1In vitro inhibition activity of polyphenol-rich extracts from Syzygium aromaticum(L.)Merr.& Perry(Clove)buds against carbohydrate hydrolyzing enzymes linked to type 2 diabetes and Fe^(2+)-induced lipid peroxidation in rat pancreas显示文摘Objective:To investigate and compare the inhibitor)'properties)of free and bound phenolic extracts of clove bud against carbohydrate hydrolyzing enzymes(alpha-amylase&alphaglucosidase)and Fe^(2+)-induced lipid peroxidation in rat pancreas in vitro.Methods:The free phenolics were extracted with 80%.(v/v)acetone,while bound phenolics were extracted from the alkaline and acid hydrolyzed residue with ethyl acetate.Then,the interaction of the extracts with alpha-amylase and alpha-glucosidase was subsequently assessed.Thereafter,the total phenolic contents and antioxidant activities of the extracts were determined.Results:The result revealed that both extracts inhibited alpha-amylase and alpha-glucosidase in a dose-dependent manner.However,the alpha-glucosidase inhibitory activity of the extracts were significantly(P<0.05)higher than their alpha-amylase inhibitory activity.The free phenolics(31.67 mg/g)and flavonoid(17.28 mg/g)contents were significantly(P<0.05)higher than bound phenolic(23.52 mg/g)and flavonoid(13.70 mg/g)contents.Both extracts also exhibited high antioxidant activities as typified by their high reducing power,LI diphenyl-2-picrylhydrazyl(DPPH)and 2,2-azinobis-3-ethylbenzo-thiazoline-6-sulfonate(ABTS)radical scavenging abilities,as well as inhibition of Fe^(2+)-induced lipid peroxidation in rat pancreas in vitro.Conclusions:This study provides a biochemical rationale by which clove elicits therapeutic effect on type 2 diabetes.Stephen Adeniyi Adefegha Ganiyu Oboh 2012Asian Pacific Journal of Tropical Biomedicine2012,2,10:11
2Phenolic composition and inhibitory activity of Mangifera indica and Mucuna urens seeds extracts against key enzymes linked to the pathology and complications of type 2 diabetes显示文摘Objective:To investigate the phenolic compounds composition and the inhibitory activity of Mangifera indica(M.indica) and Mucuna urens(M.urens) seeds extracts against some key enzymes(-amylase,-glucosidasc and aldose reductase) implicated in the pathology and complications of type 2 diabetes in vitro.Methods:Reverse phase chromatographic quantification of the major flavonoids and phenolic acids in the seeds extracts was carried out using high performance liquid chromatography coupled with diode array detection.The inhibitory activities of the seeds extracts against-amylase and-glucosidase were estimated using soluble starch and p-nitrophenylglucopyranosidc as their respective substrates.Inhibition of aldose reductase activity by the extracts was assayed using partially purified lens homogenate of normal male rat as source of enzyme;inhibition of Fe^(2+)-induced lipid peroxidation by extracts was tested in rat pancreas homogenate.Results:The chromatography result revealed that extracts of both seeds had appreciable levels of some major flavonoids and phenolic acids of pharmacological importance,including gallic acid,chlorogenic acid,caffeic acid,ellagic acid,catechin,rutin,quercitrin.quercetin and kaempferol.Extracts of both seeds effectively inhibited-amylase,-glucosidase and aldose reductase activities in a dose-dependent manner,having inhibitory preference for these enzymes in the order of aldose reductase>-glucosidase>-amylase.With lower half-maximal inhibitory concentrations(IC50)against-amylase,-glucosidase,and aldose reductase,M.indica had stronger inhibitory potency against these enzymes than M.urens.Extracts of both seeds also inhibited Fe2+-induced lipid peroxidation in a dose-dependent pattern,with M.indica being more potent than M.urens.Conclusions:The results obtained provide support for a possible use of M.indica and M.urens seeds in managing hyperglycemia and preventing the complications associated with it in type 2diabetes.Emmanuel Anyachukwu Irondi Ganiyu Oboh Afolabi Akintunde Akindahunsi Aline Augusti Boligon Margareth Linde Athayde 2014Asian Pacific Journal of Tropical Biomedicine2014,4,11:4

4Attenuation of oxidative stress and hepatic damage by some fermented tropical legume condiment diets in streptozotocin-induced diabetes in rats显示文摘Objective:To investigate the modulatory effect of fermented legume condiments diet on oxidative stress in streptozotocin(STZ) induced diabetic rats.Methods:Adult male Wistar rats were randomly divided into six groups with six animals in each group.Diabetes was induced by intraperitoneal injection of STZ(35 mg/kg b.w.).After being confirmed diabetic,the rats were fed with fermented Bambara groundnut.Locust bean and Soybean diets for 14 days.The plasma was obtained after 14-day treatment and analyzed for hepatic damage marker enzymes(AST,ALT and ALP) and in vivo antioxidant indices.Results:The diabetic untreated rats showed elevated (P<0.05) levels of AST,ALT.ALP and malondialdehyde with reduced activities of glutathiones -transferase,catalase as well as plasma reduced glutathione,vitamin C and total protein content.However,treatment of diabetic rats with lermenled legume condiments diets for 14 days significantly(P<0.05) reversed the above parameters towards normalcy,suggesting their modulation of oxidative stress,which mav be due to their high phenolic content and antioxidant capacity.Conclusions:The attenuation of oxidative stress and protection of hepatic tissue damage by the legume condiment diets in STZ induced diabetic rats compare favourably with that of metformin,a well known oral hypoglycemic drug.Adedayo O Ademiluyi Ganiyu Oboh 2012Asian Pacific Journal of Tropical Medicine2012,5,9:3
5Phenolics extract of Tetrapleura tetraptera fruit inhibits xanthine oxidase and Fe^2+-induced lipid peroxidation in the kidney,liver,and lungs tissues of rats in vitro显示文摘The phenolics composition and inhibitory activity of Tetrapleura tetraptera fruit extract on xanthine oxidase(XO)and Fe2+-induced lipid peroxidation in the kidney,liver and lungs tissues of rats were evaluated in vitro.Phenolics(flavonoids and phenolic acids)were quantified using reverse-phase high performance liquid chromatrography coupled with diode array detection(HPLC-DAD).The XO and Fe2+-induced lipid peroxidation inhibitory abilities of the extract were evaluated using spectrophotometric methods.The extract contained some flavonoids and phenolic acids,including catechin,epicatechin,rutin,quercetin,luteolin,apigenin;gallic,chlorogenic,caffeic and ellagic acids that are beneficial to health.The extract inhibited XO in the kidney,liver and lungs tissues in a dose-dependent manner.The half-maximal inhibitory concentrations(IC50)of the extract on XO from the tissues varied significantly(P<0.05),and were in the order of liver>kidney>lungs.The extract also inhibited Fe2+-induced lipid peroxidation in a dose-dependent pattern,having IC50 in the order of liver>lungs>kidney.T.tetraptera fruit extract could be a promising nutraceutical for preventing and managing hyperuricaemia and the associated disease conditions,due to its ability to inhibit XO;this bioactivity is attributable to combined effect of its flavonoids and phenolic acids.Emmanuel Anyachukwu Irondi Ganiyu Oboh Samson Olalekan Agboola Aline Augusti Boligon Margareth Linde Athayde 2016Food Science and Human Wellness2016,5,1:3

7Antioxidant properties of aqueous extracts of unripe Musa Paradisiaca on sodium nitroprusside induced lipid peroxidation in rat pancreas in vitro显示文摘Objective:To evaluate and compare antioxidant activities of the aqueous extracts of unripe plantain(Musa paradisiaca),assess their inhibitory action on sodium nitroprusside induced lipid peroxidation in rat pancreas in vitro and to cliaracterize the main phenolic constituents of the plantain products using gas chromatography analysis.Methods:Aqueous extracts of plantain products(raw,elastic pastry,roasted and boiled)(lour ol 0.1 g/mL(each)were used to determine their total phenol,total flavonoid,1,1 diphenyl-2 picrylhydrazyl(DPPH)and hydroxyl(OH)radical scavenging ability.The inhibitory effect of the extracts on sodium nitropmsside induced lipid peroxidation was also determined.Results:The results revealed that all the aqueous extracts showed antioxidant activity.The boiled Hour had highest DPPH and OH radical scavenging ability while raw Hour had the highest Fe^(2+)chelating ability,sodium nilroprusside inhibitor)effect and vitamin C content.The antioxidant results showed that elastic pastry had the highest total phenol and total flavonoid content.Characterization of the unripe plantain products for polyphenol contents using gas chromatography showed varied quantity of apigenin,myricetin.luteolin,capsaicin,isorhaemnetin.caffeic acid,kampferol,quercetin,p-hydroxybenzoic acid,shogaol,glycitein and gingerol per product on the spectra.Conclusions:Considering the antioxidant activities and ability to inhibit lipid peroxidation of unripe plantain,this could justify their traditional use in the management/prevention of diseases related to stress.Sidiqat Adamson Shodehinde Ganiyu Oboh 2013Asian Pacific Journal of Tropical Biomedicine2013,3,6:2

10Methanol extracts of Brachystegia eurycoma and Detarium microcarpum seeds flours inhibit some key enzymes linked to the pathology and complications of type 2 diabetes in vitro显示文摘The inhibitory effect of methanol extracts of Brachystegia eurycoma and Detarium microcarpum seeds flours on some key enzymes[α-amylase,α-glucosidase and aldose reductase(AR)]linked to the pathology and complications of type 2 diabetes(T2D);and their antioxidant properties were evaluated.The antioxidant properties evaluated were DPPH•and ABTS•^+scavenging abilities,reducing power,and antioxidant phytochemicals(total phenolics,tannins,total flavonoids and total saponins).Extracts of both flours inhibitedα-amylase,α-glucosidase and AR in a dose-dependent manner.The half-maximal inhibitory concentrations(IC50)of B.eurycoma onα-amylase,α-glucosidase,AR and lipid peroxidation were lower than those of D.microcarpum,indicating that it had stronger inhibitory potency than D.microcarpum.B.eurycoma also had significantly(P<0.05)higher DPPH•and ABTS•^+scavenging abilities,and reducing power than D.microcarpum.The antioxidant phytochemicals(total phenolics,tannins,total flavonoids and total saponins)were also significantly(P<0.05)higher in B.eurycoma than D.microcarpum.The inhibitory effect of B.eurycoma and D.microcarpum extracts onα-amylase,α-glucosidase and AR activities may be attributed to the combined action of their polyphenols and total saponins,and this may be a possible mechanism of action providing support for their use in managing hyperglycemia and the complications of T2D.Emmanuel Anyachukwu Irondi Ganiyu Oboh Afolabi Akintunde Akindahunsi 2015Food Science and Human Wellness2015,4,4:1
11Modulatory effects of dietary inclusion of garlic(Allium sativum) on gentamycin-induced hepatotoxicity and oxidative stress in rats显示文摘Objective:To investigate die ameliorative effect of dietary inclusion of garlic(Allium sativum)on gentamycin-induced hepatotoxicity in rats.Methods:Adult male rats were randomly divided into four groups with sis animals in each group.Groups 1 and 2 were fed basal diet while Groups 3 and 4 were fed diets containing 2%and 4%garlic respeetively for 27 d prior to gentamycin administration.Hepatotoxicity was induced by the intraperitoneal administration at gentamycin(100 mg/kg body weight)for 3 d.The liver and plasma were studied for hepatotoxicity and antioxidant indices.Results:Gentamycin induces hepatic damage as revealed by significant(P<0.05)elevation of liver damage marker enzymes(aspartate transaminase and alanine aminotransferase)and reduction in plasma albumin level.Gentamycin also eaused a significant(P<0.05)alteration in plasma and liver enzymatic(catalase,glutathione and super oxygen dehydrogenises)and non-enzymatic(glutathione and vitamin C)antioxidant indices with concomitant increase in the malondialdehyde content;however,there was a significant(P<0.05)restoration of the antioxidant status coupled with significant(P<0.05)decrease in the tissues malondialdehyde content,following consumption of diets containing garlic.Conclusions:These results suggest that dietary inclusion of garlic powder could protect against gentamycin-induced hepatotoxicity,improve antioxidant status and modulate oxidative stress;a function attributed to their phenolic constituents.Adedayo O Ademiluyi Ganiyu oboh Tosin R Owoloye Oluwaseun J Agbebi 2013Asian Pacific Journal of Tropical Biomedicine2013,3,6:1
