1Protective effect of glutamine on intestinal injury and bacterial community in rats exposed to hypobaric hypoxia environment显示文摘AIM:To investigate the protective effect of glutamine(Gln)on intestinal injury and the bacterial community in rats exposed to hypobaric hypoxia environment.METHODS:Sprague-Dawley rats were divided into control,hypobaric hypoxia(HH),and hypobaric hypoxia+Gln(5.0 g/kg BW·d)(HG)groups.On the first 3 d,all rats were placed in a normal environment.After the third day,the HH and HG groups were transferred into a hypobaric chamber at a simulated elevation of 7000m for 5 d.The rats in the HG group were given Gln by gavage daily for 8 d.The rats in the control and HH groups were treated with the same volume of saline.The intestinal morphology,serum levels of malondialdehyde(MDA),superoxide dismutase(SOD),interleukin-6 (IL-6),tumor necrosis factor-α(TNF-α),interferon-gamma(IFN-γ)and diamino oxidase(DAO)were examined.We also evaluated the expression levels of occludin,toll-like receptor 4(TLR4),nuclear factor-κB p65(NF-κB p65)and myeloid differentiation factor 88(MyD88),and examined the bacterial community in caecal contents.RESULTS:Hypobaric hypoxia induced the enlargement of the heart,liver,lung and kidney,and caused spleen atrophy.Intestinal villi damage was also observed in the HH group.Supplementation with Gln significantly alleviated hypobaric-induced damage to main organs including the intestine,increased serum SOD(1.14±0.03 vs 0.88±0.04,P<0.05)and MDA(8.35±1.60,P<0.01)levels and decreased serum IL-6(1172.13±30.49 vs 1407.05±34.36,P<0.05),TNF-α(77.46±0.78 vs 123.70±3.03,P<0.001),IFN-γ(1355.42±72.80 vs 1830.16±42.07,P<0.01)and DAO(629.30±9.15 vs 524.10±13.34,P<0.001)levels.Moreover,Gln significantly increased occludin(0.72±0.05 vs 0.09±0.01,P<0.001),TLR4(0.15±0.05 vs 0.30±0.09,P<0.05),MyD88(0.32±0.08 vs 0.71±0.06,P<0.01),and NF-κB p65(0.16±0.04 vs 0.44±0.03,P<0.01)expression levels and improved the intestinal bacterial community.CONCLUSION:Gln treatment protects from intestinal injury and regulates the gut flora imbalance in hypoxia environment.These effects may be related to the TLR4/MyD88/NF-κB signaling pathway.Chun-Lan Xu Rui Sun Xiang-Jin Qiao Cui-Cui Xu Xiao-Ya Shang Wei-Ning Niu 2014World Journal of Gastroenterology2014,20,16:29
2The lncRNA Malat1 functions as a ceRNA to contribute to berberine-mediated inhibition of HMGB1 by sponging miR-181c-5p in poststroke inflammation显示文摘Long non-coding RNAs(lncRNAs)have been identified as essential mediators in neurological dysfunction.Our previous study shows that berberine(BBR)hampers the nuclear-to-cytosolic translocation of high-mobility group box 1(HMGB1)in the process of poststroke inflammation.In this study,we explored the role of lncRNA metastasis‐associated lung adenocarcinoma transcript 1(Malat1)in the process of BBR-induced inhibition of HMGB1 in ischemic brain.Before the 60-min MCAO surgery,the mice were pretreated with BBR(50 mg·kg−1 per day,ig)for 14 days or ICV injected with specific lentiviral vector or shRNA.We showed that MCAO caused marked increase in the expression Malat1 and HMGB1 in the ipsilateral cortex,which was significantly attenuated by pretreatment with BBR.Knockdown of Malat1 attenuated the inflammatory injury after brain ischemia,whereas overexpression of Malat1 exacerbated ischemic brain inflammation.Overexpression of Malat1 also reversed BBR-induced reduction of HMGB1 and proinflammatory cytokines.The above results suggested a potential correlation between Malat1 and stroke inflammation.Based on informatics analysis we predicted that HMGB1 was a direct downstream target of miR-181c-5p,whereas Malat1 acted as a competitive endogenous RNA(ceRNA)for miR-181c-5p targeted the 3′-UTR of HMGB1 to promote inflammation after ischemic stroke.Knockdown of Malat1 significantly decreased HMGB1 level,which could be abrogated by transfection with miR-181c-5p inhibitors.Taken together,our results demonstrate for the first time that Malat1/miR-181c-5p/HMGB1 axis may be a key pathway of BBR-induced antiinflammation effects in stroke,and they may provide a novel avenue for targeted therapy.Ding-wen Cao Man-man Liu Rui Duan Yi-fu Tao Jun-shan Zhou Wei-rong Fang Jun-rong Zhu Li Niu Jian-guo Sun 2020Acta Pharmacologica Sinica2020,41,1:11

4Boxb mediate BALB/c mice corneal inflammation through a TLR4/MyD88-dependent signaling pathway in Aspergillus fumigatus keratitis显示文摘AIM: To investigate whether high-mobility group box 1(HMGB1) Boxb exacerbates BALB/c mice corneal immune responses and inflammatory through the Toll-like receptor 4(TLR4)/myeloid differentiation primary response 88(My D88)-dependent signaling pathway in Aspergillus fumigatus(A. fumigatus) keratitis.METHODS: The mice corneas were pretreated with phosphate buffer saline(PBS), Boxb before A. fumigatus infection. The abdominal cavity extracted macrophages were pretreated with PBS, Boxb, TLR4 inhibitor(CLI-095), Dimethyl sulfoxide(DMSO) separately before A. fumigatus hyphae stimulation. HMGB1 was detected in normal and infected mice corneas and macrophages by real-time reverse transcriptase polymerase chain reaction(RT-PCR), the TLR4, My D88, interleukin-1β(IL-1β), tumor necrosis factor-α(TNF-α) were detected by Western blot and PCR.RESULTS: In BALB/c mice corneas, the expressions of TLR4, HMGB1, IL-1β, TNF-α were increased after A. fumigatus infection. While pretreatment with Boxb significantly increased the expressions of TLR4, HMGB1, My D88, IL-1β, TNF-α compared with PBS control after infection. In BALB/c mice abdominal cavity extracted macrophages, pretreatment with Boxb increased the expressions of TLR4, HMGB1, My D88, IL-1β, TNF-α, while pretreatment with CLI-095 and Boxb significantly decreased the expressions of TLR4, HMGB1, My D88, IL-1β, TNF-α. 5A Low-Complexity PTS Based on Greedy and Genetic Algorithm for OFDM Systems显示文摘Partial transmit sequence(PTS) is one of effective technique to reduce high Peak-to-average power ratio(PAPR) in Orthogonal frequency division multiplexing(OFDM) system. However, the complexity of Original PTS(O-PTS) increases exponentially with the number of sub-blocks. To reduce the computational complexity while still offering a lower PAPR, a new PTS method is proposed to search for suboptimal rotating vectors in this paper. In the proposed method, the candidate rotation vectors are generated based on greedy and genetic algorithm.We also combine the proposed method and the superimposed training sequence method to get a further PAPR reduction. 7Screening and evaluation of human single-chain fragment variable antibody against hepatitis B virus surface antigen显示文摘BACKGROUND: Phage display technology has become a vital tool in studies aimed at identifying molecules binding to a specific target. It enables the rapid generation and selection of high affinity, fully human antibody product candidates to essentially any disease target appropriate for antibody therapy. In this study, we prepared the recombinant single-chain fragment variable ( ScFv) antibody to hepatitis B virus surface antigen (HBsAg) by the phage display technology for obtaining a virus-targeting mediator. METHODS: mRNA was isolated from B-lymphocytes from a healthy volunteer and converted into cDNA. The fragment variables of heavy and light chain were amplified separately and assembled into ScFv DNA with a specially constructed DNA linker by polymerase chain reaction. The ScFv DNA was ligated into the phagmid vector pCANT-AB5E and the ligated sample was transformed into competent E. coli TG1. The transformed cells were infected with M13K07 helper phage to form a human recombinant phage antibody library. The volume and recombinant rate of the library were evaluated by bacterial colony count and restriction analysis. After two rounds of panning with HBsAg. the phage clones displaying ScFv of the antibody were selected by enzyme-linked immunosorbant assay ( ELISA) from the enriched phage clones. The antigen binding affinity of the positive clone was detected by competition ELISA. HB2151 E. coli was transfected with the positive phage clone demonstrated by competition ELISA for production of a soluble form of the anti-HBsAg ScFv. ELISA assay was used to detect the antigen binding affinity of the soluble anti-HBsAg ScFv. Finally, the relative molecular mass of soluble anti-HBsAg ScFv was measured by SDS-PAGE. RESULTS: The variable heavy ( VH ) and variable light (VL) and ScFv DNAs were about 340bp, 320bp and 750bp, respectively. The volume of the library was up to 2 × 106 and 8 of 10 random clones were recombinants. Two phage clones could strongly compete with the original HBsAb for binding to HBsAg. Within 2 strong positive phage clones, the soluble anti-HBsAg ScFv from one clone was found to have the binding activity with HBsAg. SDS-PAGE showed that the relative molecular weight of soluble anti-HBsAg ScFv was 32 kDa. 8Frequency stabilization and tuning of breathing solitons in Si3N4 microresonators显示文摘Dissipative Kerr solitons offer broadband coherent and low-noise frequency combs and stable temporal pulse trains,having shown great potential applications in spectroscopy,communications,and metrology.Breathing solitons are a particular kind of dissipative Kerr soliton in which the pulse duration and peak intensity show periodic oscillation.Here we have investigated the breathing dissipative Kerr solitons in silicon nitride(Si3N4)microrings,while the breathing period shows uncertainties of around megahertz(MHz)order in both simulation and experiments.This instability is the main obstacle for future applications.By applying a modulated signal to the pump laser,the breathing frequency can be injection locked to the modulation frequency and tuned over tens of MHz with frequency noise significantly suppressed.Our demonstration offers an alternative knob for the control of soliton dynamics in microresonators and paves a new avenue towards practical applications of breathing solitons.SHUAI WAN RUI NIU ZHENG-YU WANG JIN-LAN PENG MING LI JIN LI GUANG-CAN GUO CHANG-LING ZOU CHUN-HUA DONG 2020Photonics Research2020,8,8:8
9Involvements of chloride ion in decolorization of Acid Orange 7 by activated peroxydisulfate or peroxymonosulfate oxidation显示文摘The effects of chloride anion (Cl-) (up to 1.0 mol/L) on the decolorization of a model compound,azo dye Acid Orange 7 (AO7),by sulfate radical (SO4-) based-peroxydisulfate (PS) or peroxymonosulfate (PMS) oxidation under various activated conditions (UV 254 nm /PS,Thermal (70°C/PS,UV 254 nm /PMS,Co 2+ /PMS) were investigated.Methanol and NH4 + were used as quenching reagents to determine the contributions of active chlorine species (dichloride radical (Cl2-.) and hypochlorous acid (HClO)).The results indicated that the effects of Cl- on the reaction mechanism were different under various activated conditions.For UV/PS and Thermal/PS,the inhibition tendency became more clear as the Cl- concentration increased,probably due to the reaction between Cl- and SO4-.and the generation of Cl2-.or HClO.For UV/PMS,Cl- did not exhibit inhibition when the concentration was below 0.1 mol/L.As Cl- concentration reached to 1.0 mol/L,the decolorization rate of AO7 was,however,accelerated,possibly because PMS directly reacts with Cl- to form HClO.For Co2+ /PMS,Cl- exhibited a significant inhibiting effect even at low concentration ( 0.01 mol/L).When Cl- concentration exceeded 0.1 mol/L,the activation of PMS by Co 2+ was almost completely inhibited.Under this condition,HClO maybe played a major role in decolorization of AO7.The results implicated that chloride ion is an important factor in SO4-.-based degradation of organic contamination in chloride-containing water.Ping Wang Shiying Yang Liang Shan Rui Niu Xueting Shao 2011Journal of Environmental Sciences2011,23,11:7

11Associations between haemodynamics and wall enhancement of intracranial aneurysm显示文摘Background and purpose Previous studies have reported about inflammation processes(IPs)that play important roles in aneurysm formation and rupture,which could be driven by blood flow.IPs can be identified using aneurysmal wall enhancement(AWE)on high-resolution black-blood MRI(BB-MRI)and blood flow haemodynamics can be demonstrated by four-dimensional-flow MRI(4D-flow MRI).Thus,this study investigated the associations between AWE and haemodynamics in unruptured intracranial aneurysms(IA)by combining 4D-flow MRI and high-resolution BB-MRI.Materials and methods Between April 2014 and October 2017,48 patients with 49 unruptured IA who underwent both 4D-flow MRI and high-resolution BB-MRI were retrospectively included in this study.The haemodynamic parameters demonstrated using 4D-flow MRI were compared between different AWE patterns using the Kruskal-Wallis test and ordinal regression.Results The results of Kruskal-Wallis test showed that the average wall shear stress in the IA(WSSavg-IA),maximum through-plane velocity in the adjacent parent artery,inflow jet patterns and the average vorticity in IA(vorticityavg-IA)were significantly associated with the AWE patterns.Ordinal regression analysis identified WSSavg-IA(p=0.002)and vorticityavg-IA(p=0.033)as independent predictors of AWE patterns.Conclusion A low WSS and low average vorticity were independently associated with a high AWE grade for IAs larger than 4 mm.Therefore,WSS and average vorticity could predict AWE and circumferential AWE.Miaoqi Zhang Fei Peng Xin Tong Xin Feng Yunduo Li Huijun Chen Hao Niu Baorui Zhang Guangrong Song Youxiang Li Peng Liu Aihua Liu Rui Li 2021Stroke & Vascular Neurology2021,6,3:7
12Mesenteric lymph reperfusion may exacerbate brain injury in a rat model of superior mesenteric artery occlusion shock显示文摘BACKGROUND:The intestinal lymphatic pathway and intestinal ischemia/reperfusion are mainly involved in mesenteric lymph duct ligation or drainage;moreover,intervention by reducing the lymph liquid reflux might relieve lung and other organ dysfunction induced by intestinal ischemia/reperfusion;however,research addressing mesenteric lymph reperfusion (MLR) and brain injury has not yet to be reported.OBJECTIVE:To observe the effect of MLR on brain tissue in a rat model of superior mesenteric artery occlusion (SMAO) shock,and to explore the molecular mechanism of MLR.DESIGN,TIME AND SETTING:A randomized,controlled,animal experiment at a neuro-pathophysiology level was performed at the Institute of Microcirculation,Hebei North University;Department of Pathophysiology,Basic Medical College;Department of Pathology,the First Hospital of Hebei North University between December 2007 and March 2009.MATERIALS:Adenosine triphosphate (ATP) standard was provided by the National Institute for the Control of Pharmaceutical and Biological Products;lactic acid (LA),superoxide dismutase (SOD),malonaldehyde (MDA),nitrogen monoxidum (NO),nitric oxide synthase (NOS),myeloperoxidase (MPO) and ATPase assay kits were provided by Nanjing Jiancheng Bioengineering Institute,China.METHODS:A total of 24 male Wistar rats were randomly divided into four groups.In the sham-surgery group (n=6),both the mesenteric lymph duct and the superior mesenteric artery were not blocked;in the MLR group (n=6),the mesenteric lymph duct was occluded for 1 hour followed by 2-hour reperfusion;in the SMAO group (n=6),the superior mesenteric artery was occluded for 1 hour followed by 2-hour reperfusion;in the MLR + SMAO group (n=6),both the mesenteric lymph duct and superior mesenteric artery were occluded for 1 hour followed by 2-hour reperfusion.MAIN OUTCOME MEASURES:Mean arterial blood pressure prior to and following ischemia/reperfusion;brain tissue morphology levels of LA,MDA,SOD,NO,NOS,MPO,ATPase and ATP following reperfusion.RESULTS:MLR did not cause changes in mean arterial blood pressure,brain tissue morphology,LA,MDA,NO,ATP,SOD,NOS,MPO and ATPase.However,SMAO caused a rapid decrease and gradual increase of mean arterial blood pressure.Neuronal necrosis,degeneration and swelling were observed in brain tissue.Contents of MDA,NO,LA and ATP as well as activities of NOS and MPO were significantly increased (P < 0.05),but activities of SOD and Na+-K+-ATPase were significantly decreased (P < 0.05).MLR aggravated neuronal damage in a rat model of SMAO shock.Following MLR,mean arterial blood pressure was significantly decreased (P < 0.05),contents of MDA and NO as well as activities of NOS and MPO were significantly increased (P < 0.05),but activities of Ca2+-ATPase,Mg2+-ATPase and Ca2+-Mg2+-ATPase as well as ATP content were significantly decreased (P < 0.05).CONCLUSION:MLR aggravates brain injury in a rat model of SMAO shock,which correlates with oxygen-derived free radical injury,NO synthesis and release,sequestration of neutrophilic granulocytes,decreasing activity of cell membrane pumps and energy metabolism dysfunction.Pathogenesis of the intestinal lymphatic pathway should be thoroughly investigated to prevent ischemia/reperfusion injury.Zigang Zhao Chunyu Niu Aimin Shang Jiaming Tian Rui Han Chunhui Zhang Yuping Zhang Jing Zhang 2010Neural Regeneration Research2010,5,9:7
13Endocan-expressing microvessel density as a prognostic factor for survival in human gastric cancer显示文摘AIM: To investigate the expression of endocan in tumour vessels and the relationships between endocan and the expression of vascular endothelial growth factor(VEGF) and prognosis in gastric cancer.METHODS: This study included 142 patients with confirmed gastric cancer in a single cancer centre between 2008 and 2009. Clinicopathologic features were determined, and an immunohistochemical analysis of endocan-expressing microvessel density(MVD)(endocan-MVD), VEGF and vascular endothelial growth factor receptor 2(VEGFR2) was performed. Potential relationships between endocan-MVD and clinicopathological variables were assessed using a Student's t-test or an analysis of variance test. Spearman's rank correlation was applied to evaluate the relationship between endocan-MVD and the expression of VEGF/VEGFR2. long-term survival of these patients was analysed using univariate and multivariate analyses.RESULTS: Positive staining of endocan was observed in most of the gastric cancer tissues(108/142) and in fewer of the normal gastric tissues. Endocan-MVD was not associated with gender or histological type(P > 0.05), while endocan-MVD was associated with tumour size,Borrmann type, tumour differentiation, tumour invasion, lymph node metastasis and TNM stage(P < 0.05). According to the Spearman's rank correlation analysis, endocan-MVD had a positive correlation with VEGF(r = 0.167, P = 0.047) and VEGFR2(r = 0.410, P = 0.000). The univariate analysis with a log-rank test indicated that the patients with a high level of endocan-MVD had a significantly poorer overall survival rate than those with a low level of endocan-MVD(17.9% vs 64.0%, P = 0.000). 14Research progress on the role of connective tissue growth factor in fibrosis of diabetic retinopathy显示文摘Diabetic retinopathy(DR) is one of the most important types of diabetic microangiopathy, which is a specific change of fundus lesions and is one of the most serious complications of diabetes. When DR develops to proliferative DR, the main factors of decreasing vision, and even blindness, include retinal detachment and vitreous hemorrhage caused by contraction of blood vessels by fiber membrane. Recent studies reported that the formation of fiber vascular membrane is closely related to retinal fibrosis. The connective tissue growth factor(CTGF) is a cytokine that is closely related to DR fibrosis. However, its mechanism is poorly understood. 15Plasma spray of biofunctional(Mg, Sr)-substituted hydroxyapatite coatings for titanium alloy implants显示文摘Plasma-sprayed hydroxyapatite (HA) coatings have been widely utilized in load-bearing titanium alloy implants. In this study, Mg, Sr co-substituted HA ((Mg,Sr)-HA) nano-scale powders have been synthesized, which are further used to prepare (Mg,Sr)-HA coatings on Ti-6A1-4V alloys in order to improve the biological functions. The average size of (Mg,Sr)-HA nano particles is ~75nm. The average bonding strength for (Mg,Sr)-HA coating and samples after heat treatment at 500℃ or 600℃ for 3h are 26.17±2.11 MPa, 36.07±4.48 MPa and 37.07 ±2.95 MPa, respectively. There is a significantly increase of bonding strength likely due to low residual stress after heated treatment.MC3T3-E1 cells show a high proliferation rate when cultured with (Mg,Sr)-HA coating extract compared to the normal culture medium, which also exhibit large extension and depositi on of extracellular matrices when adhered on the coating surfaces. 16Oxidation of glyoxal to glyoxylic acid by oxygen over V_2O_5/C catalyst显示文摘A novel vanadium oxide catalyst supported on active carbon was prepared by an incipient wetness impregnation method, and the precursor was obtained from oxalic acid aqueous solutions of NH4VO3. The catalyst was applied liquid phase oxidation of glyoxal to glyoxylic acid. It was found that V2O5/C catalyst exhibited obvious activity for glyoxal oxidation. Glyoxylic acid could be obtained without pH regulation during the reaction. 17Flexible ultrathin all-solid-state supercapacitors显示文摘The flexible ultrathin all-solid-state supercapacitors with good electrochemical and mechanical performance were fabricated by the facile methods. The singlewall carbon nanotubes(SWCNTs)–polyaniline(PANI)film electrodes and poly(vinyl alcohol)(PVA)/H_3PO_4 electrolyte film were prepared by spray-printing and spincoating strategies, respectively. Thus, the thickness of a supercapacitor is only 8.4 lm. When the mass ratio of SWCNT to PANI is 1:1, the tensile strength and Young's modulus of the electrode are 10.9 and 655 MPa, respectively. The interior contact resistance of the supercapacitors based on this electrode is only 15–30 Ω. Furthermore,the specific capacitance of this electrode can reach about 355.5 F·g^(-1), and the supercapacitor based on this electrode maintains 87.2% of its initial specific capacitance over5000 charging/discharging cycles. Moreover, the supercapacitor shows an excellent electrochemical stability at different bending states. 18A multicenter prospective study of next-generation sequencing-based newborn screening for monogenic genetic diseases in China显示文摘Background Newborn screening(NBS)is an important and successful public health program that helps improve the long-term clinical outcomes of newborns by providing early diagnosis and treatment of certain inborn diseases.The develop-ment of next-generation sequencing(NGS)technology provides new opportunities to expand current newborn screening methodologies.Methods We designed a a newborn genetic screening(NBGS)panel targeting 135 genes associated with 75 inborn disorders by multiplex PCR combined with NGS.With this panel,a large-scale,multicenter,prospective multidisease analysis was conducted on dried blood spot(DBS)profiles from 21,442 neonates nationwide.Results We presented the positive detection rate and carrier frequency of diseases and related variants in different regions;and 168(0.78%)positive cases were detected.Glucose-6-Phosphate Dehydrogenase deficiency(G6PDD)and phenylketonuria(PKU)had higher prevalence rates,which were significantly different in different regions.The positive detection of G6PD variants was quite common in south China,whereas PAH variants were most commonly identified in north China.In addi-tion,NBGS identified 3 cases with DUOX2 variants and one with SLC25A13 variants,which were normal in conventional NBS,but were confirmed later as abnormal in repeated biochemical testing after recall.Eighty percent of high-frequency gene carriers and 60%of high-frequency variant carriers had obvious regional differences.On the premise that there was no significant difference in birth weight and gestational age,the biochemical indicators of SLC22A5 c.1400C>G and ACADSB c.1165A>G carriers were significantly different from those of non-carriers.Conclusions We demonstrated that NBGS is an effective strategy to identify neonates affected with treatable diseases as a supplement to current NBS methods.Our data also showed that the prevalence of diseases has significant regional charac-teristics,which provides a theoretical basis for screening diseases in different regions.Ru-Lai Yang Gu-Ling Qian Ding-Wen Wu Jing-Kun Miao Xue Yang Ben-Qing Wu Ya-Qiong Yan Hai-Bo Li Xin-Mei Mao Jun He Huan Shen Hui Zou Shu-Yuan Xue Xiao-Ze Li Ting-Ting Niu Rui Xiao Zheng-Yan Zhao 2023World Journal of Pediatrics2023,19,7:4

20Rhodiola crenulata extract decreases fatty acid oxidation and autophagy to ameliorate pulmonary arterial hypertension by targeting inhibiton of acylcarnitine in rats显示文摘Pulmonary arterial hypertension(PAH)is a devastating pulmonary circulation disease lacking high-efficiency therapeutics.The present study aims to decipher the therapeutic mechanism of Rhodiola crenulata,a well-known traditional chinese medicine with cardiopulmonary protection capacity,on PAH by exploiting functional lipidomics.The rat model with PAH was successfully established for first,following Rhodiola crenulata water extract(RCE)treatment,then analysis of chemical constituents of RCE was performed,additional morphologic,hemodynamic,echocardiographic measurements were examined,further targeted lipidomics assay was performed to identify differential lipidomes,at last accordingly mechanism assay was done by combining qRT-PCR,Western blot and ELISA.Differential lipidomes were identified and characterized to differentiate the rats with PAH from healthy controls,mostly assigned to acylcarnitines,phosphatidylcholines,sphingomyelin associated with the PAH development.Excitingly,RCE administration reversed high level of decadienyl-L-carnitine by the modulation of metabolic enzyme CPT1A in mRNA and protein level in serum and lung in the rats with PAH.Furthermore,RCE was observed to reduce autophagy,confirmed by significantly inhibited PPARγ,LC3B,ATG7 and upregulated p62,and inactivated LKB1-AMPK signal pathway.Notably,we accurately identified the constituents in RCE,and delineated the therapeutic mechansim that RCE ameliorated PAH through inhibition of fatty acid oxidation and autophagy.Altogether,RCE might be a potential therapeutic medicine with multi-targets characteristics to prevent the progression of PAH.This novel findings pave a critical foundation for the use of RCE in the treatment of PAH.REN Huan-Huan NIU Zheng GUO Rui FU Min LI Hai-Ru ZHANG Xuan-Yu YAO Li 2021Chinese Journal of Natural Medicines2021,19,2:4
