1Antibacterial Mechanisms of Berberine and Reasons for Little Resistance of Bacteria显示文摘Objective To study the antibacterial mechanisms of berberine and try to understand the reasons why bacteria cells difficultly resisted to it.Methods Detecting the minimal inhibitory concentration(MIC) of bacterial cultures incubated under sub-MIC concentration of berberine,Huanglian,and Neomycin for more than 200 generations,in order to analyze the bacteria resistance.Detecting the binding kinetics of berberine to DNA,RNA,and proteins.Observing the changes in bacterial cell surface structure with scanning electron microscopy.Detecting the Ca2 +and K +released from berberine-treated bacterial cells with atomic absorption spectrum.Detection the absorption of methyl-3H-thymine (3 H-dT),3 H-uridine(3 H-U),and 3 H-tyrosine(3 H-Tyr)into berberine-treated bacterial cells.Results MICs of bacterial cultures,growing more than 200 generations in MH medium with 1/2 MIC of berberine(BA200) or Huanglian(HA200),did not increase compared to the control,while remarkably increased in MH medium with 1/2 MIC of Neomycin (NA200).In addition,from the culture NA200 it was easy to isolate resistant mutant strains which could grow in MH medium with more than four times MIC Neomycin,but from the culture BA200 and HA200 it was difficult to isolate berberine or Huanglian mutant strains could grow in MH medium with more than four times MIC berberine or Huanglian.The binding kinetics of berberine to DNA,RNA,and proteins illustrated that berberine could easily and tightly bind to DNA and RNA,and hardly dis-bind from DNA-and RNA-berberine complexes.Berberine could easily bind to protein too,but also easily dis-bind from berberine-protein complex.The bacterial cells treated with berberine sharply decreased the absorption of 3 H-dT,3 H-U,and 3 H-Tyr,as the radioactive precursors of DNA,RNA,and protein biosynthesis.Berberine could damage bacterial cell surface structure,especially for Gram-negative bacteria.Ca2 +and K + released from berberine-treated cells increased significantly compared to the control.Conclusion All of above results indicate that bacterial cells could not easily become resistant mutants to berberine.The mechanisms for the bactericidal effect of berberine include:inhibiting DNA duplication,RNA transcription,and protein biosynthesis;influencing or inhibiting enzyme activities;destructing the bacterial cell surface structure and resulting in Ca2 +and K +released from cells.All of the berberine bactericidal mechanisms are the most essential physiological functions for a live cell,if influenced any one such function,the mutation would be lethal mutation,so that it is difficult to get berberine resistant cells.The results in this paper also prefigure that berberine and its related Chinese medicines would provide a feasible way to control antibiotic resistance problem.JIN Jian-ling,HUA Guo-qiang,MENG Zhen,GAO Pei-ji State Key Laboratory of Microbial Technology,School of Life Sciences,Shandong University,Jinan 250100,China 2011Chinese Herbal Medicines2011,3,1:11
2A novel function for the cellulose binding module of cellobiohydrolase I显示文摘A homogeneous cellulose-binding module(CBM)of cellobiohydrolase I(CBHI)from Trichoderma pseudokoningii S-38 was obtained by the limited proteolysis with papain and a series of chromatographs filtration.Analysis of FT-IR spectra demonstrated that the structural changes result from a weakening and splitting of the hydrogen bond network in cellulose by the action of CBMCBHI at 40℃for 24 h.The results of molecular dynamic simulations are consistent with the experimental conclusions, and provide a nanoscopic view of the mechanism that strong and medium H-bonds decreased dramatically when CBM was bound to the cellulose surface.The function of CBMCBHI is not only limited to locating intact CBHI in close proximity with cellulose fibrils,but also is involved in the structural disruption at the fibre surface.The present studies provided considerable evidence for the model of the intramolecular synergy between the catalytic domain and their CBMs.WANG LuShan 1,2 ,ZHANG YuZhong 1 &GAO PeiJi 1 1State Key Laboratory of Microbial Technology,Shandong University,Jinan 250100,China 2Beijing Laboratory of Nanoscale Physics&Devices,Chinese Academy of Sciences,Beijing 100080,China 2008Science China(Life Sciences)2008,51,7:10
3Function of the iron-binding chelator produced by Coriolus versicolor in lignin biodegradation显示文摘An ultrafiltered low-molecular-weight preparation of chelating compounds was isolated from a wood-containing culture of the white-rot basidiomycete Coriolus versicolor. This preparation could chelate Fe3+ and reduce Fe3+ to Fe2+, demonstrating that the substance may serve as a ferric chelator, oxygen-reducing agent, and redox-cycling molecule, which would include functioning as the electron transport carrier in Fenton reaction. Lignin was treated with the iron-binding chelator and the changes in structure were investigated by 1H-NMR, 13C-NMR, difference spectrum caused by ionization under alkaline conditions and nitrobenzene oxidation. The results indicated that the iron-binding chelator could destroy the β-O-4 bonds in etherified lignin units and insert phenolic hydroxyl groups. The low-molecular-weight chelator secreted by C. versicolor resulted in new phenolic substructures in the lignin polymer, making it susceptible to attack by laccase or manganese peroxidase. 4Purification and Crystallization of Flammulin,a Basic Protein with Anti-tumor Activities from Flammulina Velutipes显示文摘Flammulin, an anti-tumor protein, was purified from the aqueous extract of basidiomes of Flammulina Velutipes to electrophoretic homogeneity and crystallized by microdialysis against a polyethylene glycol- sodium phosphate buffer. 6Comparative proteomic analysis of the response in resistant and susceptible maize inbred lines to infection by Curvularia lunata显示文摘Proteins differentially expressed from maize leaves in response to the infection by Curvularia lunata strain CX-3 were identified through a high-resolution two-dimensional gel electrophoresis (2-DE) method. Two inbred lines, 78599-1 and E28, were used, respectively, as resistant and susceptible lines to CX-3 infection. Proteins were extracted from the fourth leaves of sixor seven-leaf stage plants sampled at 24, 36, 48, 60, and 72 h after inoculation with CX-3. Twenty-seven differentially expressed protein spots resolved on the 2-DE gels were identified by MALDI-TOF MS/MS. The results showed that these proteins are associated with photosynthesis, respiration, oxidative and drought stress tolerance as well as signal transduction in maize. Among stress-related proteins, the 22 kDa drought-inducible protein, putative glutathione peroxidase (GPX), and translation initiation factor (eIF-5A) were up-regulated in the resistant inbred line and were implicated in host defense response to C. lunata infection. 7A novel approach for assessing the susceptibility of Escherichia coli to antibiotics显示文摘The dynamic growth process of Escherichia coli CVCC249 under different concentrations of antibiotics was analyzed. The results suggested that the main reason that definitive results cannot be obtained by antibiotic susceptibility testing (AST) is that the ratio of drug concentration to the population of bacteria and the combined effect of drug concentration and action time cannot be completely determined with the methods used. Based on the analysis of the growth process with a series of concentrations of gentamicin acting for a certain time, and according to the forward difference method, a novel method for AST was proposed. The net increase in turbidity of the bacterial population was used to eliminate the existing effects of resting cells, and then the recurrent coefficient for a growing sequence was used to characterize the effect of antibiotics on bacterial division, and the contour plot was used to display and analyze the combined effect of drug concentration and action time. The inhibition rate of the antibiotics can be characterized as the dynamic change in the composite function of the antibiotic concentration and action time, which indicated that the inhibition rate was dependent on the combined effect of time and concentration of antibiotics. 8Las-like quorum-sensing system negatively regulates both pyoluteorin and phenazine-1-carboxylic acid production in Pseudomonas sp. M18显示文摘A las-like quorum-sensing system in Pseudomonas sp. M18 was identified, which consisted of lasI and lasR genes encoding LuxI-LuxR type regulator. Several functions of the las system from strain M18 were investigated in this study. The chromosomal inactivation of either lasI or lasR by recombination increased the production of both pyoluteorin (Plt) and phenazine-1-carboxylic acid (PCA) by 4-5 fold and 2-3 fold over that of the wild type strain of M18, respectively. Production of both antibiotics was restored to wild-type levels after in trans complementation with the wild-type lasI or lasR gene. 9LysR family factor PltR positively regulates Pyoluteorin production in a pathway-specific manner in Pseudomonas sp. M18显示文摘The pltR gene, coding a putative LysR-type regulator, was identified upstream Plt biosynthetic gene cluster in Pseudomonas sp. M18 using bioinformatics technology. The null mutation of pltR resulted in mutant M18TRG (pltR::Gm) by recombination and its Plt (Pyoluteorin) production declined to 30% while PCA (Phenazine-1-carboxylic acid) production remained unchanged as compared with the wild-type M18 grown in King’s Medium B. After complementation, Plt production of mutant M18TRG was restored to the level in wild-type M18. Overexpression of pltR in M18 led to 13-fold enhancement of Plt produc-tion over the wild-type M18 strain. However, PCA production was unchanged under this condition. These data suggested that PltR was a positive regulator on Plt production. Plt itself, however, could not regulate expression of pltR. Expression of the plt-lacZ transcriptional fusion in mutant M18TRG de-clined obviously as compared with the wild-type M18, which further proved that PltR could regulate expression of Plt biosynthetic genes at the transcriptional level. 10Effect of physiological heterogeneity of E. coli population on antibiotic susceptivity test显示文摘According to the instantaneous growth rate (dN/dt) of E. coli CVCC249 growing in batch culture, the entire growth progress was distinguished into four phases: accelerating growth phase, constant growth phase, decelerating growth phase and declining phase, in each of which obvious variation in physiological and biochemical properties was detected, including total DNA, total protein, and MTT-dehydrogenase activity, etc., that led to difference in their antibiotic susceptivity. Antibiotic sus- ceptivity of the population sampled from each phase was tested by Concentration-killing Curve (CKC) approach following the formula N=N0/{1+exp[r?(x-BC50)]}, showing as normal distribution at the indi- vidual cell level for an internal population, in which the median bactericidal concentration BC50 repre- sents the mean level of susceptivity, while the bactericidal span BC1-99=(2lnN0)/r indicates the variation degree of the antibiotic susceptivity. Furthermore, tested by CKC approach, the antibiotic susceptivity of E. coli CVCC249 population in each physiological phase to gentamicin or enoxacin was various: susceptivity of the population in the constant growth phase and declining phase all increased com- pared with that in the accelerating growth phase for gentamicin but declined for enoxacin. The primary investigations revealed that the physiological phase should be taken into account in the context of antibiotic susceptivity and research into antimicrobial mechanism. However there are few reports concerned with this study. 11Effect of the population heterogeneity on growth behavior and its estimation显示文摘Different types of the Logistic model are constructed based on a simple assumption that the microbial populations are all composed of homogeneous members and consequently, the condition of design for the initial value of these models has to be rather limited in the case of N(t_0)=N_0. Therefore, these models cannot distinguish the dynamic behavior of the populations possessing the same N0 from heteroge-neous phases. In fact, only a certain ratio of the cells in a population is dividing at any moment during growth progress, termed as θ, and thus, ddNt not only depends on N, but also on θ. So θ is a necessary element for the condition design of the initial value. Unfortunately, this idea has long been neglected in widely used growth models. However, combining together the two factors (N0 and θ ) into the initial value often leads to the complexity in the mathematical solution. This difficulty can be overcome by using instantaneous rates (Vinst) to express growth progress. Previous studies in our laboratory sug-gested that the Vinst curve of the bacterial populations all showed a Guassian function shape and thus, the different growth phases can be reasonably distinguished. In the present study, the Gaussian dis-tribution function was transformed approximately into an analytical form (0.5x ibxYi αe=20) that can be conveniently used to evaluate the growth parameters and in this way the intrinsic growth behavior of a bacterial species growing in heterogeneous phases can be estimated. In addition, a new method has been proposed, in this case, the lag period and the double time for a bacterial population can also be reasonably evaluated. This approach proposed could thus be expected to reveal important insight of bacterial population growth. 12Cesium accumulation by bacterium Thermus sp. TibetanG7:hints for biomineralization of cesium-bearing geyserite in hot springs in Tibet,China显示文摘The bacterium Thermus sp. TibetanG7, isolated from hot springs in Tibet, China, was examined for the ability to accumulate cesium from solutions. Environmental conditions were simulated and the effects of pH, K+, Na+ and K+-regimes were then studied to determine the possible role of the bacterium in the formation of cesium-bearing geyserite around these hot springs. In despite of the inhibition of K+ and Na+, the bacterium Thermus sp. TibetanG7 revealed noticeable accumulation of cesium from solutions, with maximum accumulations of 53.49 and 40.41 μmol Cesium/g cell dry weight in Na+ and K+ inhibition experiments, respectively. The accumulation of cesium by this microorganism is rapid, with 40%―50% accumulated within the first 5 min. K+-deficient cells showed a much higher capacity of cesium accu- mulation compared with K+-sufficient cells. It is evident that the bacteria within the genus thermus play a significant role in the cesium assembly. 13Determination of Cordycepin in Cordyceps kyushuensis by Capillary Electrophoresis and its Antitumour Activity显示文摘A simple, rapid and low-cost method of determination for cordycepin in Cordyceps kyushuensis by capillary zone electrophoresis (CZE) was developed. Based on the finding that there is a high concentration of cordycepin in both natural and cultured Cordyceps kyushuensis, the in vitro antitumor activity of cordycepin and the water extracts of Cordyceps kyushuensis has been investigated.
