1Fisetin mitigates hepatic ischemia-reperfusion injury by regulating GSK3β/AMPK/NLRP3 inflammasome pathway显示文摘Background: Hepatic ischemia-reperfusion(I/R) injury(IRI) represents a crucial challenge in liver transplantation. Fisetin has anti-inflammatory, anti-aging and anti-oxidative properties. This study aimed to examine whether fisetin mitigates hepatic IRI and examine its underlying mechanisms. Methods: Sham or warm hepatic I/R operated mice were pretreated with fisetin(5, 10 or 20 mg/kg). Hepatic histological assessments, TUNEL assays and serum aminotransferase measurements were performed. An in vitro hypoxia/reoxygenation(H/R) model using RAW264.7 macrophages pretreated with fisetin(2.5, 5 or 10 μmol/L) was also used. Serum and cell supernatant concentrations of interleukin-1 β(IL-1 β), IL-18 and tumor necrosis factor-α(TNF-α) were determined by enzyme-linked immunosorbent assay(ELISA). Protein levels of p-GSK3 β, p-AMPK and NLR family pyrin domain-containing 3(NLRP3)-associated proteins were detected by Western blotting. Results: Compared with the I/R group, fisetin pretreatment reduced pathological liver damage, serum aminotransferase levels, serum concentrations of IL-1 β, IL-18 and TNF-α in the murine IRI model. Fisetin also reduced the expression of NLRP3 inflammasome-associated proteins(NLRP3, cleaved caspase-1, IL-1 β and IL-18) in I/R-operated liver. The experiments in vitro showed that fisetin decreased the release of IL-1 β, IL-18 and TNF-α, and reduced the expression of NLRP3 inflammasome-associated proteins in H/R-treated RAW264.7 cells. Moreover, fisetin increased the expressions of p-GSK3 β and p-AMPK in both models, indicating that its anti-inflammatory effects were dependent on GSK3 β/AMPK signaling. The antiinflammatory effects of fisetin were partially inhibited by the AMPK specific inhibitor compound C. 2Nutrient Composition and Distance from Point Placement to the Plant Affect Rice Growth显示文摘Point placement of urea is an efficient technology to improve urea use efficiency in transplanted rice(Oryza sativa L.), but it is largely unknown how nutrient composition in the point placement and the distance from placement site to the plant influence rice root distribution and growth, nutrient uptake, and rice grain yield. A controlled greenhouse experiment was conducted using both N-and P-deficient soil with point placement of N only or N and P together(N + P) at a distance close to or far from the plant,in comparison to an N-spilt application and a no-N control. Both nutrient composition and distance significantly affected rice root growth. Compared with the N point placement, the N + P point placement led to smaller root length and mass densities, higher specific root length(SRL) around the placement site, smaller root system, higher straw mass and grain yield, and higher N and P uptake. The difference between the N + P and N point placements was greater when close to the plant than when far from the plant. 3Estimating abundance and density of Amur tigers along the Sino- Russian border显示文摘As an apex predator the Amur tiger(Panthera tigris altaica)could play a pivotal role in maintaining the integ­rity of forest ecosystems in Northeast Asia.Due to habitat loss and harvest over the past century,tigers rapidly declined in China and are now restricted to the Russian Far East and bordering habitat in nearby China.To fa­cilitate restoration of the tiger in its historical range,reliable estimates of population size are essential to assess effectiveness of conservation interventions.Here we used camera trap data collected in Hunchun National Na­ture Reserve from April to June 2013 and 2014 to estimate tiger density and abundance using both maximum likelihood and Bayesian spatially explicit capture-recapture(SECR)methods.A minimum of 8 individuals were detected in both sample periods and the documentation of marking behavior and reproduction suggests the presence of a resident population.Using Bayesian SECR modeling within the 11400 km2 state space,den­sity estimates were 0.33 and 0.40 individuals/100 km^(2) in 2013 and 2014,respectively,corresponding to an es­timated abundance of 38 and 45 animals for this transboundary Sino-Russian population.In a maximum likeli­hood framework,we estimated densities of 0.30 and 0.24 individuals/100 km^(2) corresponding to abundances of 34 and 27,in 2013 and 2014,respectively.These density estimates are comparable to other published estimates for resident Amur tiger populations in the Russian Far East.This study reveals promising signs of tiger recovery in Northeast China,and demonstrates the importance of connectivity between the Russian and Chinese popula­tions for recovering tigers in Northeast China.Wenhong XIAO Limin FENG Pu MOU Dale GMIQUELLE Mark HEBBLEWHITE Joshua FGOLDBERG Hugh SROBINSON Xiaodan ZHAO Bo ZHOU Tianming WANG Jianping GE 2016Integrative Zoology2016,11,4:4
4Elusive cats in our backyards:persistence of the North Chinese leopard(Panthera pardus japonensis)in a human-dominated landscape in central China显示文摘The North Chinese leopard(Panthera pardus japonensis),the least-known big cat,disappeared in most historical range for decades,following the development of modern civilization.Unfortunately,we have scarce knowledge about the status of this big cat so far,apart from anecdotal reports.In this study,we investigated density,distribution,and habitat use of the leopard,the apex predator,in a complex forest landscape in the Loess Plateau.We used a camera-trapping network to obtain population estimates for leopards over 2 years through spatially explicit capture-recapture models.Our results,based on maximum likelihood and Bayesian/MCMC methods,reveal that the largest wild population of the leopard was found widely distributed in remnant forests in central Loess plateau.The population is increasing in our study area,and the density of leopards(1.70(SE=0.48)−2.40(SE=0.67)/100 km^(2))is higher than other areas of China.According to the analysis of 2 seasonal occupancy models,prey species drive partially the leopard habitat use,predicting that the big cat thrives from the recovery of prey community.However,human disturbances,especially oil wells,seem to have negative impacts on the habitat use of leopards.Specifically,it is necessary to have joint efforts by the government and researchers to improve human disturbances management and prey species population density,as well as strengthen the investment in research on the North Chinese leopard,which could all further strengthen protection ability and ensure the long-term survival of this species.Haitao YANG Bing XIE Guojing ZHAO Yinan GONG Pu MOU Jianping GE Limin FENG 2021Integrative Zoology2021,16,1:3
5IL-37b suppresses epithelial mesenchymal transition in hepatocellular carcinoma by inhibiting IL-6/STAT3 signaling显示文摘Background: Interleukin-37 b(IL-37 b), a vital negative regulator of the innate immune system, has been reported to be a tumor inhibitor in different type of cancers. However, little is known about the relationship between IL-37 b and hepatocellular carcinoma(HCC). The present study aimed to investigate the potential roles of IL-37 b in HCC progression. Methods: Subjects( n = 237) were recruited, and serum IL-37 b was measured using ELISA. The tumorsuppressive capacity and underlying mechanisms of IL-37 b in HCC were investigated in vitro and in vivo. Results: Compared to healthy controls, serum IL-37 b levels were elevated in chronic hepatitis B(CHB) patients but decreased significantly in HBV-HCC patients, especially for those with portal venous tumor thrombus. Low level serum IL-37 b in HBV-HCC patients correlated with high HCC stage and poor overall survival and disease-free survival. In vitro and in vivo, recombinant human IL-37 b inhibited proliferation and metastasis in HCC cells. Furthermore, IL-37 b inhibited epithelial mesenchymal transition in HCC cells in vitro by downregulating IL-6, pSTAT3(Y705), N-cadherin, and vimentin expression and by upregulating E-cadherin expression. These effects were partially reversed by transfection of adenovirus encoding human IL-6. Conclusions: IL-37 b inhibits HCC growth, metastasis and epithelial mesenchymal transition by regulating IL-6/STAT3 signaling. 8Evaluation of regeneration potential of Pinus koraiensis in mixed pine-hardwood forests in the Xiao Xing'an Mountains,China显示文摘Large scale harvest of Korean pine(Pinus koraiensis) seeds as a food product in the mixed Korean pine-hardwood forest of northeastern China poses a serious threat to the sustainability and restoration of this endangered regional ecosystem.Seed collection over past decades greatly reduced the seed bank and subsequent seedling and sapling recruitment,and impacting a wide array of granivorous animals that rely on the pine seeds.We surveyed Korean pine seeds,including solid seeds(SS),insect consumed seeds(ICS) and other(animal) consumed(OCS) kernels,of the seed bank(forest floor and the top 10 cm of mineral soil),the seedlings and saplings from 1 m 2 sample plots in five forest types in Liangshui Nature Reserve(LNR) of the southern Xiao Xing'an Mountains in northeastern China to provide accurate information for assessing the Korean pine regeneration potential.The average number of pine seeds in the seed bank were 11.2 seeds/m 2,9.1 seeds/m 2,4.6 seeds/m 2,1.1 seeds/m 2,and 0.2 seeds/m 2 in Korean pine-basswood forest,mixed Korean pine-hardwood forest,mixed conifer-hardwood forest,white birch forests,and oak forests,respectively.In the first three forest types,percentages of SS(potentially viable seeds) were 11.2%,3.5% and 27.8%,respectively.The percentages of ICS(not viable seeds) were consistent at around 35%.The higher but variable percentages of OCS(not viable seeds) indicated high seed predation in these forests.Compared with other studies,we recorded higher percentages of seed damage,probably due to our survey approach and the increased depth of seed bank sampled in our study.Depletion of pine seeds in the seed bank greatly reduced seedling and sapling recruitment.Densities of pine seedlings varied from about 180 trees/ha in the mixed Korean pine-hardwood forest to about 5,400 trees/ha in the mixed conifer-hardwood forests and showed a high degree of spatial variation.Saplings were rare in the mixed Korean pine-hardwood forest,but ranged in the thousands in other forests.Large scale pine seed harvest has seriously threatened the sustainability of the mixed Korean pine-hardwood forest ecosystem.Scaling down the seed harvest or supplemental planting of pine saplings are urgently needed to maintain the health of the existing Korean pine forests and to restore this endangered ecosystem.LI Yun-bin MOU Pu WANG Tian-ming GE Jianping 2012Journal of Forestry Research2012,23,4:2
9Soil carbon dynamics in a Pinus massoniana plantation following clear-cutting and slash removal显示文摘Aims Slash removal is a common practice to prepare recently harvested sites for replanting.However,little is known about its impact on soil carbon(C)dynamics in subtropical plantations.This study evaluates the effects of burning versus manual slash removal site preparation treatments on soil organic carbon(SOC),soil respiration and soil microbial community structure in a Pinus massoniana plantation in southern China.Methods Three areas within a mature P.massoniana plantation were clearcut.Two months following harvesting,slash on one-half of each area was burned(BURN),whereas slash was manually removed(MANR)on the other portion.Slash removal treatments were also compared with adjacent uncut plantation areas(UNCUT).Soil samples,and soil respiration measurements were used to characterize soil properties and microbial communities following slash removal treatments.Important Findings Mean soil respiration rates from the MANR and BURN treatments were 26%and 17%lower,respectively,than the UNCUT treatment over 1 year.The MANR and BURN treatment resulted in soils with 27%and 9%reduction in total phospholipid fatty acids(PLFAs)and 18%and 10%reduction in bacterial PLFAs,respectively,compared with the UNCUT treatment.However,no significant differences existed between slash removal treatments with respect to soil chemical properties,SOC chemical compositions,soil respiration and microbial communities;although PLFA patterns were notably different for the burned plots.Most factors affecting C dynamics and microbial communities were not sensitive to the differences imparted to the ecosystem due to manual slash removal or burning.Our results suggested that low-intensity burning after clear-cutting might have no significant effect on soil C pool and its dynamics compared with manual slash removal in subtropical plantations.Yuan Wen Jamie LSchuler Shirong Liu Pu Mou Hui Wang Haolong Yu Jingxin Wang 2016Journal of Plant Ecology2016,9,1:2

17High-static-low-dynamic stiffness isolator based on an electromagnetic negative stiffness spring with long linear stroke显示文摘Negative stiffness mechanisms can improve low-frequency vibration isolation performance and have been widely used in the vibration isolation of precision equipment. However, the negative stiffness mechanism usually introduces a nonlinear stiffness,resulting in a nonlinear response and worsening the vibration isolation performance, especially under large amplitude vibration.In this paper, an electromagnetic spring with linear negative stiffness(ESLNS) is proposed, in which the antagonistic ampere forces of the energized coils are used to generate negative stiffness within a long linear stroke. The magnetic field distribution is improved through the design of the magnetic circuit, thereby increasing the stiffness generation efficiency. The stiffness can be adjusted bidirectionally by current within the range of positive and negative stiffness. An electromagnetic stiffness model was established based on the equivalent magnetic circuit method. Experimental measurements verified the accuracy of the model and proved the linearity of the electromagnetic spring. A vibration isolator with high static and low dynamic stiffness(HSLDS) based on the ESLNS is designed and tested. The experimental results prove that the introduction of the ESLNS can effectively expand the isolation frequency band without changing the equilibrium position. Moreover, the vibration isolator with ESLNS does not produce nonlinear response. 18Is root nutrient uptake a modular function? A test using Solidago canadensis显示文摘Plant roots have been recognized to be modular, and a third order root cluster has been proposed to be the basic root module unit based upon the life cycle. This experiment examines root modularity of the nutrient-uptake function using stable isotope ^(15)N. Solidago canadensis root clusters of second or third orders—from the same third or fourth order roots, respectively—were treated with^(15)NH_4^(15)NO_3, NH_4NO_3, and de-ionized water for 15 and180 min. The δ^(15)N values of the root clusters were then analyzed and compared. The δ^(15)N values of ^(15)N-treated root clusters of both second and third orders were hundreds of times higher than that of the ^(15)N untreated root clusters.However, the differences of the δ^(15)N values among ^(15)N untreated root clusters(though expressed some significant variations), did not indicate the ^(15)N shared by the sister root clusters came from a common higher-order root. These results demonstrated functional modularity of root nutrient uptake, revealed a second order root, perhaps even a first order root to be a base module unit in terms of root nutrient uptake. The results also suggested that the concept of rootmodularity is function-specific. 19Spatial patterns nitrogen transfer models of ectomycorrhizal networks in a Mongolian scotch pine plantation显示文摘Ectomycorrhizal(EM)networks provide a variety of services to plants and ecosystems include nutrient uptake and transfer,seedling survival,internal cycling of nutrients,plant competition,and so on.To deeply their structure and function in ecosystems,we investigated the spatial patterns and nitrogen(N)transfer of EM networks using ^(15)N labelling technique in a Mongolian scotch pine(Pinus sylvestris var.mongolica Litv.)plantation in Northeastern China.In August 2011,four plots(20 × 20 m)were set up in the plantation.125 ml 5 at.%0.15 mol/L ^(15)NH_4 ^(15)NO_3solution was injected into soil at the center of each plot.Before and 2,6,30 and 215 days after the ^(15)N application,needles(current year)of each pine were sampled along four 12 m sampling lines.Needle total N and ^(15)N concentrations were analyzed.We observed needle N and^(15)N concentrations increased significantly over time after ^(15)N application,up to 31 and0.42%,respectively.There was no correlation between needle N concentration and ^(15)N/ ^(14)N ratio(R2=0.40,n=5,P=0.156),while excess needle N concentration and excess needle ^(15)N/^(14)N ratio were positively correlated across different time intervals(R^2=0.89,n=4,P\0.05),but deceased with time interval lengthening.Needle ^(15)N/^(14)N ratio increased with time,but it was not correlated with distance.Needle ^(15)N/ ^(14)N ratio was negative with distance before and 6th day and 30th day,positive with distance at 2nd day,but the trend was considerably weaker,their slop were close to zero.These results demonstrated that EM networks were ubiquitous and uniformly distributed in the Mongolian scotch pine plantation and a random network.We found N transfer efficiency was very high,absorbed N by EM network was transferred as wide as possible,we observed N uptake of plant had strong bias for ^(14)N and ^(15)N,namely N fractionation.Understanding the structure and function of EM networks in ecosystems may lead to a deeper understanding of ecological stability and evolution,and thus provide new theoretical approaches to improve conservation practices for the management of the Earth’s ecosystems.Yanbin Liu Hongmei Chen Pu Mou 2018Journal of Forestry Research2018,29,2:0
