1转录因子RORγt对妊娠期哮喘模型小鼠Th17/Treg平衡的调节作用显示文摘目的观察妊娠期哮喘模型小鼠Th17细胞相关细胞因子和维甲酸相关孤儿受体γt(RORγt)的表达变化,探讨地塞米松(DXM)对小鼠气道炎症和Th17细胞分泌功能的影响。方法 30只雌性BALB/c小鼠随机分为健康妊娠(HP)组、妊娠期哮喘(AP)组和地塞米松(DXM)组,每组10只。AP组和DXM组于实验当天(0d)和7、14d时分别给予OVA/Al(OH)3混悬液腹腔注射致敏,HP组仅给予单纯等量生理盐水(NS)。3组小鼠合笼交配至妊娠成功。于21d起,连续7d对AP组和DXM组给予50g/L OVA雾化激发(1次/d,40min/次);DXM组于雾化吸入OVA前30min给予1mg/kgDXM腹腔注射;HP组仅给予单纯等量NS进行雾化激发。各组小鼠于末次激发24h后进行气道反应性检测,结果以相应浓度乙酰胆碱(Mch)激发下增强的呼吸间歇(Penh)值与基线值的百分比[Penh值(%)]来表示;分析支气管肺泡灌洗液(BALF)中白细胞总数及分类计数;取肺组织行病理学观察;采用流式细胞仪检测外周血Th17和Treg细胞的比例并计算Th17/Treg比值;ELISA法测定外周血IL-17、IL-23、IFN-γ和RORγt含量,RT-PCR法检测前述因子在肺组织中的表达,Western blotting检测肺组织中RORγt的表达。结果雌性BALB/c小鼠以OVA致敏后气道反应性增加,BALF中白细胞总数及嗜酸性粒细胞(Eos)、淋巴细胞(Lym)和中性粒细胞(Neu)比例增加,肺组织病理学改变符合哮喘的特征。外周血标本中,AP组Th17细胞显著升高,Treg细胞显著降低(P<0.05),Th17/Treg比值上升。DXM组Th17和Treg细胞变化趋势与AP组相同,Th17/Treg比值高于HP组,但低于AP组(P<0.05)。分别在肺组织和外周血标本中检测蛋白和mRNA表达水平,结果显示,AP组IL-17、IL-23及RORγt含量均高于HP组及DXM组(P<0.05),IFN-γ含量低于HP组及DXM组(P<0.05)。AP组肺组织RORγt蛋白的表达(61.27%±3.11%)明显高于DXM组(37.51%±1.93%,P<0.01)和HP组(22.39%±0.86%,P<0.01),后两者比较差异亦有统计学意义(P<0.01)。相关分析表明,AP组肺组织RORγt蛋白表达量与Eos、Lym和Neu细胞绝对值呈显著正相关(r=0.879、0.759、0.802,P<0.01),与外周血IL-17含量、肺组织IL-17mRNA表达水平亦呈正相关(r=0.696、0.863,P<0.01)。结论 Th17细胞分泌功能及RORγt表达上调参与了妊娠期哮喘的发生发展。DXM干预在改善哮喘症状的同时可降低RORγt表达,使Th17分化和增殖功能受抑。马佳佳 Nick Lu 陈必良 2012解放军医学杂志2012,37,6:20

3The Seasonal Prediction of the Exceptional Yangtze River Rainfall in Summer 2020显示文摘During June and July of 2020,the Yangtze River basin suffered from extreme mei-yu rainfall and catastrophic flooding.This study explores the seasonal predictability and associated dynamical causes for this extreme Yangtze River rainfall event,based on forecasts from the Met Office GloSea5 operational forecast system.The forecasts successfully predicted above-average rainfall over the Yangtze River basin,which arose from the successful reproduction of the anomalous western North Pacific subtropical high(WNPSH).Our results indicate that both the Indian Ocean warm sea surface temperature(SST)and local WNP SST gradient were responsible for the westward extension of the WNPSH,and the forecasts captured these tropical signals well.We explore extratropical drivers but find a large model spread among the forecast members regarding the meridional displacements of the East Asian mid-latitude westerly jet(EAJ).The forecast members with an evident southward displacement of the EAJ favored more extreme Yangtze River rainfall.However,the forecast Yangtze River rainfall anomaly was weaker compared to that was observed and no member showed such strong rainfall.In observations,the EAJ displayed an evident acceleration in summer 2020,which could lead to a significant wind convergence in the lower troposphere around the Yangtze River basin,and favor more mei-yu rainfall.The model forecast failed to satisfactorily reproduce these processes.This difference implies that the observed enhancement of the EAJ intensity gave a large boost to the Yangtze River rainfall,hindering a better forecast of the intensity of the event and disaster mitigation.Chaofan LI Riyu LU Nick DUNSTONE Adam ASCAIFE Philip EBETT Fei ZHENG 2021Advances in Atmospheric Sciences2021,38,12:5

6Seasonal Forecasts of the Summer 2016 Yangtze River Basin Rainfall显示文摘The Yangtze River has been subject to heavy flooding throughout history,and in recent times severe floods such as those in 1998 have resulted in heavy loss of life and livelihoods.Dams along the river help to manage flood waters,and are important sources of electricity for the region.Being able to forecast high-impact events at long lead times therefore has enormous potential benefit.Recent improvements in seasonal forecasting mean that dynamical climate models can start to be used directly for operational services.The teleconnection from El Ni ?no to Yangtze River basin rainfall meant that the strong El Ni ?no in winter 2015/16 provided a valuable opportunity to test the application of a dynamical forecast system.This paper therefore presents a case study of a real-time seasonal forecast for the Yangtze River basin,building on previous work demonstrating the retrospective skill of such a forecast.A simple forecasting methodology is presented,in which the forecast probabilities are derived from the historical relationship between hindcast and observations.Its performance for2016 is discussed.The heavy rainfall in the May–June–July period was correctly forecast well in advance.August saw anomalously low rainfall,and the forecasts for the June–July–August period correctly showed closer to average levels.The forecasts contributed to the confidence of decision-makers across the Yangtze River basin.Trials of climate services such as this help to promote appropriate use of seasonal forecasts,and highlight areas for future improvements.Philip E. 9IL-17R-FcγRⅡb基因体外诱导树突状细胞表达及其对淋巴细胞增殖活化的影响显示文摘目的:构建编码人全长IL-17R与FcγRⅡb双基因表达重组腺病毒载体并对其特性进行鉴定,探讨阻断其信号调节通路对妊娠哮喘发病的影响。方法:将扩增出的拼接基因片段定向克隆于pAdTrack-CMV中经与pAdeasy-1同源重组,鉴定正确的阳性重组子通过脂质体介导转入AD293细胞,荧光显微镜观察绿色荧光(GFP)信号并采用PCR法对其进行鉴定;纯化腺病毒感染体外细胞因子协同诱导培养的妊娠哮喘模型小鼠骨髓来源树突状细胞(DC),利用RT-PCR、Western免疫印迹检测IL-17R和FcγRⅡb表达;流式细胞术、CCK-8、ELISA分析重组腺病毒的修饰对DC表面分子表达,刺激T细胞增殖抑制变化和MLR上清中IFN-γ、IL-4、IL-17分泌水平影响。结果:构建的重组腺病毒载体插入序列完全正确;经GFP表达和PCR检测证实成功包装获得高滴度重组腺病毒;感染DCs的GFP阳性细胞数达到85%;在mRNA及蛋白水平均可同时转录和表达出符合预期的IL-17R和FcγRⅡb。CD11c阳性表达率在各组间无显著差异;与对照细胞及EGFP-DCs相比,经IL-17RFcγRⅡb修饰的DC表面CD80表达增高,而CD86表达减少(P<0.05);IL-17R-FcγRⅡb-DCs相同浓度比例刺激T细胞增殖水平均明显受抑(P<0.05);转染载体共培养物上清液中Th2/Th17类细胞因子IL-4、IL-17水平下降,Th1类细胞因子IFN-γ升高(P<0.05),IFN-γ/IL-4/IL-17相对比值增加(P<0.05)。结论:IL-17R-FcγRⅡb融合基因体外阻断CD86/CD28共刺激通路可诱导DC免疫耐受状态,抑制抗原特异性T细胞增殖并影响Th1/Th2/Th17亚群分化,有望在妊娠哮喘免疫耐受缺陷和缓解气道炎症反应中发挥作用。马佳佳 Nick Lu 陈必良 2013中国免疫学杂志2013,29,11:1
10重组IL-12表达载体构建及对妊娠期哮喘小鼠气道炎症的调节与免疫应答影响显示文摘目的:构建携带IL-12重组基因的真核表达载体,探讨其对妊娠期哮喘小鼠气道炎症的免疫调节作用及其可能机制。方法:PCR扩增mIL-12基因cDNA,利用DNA重组技术将线性化片段定向插入载体pcDNA3.1(+)质粒中,经酶切和测序鉴定,通过脂质体介导瞬时转染P815细胞,观察荧光蛋白在细胞中的表达和定位。以鸡卵清蛋白(OVA)致敏和激发建立妊娠期哮喘小鼠模型,通过气道内灌注:妊娠期哮喘(AP)+pcDNA3.1(+)-mIL-12组(AP1组)、单纯哮喘(ANP)+pcD-NA3.1(+)-mIL-12组(ANP1组)给予50μl pcDNA3.1(+)-mIL-12/脂质体混合液;AP+pcDNA3.1(+)组(AP2组)、ANP+pcDNA3.1(+)组(ANP2组)给予50μl pcDNA3.1(+)/脂质体混合液;单纯妊娠组(HP组)、阴性对照组(HNP组)给予50μlPBS。于末次激发24小时后,观察肺泡灌洗液(BALF)炎性细胞计数及肺组织HE染色以评价小鼠气道炎症程度;RT-PCR、Western blot鉴定小鼠肺组织匀浆中IL-12蛋白及mRNA表达;流式细胞术检测小鼠脾细胞IL-17+及IFN-γ+分泌水平;ELISA测定小鼠外周血上清中细胞因子含量变化。结果:成功构建重组pcDNA3.1(+)-mIL-12真核表达载体,并在P815细胞中获得高效表达。与HP、HNP组相比,AP1、ANP1组BALF中白细胞总数及Eos,Neu,Lym占细胞总数百分比均显著低于AP2、ANP2组,但高于HP和HNP组,AP1与ANP1组间尤以AP1组降低显著(P<0.05),且肺组织炎症细胞浸润明显减轻,炎症反应明显轻微。肺组织标本中,AP1、ANP1组IL-12蛋白含量及mRNA表达水平在重组质粒干预后显著高于AP2、ANP2组。脾组织标本中AP1、ANP1组IFN-γ+含量明显升高,而CD4+IL-17+明显降低(P<0.05);IFN-γ+与CD4+IL-17+比例高于AP2、ANP2组(P<0.05);此变化趋势AP1组比ANP1组更为明显(P<0.05);HP组IFN-γ+及IL-17+含量变化与HNP组组间差异不显著。外周血上清中细胞因子检测结果 AP1、ANP1组IFN-γ明显升高,IL-4、IL-17显著降低(P<0.05)。结论:构建的pcD-NA3.1(+)-mIL-12真核表达载体能够在明显改善妊娠期哮喘小鼠气道炎症反应的同时上调IFN-γ表达,使Th2、Th17型细胞分化和增殖功能受抑,证实IL-12表达上调参与了妊娠期哮喘的发生发展。马佳佳 Nick Lu 陈必良 滑玮 张建芳 2013中国免疫学杂志2013,29,3:0
11Acute hepatitis of unknown aetiology among children around the world显示文摘By 26 August 2022, the number of cases of acute hepatitis of unknown etiology (AHUA) has drastically increased to 1115 distributed in 35 countries that fulfill the World Health Organization definition. Several hypotheses on the cause of AHUA have been proposed and are being investigated around the world. In the recent United Kingdom (UK) report, human adenovirus (HAdV) with adeno-associated virus (AAV) co-infection is the leading hypothesis. However, there is still limited evidence in establishing the causal relationship between AHUA and any potential aetiology. The leading aetiology continues to be HAdV infection. It is reported that HAdV genomics is not unusual among the population in the UK, especially among AUHA cases. Expanding the surveillance of HAdV and AAV in the population and the environment in the countries with AUHA cases is suggested to be the primary action. Metagenomics should be used in detecting other infectious pathogens on a larger scale, to supplement the detection of viruses in the blood, stool, and liver specimens from AUHA cases. 12人可溶性sTNFR1及胞膜外区Ig融合基因重组腺病毒的真核表达及功能活性鉴定显示文摘目的构建携带人sTNFR1与IgGFc片段融合基因的重组腺病毒,稳定表达并初步鉴定其功能活性在妊娠哮喘炎症机制中的作用。方法将PCR扩增的sTNFR1及IgGFc基因片段先后连接到穿梭质粒pAdTrack-CMV多克隆位点上,酶切线性化后在BJ5183细菌内与骨架质粒pAdEasy-1同源重组,产生获得的AdE-EGFP-sTNFR1-Ig以脂质体法转染AD293细胞进行病毒包装,以绿色荧光蛋白(GFP)监测病毒滴度及感染效率。RT-PCR、Western blot实验鉴定细胞内重组腺病毒转录及融合基因与蛋白表达;流式细胞术、ELISA实验分析混合培养上清中转染肥大细胞(MC)激活、调节同基因来源T细胞表达Th2/Treg水平及细胞因子分泌效果;MTT评估转染细胞上清中sTNFR1-Ig对TNF-α介导细胞毒效应的拮抗作用。结果成功构建编码sTNFR1-Ig基因的重组腺病毒载体经酶切和测序鉴定正确,制备的腺病毒在转染细胞中的GFP表达以及CPE效应明显,扩增后的滴度为3.7×1010~4.8×1010pfu/ml,体外感染效率可达90%以上;sTNFR1-Ig基因在mRNA及蛋白水平获得的清晰条带均证实腺病毒包装与扩增成功。表达sTNFR1-Ig的MC诱导T细胞转化为CD4+CD25+Foxp3+T细胞水平以及IL-12含量均明显升高,CD4+IL-4+Th2细胞水平下调(P<0.05),Th2/Treg所占CD4+T细胞比值下降(P<0.05)。sTNFR1-Ig封闭TNF-α对MC免疫毒性作用的浓度效应明显,而EGFP-MCs与对照细胞上清对TNF-α毒性效应的中和阻断能力不足(P<0.05)。结论携sTNFR1-Ig基因重组腺病毒参与的免疫调控与妊娠期哮喘发生发展密切相关。sTNFR1及IgG-Fc亚基胞膜外区结合的表达和引起免疫耐受的作用研究,为妊娠哮喘发病机制在理论上的新突破,为以sTNFRI-Ig为靶点的妊娠哮喘干预治疗奠定基础。马佳佳 Nick Lu 陈必良 2013中国药理学通报2013,29,8:0
13Prediction of the Western North Pacific Subtropical High in Summer without Strong ENSO Forcing显示文摘The western North Pacific subtropical high(WNPSH) is one of the deterministic predictors of the East Asian summer climate, and a better prediction of the WNPSH favors more reasonable forecast of the East Asian summer climate. This study focuses on seasonal prediction of the WNPSH during neutral summers without strong El Ni?o–Southern Oscillation(ENSO) forcing, and explores the associated predictable sources, using the one-month lead time retrospective forecasts from the Ensembles-Based Predictions of Climate Changes and Their Impacts(ENSEMBLES) project during 1960–2005. The results indicate that the ENSEMBLES atmosphere–ocean–land coupled models exhibit considerable prediction skill for the WNPSH during neutral summers, with successful reproduction of the WNPSH in the majority of neutral summers. The anomalous WNPSH in neutral summers, which corresponds to cyclonic/anticyclonic anomalies in the lower troposphere, is highly correlated with an east–west dipole local sea surface temperature(SST) distribution over the tropical WNP, suggesting an intimate local air–sea coupling. Further diagnosis of the local SST–rainfall relationship and surface heat flux indicates that the anomalous local SST plays an active role in modulating the variation of the WNPSH during neutral summers, rather than passively responding to the atmospheric change. The local SST anomalies and relevant air–sea coupling over the tropical WNP are reasonably well reproduced in the model predictions, and could act as primary predictable sources of the WNPSH in neutral summers. 15靶向沉默Gata3对妊娠期哮喘小鼠树突状细胞表型及效应T细胞失衡表达的影响显示文摘目的:研究慢病毒介导siRNA靶向沉默Gata3基因表达对妊娠哮喘模型小鼠骨髓来源树突状细胞(DC)抗原递呈及其免疫生物学特性的影响。方法:以脂质体DNA沉淀法将重组慢病毒干扰质粒与包装质粒共转染AD293细胞,包装生成慢病毒感染体外诱导DC,RT-PCR、Western blot分析其对siGata3基因的整合转录效果;流式细胞术观察细胞表型变化。通过尾静脉注射将pSH1/siGata3转移至小鼠体内,ELISA法测定其肺泡灌洗液(BALF)中的炎性因子含量;MTT法验证混合淋巴细胞反应(MLR)后T细胞增殖能力。结果:经重组、包装携有Gata3特异siRNA慢病毒感染DC成功,其siRNA靶点Gata3 mRNA及蛋白的转录表达分别下调87.30%和81.33%(P<0.05);同时降低DCs膜表面CD86荧光强度(P<0.05)。减少pSH1/siGata3组小鼠BALF中IL-5、IL-17分泌而提升IL-12水平(P<0.05);MLR证实,共培养感染DC激活T细胞的能力明显不足(P<0.05)。结论:siRNA干扰可有效抑制模型小鼠免疫微环境特定Gata3通路,阻遏T细胞及其亚群分化由此来诱导DC免疫耐受,为妊娠期哮喘防治奠定基础并提供新的思路和手段。马佳佳 陈必良 Nick Lu 2014现代妇产科进展2014,23,7:0
