| 1 | Enhancing the antihepatitis B virus immune response by adefovir dipivoxil and entecavir therapies显示文摘Chronicity of hepatitis B(CHB)infection is characterized by a weak immune response to the virus.Entecavir(ETV)and adefovir dipivoxil(ADV)are effective in suppressing hepatitis B virus(HBV)replication.However,the underlying immune mechanism in the antiviral response of patients treated with nucleoside or nucleotide analogs is not clearly understood.In this study,regulatory T cells(Tregs)and intracellular cytokines,including IL-2,interferon(IFN)-γ,tumor-necrosis factor(TNF)-α and IL-4,were measured prior to and at 12,24,36 and 48 weeks after treatment with ETV or ADV.The cytokines were increased from 24 to 48 weeks after treatment.Higher levels of Th1 cytokines were observed with ETV(n=29)versus ADV(n=28)treatment.By contrast,the numbers of Tregs in both groups were decreased.The altered cytokine profile and cellular component was accompanied by a decrease in HBV DNA levels in both groups,which may contribute to their therapeutic effect in CHB infection.Our findings suggest that the antiviral effect of the drugs may be attributed not only to their direct effect on virus suppression but also to their immunoregulatory capabilities. | Yanfang Jiang Wanyu Li Lei Yu Jingjing Liu Guijie Xin Hongqing Yan Pinghui Sun Hong Zhang Damo Xu Junqi Niu | 2011 | Cellular & Molecular Immunology2011,8,1: | 16 |
| 2 | The effect of the fibre orientation of electrospun scaffolds on the matrix production of rabbit annulus fibrosus-derived stem cells显示文摘Annulus fibrosus(AF) tissue engineering has recently received increasing attention as a treatment for intervertebral disc(IVD) degeneration; however, such engineering remains challenging because of the remarkable complexity of AF tissue. In order to engineer a functional AF replacement, the fabrication of cell-scaffold constructs that mimic the cellular, biochemical and structural features of native AF tissue is critical.In this study, we fabricated aligned fibrous polyurethane scaffolds using an electrospinning technique and used them for culturing AF-derived stem/progenitor cells(AFSCs). Random fibrous scaffolds, also prepared via electrospinning, were used as a control. We compared the morphology, proliferation, gene expression and matrix production of AFSCs on aligned scaffolds and random scaffolds. There was no apparent difference in the attachment or proliferation of cells cultured on aligned scaffolds and random scaffolds. However, compared to cells on random scaffolds, the AFSCs on aligned scaffolds were more elongated and better aligned, and they exhibited higher gene expression and matrix production of collagen-I and aggrecan. The gene expression and protein production of collagen-II did not appear to differ between the two groups. | 5 | Substrate stiffness modulates bone marrow-derived macrophage polarization through NF-κB signaling pathway显示文摘The stiffness of the extracellular matrix(ECM)plays an important role in regulating the cellular programming.However,the mechanical characteristics of ECM affecting cell differentiation are still under investigated.Herein,we aimed to study the effect of ECM substrate stiffness on macrophage polarization.We prepared polyacrylamide hydrogels with different substrate stiffness,respectively.After the hydrogels were confirmed to have a good biocompatibility,the bone marrow-derived macrophages(BMMs)from mice were incubated on the hydrogels.With simulated by the low substrate stiffness,BMMs displayed an enhanced expression of CD86 on the cell surface and production of reactive oxygen species(ROS)in cells,and secreted more IL-1βand TNF-αin the supernatant.On the contrary,stressed by the medium stiffness,BMMs expressed more CD206,produced less ROS,and secreted more IL-4 and TGF-β.In vivo study by delivered the hydrogels subcutaneously in mice,more CD68+CD86+cells around the hydrogels with the low substrate stiffness were observed while more CD68+CD206+cells near by the middle stiffness hydrogels.In addition,the expressions of NIK,phosphorylated p65(pi-p65)and phosphorylated IκB(pi-IκB)were significantly increased after stimulation with low stiffness in BMMs.Taken together,these findings demonstrated that substrate stiffness could affect macrophages polarization.Low substrate stiffness promoted BMMs to shift to classically activated macrophages(M1)and the middle one to alternatively activated macrophages(M2),through modulating ROS-initiated NF-κB pathway.Therefore,we anticipated ECM-based substrate stiffness with immune modulation would be under consideration in the clinical applications if necessary. | Mimi Chen Yu Zhang Pinghui Zhou Xingzhi Liu Huan Zhao Xichao Zhou Qiaoli Gu Bin Li Xuesong Zhu Qin Shi | 2020 | Bioactive Materials2020,5,4: | 7 |
| 6 | Hydrogen production by reforming methane in a corona inducing dielectric barrier discharge and catalyst hybrid reactor显示文摘A novel corona inducing dielectric barrier discharge(CIDBD) and catalyst hybrid reactor was developed for reforming methane. | 7 | Study of the mechanisms of the flame propagation and stabilization in porous media显示文摘The CH4/air premixed gas combustion processes in porous media were numerically studied using the two-temperature reacting fluid model with dispersions and detailed chemical reaction mechanism GRI 3.0. The mechanisms of the propagation and stabilization of submerge flames and surface flames in porous media were illuminated distinctly by considering the magnitude of the terms in the two energy equations, analyzing the sensibility of flame propagation speed to flame location, heat exchange coefficient between gas and solid, thermal conductivity and radiative extinction coefficient of porous media. | 8 | Numerical investigation on hydrogen production by CH_4-Rich combustion in an alumina foam显示文摘The two-phase volume-averaged model with the detailed chemistry reaction mechanism GRI 3.0 was adopted in the quasi-steady-state simulation of hydrogen production by CH4-rich filtration combustion in an alumina foam under fully developed conditions. The relations among the combustion wave velocity, the inlet gas velocity and the equivalence ratio were discussed, and their influences on the distributions of temperature and species in the alumina foam and on H2 yield, CH4 conversion, H2 selectivity and CO selectivity were analyzed in detail. The results show that the combustion wave velocity increases with the increase of equivalence ratio or inlet gas velocity. The H2 yield exceeded 50% with equivalence ratio between 2.0 and 3.0 and combustion wave velocity larger than 0.4 mm/s. | 13 | Phenylfluorenamine-functionalized poly(N-vinylcarbazole)s as dopant-free polymer hole-transporting materials for inverted quasi-2D perovskite solar cells显示文摘In order to improve the efficiency and stability of inverted three-dimensional(3D) or quasi-2D perovskite solar cells(PSCs) for future commercialization, exploring high efficient dopant-free polymer holetransporting materials(HTMs) is still desired and meaningful. One simple and efficient way to achieve high performance dopant-free HTMs is to synthesize novel non-conjugated side-chain polymers via rational molecular design. In this work, N-(4-methoxyphenyl)-9,9-dimethyl-9H-fluoren-2-amine(FMeNPh) groups are introduced into the poly(N-vinylcarbazole)(PVK) side chains to afford two nonconjugated polymers PVCz-DFMeNPh and PVCz-FMeNPh as dopant-free HTMs in inverted quasi-2D PSCs. Benefited from the flexible properties of polyethylene backbone and excellent optoelectronic natures of FMeNPh side-chain groups, PVCz-DFMeNPh with more FMeNPh units exhibited excellent thermal stability, well-matched energy levels and improved charge mobility as compared to PTAA and PVCzFMeNPh. Moreover, the morphologies investigation of quasi-2D perovskite on PVCz-DFMeNPh shows more compact and homogeneous perovskite films than those on PTAA and PVCz-FMeNPh. As a result,the dopant-free PVCz-DFMeNPh based inverted quasi-2D PSCs deliver power conversion efficiency(PCE) up to 18.44% as well as negligible hysteresis and favorable long-term stability, which represents as excellent performance reported to date for inverted quasi-2D PSCs.
