1Antigenic variation of the human influenza A(H3N2) virus during the 2014–2015 winter season显示文摘The human influenza A(H3N2) virus dominated the 2014–2015 winter season in many countries and caused massive morbidity and mortality because of its antigenic variation. So far, very little is known about the antigenic patterns of the recent H3N2 virus. By systematically mapping the antigenic relationships of H3N2 strains isolat ed since 2010, we discov ered that two groups with obvious antigenic divergence, named SW13(A/Switzerland/9715293/2013-like strains) and HK14(A/Hong Kong/5738/2014-like strains), co-circulated during the 2014–2015 winter season. HK14 group co-circulated with SW13 in Europe and the United States during this season, while there were few strains of HK14 in China's Mainland, where SW13 has dominated since 2012. Furthermore, we found that substitutions near the receptor-binding site on hemagglutinin played an important role in the antigenic variation of both the groups. 2Clinical usefulness of ankle brachial index and brachial-ankle pulse wave velocity in patients with ischemic stroke显示文摘Ankle brachial index(ABI) and brachial-ankle pulse wave velocity(baPWV) are widely used noninvasive modalities to evaluate atherosclerosis.Recently,evidence has increased supporting the use of ABI and baPWV as markers of cerebrovascular disease.This study sought to examine the relationship between ABI and baPWV with ischemic stroke.This study also aimed to determine which pathogenic mechanism,large artery disease(LAD) or small vessel disease(SVD),is related to ABI or baPWV.Retrospectively,121 patients with ischemic stroke and 38 subjects with no obvious ischemic stroke history were recruited.First,ABI and baPWV were compared between the groups.Then,within the stroke group,the relevance of ABI and baPWV with regard to SVD and LAD,which were classified by brain magnetic resonance image(MRI) and magnetic resonance angiography(MRA) or computed tomography angiography(CTA) findings,was assessed.The baPWV was higher in the stroke group than non-stroke group(1,944.18±416.6 cm/s vs.1,749.76±669.6 cm/s,P< 0.01).Regarding LAD,we found that mean ABI value was lower in the group with extracranial large artery stenosis(P< 0.01),and there was an inverse linear correlation between ABI and the grade of extracranial large artery stenosis(P<0.01).For SVD,there was a significant correlation between SVD and baPWV(2,057.6±456.57 cm/s in the SVD(+) group vs.1,491 ±271.62 cm/s in the SVD(-) group;P < 0.01).However,the grade of abnormalities detected in SVD did not correlate linearly with baPWV.These findings show that baPWV is a reliable surrogate marker of ischemic stroke.Furthermore,baPWVand ABI can be used to indicate the presence of small vessel disease and large arterial disease,respectively.Hyung-Suk Lee Hye Lim Lee Ho-seong Han Minju Yeo Ji Seon Kim Sung-Hyun Lee Sang-Soo Lee Dong-Ick Shin 2016The Journal of Biomedical Research2016,30,4:8
3Epidemiological and Virological Surveillance of Seasonal Influenza Viruses—China,2020–2021显示文摘Introduction:During the coronavirus disease 2019(COVID-19)pandemic,the circulation of seasonal influenza virus declined globally and remained below previous seasonal levels.We analyzed the results of the epidemiology,antigenic,and genetic characteristics,and antiviral susceptibilities of seasonal influenza viruses isolated from the mainland of China during October 5,2020 through September 5,2021,to better assess the risk of influenza during subsequent influenza season in 2021–2022.Methods:Positive rates of influenza virus detection during this period were based on real-time polymerase chain reaction(PCR)detection by the Chinese National Influenza Surveillance Network laboratories,and isolated viruses from influenza positive samples were submitted to the Chinese National Influenza Center.Antigenic analyses for influenza viruses were conducted using the hemagglutination inhibition assay.Next-generation sequencing was used for genetic analyses.Viruses were tested for resistance to antiviral medications using a phenotypic assay and next-generation sequencing.Results:In southern China,the influenza positivity rate was elevated especially after March 2021 and was higher than the same period the previous year with the COVID-19 pandemic.In northern China,influenza positive rate peaked at Week 18 in 2021 and has declined since then.Nearly all isolated viruses were B/Victoria lineage viruses during the study period,and 37.3%of these viruses are antigenically similar to the reference viruses representing the vaccine components for the 2020–2021 and 2021–2022 Northern Hemisphere influenza season.All seasonal influenza viruses were susceptible to neuraminidase inhibitors and endonuclease inhibitors.Conclusions:Influenza activity has gradually increased in the mainland of China in 2021,although the intensity of activity is still lower than before the COVID-19 pandemic.The diversity of circulating influenza types/subtypes decreased,with the vast majority being B/Victoria lineage viruses.The surveillance data from this study suggest that we should strengthen influenza surveillance during the upcoming traditional influenza season.It also provided evidence for vaccine recommendations and prevention and control of influenza and clinical use of antiviral drugs.Weijuan Huang Xiyan Li Minju Tan Yanhui Cheng Tao Chen Hejiang Wei Xiaoxu Zeng Yiran Xie Jia Liu Ning Xiao Lei Yang Dayan Wang 2021China CDC weekly2021,3,44:4

5Characterization of Influenza Viruses-China, 2019−2020显示文摘Introduction:Influenza surveillance is necessary for detection of emerging variants of influenza viruses and determining their epidemiological and clinical significance.Vaccination and antiviral drugs remain the most useful ways to protect against seasonal influenza and its potentially severe consequences.This study describes the epidemiology,antigenic and genetic characteristics,and antiviral susceptibilities of influenza viruses isolated from the mainland of China during the April 1,2019 through October 4,2020.Methods:All viruses analyzed were isolated and submitted by Chinese National Influenza Surveillance Network laboratories.The Chinese National Influenza Center performed antigenic analysis,sequencing,and antiviral resistance testing after propagation of the viruses.Antigenic characterizations were determined by hemagglutinin inhibition assay;next-generation sequencing was used for genetic analyses;phenotypic assay and next-generation sequencing were used for determining antiviral resistance.Results:The influenza positivity rate declined significantly starting in late January 2020 and has remained low.There was no summer influenza peak season in south China.Influenza A(H3N2)and B/Victoria lineage viruses were the dominant subtype/lineage during April 1,2019 through October 4,2020.The majority of influenza viruses were antigenically and genetically similar to reference viruses representing components of vaccines for the 2020–2021 northern hemisphere influenza season.Nearly all seasonal influenza viruses were susceptible to oseltamivir and zanamivir.Conclusions and Implications for Public Health Practice:Since the outbreak of COVID-19,the influenza positivity rate declined with implementation of strong COVID-19 control measures.The majority of circulating viruses are well matched with the current 2020–2021 northern hemisphere influenza vaccine viruses.Circulating seasonal influenza viruses were sensitive to neuraminidase inhibitors and Baloxavir marboxil.This study provided evidence for World Health Organization(WHO)recommendations for vaccine viruses,for prevention and control of influenza,and for clinical use of antiviral medications.Weijuan Huang Yanhui Cheng Tao Chen Xiyan Li Minju Tan Hejiang Wei Xiaoxu Zeng Yiran Xie Jia Liu Ning Xiao Lei Yang Dayan Wang 2020China CDC weekly2020,2,44:2

8Mutations associated with egg adaptation of influenza A(H1N1)pdm09 virus in laboratory based surveillance in China, 2009–2016显示文摘Mutations of influenza virus associated with adaptation occurring during passage in embryonated chicken eggs could result in antigenic change or reduced vaccine effectiveness.In this study,we investigated the mutations of influenza A(H1N1)pdm09 egg isolates from the Chinese National Influenza Surveillance Network between 2009 and 2016.Thirteen mutations were identified in the hemagglutinin(HA)protein from viruses passaged in eggs,in comparing to those in cells.After scanning public database,four mutations,D127E,L191I,D222G/N and Q223R in HA1,which may alter the receptor-binding specificity,were observed frequently.Although the A(H1N1)pdm09 virus has evolved in human for nearly ten years,most egg-cultured viruses acquired one or more further mutations.Using the egg isolates for influenza surveillance requires extra caution because of these selected mutations,and their impacts on antigenicity and receptor-binding property need further evaluation.Currently,most of the influenza vaccines are produced using egg isolates,particularly in China.Thus,there is an urge to promote the establishment of an alternative influenza vaccine production platform.Lei Yang Yanhui Cheng Xiang Zhao Hejiang Wei Minju Tan Xiyan Li Wenfei Zhu Weijuan Huang Wenbing Chen Jia Liu Zi Li Yuelong Shu Dayan Wang 2019Biosafety and Health2019,1,1:1

13Fragmentation stability and retention time-shift obtained by LC-MS/MS to distinguish sialylated N-glycan linkage isomers in therapeutic glycoproteins显示文摘Sialylated N-glycan isomers withα2-3 orα2-6 linkage(s)have distinctive roles in glycoproteins,but are difficult to distinguish.Wild-type(WT)and glycoengineered(mutant)therapeutic glycoproteins,cytotoxic T lymphocyte-associated antigen-4-immunoglobulin(CTLA4-Ig),were produced in Chinese hamster ovary cell lines;however,their linkage isomers have not been reported.In this study,N-glycans of CTLA4-Igs were released,labeled with procainamide,and analyzed by liquid chromatography-tandem mass spectrometry(MS/MS)to identify and quantify sialylated N-glycan linkage isomers.The linkage isomers were distinguished by comparison of 1)intensity of the N-acetylglucosamine ion to the sialic acid ion(Ln/Nn)using different fragmentation stability in MS/MS spectra and 2)retention time-shift for a selective m/z value in the extracted ion chromatogram.Each isomer was distinctively identified,and each quantity(>0.1%)was obtained relative to the total N-glycans(100%)for all observed ionization states.Twenty sialylated N-glycan isomers with onlyα2-3 linkage(s)in WT were identified,and each isomer's sum of quantities was 50.4%.Furthermore,39 sialylated N-glycan isomers(58.8%)in mono-(3 N-glycans;0.9%),bi-(18;48.3%),tri-(14;8.9%),and tetra-(4;0.7%)antennary structures of mutant were obtained,which comprised mono-(15 N-glycans;25.4%),di-(15;28.4%),tri-(8;4.8%),and tetra-(1;0.2%)sialylation,respectively,with onlyα2-3(10 N-glycans;4.8%),bothα2-3 andα2-6(14;18.4%),and onlyα2-6(15;35.6%)linkage(s).These results are consistent with those forα2-3 neuraminidase-treated N-glycans.This study generated a novel plot of Ln/Nn versus retention time to distinguish sialylated N-glycan linkage isomers in glycoprotein.Chi Soo Park Minju Kang Ahyeon Kim Chulmin Moon Mirae Kim Jieun Kim Subin Yang Leeseul Jang Ji Yeon Jang Ha Hyung Kim 2023Journal of Pharmaceutical Analysis2023,13,3:0

15Lactobacillus salivarius HHuMin-U Activates Innate Immune Defense against Norovirus Infection through TBK1-IRF3 and NF-κB Signaling Pathways显示文摘The composition of commensal bacteria plays a critical role in controlling immune responses in the intestine.Studies have shown that specific bacterial strains may have the capacity to enhance host immune defense against gastrointestinal viral infections.While norovirus is known to be the most common cause of gastroenteritis,leading to an estimated 200,000 deaths every year,identification of bacterial strains with protective effects against norovirus infection remains elusive.Here,we discovered Lactobacillus salivarius HHuMin-U(HHuMin-U)as a potent antiviral strain against norovirus infection.HHuMin-U significantly suppressed murine norovirus replication and lowered viral RNA titers in macrophages.The transcriptome sequencing(RNA sequencing)analysis revealed that HHuMin-U markedly enhanced the expression level of antiviral interferon-stimulated genes compared to mock treatment.HHuMin-U treatment dose-dependently induced type I interferons(IFN-αand IFN-β)and tumor necrosis factor-αproduction in mouse and human macrophages,promoting antiviral innate responses against norovirus infection.Investigation on the molecular mechanism demonstrated that HHuMin-U can activate nuclear factorκB and TANK-binding kinase 1(TBK1)–interferon regulatory factor 3 signaling pathways,leading to the phosphorylation of signal transducer and activator of transcription 1 and signal transducer and activator of transcription 2,the key mediators of interferon-stimulated genes.Finally,oral administration of HHuMin-U increased IFN-βlevels in the ileum of mice and altered the gut microbiome profile.These results suggest the species/strain-specific importance of gut microbial composition for antiviral immune responses and the potential use of HHuMin-U as a probiotic agent.Da Hyun Kim Minju Jeong Jae Hwan Kim Joe Eun Son John JYLee Sang-jun Park Juyeon Lee Minwoo Kim Jong-Won Oh Myeong Soo Park Sanguine Byun 2023Research2023,,3:0
16Current treatment for Wilms tumor:COG and SIOP standards显示文摘background Wilms tumor(WT)is the most common renal malignant tumor in children.It occurs primarily at preschool age.The purpose of this review is to present current standards of diagnosis and treatment of WT around the world.Data sources All the recent literature on diagnosis and treatment of WT were searched and reviewed.results Most cases with WT are sporadic.The current survival in patients with WT is high(90%).Involvement of mutidisciplinary collaborative groups in the diagnosis and treatment of WT.National Wilms Tumor Study Group(NWTSG)/Children’s Oncology Group(COG)and The International Society of Paediatric Oncology(SIOP)are two major guidelines used for the current management of WT worldwide.The major difference exists in the two guidelines is the timing of surgery:SIOP recommends using preoperative chemotherapy,and NWTSG/COG prefers using primary surgery before any adjuvant treatments.Conclusions Most patients with WT have good overall survival outcomes.Further studies should be highlighted on how to use chemotherapy and radiotherapy under more accurate risk-stratified strategies.Surgeons must be more focusing on how to maximize preoperative and postoperative treatment possibilities for achieving optimal results of patients with WT.Jinhu Wang Minju Li Daxing Tang Weizhong Gu Junqing Mao Qiang Shu 2019World Journal of Pediatric Surgery2019,2,3:0
17Different Starting Dominant Strain of Seasonal Influenza in China and Other Neighboring Asian Countries in 2019−2020 Winter Season显示文摘Introduction:Seasonal influenza is a prevalent and highly contagious acute respiratory disease that causes a major global disease burden.New strains of influenza viruses also have the potential to cause an influenza pandemic.Due to the high variability and the consequent uncertainty about virus strains,the prevention and control of influenza are faced with many challenges.Surveillance is considered a key strategy to lead to the prevention and control of influenza,and influenza is one of first infectious diseases to be monitored globally.Methods:The Chinese National Influenza Surveillance Network conducts routine influenza surveillance and includes 410 network laboratories and 554 sentinel hospitals based on the requirements of National Influenza Surveillance Guidelines.China and other countries submit epidemiological and virological data to the WHO FluMart database,which can be viewed online.Results:Seasonal influenza H3N2 virus was the dominant subtype in China when entering the 2019–2020 winter influenza season,while neighboring countries such as Japan and Republic of Korea reported the dominance of H1N1pdm09 and Mongolia and Russia reported a higher proportion of type B virus than type A virus.Conclusions and Implications for Public Health Practice:Asian countries entered the 2019–2020 winter influenza season with different type/subtypes of influenza virus dominant in different areas.Influenza surveillance needs to be strengthened to closely monitor changes in the antigenicity and genetic characteristics of emerging viruses.Jing Yang Weijuan Huang Yanhui Cheng Tao Chen Minju Tan Qian Guo Hejiang Wei Fei Zhu Tomoko Kuwahara Xiyan Li Xiang Zhao Hideki Hasegawa George F.Gao Dayan Wang 2020China CDC weekly2020,2,4:0
