| 8 | Structure and mechanics of native and decellularized porcine cranial dura mater显示文摘The dura mater is the outermost layer of meninges and consists of a dense elastic membrane that keeps cere-brospinal fluid inside the cavity. In most cranial surgical interventions, the dura mater is incised and needs to be repaired with a graft replacement. We assessed decellularized porcine dura mater as a novel graft material by quantifying the mechanical and structural properties of the dura membrane.Porcine dura mater was decellular-ized using the Sodium Dodecyl Sulfate(SDS)technique and subjected to uniaxial tensile testing, micro indentation testing, histological analysis, and Transmission Electron Microscopy(TEM).For native dura,we found the tensile modulus in the linear region(15%-25%strain)to be 19.31±1.23 MPa,with an initial tensile modulus(0%-3.5%strain range)of 451±0.30 kPa,and the failure stress as 4.61±1.50 MPa at 35%strain.For decellularized dura, the tensile modulus in the linear region was 10.81±0.88 MPa,the initial tensile modulus was 226±22 kPa,and the failure stress was 4.55±1.05 MPa at 55%strain.The effective compressive modulus was 7 to 19 kPa and 19-57 kPa for the native dura and the decellularized dura, respectively. Our histological and TEM observations showed that the orientation of fibers within the dura was maintained after decellularization. In short, our study demonstrated that decellularized porcine dura was able to maintain its overall morphological/structural integrity and preserve the native dura’s mechanical behavior, which provides a solid foundation for its use as a functional grafting material. | Ashma Sharma Jun Liao Lakiesha N.Williams | 2023 | Engineered Regeneration2023,4,2: | 0 |
| 9 | Structural and biomechanical characterizations of acellular porcine mitral valve scaffolds:anterior leaflets, posterior leaflets, and chordae tendineae显示文摘Mitral valve(MV)tissue engineering is still in its early stage, and one major challenge in MV tissue engineering is to identify appropriate scaffold materials. With the potential of acellular MV scaffolds being demonstrated recently,it is important to have a full understanding of the biomechanics of the native MV components and their acellular scaffolds. In this study,we have successfully characterized the structural and mechanical properties of porcine MV components, including anterior leaflet(AL),posterior leaflet(PL),strut chordae, and basal chordae, before and after decellularization.Quantitative DNA assay showed more than 90%reduction in DNA content, and Griffonia simplicifolia(GS)lectin immunohistochemistry confirmed the complete lack of porcine-Gal antigen in the acellular MV components. In the acellular AL and PL,the atrialis, spongiosa, and fibrosa trilayered structure, along with its ECM constitutes,i.e.,collagen fibers, elastin fibers, and portion of GAGs,were preserved.Never-theless,the ECM of both AL and PL experienced a certain degree of disruption, exhibiting a less dense, porous ECM morphology. The overall anatomical morphology of the strut and basal chordae were also maintained af-ter decellularization, with longitudinal morphology experiencing minimum disruption, but the cross-sectional morphology exhibiting evenly-distributed porous structure. In the acellular AL and PL,the nonlinear anisotropic biaxial mechanical behavior was overall preserved;however, uniaxial tensile tests showed that the removal of cellular content and the disruption of structural ECM did result in small decreases in maximum tensile modulus, tissue extensibility, failure stress, and failure strain for both MV leaflets and chordae. | Bo Wang Leslie N.Sierad Jeremy J.Mercuri Agneta Simionescu Dan T.Simionescu Lakiesha N.Williams Ryan Vela Pietro Bajona Matthias Peltz Sharan Ramaswamy Yi Hong Jun Liao | 2022 | Engineered Regeneration2022,3,4: | 0 |
| 10 | Burn recidivism:a 10-year retrospective study characterizing patients with repeated burn injuries at a large tertiary referral burn center in the United States显示文摘Background:Psychiatric and substance use disorders are common among trauma and burn patients and are known risk factors for repeat episodes of trauma, known as trauma recidivism. The epidemiology of burn recidivism, specifically, has not been described. This study aimed to characterize cases of burn recidivism at a large US tertiary care burn center and compare burn recidivists(RCs)with non-recidivists(NRCs).Methods:A 10-year retrospective descriptive cohort study of adult burn patients admitted to the North Carolina Jaycee Burn Center was conducted using data from an electronic burn registry and the medical record.Continuous variables were reported using medians and interquartile ranges(IQR).Chi-square and Wilcoxon-Mann-Whitney tests were used to compare demographic, burn, and hospitalization characteristics between NRCs and RCs.Results:A total of 7134 burn patients were admitted, among which 51(0.7%)were RCs and accounted for 129(1.8%)admissions.Of the 51 RCs,37 had two burn injuries each, totaling 74 admissions as a group, while the remaining 14 RCs had between three and eight burn injuries each, totaling 55 admissions as a group.Compared to NRCs,RCs were younger(median age 36 years vs.42 years,p=0.02)and more likely to be white(75%vs.60%,p=0.03),uninsured(45%vs.30%,p=0.02),have chemical burns(16%vs.5%,p<0.0001),and have burns that were≤10%total body surface area(89%vs.76%,p=0.001).The mortality rate for RCs vs.NRCs did not differ(0%vs.1.2%,p=0.41).Psychiatric and substance use disorders were approximately five times greater among RCs compared to NRCs(75%vs.15%,p<0.001).Median total hospital charges per patient were nearly three times higher for RCs vs.NRCs($85,736 vs.$32,023,p<0.0001).Conclusions:Distinct from trauma recidivism, burn recidivism is not associated with more severe injury or increased mortality.Similar to trauma recidivists, but to a greater extent, burn RCs have high rates of comorbid psychiatric and medical conditions that contribute to increased health care utilization and costs.Studies involving larger samples from multiple centers can further clarify whether these findings are generalizable to national burn and trauma populations. | Sarah L.Laughon Bradley N.Gaynes Lori P.Chrisco Samuel W.Jones Felicia N.Williams Bruce A.Cairns Gary J.Gala | 2019 | Burns & Trauma2019,7,1: | 0 |
