1A novel peptide, selected from phage display library of random peptides, can efficiently target into human breast cancer cell显示文摘To develop a targeting vector for breast cancer biotherapy, MDA-MB-231 cell, a human breast cancer cell line, was co-cultured with pC89 (9 aa) phage display library of random peptides. In multiple inde- pendent peptide-presenting phage screening trials, subtilisin was used as a protease to inactivate ex- tra-cellular phages. The internalized phages were collected by cell lysising and amplified in E. coli XLI-Blue. Through five rounds of selection, the peptide-presenting phages which could be internalized in MDA-MB-231 cells were isolated. A comparison was made between internalization capacities of pep- tide-presenting phages isolated from MDA-MB-231 cells and RGD-integrin binding phage by cocultur- ing them with other human tumor cell lines and normal cells. The nucleotide sequences of isolated peptide-presenting phages were then determined by DNA sequencing. To uncover whether phage coat protein or amino acid order was required for the character of the peptide to MDA-MB-231 cells, three peptides were synthesized. They are CASPSGALRSC, ASPSGALRS and CGVIFDHSVPC (the shifted sequence of CASPSGALRSC), and after coculturing them with different cell lines, their targeting ca- pacities to MDA-MB-231 cells were detected. These data suggested that the internalization process was highly selective, and capable of capturing a specific peptide from parent peptide variants. Moreover, the targeting internalization event of peptides was an amino acid sequence dependent manner. 2Hydrogen peroxide modulates abscisic acid signaling in root growth and development in Arabidopsis显示文摘Exogenous abscisic acid(ABA)can inhibit root growth and promote formation of more root hairs in the root tip of Arabidopsis.However,the molecular mechanisms that underlie root ABA signaling are largely unknown.We report here that hydrogen peroxide(H2O2)reduces the root growth of wild type, and the phenotype of H2O2 on the root growth is similar to ABA response.Meanwhile ABA-induced changes in the morphology of root system can be partly reversed by ascorbic acid in wild type and abolished in NADPH oxidase defective mutant atrbohF and atrbohC.Further,ABA can induce H2O2 accumulation in the root cells and enhance transcription level of OXI1,which is necessary for many more AOS-dependent processes such as root hair growth in Arabidopsis.Our results suggest that H2O2 as an important signal molecule is required for the ABA-regulated root growth and development in Arabidopsis.BAI Ling ZHOU Yun ZHANG XiaoRan SONG ChunPeng Cao MingQing 2007Chinese Science Bulletin2007,52,8:12

4Three independent parameters to describe conventional triaxial compressive strength of intact rocks显示文摘The strengths of 12 rocks cited from literatures increase in a nonlinear way with increasing confining pressure against the Coulomb criterion. The criteria with power forms like the generalized Hoek-Brown criterion are not available for describing the strength properties in the whole test range for Indiana limestone, Yamaguchi marble and Vosges sandstone, of which the differential stresses are approximately constant at high confining pressures. The exponential criterion with three parameters fits the test data of those 12 rocks well with a low misfit. 5A combination extract of Renshen(Panax Ginseng),Yinyanghuo(Herba Epimedii Brevicornus),Yuanzhi(Radix Palygalae) and Jianghuang(Rhizoma Curcumae Longae) decreases glycogen synthase kinase 3β expression in brain cortex of APPV717I transgenic mice显示文摘OBJECTIVE:To investigate the neuroprotective mechanism of combination extract of Renshen(Panax Ginseng),Yinyanghuo(Herba Epimedii Brevicornus),Yuanzhi(Radix Palygalae) and Jianghuang(Rhizoma Curcumae Longae)(GEPT) in treating Alzheimer's disease on the target of glycogen synthase kinase 3β(GSK-3β).METHODS:Three-month-old APPV717I transgenic mice were randomly divided into ten groups(n=12 per group) and intragastrically administrated vehicle or medicines:APP group was given 0.5% CMC,donepezil group was given donepezil(APP + D group)(0.92 mg/kg-1·day-1),and GEPT groups were given small dose of GEPT(APP+Gs group)(0.075 g/kg-1·day-1),medium dose(APP+Gm group)(0.15 g/kg-1·day-1),and large dose(APP+Gl group)(0.30 g/kg-1·day-1) for 4 or 8 months,respectively.Three-month-old C57BL/6J mice as vehicle controls(n=12) were given 0.5% CMC for 4 or 8 months as well.The GSK-3β expression in the cortex of 7-and 11-month-old APPV717I transgenic mice with and without GEPT or donepezil treatment and normal C57BL/6J mice were measured via Western blotting and Immunohistochemistry.RESULTS:Immunohistochemistry analysis showed significant increase of GSK-3β in the cerebral cortex of 7-month-old APP group(compare to control group P=0.003),while the GSK-3β expression of donepezil or GEPT group were all significantly decreased(Donepezil vs APP:P=0.041;Gl vs APP:P= 0.049;Gm vs APP:P=0.029;Gh vs APP:P=0.036).Western blot analysis showed similar results.The densitometric measures of GSK-3β in APP mice increased significantly as compared with the control group(P=0.008).And the GSK-3β expression in donepezil and GEPT groups were all decreased.There was significant difference between Gh group or donepezil group and the control group(P=0.05).Similar findings were shown in the 11-month-old mice in each group,except for greater decrease of GSK-3β in the GEPT group.CONCLUSION:GEPT can effectively decrease the level of GSK-3β expression in the brain cortex of APPV717I transgenic mice,and such effect is more significant in 11-month-old mice.This partially explains the neuroprotecting mechanism of GEPT in preventing and treating of AD.Jing Shi Jinzhou Tian Xuekai Zhang Chuiyou Zeng Mingqing Wei Pengwen Wang Yongyan Wang 2013Journal of Traditional Chinese Medicine2013,33,2:6
6Effect of rice-rice-rape rotation on physicochemical property and bacterial community of rhizosphere soil显示文摘Through collecting rhizosphere soil sample from a 30-year long-term fixed location test site that use“rice-ricerape”crop rotation(RRR)and“rice-rice-fallow”continuous cropping systems(RRF),this paper investigated effects of long-term crop rotation on physicochemical property and bacterial community of rhizosphere soil.Results showed that total nitrogen(TN),total phosphorus(TP)and available potassium(AK)contents in rhizosphere soil under long-term RRR were decreased by 28.09%,15.69%and 6.25%respectively.Alkali-hydrolyzable nitrogen(AN)and available phosphorus(AP)contents were 10.59%and 13.25%higher than those of soil in RRF respectively.Three soil samples collected during different periods also showed that RRR resulted in a lower rhizosphere soil pH than RRF.Clone library analysis revealed that significant difference in rhizosphere soil bacterial community was observed between RRR and RRF continuous cropping.Abundance ofα-Proteobacteria,β-Proteobacteria andγ-Proteobacteria were higher in rhizosphere soil of RRR compared to RRF.pH of rhizosphere soil was significantly correlated with Acidobacteria level,while total organic carbon(TOC)content was significantly correlated with Proteobacteria level.Long-term RRR enhanced conversion of N and P in rhizosphere soil,increased bio-availability to crop,and promoted diversity of soil bacterial community.Bacterial diversity in RRR could be ecological significance in maintaining soil fertility and functionality.Licheng Zhang Juan Li Mingqing Zhang 2020Oil Crop Science2020,5,3:4
7Strength and damage of marble in ductile failure显示文摘To study the effects of loading paths and stress states on rock strength and deformation, marble specimens were axially compressed to various displacements under a confining pressure (CP) firstly, and then the damaged specimens were recompressed under another CP. The bearing capacity of a marble specimen depends merely on CP at the stage of ductile deformation, and it has no relation with the loading history when CP keeps constant or increases. However, the damaged specimen turns into brittle when it is recompressed uniaxially or at a lower CP, and the Young's modulus and strength are lower than those of a dense specimen. The increasing ratio of triaxial strength to CP has a close relation with the areas of fissures in the damaged specimens but not the internal friction angle. Material strength and bearing capacity are two different conceptions for rocks. 9Prevalence and trends of aminoglycoside resistance in Shigella worldwide, 1999-2010显示文摘Shigellosis causes diarrheal disease in humans in both developed and developing countries, and multi-drug resistance in Shigella is an emerging problem. Understanding changing resistance patterns is important in determining appropriate antibiotic treatments. This meta-analysis systematically evaluated aminoglycoside resist-ance in Shigella. A systematic review was constructed based on MEDLINE and EMBASE databases. Randomeffect models or fixed-effect models were used based on P value considering the possibility of heterogeneity between studies for meta-analysis. Data manipulation and statistical analyses were performed using software STATA 11.0. By means of meta-analysis, we found a lower resistance to three kinds of aminoglycosides in the Europe-America areas during the 12 year study period than that of the Asia-Africa areas. Kanamycin resistance was observed to be the most common drug resistance among Shigella isolates with a prevalence of 6.88% (95%CI: 6.36%-7.43%). Comparison of data from Europe-America and Asia-Africa areas revealed that Shigella flexneri resistance was greater than the resistance calculated for Shigella sonnei. Importantly,Shigella sonnei has played a significant role in aminoglycoside-resistance in recent years. Similarly, data showed that resistance to these drugs in children was higher than the corresponding data of adults. In conclusion, aminoglycoside-resistant Shigella is not an unusual phenomenon worldwide. Distribution in Shigella resistance differs sharply based on geographic areas, periods of time and subtypes. 10Abnormal regulation of chemokine TECK and its receptor CCR9 in the endometriotic milieu is involved in pathogenesis of endometriosis by way of enhancing invasiveness of endometrial stromal cells显示文摘The chemokine thymus-expressed chemokine(TECK),which regulates T-cell development and tissue-specific homing,has been identified as a potential contributor to the pathogenesis and progression of endometriosis.Dioxin(2,3,7,8-tetrachlorodibenzo-p-dioxin,TCDD),an air pollutant,and estrogen also appear to be involved in endometriosis.Both endometrial stromal cells(ESCs)and the combination of 17b-estradiol and TCDD increase the secretion of TECK in the endometriosis-associated cells and promote the invasiveness of ESCs by increasing expression of matrix metalloproteinase(MMP)-2 and MMP-9.Anti-TECK neutralizing antibodies can effectively inhibit the invasiveness of ESCs and the expression of MMP-2 and MMP-9 in the cells.Interestingly,the expression of chemokine C receptor 9(CCR9)and its ligand TECK increases significantly in the endometriotic milieu of patients with endometriosis.Therefore,the over-expressed TECK interacts with CCR9 on the ESCs in the endometriotic milieu,which may contribute to the onset and progression of endometriosis.Yun Wang Jing Yu Xuezhen Luo Xiaoqiu Wang Mingqing Li Ling Wang Dajin Li 2010Cellular & Molecular Immunology2010,7,1:3
