| 1 | Alteration of p53 and p21 during hepatocarcinogenesis in tree shrews显示文摘AIM: To investigate p53 mutation and p21 expression in hepatocarcinogenesis induced by hepatitis B virus (HBV) and aflatoxin B1 (AFB1) in tree shrews, and to reveal the role of these genes in hepatocarcinogenesis.METHODS: Tree shrews were divided into four groups:group A, those infected with HBV and fed with AFB1 (n = 39);group B, those infected with HBV alone (n = 28); group C,those fed with AFB1 alone (n = 29); and group D, normal controls (n = 20). The tree shrews underwent liver biopsies once every 15 wk. Expression of p53 and p21 proteins and genes in the biopsies and tumor tissues of the experimental tree shrews was detected, respectively, by immunohistochemistry,and by Southem blotting and reverse transcription-polymerase chain reaction and sequencing.RESULTS: The incidence of hepatocellular carcinomas (HCC) was higher in group A (66.7%) than that in group B (3.57%) and C (30%). The time of HCC occurrence was also earlier in group A than that in group C (120.0±16.6 wk vs 153.3±5.8 wk, respectively, P<0.01). p53 protein was not detected by immunohistochemistry in all groups before the 75^th wk of the experiment. At the 105^th wk, the positive rates fo p53 were 78.6%, 60% and 71.4% in groups A, B and C, respectively, which were significantly higher than that in group D (10%) (all P<0.05). An abnormal band of p53 gene was observed in groups A and C. The mutation points of p53gene in tree shrews with HCC were at codons 275, 78 and 13. The nucleotide sequence and amino acid sequence of tree shrew's wild-type p53 showed 91.7% and 93.4% homologies with those of human p53,respectively. The immunopositivity for p21 was found before HCC development. | 2 | Effect of longer battery life on small bowel capsule endoscopy显示文摘AIM:To determine if longer battery life improves capsule endoscopy(CE) completion rates.METHODS:A retrospective study was performed at a tertiary,university-affiliated hospital in Vancouver,Canada.Patients who underwent CE with either PillCamTM SB2 or SB2 U between 01/2010 and 12/2013 were considered for inclusion.SB2 and SB2 U share identical physical dimensions but differ in their battery lives(8 h vs 12 h).Exclusion criteria included history of gastric or small bowel surgery,endoscopic placement of CE,interrupted view of major landmarks due to technical difficulty or significant amount of debris,and repeat CE using same system.Basic demographics,comorbidities,medications,baseline bowel habits,and previous surgeries were reviewed.Timing of major landmarks in CE were recorded,and used to calculate gastric transit time,small bowel transit time,and total recording time.A complete CE study was defined as visualization of cecum.Transit times and completion rates were compared.RESULTS:Four hundred and eight patients,including 208(51.0%) males,were included for analysis.The mean age was 55.5 ± 19.3 years.The most common indication for CE was gastrointestinal bleeding(n =254,62.3%),followed by inflammatory bowel disease(n =86,21.1%).There was no difference in gastric transit times(group difference 0.90,95%CI:0.72-1.13,P =0.352) and small bowel transit times(group difference 1.07,95%CI:0.95-1.19,P =0.261) between SB2 U and SB2,but total recording time was about 14% longer in the SB2 U group(95%CI:10%-18%,P < 0.001) and there was a corresponding trend toward higher completion rate(88.2% vs 93.2%,OR =1.78,95%CI 0.88-3.63,P =0.111).There was no statistically significant difference in the rates of positive findings(OR =0.98,95%CI:0.64-1.51,P =0.918).CONCLUSION:Extending the operating time of CE may be a simple method to improve completion rate although it does not affect the rate of positive findings. | George Ou Neal Shahidi Cherry Galorport Oliver Takach Terry Lee Robert Enns | 2015 | World Journal of Gastroenterology2015，21，9： | 13 |
| 3 | Current systemic treatment of hepatocellular carcinoma:A review of the literature显示文摘Hepatocellular carcinoma(HCC) is the fifth most common form of human cancer worldwide and the third most common cause of cancer-related deaths. The strategies of various treatments for HCC depend on the stage of tumor,the status of patient's performance and the reserved hepatic function. The Barcelona Clinic Liver Cancer(BCLC) staging system is currently used most for patients with HCC. For example,for patients with BCLC stage 0(very early stage) and stage A(early stage) HCC,the curable treatment modalities,including resection,transplantation and radiofrequency ablation,are taken into consideration. If the patients are in BCLC stage B(intermediate stage) and stage C(advanced stage) HCC,they may need the palliative transarterial chemoembolization and even the target medication of sorafenib. In addition,symptomatic treatment is always recommended for patients with BCLC stage D(end stage) HCC. | 9 | Mechanical and low-cycle fatigue behavior of stainless reinforcing steel for earthquake engineering applications显示文摘Use of stainless reinforcing steel (SRS) in reinforced concrete (RC) structures is a promising solution to corrosion issues.However, for SRS to be used in seismic applications, several mechanical properties need to be investigated.These include specifi ed and actual yield strengths, tensile strengths, uniform elongations and low-cycle fatigue behavior.Three types of SRSs (Talley S24100, Talley 316LN and Talley 2205) were tested and the results are reported in this paper.They were compared with the properties of A706 carbon reinforcing steel (RS), which is typical for seismic applications, and MMFX II, which is a high strength, corrosion resistant RS.Low-cycle fatigue tests of the RS coupons were conducted under strain control with constant amplitude to obtain strain life models of the steels.Test results show that the SRSs have slightly lower moduli of elasticity, higher uniform elongations before necking, and better low-cycle fatigue performance than A706 and MMFX II.All five types of RSs tested satisfy the requirements of the ACI 318 code on the lower limit of the tensile to yield strength ratio.Except Talley 2205, the other four types of RSs investigated meet the ACI 318 requirement that the actual yield strength does not exceed the specified yield strength by more than 18 ksi (124 MPa).Among the three types of SRSs tested, Talley S24100 possesses the highest uniform elongation before necking, and the best low-cycle fatigue performance. | Yihui Zhou Yu-Chen OU George C.
