| 2 | Application of platelet-rich plasma with stem cells in bone and periodontal tissue engineering显示文摘Presently, there is a high paucity of bone grafts in the United States and worldwide. Regenerating bone is of prime concern due to the current demand of bone grafts and the increasing number of diseases causing bone loss. Autogenous bone is the present gold standard of bone regeneration. However, disadvantages like donor site morbidity and its decreased availability limit its use. Even allografts and synthetic grafting materials have their own limitations. As certain specific stem cells can be directed to differentiate into an osteoblastic lineage in the presence of growth factors(GFs), it makes stem cells the ideal agents for bone regeneration.Furthermore, platelet-rich plasma(PRP), which can be easily isolated from whole blood, is often used for bone regeneration, wound healing and bone defect repair. When stem cells are combined with PRP in the presence of GFs, they are able to promote osteogenesis. | 3 | Effect of wastewater COD/N ratio on aerobic nitrifying sludge granulation and microbial population shift显示文摘The effect of COD/N ratio on the granulation process and microbial population succession was investigated.Four identical sequencing batch reactors,R1,R2,R3 and R4,were operated with various initial COD/N ratios ranging from 0/200 to 800/200(m/m).Ethanol was fed as the source of COD.Aerobic granules were successfully cultivated in R2 and R3,operating with the COD/N ratio of 200/200 and 400/200,respectively.Scanning electron microscope observations indicated that short rod-shaped and spherical bacteria were dominant in R2,while granules produced in R3 were surrounded with a large amount of filamentous bacteria.The average specific nitritation rate in R2 and R3 were 0.019 and 0.008 mg N/(mg MLVSS.hr),respectively.Fluorescence in situ hybridization results demonstrated that nitrifying bacteria population was enriched remarkably in R2.It indicated that nitrification ability and nitrifying bacteria population were enriched remarkably at low COD/N ratio.However,no granules were formed in R1 and R4 which might attribute to either limited or excessive extracellular polymeric substances production.This study contributed to a better understanding of the role of COD/N ratio in nitrifying sludge granulation. | Lei Wu Chengyao Peng Yongzhen Peng Lingyun Li Shuying Wang Yong Ma | 2012 | Journal of Environmental Sciences2012,24,2: | 29 |
| 4 | Denitrifying phosphorus removal in a step-feed CAST with alternating anoxic-oxic operational strategy显示文摘A bench-scale cyclic activated sludge technology(CAST) was operated to study the biological phosphorus removal performance and a series of batch tests was carried out to demonstrate the accumulation of denitrifying polyphosphate-accumulating organisms(DNPAOs) in CAST system.Under all operating conditions,step-feed CAST with enough carbon sources in influent had the highest nitrogen and phosphorus removal effciency as well as good sludge settling performance.The average removal rate of COD,NH4+-N,PO43--P and total nitrogen(TN) was 88.2%,98.7%,97.5% and 92.1%,respectively.The average sludge volume index(SVI) was 133 mL/g.The optimum anaerobic/aerobic/anoxic(AOA) conditions for the cultivation of DNPAOs could be achieved by alternating anoxic/oxic operational strategy,thus a significant denitrifying phosphorus removal occurred in step-feed CAST.The denitrification of NOx--N completed quickly due to step-feed operation and enough carbon sources,which could enhance phosphorus release and further phosphorus uptake capability of the system.Batch tests also proved that polyphosphate-accumulating organisms(PAOs) in the step-feed process had strong denitrifying phosphorus removal capacity.Both nitrate and nitrite could be used as electron acceptors in denitrifying phosphorus removal.Low COD supply with step-feed operation strategy would favor DNPAOs accumulation. | MA Juan PENG Yongzhen WANG Shuying WANG Li LIU Yangt MA Ningpin | 2009 | Journal of Environmental Sciences2009,21,9: | 27 |
| 5 | Relationship between Cognition Function and Hippocampus Structure after Long-term Microwave Exposure显示文摘Objective To analyze the effects of long-term microwave exposure on hippocampal structure and function in the rat.Methods Experiments were performed on 184 male Wistar rats(three exposure groups and a sham group).Microwaves were applied daily for 6 min over 1 month at average power densities of 2.5,5,and 10 mW/cm2.Learning and memory abilities were assessed by Morris water maze.High performance liquid chromatography was used to detect neurotransmitter concentrations in the hippocampus.Hippocampal structures were observed by histopathological analysis.Results Following long-term microwave exposure there was a significant decrease in learning and memory activity in the 7 d,14 d,and 1 m in all three microwave exposure groups.Neurotransmitter concentrations of four amino acids(glutamate,aspartic acid,glycine,and gamma-aminobutyric acid) in hippocampus were increased in the 2.5 and 5 mW/cm2 groups and decreased in the 10 mW/cm2 group.There was evidence of neuronal degeneration and enlarged perivascular spaces in the hippocampus in the microwave exposure groups.Further,mitochondria became swollen and cristae were disordered.The rough endoplasmic reticulum exhibited sacculated distension and there was a decrease in the quantity of synaptic vesicles.Conclusion These data suggest that the hippocampus can be injured by long-term microwave exposure,which might result in impairment of cognitive function due to neurotransmitter disruption. | ZHAO Li PENG Rui Yun WANG Shui Ming WANG Li Feng GAO Ya Bing DONG Ji LI Xiang SU Zhen Tao | 2012 | Biomedical and Environmental Sciences2012,25,2: | 27 |
| 6 | Diagnostic performance of contrast-enhanced ultrasound and enhanced magnetic resonance for breast nodules显示文摘In the current study, we sought to evaluate the diagnostic efficacies of conventional ultrasound(US), contrastenhanced US(CEUS), combined US and CEUS and magnetic resonance imaging(MRI) in detecting focal solid breast lesions. Totally 117 patients with 120 BI-RADS category 4A-5 breast lesions were evaluated by conventional US and CEUS, and MRI, respectively. SonoVue was used as contrast agent in CEUS and injected as an intravenous bolus; nodule scan was performed 4 minutes after bolus injection. A specific sonographic quantification software was used to obtain color-coded maps of perfusion parameters for the investigated lesion, namely the time-intensity curve.The pattern of contrast enhancement and related indexes regarding the time-intensity curve were used to describe the lesions, comparatively with pathological results. Histopathologic examination revealed 46 benign and 74 malignant lesions. Sensitivity, specificity, and accuracy of US in detecting malignant breast lesions were 90.14%, 95.92%, and 92.52%, respectively. Meanwhile, CE-MRI showed sensitivity, specificity, and accuracy of 88.73%, 95.92%, and91.67%, respectively. The area under the ROC curve for combined US and CEUS in discriminating benign from malignant breast lesions was 0.936, while that of MRI was 0.923, with no significant difference between them, as well as among groups. The time-intensity curve of malignant hypervascular fibroadenoma and papillary lesions mostly showed a fast-in/fast-out pattern, with no good correlation between them(kappa <0.20). This new method is a combination of the unscented transformation and the extended Kalman filter (EKF). The extended Kalman filter is similar to that in a conventional EKF. However, in every running step of the EKF the unscented transformation is running, the deterministic sample is caught by unscented transformation, then posterior mean of nonlinearity is caught by propagating, but the posterior covariance of nonlinearity is caught by linearizing. | 10 | Fractionation of heavy metals in shallow marine sediments from Jinzhou Bay,China显示文摘This work investigated the distribution and speciation of Cd, Cu, Pb, Fe and Mn in the shallow sediments of Jinzhou Bay, Northeast China, which has been heavily contaminated by nonferrous smelting activities. The concentrations of Cd, Cu and Pb in sediments were found to be 100, 13 and 7 times, respectively, being higher than the national guideline (GB 18668-2002). Sequential extraction test showed that 39%-61% of Cd were exchangeable fractions, indicating that Cd in the sediments posed a high risk to local environments. While Cu and Pb were at moderate risk levels. | 11 | Achieving nitritation at low temperatures using free ammonia inhibition on Nitrobacter and real-time control in an SBR treating landfill leachate显示文摘Free ammonia(FA) inhibition on nitrite-oxidized bacteria(NOB) and real-time control are used to achieve nitrogen removal from landfill leachate via nitrite pathway at low temperatures in sequencing batch reactor. The inhibition of FA on NOB activity during the aerobic period was prolonged using real-time control. The degree of nitrite accumulation was monitored along with variations of the ammonia-oxidizing bacteria and NOB population using fluorescence in situ hybridization techniques. It is demonstrated that the end-point of ammonia oxidization is detected from the on-line measured dissolved oxygen,oxidization–reduction potential, and p H signals, which could avoid the loss the FA inhibition on NOB caused by excess aeration. At low temperature(13.0–17.6°C), the level of nitrite pathway rapidly increased from 19.8% to 90%, suggesting that nitritation was successfully started up at low temperature by applying syntrophic association of the FA inhibition and real-time control, and then this high level of nitrite pathway was stably maintained for as long as 233 days. Mechanism analysis shows that the establishment of nitritation was primarily the result of predominant ammonia-oxidizing bacteria developed in the nitrifying bacteria population compared to NOB. | 12 | Achieving partial nitrification by inhibiting the activity of Nitrospira-like bacteria under high-DO conditions in an intermittent aeration reactor显示文摘It is generally accepted that a low dissolved oxygen(DO) concentration is more beneficial for achieving partial nitrification than high-DO. In this study, partial nitrification was not established under low-DO conditions in an intermittent aeration reactor for treating domestic wastewater. During the operational period of low-DO conditions(DO: 0.3 ±0.14 mg/L), stable complete nitrification was observed. The abundance of Nitrospira-like bacteria, which were the major nitrite-oxidizing bacteria, increased from 1.03 × 10~6to2.64 × 10~6cells/m L. At the end of the low-DO period, the batch tests showed that high-DO concentration(1.5, 2.0 mg/L) could inhibit nitrite oxidation, and enhance ammonia oxidation. After switching to the high-DO period(1.8 ± 0.32 mg/L), partial nitrification was gradually achieved. Nitrospira decreased from 2.64 × 10~6 to 8.85 × 10~5cells/m L. | 13 | Wilson disease: Identification of two novel mutations and clinical correlation in Eastern Chinese patients显示文摘AIM: To study mutations in the P-type ATPase (ATP7B) gene responsible for Wilson disease (WD) in the Eastern Chinese population, and the possible correlation of specific mutations with clinical characteristics. METHODS: Mutations of the ATP7B gene were sought by means of direct sequencing in 50 Eastern Chinese WD patients of Han ethnic origin. RESULTS: Two novel mutations, Asp96Gly and Asp196Glu, were first identified. We also compared the characterization of mutations in ATP7B with the clinical findings, and a significant correlation with hepatic manifestations between patients carrying the Arg778Leu mutation and those without was found. CONCLUSION: Gene sequencing analysis was shown to have a high detection rate and accuracy. | 14 | Ecological and Economic Benefits of Vegetation Management Measures in Citrus Orchards on Red Soils显示文摘A three-year experiment was conducted to investigate and compare the economic and ecological benefits of six types of vegetation management measures in citrus orchards of the hilly red soil region of the eastern part of China.Six vegetation treatments,including tillage without herbicide(clean tillage)and no tillage without herbicide(sod culture) and with herbicide paraquat(paraquat),glyphosate(glyphosate),glyphosate-glyphosate-paraquat(G-G-P),and paraquat- paraquat-glyphosate(P-P-G),were applied in the citrus orchards on a clayey red soil with slopes of 8°and 13°and a sandy soil with slope of 25°.The results showed that the sod culture,paraquat,glyphosate,G-G-P,and P-P-G treatments reduced surface runoff by 38.8%,42.5%,18.7%,28.7%,and 37.5%,then the soil-water losses by 55.5%,51.7%,39.9%,46.8%, and 50.0%,and the N,P,and K nutrient losses by 60.3%,50.2%,37.0%,41.8%,and 45.4%,respectively,as compared with the clean tillage treatment.The weed regeneration ratios with the treatments of clean tillage without herbicide,paraquat, glyphosate,G-G-P,and P-P-G were reduced by 55.1%,67.2%,30.3%,36.8%,and 51.2%,respectively,as compared with the sod culture.The sod culture,paraquat,glyphosate,G-G-P,and P-P-G treatments could increase the soil fertility (annual accumulation of N,P,K,and OM)by 7.1%,6.9%,5.3%,6.2%,and 6.6%,respectively,whereas the clean tillage treatment without herbicide reduced soil fertility by 4.4% after the three-year experiment.The citrus fruit yields in the treatments of paraquat,glyphosate,G-G-P,and P-P-G increased by 7%-10%;the soluble solid,total sugar,total acidity,sugar-acid ratio,and single fruit weight of citrus fruits of all treatments except sod culture significantly(P>0.05) exceeded that of the clean tillage treatment.In general,the paraquat treatment showed the best economic and ecological benefits among the six treatments;therefore,it could be regarded as the best available vegetation management measure in citrus orchards of hilly red soil region to retain water and soil,enhance soil fertility,and improve the yield and quality of citrus. | SHUI Jian-Guo WANG Qiu-Zhen LIAO Gen-Qing J. | 15 | Microwave Exposure Impairs Synaptic Plasticity in the Rat Hippocampus and PC12 Cells through Over-activation of the NMDA Receptor Signaling Pathway显示文摘Objective The aim of this study is to investigate whether microwave exposure would affect the N-methyl-D-aspartate receptor(NMDAR) signaling pathway to establish whether this plays a role in synaptic plasticity impairment. Methods 48 male Wistar rats were exposed to 30 m W/cm2 microwave for 10 min every other day for three times. Hippocampal structure was observed through H&E staining and transmission electron microscope. PC12 cells were exposed to 30 m W/cm2 microwave for 5 min and the synapse morphology was visualized with scanning electron microscope and atomic force microscope. The release of amino acid neurotransmitters and calcium influx were detected. The expressions of several key NMDAR signaling molecules were evaluated. Results Microwave exposure caused injury in rat hippocampal structure and PC12 cells, especially the structure and quantity of synapses. The ratio of glutamic acid and gamma-aminobutyric acid neurotransmitters was increased and the intracellular calcium level was elevated in PC12 cells. A significant change in NMDAR subunits(NR1, NR2 A, and NR2B) and related signaling molecules(Ca2+/calmodulin-dependent kinase II gamma and phosphorylated c AMP-response element binding protein) were examined. Conclusion 30 m W/cm2 microwave exposure resulted in alterations of synaptic structure, amino acid neurotransmitter release and calcium influx. | 16 | A structural view of the antibiotic degradation enzyme NDM-1 from a superbug显示文摘Gram-negative Enterobacteriaceae with resistance to carbapenem conferred by New Delhi metallo-β-lactamase 1(NDM-1)are a type of newly discovered antibioticresistant bacteria.The rapid pandemic spread of NDM-1 bacteria worldwide(spreading to India,Pakistan,Europe,America,and Chinese Taiwan)in less than 2 months characterizes these microbes as a potentially major global health problem.The drug resistance of NDM-1 bacteria is largely due to plasmids containing the blaNDM-1 gene shuttling through bacterial populations.The NDM-1 enzyme encoded by the blaNDM-1 gene hydrolyzes β-lactam antibiotics,allowing the bacteria to escape the action of antibiotics.Although the biological functions and structural features of NDM-1 have been proposed according to results from functional and structural investigation of its homologues,the precise molecular characteristics and mechanism of action of NDM-1 have not been clarified.Here,we report the threedimensional structure of NDM-1 with two catalytic zinc ions in its active site.Biological and mass spectroscopy results revealed that D-captopril can effectively inhibit the enzymatic activity of NDM-1 by binding to its active site with high binding affinity.The unique features concerning the primary sequence and structural conformation of the active site distinguish NDM-1 from other reported metallo-β-lactamases(MBLs)and implicate its role in wide spectrum drug resistance.We also discuss the molecular mechanism of NDM-1 action and its essential role in the pandemic of drug-resistant NDM-1 bacteria.Our results will provide helpful information for future drug discovery targeting drug resistance caused by NDM-1 and related metallo-β-lactamases. | Yu Guo Jing Wang Guojun Niu Wenqing Shui Yuna Sun Honggang Zhou Yaozhou Zhang Cheng Yang Zhiyong Lou Zihe Rao | 2011 | Protein & Cell2011,2,5: | 16 |

In this review, we discuss the immune characteristics of liver cancer, the obstacles encountered during the application of CAR-T therapy, and preclinical and clinical progress in the use of CAR-T therapy in patients with liver cancer.Data sources: The data on CAR-T therapy related to liver cancers were collected by searching Pub Med and the Web of Science databases prior to December 2017 with the keywords 'chimeric antigen receptor','CAR-T', 'liver cancer', 'hepatocellular carcinoma', and 'solid tumor'. Additional articles were identified by manual search of references found in the primary articles. The data for clinical trials were collected by searching Clinical Trials.gov.Results: The liver has a tolerogenic nature in the intrahepatic milieu and its tumor microenvironment significantly affects tumor progression. The obstacles that reduce the efficacy of CAR-T therapy in solid tumors include a lack of specific tumor antigens, limited trafficking and penetration of CAR-T cells to tumor sites, and an immunosuppressive tumor microenvironment. To overcome these obstacles, several strategies have emerged. In addition, several strategies have been developed to manage the side effects of CAR-T, including enhancing the selectivity of CARs and controlling CAR-T activity. To date, no clinical trials of CAR-T therapy against HCC have been completed. However, preclinical studies in vitro and in vivo have shown potent antitumor efficacy. Glypican-3, mucin-1, epithelial cell adhesion molecule, carcinoembryonic antigen, and other targets are currently being studied.Conclusions: The application of CAR-T therapy for liver cancer is just beginning to be explored and more research is needed. | 19 | Accurate authentication of Dendrobium officinale and its closely related species by comparative analysis of complete plastomes显示文摘Owing to its great medicinal and ornamental values, Dendrobium officinale is frequently adulterated with other Dendrobium species on the market. Unfortunately, the utilization of the common DNA markers ITS,ITS2, and mat Ktrbc L is unable to distinguish D. officinale from 5 closely related species of it(D. tosaense, D.shixingense, D. flexicaule, D. scoriarum and D. aduncum). Here, we compared 63 Dendrobium plastomes comprising 40 newly sequenced plastomes of the 6 species and 23 previously published plastomes. The plastomes of D. officinale and its closely related species were shown to have conserved genome structure and gene content. Comparative analyses revealed that small single copy region contained higher variation than large single copy and inverted repeat regions, which was mainly attributed to the loss/retention of ndh genes.Furthermore, the intraspecific sequence variability among different Dendrobium species was shown to be diversified, which necessitates a cautious evaluation of genetic markers specific for different Dendrobium species.By evaluating the maximum likelihood trees inferred from different datasets, we found that the complete plastome sequence dataset had the highest discriminatory power for D. officinale and its closely related species,indicating that complete plastome sequences can be used to accurately authenticate Dendrobium species.& 2018 Chinese Pharmaceutical Association and Institute of Materia Medica, Chinese Academy of Medical Sciences.
