2KIT exon 11 codon 557/558 deletion/insertion mutations define a subset of gastrointestinal stromal tumors with malignant potential显示文摘AIM: To study the association of the frequency and pattern of KIT and PDGFRA mutations and clinicopathological factors in a group of patients with gastrointestinal stromal tumors (GIST). METHODS: Thirty patients with GIST were examined. Exons 9, 11, 13, and 17 of the KIT and exons 12 and 18 of the PDGFRA gene were analyzed for the presence of mutations by PCR amplification and direct sequencing. RESULTS: KIT or PDGFRA mutations were detected in 21 of the 30 patients (70%). Sixteen patients had mutations within KIT exon 11, three within KIT exon 9, and two within PDGFRA exon 18. GISTs with KIT exon 9 mutations were predominantly located in the small intestine, showed a spindle cell phenotype, and were assessed as potentially malignant. GISTs with KIT exon 11 mutations were located in the stomach and intestine, showed mainly a spindle cell phenotype, and were scored as potentially malignant (P < 0.05). Tumors with KIT exon 11 codon 557/558 deletion/insertion mutations were found to be associated with a potentially malignant clinical behaviour (P < 0.003). GISTs with PDGFRA mutations located in stomach showed a mixedcell phenotype and were classified as of very low or low moderate malignant potential. CONCLUSION: Determination of KIT and PDGFRA mutations should be additional parameters for the better prediction of GISTs clinical behaviour. 3Restoring the dampened expression of the core clock molecule BMAL1 protects against compression-induced intervertebral disc degeneration显示文摘The circadian clock participates in maintaining homeostasis in peripheral tissues,including intervertebral discs(IVDs).Abnormal mechanical loading is a known risk factor for intervertebral disc degeneration(IDD).Based on the rhythmic daily loading pattern of rest and activity,we hypothesized that abnormal mechanical loading could dampen the IVD clock,contributing to IDD.Here,we investigated the effects of abnormal loading on the IVD clock and aimed to inhibit compression-induced IDD by targeting the core clock molecule brain and muscle Arnt-like protein-1(BMAL1).In this study,we showed that BMAL1 KO mice exhibit radiographic features similar to those of human IDD and that BMAL1 expression was negatively correlated with IDD severity by systematic analysis based on 149 human IVD samples.The intrinsic circadian clock in the IVD was dampened by excessive loading,and BMAL1 overexpression by lentivirus attenuated compression-induced IDD.Inhibition of the RhoA/ROCK pathway by Y-27632 or melatonin attenuated the compression-induced decrease in BMAL1 expression.Finally,the two drugs partially restored BMAL1 expression and alleviated IDD in a diurnal compression model.Our results first show that excessive loading dampens the circadian clock of nucleus pulposus tissues via the RhoA/ROCK pathway,the inhibition of which potentially protects against compression-induced IDD by preserving BMAL1 expression.These findings underline the importance of the circadian clock for IVD homeostasis and provide a potentially effective therapeutic strategy for IDD.Dong Wang Pandi Peng Michal Dudek Xueyu Hu Xiaolong Xu Qiliang Shang Di Wang Haoruo Jia Han Wang Bo Gao Chao Zheng Jianxin Mao Chu Gao Xin He Pengzhen Cheng Huanbo Wang Jianmin Zheng Judith A.Hoyland Qing-Jun Meng Zhuojing Luo Liu Yang 2022Bone Research2022,10,2:2

5Liana community and functional trait analysis in tropical dry evergreen forest of India显示文摘Aim lianas are abundant and diverse throughout the world and consti-tute an important structural and functional component of tropical forests.This study aims to investigate liana diversity,abundance and their functional traits in Indian tropical dry evergreen forest(TDEF).Methods a total of ten 1-ha plots,one each in 10 Indian TDEF sites were demarcated.Each 1-ha plot was divided into one-hundred 10-×10-m quadrats to facilitate woody species inventory.all lianas≥1 cm diameter measured at 130 cm from the rooting point and all trees≥10-cm girth at breast height(gbh)were recorded from the study sites to analyze the patterns of liana diversity and abundance and also to compare the contribution of lianas to the total woody species richness,density and basal area.liana variables across the study sites were compared using one-way analysis of variance.The qualitative functional traits of inventoried lianas and trees were assessed on the field and referring to pertinent field manuals.Important Findingsa total of 9237 liana individuals(ranged from 408-1658 individu-als ha^(−1))representing 52 species,45 genera and 28 families were encountered from the 10 study sites.liana species richness ranged from 11-31 species ha^(−1) in 10 sites,which averaged 23.4(±5.7)species ha^(−1).The total basal area of lianas in the study sites was 7.3 m^(2)(0.20-1.76 m^(2) ha^(−1)).There was a significant variation in liana species richness,density and basal area across the studied sites.on the whole,lianas contributed 52%,49.3%and 4.1%to the total woody species(lianas and trees)richness,density and basal area,respectively.liana trait analysis revealed the majority(50%)of lianas belonged to brevi-deciduous type.stem twining was the chief climbing mechanism,exhibited by 21 species(52.6%of total abundance).more than half of the liana species(34 species;6925 individuals)had microphyllous leaves.Fleshy-fruited lianas mostly bearing berries and drupes constituted the major fruit type in the studied sites.Zoochory was the predominant dispersal mode observed in 63.4% of species.Considering the ecological and func-tional role of lianas in Indian TDEF,the need for conservation is emphasized.Pandi Vivek Narayanaswamy Parthasarathy 2015Journal of Plant Ecology2015,8,5:2
