1The deubiquitinase OTUB1 augments NF-κB-dependent immune responses in dendritic cells in infection and inflammation by stabilizing UBC13显示文摘Dendritic cells(DCs)are indispensable for defense against pathogens but may also contribute to immunopathology.Activation of DCs upon the sensing of pathogens by Toll-like receptors(TLRs)is largely mediated by pattern recognition receptor/nuclear factor-κB(NF-κB)signaling and depends on the appropriate ubiquitination of the respective signaling molecules.However,the ubiquitinating and deubiquitinating enzymes involved and their interactions are only incompletely understood.Here,we reveal that the deubiquitinase OTU domain,ubiquitin aldehyde binding 1(OTUB1)is upregulated in DCs upon murine Toxoplasma gondii infection and lipopolysaccharide challenge.Stimulation of DCs with the TLR11/12 ligand T.gondii profilin and the TLR4 ligand lipopolysaccharide induced an increase in NF-κB activation in OTUB1-competent cells,resulting in elevated interleukin-6(IL-6),IL-12,and tumor necrosis factor(TNF)production,which was also observed upon the specific stimulation of TLR2,TLR3,TLR7,and TLR9.Mechanistically,OTUB1 promoted NF-κB activity in DCs by K48-linked deubiquitination and stabilization of the E2-conjugating enzyme UBC13,resulting in increased K63-linked ubiquitination of IRAK1(IL-1 receptor-associated kinase 1)and TRAF6(TNF receptor-associated factor 6).Consequently,DC-specific deletion of OTUB1 impaired the production of cytokines,in particular IL-12,by DCs over the first 2 days of T.gondii infection,resulting in the diminished production of protective interferon-γ(IFN-γ)by natural killer cells,impaired control of parasite replication,and,finally,death from chronic T.encephalitis,all of which could be prevented by low-dose IL-12 treatment in the first 3 days of infection.In contrast,impaired OTUB1-deficient DC activation and cytokine production by OTUB1-deficient DCs protected mice from lipopolysaccharide-induced immunopathology.Collectively,these findings identify OTUB1 as a potent novel regulator of DCs during infectious and inflammatory diseases.Floriana Mulas Xu Wang Shanshan Song Gopala Nishanth Wenjing Yi Anna Brunn Pia-Katharina Larsen Berend Isermann Ulrich Kalinke Antonio Barragan Michael Naumann Martina Deckert Dirk Schlüter 2021Cellular & Molecular Immunology2021,18,6:4

12Analysis of flow-field in a dual mode ramjet combustor with boundary layer bleed in isolator显示文摘A two-dimensional Reynolds averaged Navier Stokes(RANS)simulation of a dual mode ramjet(DMRJ)combustor is performed,modeling the University of Michigan dual-mode combustor experimental setup operating in reacting mode with different equivalence ratios(4).The simulations are carried out using a k-u SST turbulence model and a steady diffusion flamelet model for non-premixed combustion.Air enters the isolator at Mach 2.2,stagnation pressure and temperature of 549.2 kPa and 1400 K respectively.Hydrogen is injected transverse to the flow direction and upstream of the cavity flame holder to simulate ramjet(4 Z 0.29)and scramjet(4 Z 0.19)modes of operation.Wall static pressure plots are used to validate numerical results against experimental data.Analysis of flow separation in ramjet mode due to the presence of a shock train in the isolator is carried out by means of numerical Schlieren images overlapped with contours of negative axial velocity,showing the effects of shock wave boundary layer interaction(SWBLI).Active control through wall normal boundary layer bleed in the separated flow region is implemented,which weakens the shock train and moves it downstream closer to the cavity.Bleed results in an improved stagnation pressure recovery in ramjet mode,with a marginal increase in combustion efficiency.Nishanth Thillai Amit Thakur Srikrishnateja K. 14Design of bio-oil additives via molecular signature descriptors using a multi-stage computer-aided molecular design framework显示文摘Direct application of bio-oil from fast pyrolysis as a fuel has remained a challenge due to its undesirable attributes such as low heating value,high viscosity,high corrosiveness and storage instability.Solvent addition is a simple method for circumventing these disadvantages to allow further processing and storage.In this work,computer-aided molecular design tools were developed to design optimal solvents to upgrade bio-oil whilst having low environmental impact.Firstly,target solvent requirements were translated into measurable physical properties.As different property prediction models consist different levels of structural information,molecular signature descriptor was used as a common platform to formulate the design problem.Because of the differences in the required structural information of different property prediction models,signatures of different heights were needed in formulating the design problem.Due to the combinatorial nature of higher-order signatures,the complexity of a computer-aided molecular design problem increases with the height of signatures.Thus,a multi-stage framework was developed by developing consistency rules that restrict the number of higher-order signatures.Finally,phase stability analysis was conducted to evaluate the stability of the solvent-oil blend.As a result,optimal solvents that improve the solvent-oil blend properties while displaying low environmental impact were identified.Jia Wen Chong Suchithra Thangalazhy-Gopakumar Kasturi Muthoosamy Nishanth G.Chemmangattuvalappil 2022Frontiers of Chemical Science and Engineering2022,16,2:1
