20Anti-miR153对HEK293细胞增殖、凋亡和周期影响的实验研究显示文摘目的 在既往通过microRNA芯片和生物学信息技术,已筛查出miR153的基础上,进一步探讨其对HEK293细胞(人胚胎肾细胞,Human Embryonic Kidney 293 cells)增殖和凋亡的影响.方法 通过脂质体转染anti-miR153进入HEK293细胞以降低miR153的表达.采用realtime PCR的方法检测转染前后miR153在HEK293细胞中的表达变化;记录转染后细胞数量和形态学的变化.描制转染后HEK293细胞的生长曲线.流式细胞仪检测转染后细胞死亡和细胞周期的改变.结果 ①miR153在HEK293细胞中有表达;②在anti-miR153组中,HEK293细胞的miR153的表达明显低于阴性对照组,基因敲低率达80.5%~84.3%,证实转染的有效性;③anti-miR153组中细胞的生长明显低于阴性对照组,转染后24 h起增殖活性受到抑制,在48 h达到抑制高峰,较阴性对照组降低38.3%;④anti-miR153组中细胞离散,变圆,失去突触,容易脱壁,可见坏死漂浮的细胞;⑤流式细胞仪检测,anti-miR153组的细胞死亡和凋亡均增加(9.39%),明显高于阴性对照组(0.7%).细胞周期的检测,发现在anti-miR153组中G2/M期消失,更多的细胞进入了G0/G1期.结论 针对miR153的anti-miR的转染可以显著影响HEK293细胞的功能,包括细胞坏死、凋亡,停止生长,进入G0/G1期.Anti-miR153代表了一种新的肿瘤治疗方式,但还需要更多的进一步研究.李凯 John Perentesis 高解春 Gretchen Radloff 李军 肖现民 2010中华小儿外科杂志2010,31,12:1
