8Initial steroid-free immunosuppression after liver transplantation in recipients with hepatitis c virus related cirrhosis显示文摘AIM: Steroids can increase hepatitis C virus (HCV) replication. After liver transplantation (LTx), steroids are commonly used for immunosuppression and acute rejection is usually treated by high steroid dosages. Steroids can worsen the outcome of recurrent HCV infection. Therefore,we evaluated the outcome of HCV infected liver recipients receiving initial steroid-free immunosuppression.METHODS: Thirty patients undergoing LTx received initial steroid-free immunosuppression. Indication for LTx included 7 patients with HCV related drrhosis. Initial immunosuppression consisted of tacrolimus 2x0.05 mg/kg.d po and mycophenolate mofetil (MMF) 2x15 mg/kg.d po. The tacrolimus dosage wasa djusted to trough levels in the target range of 10-15 μg/Ldudng the first 3 mo and 5-10 μg/L thereafter. Manifestations of acute rejection were verified histologically.RESULTS: Patient and graft survival of 30 patients receiving initial steroid-free immunosuppression was 86% and 83% at 1 and 2 years. Acute rejection occurred in 8/30 patients,including 1 HCV infected recipient. All HCV-infected patients had HCV genotype II (lb). HCV seropositivity occurred within the first 4 mo after LTx. The virus load was not remarkably increased during the first year after LTx. Histologically, grafts had no severe recurrent hepatitis.CONCLUSION: From our experience, initial steroid-free immunosuppression does not increase the risk of acute rejection in HCV infected liver recipients. Furthermore, none of the HCV infected patients developed serious chronic liver diseases. 9Partial external biliary diversion in bile salt export pump deficiency: Association between outcome and mutation显示文摘AIM To investigate the relation of two different mutations to the outcome of partial external biliary diversion(PEBD)in severe bile salt export pump(BSEP) deficiency.METHODS Mutations in the gene encoding BSEP leading to severe BSEP deficiency in two unrelated patients were identified by genomic sequencing. Native liver biopsies and transiently transfected human embryonic kidney(HEK) 293 cells expressing either wild-type or mutated BSEP were subjected to immunofluorescence analysis to assess BSEP transporter localization. Bile acid profiles of patient and control bile samples were generated by ultra-performance liquid chromatographytandem mass spectrometry. Wild-type and mutant BSEP transport of [~3H]-labeled taurocholate(TC) and taurochenodeoxycholate(TCDC) was assessed by vesicular transport assays.RESULTS A girl(at 2 mo) presented with pruritus, jaundice and elevated serum bile salts(BS). PEBD stabilized liver function and prevented liver transplantation. She was heterozygous for the BSEP deletion p.T919 del and the nonsense mutation p.R1235 X. At the age of 17 years relative amounts of conjugated BS in her bile were normal, while total BS were less than 3% as compared to controls. An unrelated boy(age 1.5 years) presenting with severe pruritus and elevated serum BS was heterozygous for the same nonsense and another missense mutation, p.G1032 R. PEBD failed to alleviate pruritus, eventually necessitating liver transplantation. BS concentration in bile was about 5% of controls. BS were mainly unconjugated with an unusual low amount of chenodeoxycholate derivatives(< 5%). The patients' native liver biopsies showed canalicular BSEP expression. Both BSEP p.T919 del and p.G1032 R were localized in the plasma membrane in HEK293 cells. In vitro transport assays showed drastic reduction of transport by both mutations. 13Characterization of novel Brown midrib 6 mutations affecting lignin biosynthesis in sorghum显示文摘The presence of lignin reduces the quality of lignocellulosic biomass for forage materials and feedstock for biofuels. In C4 grasses,the brown midrib phenotype has been linked to mutations to genes in the monolignol biosynthesis pathway. For example,the Bmr6 gene in sorghum(Sorghum bicolor) has been previously shown to encode cinnamyl alcohol dehydrogenase(CAD),which catalyzes the final step of the monolignol biosynthesis pathway. Mutations in this gene have been shown to reduce the abundance of lignin,enhance digestibility,and improve saccharification efficiencies and ethanol yields. Nine sorghum lines harboring five different bmr6 alleles were identified in an EMS-mutagenized TILLING population. DNA sequencing of Bmr6 revealed that the majority of the mutations impacted evolutionarily conserved amino acids while three-dimensional structural modeling predicted that all of these alleles interfered with the enzyme's ability to bind with its NADPH cofactor. All of the new alleles reduced in vitro CAD activity levels and enhanced glucose yields following saccharification. Further,many of these lines were associated with higher reductions in acid detergent lignin compared to lines harboring the previously characterized bmr6-ref allele. 15Challenges and perspectives for structural biology of IncRNAs-the example of the Xist IncRNA A-repeats显示文摘Following the discovery of numerous long non-coding RNA(IncRNA)transcripts in the human genome,their important roles in biology and human disease are emerging.Recent progress in experimental methods has enabled the identification of structural features of IncRNAs.However,determining high-resolution structures is challenging as IncRNAs are expected to be dynamic and adopt multiple conformations,which may be modulated by interaction with protein binding partners.The X-inactive specific transcript(Xist)is necessary for X inactivation during dosage compensation in female placental mammals and one of the beststudied IncRNAs.Recent progress has provided new insights into the domain organization,molecular features,and RNA binding proteins that interact with distinct regions of Xist.The A-repeats located at the 5'end of the transcript are of particular interest as they are essential for mediating silencing ofthe inactive X chromosome.Here,we discuss recent progress with elucidating structural features of the Xist IncRNA,focusing on the A-repeats.We discuss the experimental and computational approaches employed that have led to distinct structural models,likely reflecting the intrinsic dynamics of this RNA.The presence of multiple dynamic conformations may also play an important role in the formation ofthe associated RNPs,thus influencing the molecular mechanism underlying the biological function of the Xist A-repeats.We propose that integrative approaches that combine biochemical experiments and high-resolution structural biology in vitro with chemical probing and functional studies in vivo are required to unravel the molecular mechanisms of IncRNAs.Alisha NJones Michael Sattler 2019Journal of Molecular Cell Biology2019,11,10:2
