| 1 | Direct injection HILIC-MS/MS analysis of darunavir in rat plasma applying supported liquid extraction显示文摘A novel bioanalytical method was developed and validated for the quantitative determination of darunavir(DRV) in rat plasma by employing hydrophilic interaction chromatography and tandem mass spectrometry(HILIC-MS/MS) with supported liquid extraction(SLE).lrbesartan(IRB) was used as an internal standard(IS).The analyte in rat plasma(200 μL) was isolated through SLE using ethyl acetate as the eluting solvent.The chromatographic separation was achieved on Luna-HILIC(250 mm × 4.6 mm,5 μm)column with a mobile phase of 0.1% of formic acid in water:acetonitrile(5:95,v/v),at a constant flow rate of 1.0mL/min.The MS/MS ion transitions for DRV(548.1→392.0) and IS(429.2→207.1) were monitored on an ion trap mass spectrometer,operating in the multiple reaction monitoring(MRM) mode.The lower limit of quantitation(LLOQ) was 0.2 ng/mL and quantitation range was 0.2-5000 ng/mL.The method was validated for its selectivity,sensitivity,carryover,linearity,precision,accuracy,recovery,matrix effect and stability.The method was successfully applied to pharmacokinetic study in rats. | Bokka Ramesh Nemali Manjula Sistla Ramakrishna Potturi Sita Devi | 2015 | Journal of Pharmaceutical Analysis2015,5,1: | 2 |

| 5 | LC–HRMS determination of piperine on rat dried blood spots: A pharmacokinetic study显示文摘A liquid chromatography–high resolution mass spectrometry(LC–HRMS) method was developed and validated for the determination of piperine(PPR) on dried blood spots(DBS). DBS samples were prepared by spiking the whole blood with analyte to produce 30 m L of blood spots on specimen collection cards.Chromatographic separation was achieved on an Atlantis d C18 column using acetonitrile and water(0.1%formic acid)(85:15, v/v) as mobile phase in an isocratic mode of elution at a flow rate of 0.75 m L/min. MS detection was carried out in electrospray positive ion mode for the target ions and monitored at m/z286.1465 for PPR and 272.1303 for the internal standard(IS). The developed method exhibited a linear dynamic range over 0.01–2000 ng/m L for PPR on DBS. The overall extraction recovery of PPR from DBS was 92.5%. Influence of hematocrit and spot volume on DBS was also evaluated and found to be well within the acceptable limits. The method was successfully applied to pharmacokinetic studies of PPR in rats. | Bokka Ramesh P. | 11 | Duodenal neuroendocrine tumor-tertiary care centre experience:A case report显示文摘BACKGROUND Neuroendocrine neoplasms(NENs)are a heterogeneous group of neoplasms arising from neuroendocrine cells,which contribute a small fraction of gastrointestinal malignancies.Duodenal neuroendocrine tumors(dNETs)represent 2%of all gastroenteropancreatic NENs.NENs are heterogeneous in terms of clinical symptoms,location,and prognosis.Non-functional NETs are mostly asymptomatic and need a high degree of clinical suspicion.Diagnosis of NETs is by endoscopic,endosonographic biopsy,and histopathological examination with immunohistochemistry staining for synaptophysin and chromogranin A.CASE SUMMARY We present case reports of 5 patients obtained over a period of 10 years in our center with dNETs.One patient had moderately differentiated NET and the remaining four had well-differentiated NET.Surveillance endoscopy was recommended in all the patients and is kept under regular follow-up after performing endoscopic therapy using endoscopic mucosal resection in 4 of them and one patient was advised to undergo a Whipple procedure.CONCLUSION Recently,the number of reported cases of NETs has increased due to advancements in diagnostic modalities and prevalence because of longer survival duration.The management differs based on the site,size,proliferation grade,and locally invasive pattern.They are slow-growing tumors with a good overall prognosis.The prognosis correlates with local lymph node status and metastasis. | Uma Devi Malladi Suraj Kumar Chimata Ramesh Kumar Bhashyakarla Sahitya Reddy Lingampally Vikas Reddy Venkannagari Zeeshan Ali Mohammed Rahul Vijay Vargiya | 2023 | World Journal of Translational Medicine2023,11,1: | 0 |
| 12 | Kinetics study of metaxalone degradation under hydrolytic, oxidative and thermal stress conditions using stability-indicating HPLC method显示文摘An isocratic stability indicating RP-HPLC-UV method is presented for the determination of metaxalone (MET) in the presence of its degradation products. The method uses Dr. Maisch C18 column (250 mm 4.6 mm, 5 mm) with mobile phase consisting of acetonitrile-potassium dihydrogen orthophosphate buffer with 4 mL of 0.4% triethyl amine (pH 3.0; 10 mM) (58:42, v/v) at a flow rate of 1.0 mL/min. pH of the buffer was adjusted with o-phosphoric acid. UV detection was performed at 225 nm. The method was validated for specificity, linearity, precision, accuracy, limit of detection, limit of quantification and robustness. The calibration plot was linear over the concentration range of 1-100 mg/mL having a correlation coefficient (r2) of 0.999. Limits of detection and quantification were 0.3 and 1 mg/mL, respectively. Intra-day and inter-day precision (% RSD) was 0.65 and 0.79 respectively. The proposed method was used to investigate the degradation kinetics of MET under different stress conditions employed. Degradation of MET followed a pseudo-first-order kinetics, and rate constant (K), time left for 50% potency (t1/2), and time left for 90% potency (t90) were calculated. | Vamsi Krishna Marothu Rajendra N.
