Antimicrobial chemical constituents from endophytic fungus Phoma sp.显示文摘Objective:To evaluate the antiniicrobial polenlial of different extracts of the endophytic fungus Phoma sp.and the tentative identification of their active constituents. Methods:The extract and compounds were screened for antimicrobial activity using the Agar Well Diffusion Method.Four compounds were purified using column chromatography and tlieir structures were assigned using~1H and~(13)C NMR spectra,DEPT,2D COSY,HMQC and HMBC experiments. Results:The ethyl acetate fraction of Phoma sp.showed good antifungal, antibacterial, and algicidal properties. One new dihydrofuran derivative, named phomafuranol(1),together with tliree known compounds,phomalacton(2),(3R)-5-hydroxymellein(3)and emodin(4)were isolated from the ethyl acetate fraction of Phoma sp.Preliminary studies indicated that phomalacton(2)displayed strong antibacterial, good antifungal and antialgal activities.Similarly(3R)-5-hydroxymellein(3)and emodin(4)showed good antifungal, antibacterial and algicidal properties. Conclusions:Antimicrobial activities of the ethyl acetate fraction of the endophytic fungus Phoma sp.and isolated compounds clearly demonstrate that Phoma sp.and its active compounds represent a great potential for the food, cosmetic and pharmaceutical industries. | Hidayat Hussain Ines Kock Ahmed Al-Harrasi Ahmed Al-Rawahi Ghulam Abbas Ivan R.Green Afzal Shah Amin Badshah Muhammad Saleem Siegfried Draeger Barbara Schulz Karsten Krohn | 2014 | Asian Pacific Journal of Tropical Medicine2014,7,9: | 3

Seimatoric acid and colletonoic acid:Two new compounds from the endophytic fungi,Seimatosporium sp.and Colletotrichum sp.显示文摘A new metabolite, named seimatoric acid(1),representing a new oxobutanoic acid derivative has been isolated from Seimatosporium sp., in addition to four known compounds viz.,2-hydroxymethyl-4β,5α,6β-trihydroxycyclohex-2-enone(2),(-)-phyllostine(3),(+)-epiepoxydon(4) and(+)-epoxydon monoacetate(5).Similarly one new benzoic acid derivative, named colletonoic acid(6) was isolated from the ethyl acetate fraction of Colletotrichum sp.The structures of the new compounds were elucidated by detailed ~1 H NMR,^(13)C NMR,COSY,HMQC.HMBC spectroscopic analysis, and HR-E1-MS.Seimatoric acid(1)was also isolated from another taxonomical unidentified fungal strain 4295 in ourgroup. The structures of the known compounds were elucidated by their spectral data comparison to literature data.Preliminary studies showed that colletonoic acid(6) showed good antibacterial, antifungal, and antialgal activities. | Hidayat Hussain Natalia Root Farah Jabeen Ahmed Al-Harrasi Ahmed Al-Rawahi Manzoor Ahmad Zahid Hassan Ghulam Abbas Fazal Mabood Afzal Shah Amin Badshah Amjad Khan Riaz Ahmad Ivan R.Green Siegfried Draeger Barbara Schulz Karsten Krohn | 2014 | Chinese Chemical Letters2014,25,12: | 1

Glia and hemichannels: key mediators of perinatal encephalopathy显示文摘Perinatal encephalopathy remains a major cause of disability, such as cerebral palsy. Therapeutic hypothermia is now well established to partially reduce risk of disability in late preterm/term infants. However, new and complementary therapeutic targets are needed to further improve outcomes. There is increasing evidence that glia play a key role in neural damage after hypoxia-ischemia and infection/inflammation. In this review, we discuss the role of astrocytic gap junction(connexin) hemichannels in the spread of neural injury after hypoxia-ischemia and/or infection/inflammation. Potential mechanisms of hemichannel mediated injury likely involve impaired intracellular calcium handling, loss of blood-brain barrier integrity and release of adenosine triphosphate(ATP) resulting in over-activation of purinergic receptors. We propose the hypothesis that inflammation-induced opening of connexin hemichannels is a key regulating event that initiates a vicious cycle of excessive ATP release, which in turn propagates activation of purinergic receptors on microglia and astrocytes. This suggests that developing new neuroprotective strategies for preterm infants will benefit from a detailed understanding of glial and connexin hemichannel responses. | Robert Galinsky Joanne O.Davidson Justin M.Dean Colin R.Green Laura Bennet Alistair J.Gunn | 2018 | Neural Regeneration Research2018,13,2: | 1

Connexin therapeutics:blocking connexin hemichannel pores is distinct from blocking pannexin channels or gap junctions显示文摘Compounds that block the function of connexin and pannexin protein channels have been suggested to be valuable therapeutics for a range of diseases. Some of these compounds are now in clinical trials, but for many of them, the literature is inconclusive about the molecular effect on the tissue, despite evidence of functional recovery.Blocking the different channel types has distinct physiological and pathological implications and this review describes current knowledge of connexin and pannexin protein channels, their function as channels and possible mechanisms of the channel block effect for the latest therapeutic compounds. We summarize the evidence implicating pannexins and connexins in disease, considering their homeostatic versus pathological roles, their contribution to excesive ATP release linked to disease onset and progression. | Monica L.Acosta Mohd N.Mat Nor Cindy X.Guo Odunayo O.Mugisho Frazer P.Coutinho Ilva D.Rupenthal Colin R.Green | 2021 | Neural Regeneration Research2021,16,3: | 1

A behavior and physiology-based decision support tool to predict thermal comfort and stress in non-pregnant,mid-gestation,and late-gestation sows显示文摘Background:Although thermal indices have been proposed for swine, none to our knowledge differentiate by reproductive stage or predict thermal comfort using behavioral and physiological data. The study objective was to develop a behavior and physiology-based decision support tool to predict thermal comfort and stress in multiparous(3.28±0.81)non-pregnant(n=11),mid-gestation(n=13),and late-gestation(n=12)sows.Results:Regression analyses were performed using PROC MIXED in SAS 9.4 to determine the optimal environmental indicator[dry bulb temperature(TDB)and dew point]of heat stress(HS)in non-pregnant,mid-gestation,and lategestation sows with respiration rate(RR)and body temperature(TB)successively used as the dependent variable in a cubic function.A linear relationship was observed for skin temperature(T_(S))indicating that TDB rather than the sow HS response impacted T_(S)and so T_(S)was excluded from further analyses.Reproductive stage was significant for all analyses(P<0.05).Heat stress thresholds for each reproductive stage were calculated using the inflections points of RR for mild HS and TB for moderate and severe HS.Mild HS inflection points differed for non-pregnant,mid-gestation,and late gestation sows and occurred at 25.5,25.1,and 24.0℃,respectively.Moderate HS inflection points differed for non-pregnant,mid-gestation,and late gestation sows and occurred at 28.1,27.8,and 25.5℃,respectively.Severe HS inflection points were similar for non-pregnant and mid-gestation sows(32.9℃)but differed for late-gestation sows(30.8℃).These data were integrated with previously collected behavioral thermal preference data to estimate the TDB that non-pregnant,mid-gestation,and late-gestation sows found to be cool(TDB | Betty R.McConn Allan P.Schinckel Lindsey Robbins Brianna N.Gaskill Angela R.Green‑Miller Donald C.Lay Jr Jay S.Johnson | 2023 | Journal of Animal Science and Biotechnology2023,14,2: | 0
