| 1 | Magnesium deficiency in plants:An urgent problem显示文摘Although magnesium(Mg) is one of the most important nutrients, involved in many enzyme activities and the structural stabilization of tissues, its importance as a macronutrient ion has been overlooked in recent decades by botanists and agriculturists, who did not regard Mg deficiency(MGD) in plants as a severe health problem. However, recent studies have shown,surprisingly, that Mg contents in historical cereal seeds have markedly declined over time, and two thirds of people surveyed in developed countries received less than their minimum daily Mg requirement. Thus, the mechanisms of response to MGD and ways to increase Mg contents in plants are two urgent practical problems. In this review, we discuss several aspects of MGD in plants, including phenotypic and physiological changes, cell Mg2+homeostasis control by Mg2+transporters, MGD signaling, interactions between Mg2+and other ions, and roles of Mg2+in plant secondary metabolism. | 2 | Whole-Genome Resequencing of a Worldwide Collection of Rapeseed Accessions Reveals the Genetic Basis of Ecotype Divergence显示文摘Rapeseed (Brassica napus),an important oilseed crop,has adapted to diverse climate zones and latitudes by forming three main ecotype groups,namely winter,semiwinter,and spring types. However,genetic variations underlying the divergence of these ecotypes are largely unknown. Here,we report the global pattern of genetic polymorphisms in rapeseed determined by resequencing a worldwide collection of 991 germplasm accessions.A total of 5.56 and 5.53 million singlenucleotide polymorphisms (SNPs)as Well as 1.86 and 1.92 million InDels were identified by mapping reads to the reference genomes of 'Darmor-bzh'and 'Tapidor,'respectively.We generated a map of allelic drift paths that shows splits and mixtures of the main populations,and revealed an asymmetric evolution of the two subgenomes of B.napus by calculating the genetic diversity and linkage disequilibrium parameters.Selective-sweep analysis revealed genetic changes in genes orthologous to those regulating various aspects of plant development and response to stresses.A genome-wide association study identified SNPs in the promoter regions of FLOWERING LOCUS T and FLOWERING LOCUS C orthologs that corresponded to the different rapeseed ecotype groups. | 3 | Detection of Tocopherol in Oilseed Rape (Brassica napus L.) Using Gas Chromatography with Flame Ionization Detector显示文摘The variation among Chinese genotypes of Brassica napus L.for seed tocopherols content and their analysis using gas chromatography has not been comprehensively reported till to date.In the present study,the tocopherol contents of four Chinese genotypes of Brassica napus L.,namely,Gaoyou 605,Zhejiang 619,Zheshuang 758,and Zheshuang 72,were evaluated using three modified sample preparation protocols(P1,P2,and P3) for tocopherol extraction.These methods were distinguished as follows.Protocol one(P1) included the evaporation of solvent after extraction without silylation.Protocol two(P2) followed the direct supernatant collection after overnight extraction without drying and silylation.Protocol three(P3) included trimethylsilylation with N,O-bis(trimethylsilyl) trifluoroacetamide.Genotypic comparison of tocopherol and its isoforms revealed that Gaoyou 605 was dominant over the other genotypes with(140.5±10.5),(316.2± 9.2),and(559.1±24.3) μg g-1 of seed meal α-,γ-,and total(T-) tocopherol,respectively,and a 0.44±0.04 α-to γ-tocopherol ratio.The comparison of the sample preparation protocols,on the other hand,suggests that P3 is the most suitable method for the tocopherol extraction from Brassica oilseeds and for the analysis of tocopherols using gas chromatography flame ionization detector(GC-FID).Trimethylsilylation is the key step differentiating P3 from P1 and P2.Variations detected in tocopherol contents among the Chinese rapeseed(B.napus) genotypes signify the need to quantify a wide range of rapeseed germplasm for seed tocopherol dynamics in short and crop improvement in long. | Nazim Hussain Zahra Jabeen LI Yuan-long CHEN Ming-xun LI Zhi-lan GUO Wan-li Imran Haider Shamsi CHEN Xiao-yang JIANG Li-xi | 2013 | Journal of Integrative Agriculture2013,12,5: | 6 |
| 4 | Uncertainty quantification of stability and damage detection parameters of coupled hydrodynamic-ground motion in concrete gravity dams显示文摘In this paper, models of the global system of the Koyna dam have been created using ABAQUS software considering the dam-reservoir-fbundation interaction. Non coupled models and the coupled models were compared regarding the horizontal displacement of the dam crest and the differential settlement of the dam base in clay foundation. Meta models were constructed and uncertainty quantification process was adopted by the support of Sobol's sensitivity in dices considering five un certain parameters by exploiting Box-Behnken experimental method. The non coupled models results determined overestimated predicted stability and damage detection in the dam. The rational effects of the reservoir height were very sensitive in the variation of the horizontal displacement of the dam crest with a small interaction effect with the beta viscous damping coefficient of the clay fbundation. The modulus of elasticity of the clay foundation was the decisive parameter regarding the variation of the differential settlement of the dam base. The XFEM approach has been used for damage detection in relation with both minimum and maximum values of each uncertain parameter. | 5 | Developing Wheat for Improved Yield and Adaptation Under a Changing Climate: Optimization of a Few Key Genes显示文摘世界上雨养无灌溉条件下种植的小麦常常受到干旱的影响。针对未来气候的模拟预测显示,除非采用适当的品种,否则温度上升和降雨模式的联合作用将加剧这种干旱情况,并可能造成小麦大幅度减产。由于物候基因的多样性,小麦具有广泛的适应性。小麦物候学提供了一个根据目标环境中可用水改变作物的发育阶段从而提高抗旱性。本文综述了春化(Vrn)、光周期(Ppd)和矮秆(Rht)基因等小麦物候学研究的最新进展。在不同气候条件下Vrn和Ppd基因的等位基因、单倍型和拷贝数有不同的反应,因而不仅可以改变发育阶段,而且可以改良产量。与模式植物拟南芥(Arabidopsis thaliana)相比,还有更多尚未发现的小麦物候基因,在目标环境中量化其效应将有助于通过育种提高小麦的耐旱性。因此,通过配置适当的物候基因、Rht基因和其他耐旱性相关的重要生理性状的组合,可以在水资源有限的环境中使产量最大化。 | M.A.N. | 9 | Biofilter:a promising tool for mitigating methane emission from manure storage显示文摘Liquid manure storage may contribute to methane(CH4) emission and this emission can be greatly reduced if appropriate management practices are applied. Biofiltration has been used in other fields for mitigating greenhouse gas(GHG) emission(e.g.,landfill) and shown promise for mitigation CH4 emission from liquid manure storage. It has been reported that biofilter was capable of reducing 80% of CH4 emissions from manure storage. The CH4 removal efficiency is influenced by many factors,including CH4 and O2 concentrations,temperature,moisture,composition of the filter bed,nutrient,and empty bed residency time(EBRT) . Biological conversion of methane of a biofilter is a slow process due to the low water solubility of methane. The residence times(EBRT) between 5 min and 5 h have been used,whereas a typical EBRT of 25 s is used for common biofilter applications. Temperature at which methanotrophic bacteria are active ranges from 10oC to 45oC. The maximum activity is found at around 30oC. The optimal filter bed water content depends on both the gas flow rate and the type of filter bed(soil,compost,etc.) and ranges from 30%-70% of the water holding capacity. Compost is the best material for filter bed. The optimal pH for methanotrophic bacteria is neutral to slightly acidic. Copper and nitrogen compounds especially nitrate are important nutrients to methanotrophic bacteria but their optimal concentrations have not been founded.
