important determinants of transplant compatibility. At present, several different genotyping techniques(e.g., sequence specific primer and sequence based typing) can be used to characterize blood group, HLA, MICA and KIR and loci. These molecular techniques have several advantages because they do not depend on the availability of anti-sera, cellular expression and have greater specificity and accuracy compared with the antibody-antigen based typing. Nonetheless, these molecular techniques have limited capability to capture increasing number of markers which have been demonstrated to determine donor and recipient compatibility. It is now possible to genotype multiple markers and to the extent of a complete sequencing of the human genome using next generation sequencer(NGS). This high throughput genotyping platform has been tested for HLA, and it is expected that NGS will be used to simultaneously genotype a large number of clinically relevant transplantation genes in near future. This is not far from reality due to the bioinformatics support given by the immunogenetics community and the rigorous improvement in NGS methodology. 17Fabrication of multi-level branched metal nanowires by AAO template electro-deposition显示文摘Anodic aluminum oxide (AAO) tem- plates with various branched pores were prepared through one- to multi-step varying the anodic voltage by a factor of 1/ 2 during an anodization of alumi- num. The corresponding branched Fe nanowires (i.e. Y-branched, two-level branched and more complex tree-like nanostructures) were achieved by electrodepositing Fe into the branched pores of the AAO template. Selecting area electron diffraction and high-resolution TEM images show that the stem and branches of the branched Fe nanowires are single- crystalline with a face-centered cubic lattice. 18Spiral CT localization of pancreatic function-ing islet cell tumors显示文摘BACKGROUND: Surgeons are always concerned about the localization of pancreatic functioning islet cell tumor. If the tumor is accurately localized before operation, resection of the pancreatic body and tail without intention can be avoided. The purpose of this study was to evaluate spiral CT localization of pancreatic functioning islet cell tumors and CT techniques. METHODS: CT manifestations in 6 patients with clinically and pathologically proved pancreatic functioning islet cell tumors ware analyzed retrospectively. RESULTS: In 4 patients with insulinomas and 2 patients with glucagonomas, 5 were localized accurately by CT be- fore surgery and 1 was detected retrospectively. The en- hancement of tumors was greater than that of normal pan- creas in arterial phase and pancreatic parenchymal phase. Four patients showed mild high-density and 2, iso-density in the portal venous phase. 19Synthesis, Characterization and X-ray Structure of N-(4-bromobutanoyl-N'-(2-3-and 4-methylphenyl)thiourea显示文摘N-4-(bromobutanoyl)-N'-(o-, m- and p-tolyl)thioureas 3a, 3b and 3c respectively, were synthesized by the reaction of 4-bromobutanoylisothiocyanate with p-, m- and o-toludine. The products were characterized by IR, and NMR spectroscopic techniques. The two carbonoylthiourea isomers N-(4-bromobutanoyl)-N'-(3-methylphenyl)thiourea(3b) and N-(4-bromobutanoyl)-N'-(4-methylphenyl)thiourea(3c) were obtained in crystalline form by recrystallization from DMSO. Xray crystallographic studies showed that both compounds 3b and 3c crystallize in triclinic system with space group of P1. The molecules adopt trans-cis configuration with respect to the positions of 4-bromobutanoyl and tolyl groups respectively, against the thiono C=S bond across their C–N bonds. The configuration is attributed by the intrahydrogen bond between the carbonyl oxygen and amide hydrogen atoms.
