| 1 | The epigallocatechin gallate derivative Y_6 reverses drug resistance mediated by the ABCB1 transporter both in vitro and in vivo显示文摘Previously, we reported that Y_6, a new epigallocatechin gallate derivative, is efficacious in reversing doxorubicin(DOX)–mediated resistance in hepatocellular carcinoma BEL-7404/DOX cells. In this study, we evaluated the efficacy of Y_6 in reversing drug resistance both in vitro and in vivo by determining its effect on the adenosine triphosphate-binding cassette protein B1 transporter(ABCB1 or P-glycoprotein, P-gp). Our results showed that Y_6 significantly sensitized cells overexpressing the ABCB1 transporter to anticancer drugs that are ABCB1 substrates. Y_6 significantly stimulated the adenosine triphosphatase activity of ABCB1. Furthermore, Y_6 exhibited a higher docking score as compared with epigallocatechin gallate inside the transmembrane domain of ABCB1. In addition, in the nude mousetumor xenograft model, Y_6(110 mg/kg, intragastric administration), in combination with doxorubicin(2 mg/kg, intraperitoneal injection), significantly inhibited the growth of BEL-7404/DOX cell xenograft tumors, compared to equivalent epigallocatechin gallate. | 2 | Simultaneous quantification of prodrug oseltamivir and its metabolite oseltamivir carboxylate in human plasma by LC-MS/MS to support a bioequivalence study显示文摘A simple,precise and rapid liquid chromatography-tandem mass spectrometry(LC-MS/MS) method has been developed and validated for the simultaneous determination of oseltamivir and oseltamivir carboxylate,a neuraminidase inhibitor,using their deuterated analogs as internal standards(ISs).The method involved solid phase extraction of the analytes and ISs from 200 μL human plasma with no reconstitution and drying steps.The chromatographic separation was achieved on a Symmetry C18(100 mm 4.6 mm,5 μm) column using 10 mM ammonium formate and acetonitrile(30:70,v/v) as the mobile phase in a run time of 2.0 min.Quantitation of analytes and ISs were done by multiple reaction monitoring on a triple quadrupole mass spectrometer in the positive ionization mode.The linearity of the method was established in the concentration range of 0.5-200 ng/mL and 2.0-800 ng/mL for oseltamivir and oseltamivir carboxylate respectively.The mean extraction recovery for oseltamivir(94.4%) and oseltamivir carboxylate(92.7%) from spiked plasma samples was consistent and reproducible.The application of this method was demonstratedby a bioequivalence study in 42 healthy Indian subjects with 75 mg oseltamivir phosphate capsules.The assay reproducibility was established by reanalysis of 151 incurred subject samples. | Ajay Gupta Swati Guttikar Pranav S. | 3 | Evaluation of mannitol effect in patients with acute hepatic failure and acute-on-chronic liver failure using conventional MRI,diffusion tensor imaging and in-vivo proton MR spectroscopy显示文摘AIM:To evaluate the effect of an intravenous bolus of mannitol in altering brain metabolites, brain water content, brain parenchyma volume, cerebrospinal fluid (CSF) volume and clinical signs in controls and in patients with acute liver failure (ALF) and acute-on-chronic liver failure (ACLF), by comparing changes in conventional magnetic resonance imaging (MRI), in vivo proton magnetic resonance spectroscopy (PMRS) and diffusion tensor imaging (DTI) before and after its infusion.METHODS: Five patients each with ALF and ACLF in grade 3 or 4 hepatic encephalopathy and with clinical signs of raised intracranial pressure were studied along with five healthy volunteers. After baseline MRI, an intravenous bolus of 20% mannitol solution was given over 10 min in controls as well as in patients with ALF and ACLF. Repeat MRI for the same position was acquired 30 min after completing the mannitol injection. RESULTS: No statistically significant difference was observed between controls and patients with ALF and ACLF in metabolite ratios, DTI metrics and brain volume or CSF volume following 45 min of mannitol infusion. There was no change in clinical status at the end of post-mannitol imaging. | 7 | Selective and rapid determination of raltegravir in human plasma by liquid chromatography–tandem mass spectrometry in the negative ionization mode显示文摘A selective and rapid high-performance liquid chromatography–tandem mass spectrometry method was developed and validated for the quantification of raltegravir using raltegravir-d3 as an internal standard(IS). The analyte and IS were extracted with methylene chloride and n-hexane solvent mixture from 100 mL human plasma. The chromatographic separation was achieved on a Chromolith RP-18 e endcapped C18(100 mm 4.6 mm) column in a run time of 2.0 min. Quantitation was performed in the negative ionization mode using the transitions of m/z 443.1-316.1 for raltegravir and m/z 446.1-319.0 for IS. The linearity of the method was established in the concentration range of 2.0–6000 ng/m L.The mean extraction recovery for raltegravir and IS was 92.6% and 91.8%, respectively, and the IS-normalized matrix factors for raltegravir ranged from 0.992 to 0.999. | 8 | A general-purpose material property data extraction pipeline from large polymer corpora using natural language processing显示文摘The ever-increasing number of materials science articles makes it hard to infer chemistry-structure-property relations from literature.We used natural language processing methods to automatically extract material property data from the abstracts of polymer literature.As a component of our pipeline,we trained MaterialsBERT,a language model,using 2.4 million materials science abstracts,which outperforms other baseline models in three out of five named entity recognition datasets.Using this pipeline,we obtained~300,000 material property records from~130,000 abstracts in 60 hours.The extracted data was analyzed for a diverse range of applications such as fuel cells,supercapacitors,and polymer solar cells to recover non-trivial insights.The data extracted through our pipeline is made available at polymerscholar.org which can be used to locate material property data recorded in abstracts.This work demonstrates the feasibility of an automatic pipeline that starts from published literature and ends with extracted material property information. | Pranav Shetty Arunkumar Chitteth Rajan Chris Kuenneth Sonakshi Gupta Lakshmi Prerana Panchumarti Lauren Holm Chao Zhang Rampi Ramprasad | 2023 | npj Computational Materials2023,,1: | 0 |
