| 1 | 脂肪干细胞分离、纯化和保存:研究进展与未来方向显示文摘背景:2001年,Zuk等从吸脂术抽出的脂肪中发现了脂肪干细胞,揭开了干细胞研究的新篇章。近年来的研究证实,干细胞广泛存在于体内各组织,但脂肪干细胞以其来源广泛、获取简单等优点,一直以来是整形修复科、组织工程和再生医学等相关学科的研究重点。目的:从脂肪组织的取材、脂肪干细胞的分离、纯化、传代培养和冻存等方面进行综述,讨论上述过程中影响脂肪干细胞产量、增殖和分化能力的主要因素,同时提出目前的问题和未来的研究方向。方法:于2015年9月10日在PubM ed中以(adipose stem cells[Title])OR(adipose-derived stem cells[Title])OR(adipose-derived mesenchymal stem cells[Title])作为检索式进行检索,在SinoM ed中以('脂肪干细胞'[中文标题:智能])or('脂肪间充质干细胞'[中文标题:智能])作为检索式进行检索。通过标题及摘要判断其主要内容,最终选取81篇代表性文献。文章还介绍了美国德克萨斯大学MD安德森肿瘤中心整形科和TRAMCEL实验室(Tissue Regeneration and Molecular Cell Engineering Lab)的相关经验。结果与结论:脂肪组织分布广泛,给脂肪干细胞提供了充足的来源。利用吸脂术获得脂肪组织,患者痛苦小,供区损伤低。胶原酶消化法操作简单,产量高,仍然是基础实验中脂肪干细胞的主要分离方法,但是临床应用中一般不分离脂肪干细胞,或者利用临床级的无异种胶原酶甚至无酶的分离方法。脂肪干细胞的产量、表型、增殖和分化能力等生物学特性会受到取材和分离过程中多种因素的影响,因此目前需要制定标准化的脂肪干细胞分离准则。 | 陈犹白 陈聪慧 Qixu Zhang 韩岩 | 2016 | 中国组织工程研究2016,20,10: | 34 |
| 2 | 脂肪干细胞成骨分化的研究进展显示文摘创伤或肿瘤导致的骨缺损是骨科、整形修复科、口腔颌面外科等科室的常见疾病,现有的治疗手段均有一定局限性。脂肪干细胞(ASC)是来自脂肪组织的多能干细胞,基于ASC的干细胞疗法和骨组织工程,为骨缺损的修复和再生提供了新的思路。ASC成骨分化是多种基因、蛋白和信号通路相互作用的复杂过程,其中Runx2和Osterix、Wnt、骨形成蛋白、Notch、成纤维细胞生长因子、环磷腺苷/蛋白激酶A、Hedgehog、丝裂原活化蛋白激酶等均扮演了重要角色。体外化学诱导ASC的成骨分化已经非常成熟,利用细胞共培养和各种支架也可诱导ASC的成骨分化。验证ASC分化成骨的方法包括茜素红染色和相关基因与蛋白的检测。影响ASC成骨分化的因素包括供体种属、年龄、脂肪获取部位等供体因素,培养液糖浓度、ASC的代数、冻存等实验因素,血管内皮细胞生长因子、生长分化因子、胰岛素样生长因子、转化生长因子β、尼尔样1型分子、LIM矿化蛋白1、低氧诱导因子1、肿瘤坏死因子α、干扰素γ、IL-6等生长因子,糖皮质激素、雌激素、胰岛素、褪黑素、瘦素等激素,一氧化氮、组蛋白H1、白藜芦醇、曲古抑菌素A、小檗碱、硼替佐米、阿司匹林、辛伐他汀等化学因子及药物,电磁场和超声波等物理因素,拉伸力和剪切力等生物力学因素,钙、锌、锂、锶、硒等金属或非金属离子,微小RNA以及富血小板血清和富血小板纤维蛋白、降钙素基因相关肽、中药和咖啡因等其他因素。本文总结了ASC成骨分化的过程,分析了相关基因和信号通路,回顾了诱导和验证方法,讨论了主要影响因素及其机制,并展望了未来研究方向。 | 陈犹白 陈聪慧 Qixu Zhang 韩岩 | 2016 | 中华损伤与修复杂志(电子版)2016,11,2: | 19 |
| 3 | 脂肪干细胞表型和标记物的研究进展显示文摘由于脂肪供体、提取方法、分离、培养、体内外微环境、鉴定时的细胞代数和检测方法等诸多因素的差异,迄今为止尚未发现脂肪干细胞(adipose-derived stem cell,ASCs)的特异性标记物。本文总结了所有ASCs的相关标记物,发现阳性标记物集中在基质细胞、间充质干细胞和成纤维细胞标记物上,而阴性标记物主要是造血系细胞标记物,ASCs的争议标记物主要以骨髓间充质干细胞(bone marrow stem cells,BMSCs)、血管内皮细胞和周细胞标记物为主,这也体现了关于ASCs体内来源的争议。体内、刚分离的和体外培养初期的ASCs表达CD34,但经长时间体外培养后,其表达逐渐降低最后完全消失。这证实了ASCs体内外的表型差异,因此不能通过检测体外长期培养后的ASCs来确定体内的ASCs表型。Stro-1等干性标记物在ASCs的表达对鉴别ASCs有重要意义,但能否作为ASCs的常规标记物用于鉴定ASCs,尚需要进一步的研究。基质血管成分(Stromal vascular fraction,SVF)和ASCs不同亚群的表型差异提示了ASCs的体内起源,即BMSCs、周细胞或成纤维细胞,但尚无定论。ASCs表达CD36、CD49d,不表达CD48f、CD104,BMSCs则相反,这是鉴别ASCs和BMSCs的表型依据。本文还介绍了美国德克萨斯大学MD安德森肿瘤中心整形科组织再生与分子细胞工程实验室(Tissue Regeneration and Molecular Cell Engineering Lab,TRAMCEL)在ASCs表型方面的研究经验,探讨了ASCs表型研究目前存在的问题和未来的发展方向。 | 陈犹白 陈聪慧 Qixu Zhang 韩岩 | 2016 | 中国美容医学2016,25,3: | 18 |

| 7 | A static technique for detecting input validation vulnerabilities in Android apps显示文摘Input validation vulnerabilities are common in Android apps, especially in inter-component communications. Malicious attacks can exploit this kind of vulnerability to bypass Android security mechanism and compromise the integrity, confidentiality and availability of Android devices. However, so far there is not a sound approach at the source code level for app developers aiming to detect input validation vulnerabilities in Android apps. In this paper, we propose a novel approach for detecting input validation flaws in Android apps and we implement a prototype named Easy IVD, which provides practical static analysis of Java source code.Easy IVD leverages backward program slicing to extract transaction and constraint slices from Java source code.Then Easy IVD validates these slices with predefined security rules to detect vulnerabilities in a known pattern.To detect vulnerabilities in an unknown pattern, Easy IVD extracts implicit security specifications as frequent patterns from the duplicated slices and verifies them. Then Easy IVD semi-automatically confirms the suspicious rule violations and reports the confirmed ones as vulnerabilities. We evaluate Easy IVD on four versions of original Android apps spanning from version 2.2 to 5.0. It detects 58 vulnerabilities including confused deputy attacks and denial of service attacks. | 9 | 3D-printed models improve surgical planning for correction of severe postburn ankle contracture with an external fixator显示文摘Gradual distraction with an external fixator is a widely used treatment for severe postburn ankle contracture(SPAC).However,application of external fixators is complex,and conventional two-dimensional(2D)imaging-based surgical planning is not particularly helpful due to a lack of spatial geometry.The purpose of this study was to evaluate the surgical planning process for this procedure with patient-specific three-dimension-printed models(3DPMs).In this study,patients coming from two centers were divided into two cohorts(3DPM group vs.control group)depending on whether a 3DPM was used for preoperative surgical planning.Operation duration,improvement in metatarsal-tibial angle(MTA),range of motion(ROM),the American Orthopedic Foot and Ankle Society(AOFAS)scores,complications,and patient-reported satisfaction were compared between two groups.The 3DPM group had significantly shorter operation duration than the control group((2.0±0.3)h vs.(3.2±0.3)h,P<0.01).MTA,ROM,and AOFAS scores between the two groups showed no significant differences pre-operation,after the removal of the external fixator,or at follow-up.Plantigrade feet were achieved and gait was substantially improved in all patients at the final follow-up.Pin-tract infections occurred in two patients(one in each group)during distraction and were treated with wound care and oral antibiotics.Patients in the 3DPM group reported higher satisfaction than those in the control group,owing to better patient-surgeon communication.Surgical planning using patient-specific 3DPMs significantly reduced operation duration and increased patient satisfaction,while providing similar improvements in ankle movement and function compared to traditional surgical planning for the correction of SPAC with external fixators. | Youbai CHEN Zehao NIU Weiqian JIANG Ran TAO Yonghong LEI Lingli GUO Kexue ZHANG Wensen XIA Baoqiang SONG Luyu HUANG Qixu ZHANG Yan HAN | 2021 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2021,22,10: | 3 |

| 12 | Harnessing the synergy of perfusable muscle flap matrix and adipose-derived stem cells for prevascularization and macrophage polarization to reconstruct volumetric muscle loss显示文摘Muscle flaps must have a strong vascular network to support a large tissue volume and ensure successful engraftment.We developed porcine stomach musculofascial flap matrix(PDSF)comprising extracellular matrix(ECM)and intact vasculature.PDSF had a dominant vascular pedicle,microcirculatory vessels,a nerve network,well-retained 3-dimensional(3D)nanofibrous ECM structures,and no allo-or xenoantigenicity.In-depth proteomic analysis demonstrated that PDSF was composed of core matrisome proteins(e.g.,collagens,glycoproteins,proteoglycans,and ECM regulators)that,as shown by Gene Ontology term enrichment analysis,are functionally related to musculofascial biological processes.Moreover,PDSF��human adipose-derived stem cell(hASC)synergy not only induced monocytes towards IL-10��producing M2 macrophage polarization through the enhancement of hASCs’paracrine effect but also promoted the proliferation and interconnection of both human skeletal muscle myoblasts(HSMMs)and human umbilical vein endothelial cells(HUVECs)in static triculture conditions.Furthermore,PDSF was successfully prevascularized through a dynamic perfusion coculture of hASCs and HUVECs,which integrated with PDSF and induced the maturation of vascular networks in vitro.In a xenotransplantation model,PDSF demonstrated myoconductive and immunomodulatory properties associated with the predominance of M2 macrophages and regulatory T cells.In a volumetric muscle loss(VML)model,prevascularized PDSF augmented neovascularization and constructive remodeling,which was characterized by the predominant infiltration of M2 macrophages and significant musculofascial tissue formation.These results indicate that hASCs’integration with PDSF enhances the cells’dual function in immunomodulation and angiogenesis.Owing in part to this PDSF-hASC synergy,our platform shows promise for vascularized muscle flap engineering for VML reconstruction. | Qixu Zhang Yulun Chiu Youbai Chen Yewen Wu Lina W.Dunne Rene D.Largo Edward I.Chang David M.Adelman Mark V.Schaverien Charles E.Butler | 2023 | Bioactive Materials2023,,4: | 1 |

| 16 | Selectivity Estimation for String Predicates Based on Modified Pruned Count-Suffix Tree显示文摘The accuracy of predicates selectivity estimation is one of the important factors affecting query optimization performance. State-of-art selectivity estimation algorithms for string predicates based on Pruned countsuffix tree(PST) often suffer severe underestimating and overestimating problems, thus their average relative errors are not good. We analyze the main causes of the underestimating and overestimating prob-lems, propose a novel Restricted pruned count-suffix tree(RPST) and a new pruning strategy. Based on these, we present the EKVI algorithm and the EMO algorithm which are extended from the KVI algorithm and the MO algorithm respectively. The experiments compare the EKVI algorithm and the EMO algorithm with the traditional KVI algorithm and the MO algorithm, and the results show that the average relative errors of our selectivity estimation algorithms are significantly better than the traditional selectivity estimation algorithms. | 17 | XAS:Cross-API scripting attacks in social ecosystems显示文摘With the rapid development of online social networks, various Web application programming interfaces(APIs) on social platforms are released to share profitable social data with all kinds of third-party online services. However, it also brings new risks to social networks once Web APIs are insecurely designed,implemented, and invoked. The focused topic in this paper is security analysis of a new type of cross-site scripting(XSS) which is based on Web APIs in new complicated social ecosystems which consist of social networks,third-party apps, and other online services. In this paper, we refer to Web API-based XSS as cross-API scripting(XAS). For the first time, we take typical XAS attacks in diversified context as cases to demonstrate the new exploiting opportunities and threats in social ecosystems. Also, we design a tool to identify the design and implementation flaws of Web APIs in 11 popular social networks. We discover several security flaws of API via our experiment. According to the results, we conclude causes of XAS flaws in depth. We also examined 143Web-based apps and verified the prevalence of XAS flaws. | 18 | An adaptive system for detecting malicious queries in web attacks显示文摘Web request query strings(queries), which pass parameters to a referenced resource, are always manipulated by attackers to retrieve sensitive data and even take full control of victim web servers and web applications. However, existing malicious query detection approaches in the literature cannot cope with changing web attacks. In this paper, we introduce a novel adaptive system(AMOD) that can adaptively detect web-based code injection attacks, which are the majority of web attacks, by analyzing queries. We also present a new adaptive learning strategy, called SVM HYBRID, leveraged by our system to minimize manual work. In the evaluation, an up-to-date detection model is trained on a ten-day query dataset collected from an academic institute's web server logs. The evaluation shows our approach overwhelms existing approaches in two respects. Firstly, AMOD outperforms existing web attack detection methods with an F-value of 99.50%and FP rate of 0.001%. Secondly, the total number of malicious queries obtained by SVM HYBRID is3.07 times that by the popular support vector machine adaptive learning(SVM AL) method.
