1Angiogenic synergistic effect of basic fibroblast growth factor and vascular endothelial growth factor in an in vitro quantitative microcarrier-based three-dimensional fibrin angiogenesis system显示文摘AIM: To develop an in vitro three-dimensional (3-D) angiogenesis system to analyse the capillary sprouts induced in response to the concentration ranges of basic fibroblast growth factor (bFGF) and vascular endothelial growth factor (VEGF) and to quantify their synergistic activity.METHODS: Microcarriers (MCs) coated with human microvascular endothelial cells (HMVECs) were embedded in fibrin gel and cultured in 24-well plates with assay media. The growth factors bFGF, or VEGF, or both were added to the system. The wells (n = 8/group) were digitally photographed and the average length of capillary-like sprouts (ALS) from each microcarrier was quantitated.RESULTS: In aprotinin-stabilized fibrin matrix, human microvascular endothelial cells on the MCs invaded fibrin,forming sprouts and capillary networks with lumina. The angiogenic effects of bFGF or VEGF were dose-clependent in bhe range from 10 to 40 ng/mL. At d 1, 10 ng/mL of bFGF and VEGF induced angiogenesis with an ALS of 32.13±16.6 μm and 43.75±27.92 μm, respectively, which were significantly higher than that of the control (5.88±4.45 μm, P<0.01),and the differences became more significant as the time increased. In addition, the combination of 10 ng/mL of bFGF and VEGF each induced a more significant effect than the summed effects of bFGF (10 ng/mL) alone and VEGF (10 ng/mL) alone when analyzed using SPSS system for general linear model (GLM) (P= 0.011), and bhat also exceeded the effects by 20 ng/mL of either bFGF or VEGF.CONCLUSION: A microcarrier-based in vitro threedimensional angiogenesis model can be developed in fibrin.It offers a unique system for quantitative analysis of angiogenesis. 2Luobusaite:A New Mineral显示文摘A group of mantle minerals including about 70-80 subtypes of minerals are discovered from a podiform chromitite in Tibet,China.Recovered minerals include diamond,coesite,moissanite,wustite,Fe-silides and a new mineral,luobusaite.All of these minerals were hand-picked from heavy-mineral separates of the podiform chromitite in the mantle peridotite of an ophiolite.The grains of luobusaite are as host mineral with inclusions of native silicon or as an intergrowth with native silicon and Fe-Si phase.Luobusaite occurs as irregular grains,with 0.1-0.2 mm in size,consisting of very fine-grained aggregates.The mineral is steel-grey in color,metallic luster,and opaque.The empirical formula(based on 2 for Si)is Fe0.83Si2,according to the chemical compositions of luobusaite.X-ray powder-diffraction data:orthorhombic system,space group Cmca,a = 9.874(14)?,b = 7.784(5)?,c= 7.829(7)?,Z=16.BAI Wenji SHI Nicheng FANG Qingsong LI Guowu XIONG Ming YANG Jingsui RONG He 2006Acta Geologica Sinica(English Edition)2006,80,5:10
3Titanium, Ti, A New Mineral Species from Luobusha, Tibet, China显示文摘We describe the new mineral species titanium,ideally Ti,found in the podiform chromitites of the Luobusha ophiolite in Tibet,People's Republic of China.The irregular crystals range from 0.1 to 0.6 mm in diameter and form an intergrowth with coesite and kyanite.Titanium is silver grey in colour,the luster is metallic,it is opaque,the streak is grayish black,and it is non-fluorescent.The mineral is malleable,has a rough to hackly fracture and has no apparent cleavage.The estimated Mohs hardness is 4,and the calculated density is 4.503 g/cm3.The composition is Ti 99.23-100.00 wt%.The mineral is hexagonal,space group P63 /mmc.Unit-cell parameters are a 2.950(2),c 4.686(1),V 35.32(5) 3,Z = 2.The five strongest powder diffraction lines [d in(hkl)(I/I0)] are: 2.569(010)(32),2.254(011)(100),1.730(012)(16),1.478(110)(21),and 0.9464(121)(8).The species and name were approved by the CNMNC(IMA 2010–044).FANG Qingsong BAI Wenji YANG Jingsu RONG He SHI Nicheng LI Guowu XIONG Ming MA Zhesheng 2013Acta Geologica Sinica(English Edition)2013,87,5:7
4Differential gene expression during capillary morphogenesis in a microcarrier-based three-dimensional in vitro model of angiogenesis with focus on chemokines and chemokine receptors显示文摘AIM: To globally compare the gene expression profiles during the capillary morphogenesis of human microvascular endothelial cells (HMVECs) in an in vitro angiogenesis system with affymetrix oligonucleotide array.METHODS: A microcarrier-based in vitro angiogenesis system was developed, in which ECs migrated into the matrix,proliferated, and formed capillary sprouts. The sprouts elongated, branched and formed networks. The total RNA samples from the HMVECs at the selected time points (0.5,24, and 72 h) during the capillary morphogenesis were used for microarray analyses, and the data were processed with the softwares provided by the manufacturers. The expression patterns of some genes were validated and confirmed by semi-quantitative RT-PCR. The regulated genes were grouped based on their molecular functions and expression patterns, and among them the expression of chemokines and chemokine receptors was specially examined and their functional implications were analyzed.RESULTS: A total of 1 961 genes were up- or downregulated two-folds or above, and among them, 468 genes were up- or down-regulated three-folds or above. The regulated genes could be grouped into categories based on their molecular functions, and were also clustered into six groups based on their patterns of expression. As for chemokines and chemokine receptors, CXCL1/GRO-α,CXCL2/GRO-β, CXCLS/ENA-78, CXCL6/GCP2, IL-8/CXCL8,CXCL12/SDF-1, CXCL9/Mig, CXC11/ITAC, CX3CL1/fractalkine,CCL2/MCP-1, CCL3, CCLS/RANTES, CCL7, CCL15, CCL21,CCL23, CCL28, and CCR1, CCR9, CXCR4 were identified.Moreover, these genes demonstrated different changing patterns during the capillary morphogenesis, which implied that they might have different roles in the sequential process. Among the chemokines identified, CCL2/MCP-1,CCL5/RANTES and CX3CL1 were specially up-regulated at the 24-h time point when the sprouting characterized the morphological change. 6Mineralogy and crystal structure determination of Mg-fillowite显示文摘Accurate crystal structure determination of complex phosphate Mg-fillowite ? Na2Ca(Mn4Mg2Fe)7[PO4]6 has been finished. Mg-fillowite occurs in muscovite-pegmatite in the Altay area, Xinjiang, China. It is of the trigonal lattice, with space group R-3, unit cell a=1.5143(3) nm, c=4.3191(2) nm, V=8.5736 nm3, Z = 18. The R-factor of the determination accuracy is R (I > 2σ ( I )) = 0.0776. The cationic polyhedra consist of two kinds of structural units: compound col- umn and screw column. In three-dimensional space, they are linked with each other through corner sharing or edge sharing to build the entire structure. There are 45 atoms in an asymmetric unit and 720 atoms in a unit cell. In terms of types and connection manners of the coordination polyhedra, the degree of complicity is rarely seen in all mineral crystal structures. 8Point Groups and Single Forms of Quasicrystals with Eightfold and Twelvefold Symmetry显示文摘This paper mainly deals with the point groups and single forms of octagonal quasicrystals and the description of one-dimensional quasilattice. The authors present a new sequence for describing the arrangement of quasiperiods in one-dimensional quasilattice. The first ten numbers of quasiperiods of this sequence are 1, 1, 2, 5, 12, 29, 70, 169, 408 and 985. The arrangement of quasiperiods in the first five steps are a, b, ab. babab and babababbabab. Seven p(?)nt groups and nine single forms for the octagonal system have been deduced, They are as follows: Point groups: 8.8m, 82, 8/m, 8/mmm, 8 and 82m; single forms: octagonal prism, dioctagonal prism. octagonal pyramid. dioctagonal pyramid. octagonal dipyramid, dioctagonal dipyramid, octagonal scalenohedron, dioctagonal scalenohedron and octagonal trapezohedron. 9Determination of crystal structure of omphacite显示文摘The chemical formula of omphacite was expressed with (MM)(Si, AI)2O6.Cations that occupied the M site were large cations such as Ca or Na, and Na/(Na+Ca) ratio ranged from 0.2 to 0.8; the 6-coordinated M site accommodated cations such as Mg, Fe2+,Al, Fe3+ ,and Al/(Al+Fe3+) ratio was more than 0.5 Omphacite space groups reported were C2/c, P2, P2/n, P2/c, cell parameters are a0 = 0.9600—0.9630 nm, b0 = 0.8750—0.8830 nm, C0 =0.5230—0.5290 nm,β = 106°40’—107°10’. The sample was picked up from the eclogites in Zhucheng County, Shangdong Province. The intensity data were collected with the RIGAKU RASA Ⅱ -S four-circle single crystal diffractometer. The correct structure model was obtained by using the Patterson method and Fourier synthesis, SHELX-76 program, structure refinement with 905 independent diffraction points (|F0|> 3σ|F0 |). After the structure parameter refinement, the R-factor reduced to 0.0515.
