1Reliable cell purification and determination of cell purity:crucial aspects of olfactory ensheathing cell transplantation for spinal cord repair显示文摘Transplantation of olfactory ensheathing cells,the glia of the primary olfactory nervous system,has been trialed for spinal cord injury repair with promising but variable outcomes in animals and humans.Olfactory ensheathing cells can be harvested either from the lamina propria beneath the neuroepithelium in the nasal cavity,or from the olfactory bulb in the brain.As these areas contain several other cell types,isolating and purifying olfactory ensheathing cells is a critical part of the process.It is largely unknown how contaminating cells such as fibroblasts,other glial cell types and supporting cells affect olfactory ensheathing cell function post-transplantation;these cells may also cause unwanted side-effects.It is also,however,possible that the presence of some of the contaminant cells can improve outcomes.Here,we reviewed the last decade of olfactory ensheathing cell transplantation studies in rodents,with a focus on olfactory ensheathing cell purity.We analyzed how purification methods and resultant cell purity differed between olfactory mucosa-and olfactory bulb-derived cell preparations.We analyzed how the studies reported on olfactory ensheathing cell purity and which criteria were used to define cells as olfactory ensheathing cells.Finally,we analyzed the correlation between cell purity and transplantation outcomes.We found that olfactory bulb-derived olfactory ensheathing cell preparations are typically purer than mucosa-derived preparations.We concluded that there is an association between high olfactory ensheathing cell purity and favourable outcomes,but the lack of olfactory ensheathing cell-specific markers severely hampers the field.Ronak Reshamwala Megha Shah Lucy Belt Jenny A.K.Ekberg James A.St John 2020Neural Regeneration Research2020,15,11:3
2The link between olfactory ensheathing cell survival and spinal cord injury repair: a commentary on common limitations of contemporary research显示文摘Olfactory ensheathing cells(OECs)are crucial players in the continuous regeneration of the olfactory nervous system that occurs through out life and are thought to have unique growth-promoting properties.For this reason,OEC transplantation has been thoroughly explored for the potential to promote neural repair after both central and peripheral nervous system injuries.Numerous studies have shown that OEC transplantation is safe and can promote recovery after spinal cord injury(SCI),both in animal models and in human clinical trials.To date,a variety of injury types and time-points after injury,as well as different delivery methods,have been tested.Outcomes have been encouraging(in rodent models including,for example,restoration of locomotion,breathing and climbing ability along with induction of axonal sprouting and some axonal regeneration)but highly variable(Barnett and Riddell,2007;Gomez et al.,2018).Ronak Reshamwala Megha Shah James St.John Jenny Ekberg 2020Neural Regeneration Research2020,15,10:1

10Extreme hyperbilirubinemia: An indicator of morbidity and mortality in sickle cell disease显示文摘BACKGROUND Sickle cell disease(SCD) is a disorder that results in increased hospitalizations and higher mortality. Advances in management have resulted in increases in life expectancy and led to increasing awareness of sickle cell hepatopathy(SCH).However, its impact in patients on the natural history and outcomes of SCD is not known. Our study aims to describe the prevalence of extreme hyperbilirubinemia(EH), one form of SCH, its effect on morbidity and mortality,and correlations between sickle cell genotype and SCH type. We hypothesize that EH is associated with higher morbidity and mortality.AIM To investigate the effects of EH on morbidity and mortality among patients with SCD.METHODS This retrospective cohort study was performed using a database of patients with SCD treated at Grady Memorial Hospital between May 2004 and January 2017.Patients with EH(defined as total bilirubin above 13.0 mg/dL) were identified. A control group was identified from the same database with patients with total serum bilirubin ≤ 5.0 mg/dL. Electronic medical records were used to extract demographic information, laboratory values, radiology results, current medications, need for transfusions and mortality data. Two samples T-test, chi-squared test and Fisher's exact test were then used to compare the parameters between the two groups.RESULTS Out of the database, fifty-seven charts were found of patients with bilirubin > 13 mg/dL. Prevalence of severe SCH as defined by EH was 4.8%(57/1172). There were no demographic differences between patients with and without EH.Significant genotypic differences existed between the two groups, with hemoglobin SS SCD being much higher in the EH group(P < 0.001). Patients with severe EH had a significant elevations in alanine aminotransferase(157.0 ± 266.2 IU/L vs 19.8 ± 21.3 IU/L, P < 0.001), aspartate aminotransferase(256.5 ± 485.9 U/L vs 28.2 ± 14.7 U/L, P < 0.001) and alkaline phosphatase(218.0 ± 176.2 IU/L vs 85.9 ± 68.4 IU/L, P < 0.001). Patients with EH had significantly higher degree of end organ failure measured with quick Sequential Organ Failure Assessment scores(0.42 ± 0.68 vs 0.01 ± 0.12, P < 0.001), increased need for blood products(63% vs 5%, P < 0.001), and exchange transfusions(10.5% vs 1.3%, P = 0.022).CONCLUSION Among patients with SCD, elevated levels of total bilirubin are rare, but indicative of elevated morbidity, mortality, and need for blood transfusions.Large differences in sickle cell genotype also exist, but the significance of this is unknown.John Paul Haydek Cesar Taborda Rushikesh Shah Preeti A Reshamwala Morgan L McLemore Fuad El Rassi Saurabh Chawla 2019World Journal of Hepatology2019,11,3:0
