1miR-25 modulates triacylglycerol and lipid accumulation in goat mammary epithelial cells by repressing PGC-1beta显示文摘Background: The goat(Caprahircus) is one of the most important livestock animals. Goat milk fat is an important component in the nutritional quality of goat milk. Growing evidence points to the critical roles of microRNAs(miRNAs) in lipid metabolism.Results: Using a highly sensitive method of S-poly(T) plus for miRNAs detection, we analyze the expression patterns of 715 miRNAs in goat mammary gland tissues at different stages of lactation. We observed that miR-25 expression had an inverse relationship with milk production. Overexpression of miR-25 significantly repressed triacylglycerol synthesis and lipid droplet accumulation. To explore the regulatory mechanism of miR-25 in milk lipid metabolism,we analyzed its putative target genes with bioinformatics analysis followed by 3′-UTR assays. Peroxisome proliferative activated receptor gamma coactivator 1 beta(PGC-1 beta), a key regulator of lipogenics was identified as a direct target of miR-25 with three specific sites within its 3′-UTR. 2Characteristics and tectonic setting of volcanic rocks in Early Cretaceous Baiyingaolao Formation of Keyouzhongqi area,Inner Mongolia显示文摘The authors studied zircon LA-ICP-MS U-Pb dating,the zircon Hf isotope and geochemistry of acidic volcanic rocks in Baiyingaolao Formation of Keyouzhongqi area,Inner Mongolia,and discussed the chronology,source region and tectonic setting of the volcanic rocks in the studied area. The clear oscillatory zoning of zircons indicates a typical magmatic origin,and the results of dating show that the volcanic rocks of Baiyingaolao Formation were formed in Early Cretaceous( 121. 5 ± 1. 0 Ma). The features of major and trace elements show that the rocks are alkali-rich,poor in calcium and magnesium with enrichment in LILEs like Th,U,K and Gd and depletion in HFSEs,e. g. Nb,Ta,Sr and Ti. The fact implies that they were the products of partial melting of the crust. εHf( t) =( +6.30--+9.06) and TDM2= 600--835 Ma,suggest the magma originated from partial melting of the young crust. 3Vibration Deformation Monitoring of Offshore Wind Turbines Based on GBIR显示文摘In view of the disadvantages of vibration safety monitoring technology for offshore wind turbines,a new method is proposed to obtain deformation information of towering and dynamic targets in real-time by the ground-based interferometric ra-dar(GBIR).First,the working principle and unique advantages of the GBIR system are introduced.Second,the offshore wind turbines in Rongcheng,Shandong Province are selected as the monitoring objects for vibration safety monitoring,and the GPRI-II portable radar interferometer is used for the health diagnosis of these wind turbines.Finally,the interpretation method and key processing flow of data acquisition are described in detail.This experiment shows that the GBIR system can accurately identify the millimeter-scale vibration deformation of offshore wind turbines and can quickly obtain overall time series deformation images of the target bodies,which demonstrate the high-precision deformation monitoring ability of the GBIR technology.The accuracy meets the requirements of wind turbine vibration monitoring,and the method is an effective spatial deformation monitoring means for high-rise and dynamic targets.This study is beneficial for the further enrichment and improvement of the technical system of wind turbine vibration safety monitoring in China.It also provides data and technical support for offshore power engineering management and control,health diagnosis,and disaster prevention and mitigation.MA Deming LI Yongsheng LIU Yanxiong CAI Jianwei ZHAO Rui 2021Journal of Ocean University of China2021,20,3:2

6Petrogenesis and tectonic setting of granite porphyry in Keyouzhongqi area,Inner Mongolia显示文摘The authors studied the geochemistry,zircon U-Pb ages,Hf isotope of the granite porphyry and its petrogenesis and tectonic setting in the studied area. The zircon U-Pb dating indicates the formation of the granite porphyry is in Early Cretaceous( 125. 1 ± 1. 5 Ma). The granite porphyry has high-SiO2and alkali-rich features,which belongs to high-K calc-alkaline series rocks( A / CNK = 0. 95%--1. 25%). The analyses of trace elements show the characteristics of a swallow-shaped REE distribution pattern with enrichment in LREEs and most of LILEs and HFSEs,depletion in Ba,Sr,Nb,P and Ti,and especially strong depletion in Eu,which indicates the granite porphyry belongs to the aluminous A-type granite. Their εHf( t)range from 5. 94 to 8. 80with Hf two-stage model ages( TDM2) of 620 Ma to 803 Ma. 9Elevational Variation in Reproductive Strategy of a Widespread Lizard:High-Elevation Females Lay Fewer but Larger Eggs显示文摘Identifying how reproductive strategies such as the trade-off between clutch size versus egg mass vary with elevational gradients is essential for our understanding of life-history evolution.We studied lacertid lizards(Eremias argus)in China,from six populations at different altitudes,to assess elevational variation in reproductive strategy.We found significant between-population variation in maternal body size and clutch mass,but these variations were not explained by elevational differences.However,high-elevation females tended to produce smaller clutches of larger eggs compared with their low-elevation counterparts,demonstrating an elevational change in the trade-off between egg size and number.The egg size-number trade-off is a reproductive strategy that may favor large offspring,better enabling them to survive severe and unpredictable environments found at high elevations.Gideon Gywa DEME Xin HAO Liang MA Baojun SUN Weiguo DU 2022Asian Herpetological Research2022,13,3:0

12Upregulated adenosine 2A receptor accelerates post-infectious irritable bowel syndrome by promoting CD4+T cells’T helper 17 polarization显示文摘BACKGROUND Post-infectious irritable bowel syndrome(PI-IBS)is generally regarded as a functional disease.Several recent studies have reported the involvement of lowgrade inflammation and immunological dysfunction in PI-IBS.T helper 17(Th17)polarization occurs in IBS.Adenosine and its receptors participate in intestinal inflammation and immune regulation.AIM To investigate the role of Th17 polarization of CD4+T cells regulated by adenosine 2A receptor(A2AR)in PI-IBS.METHODS A PI-IBS model was established by infecting mice with Trichinella spiralis.The intestinal A2AR and CD4+T lymphocytes were detected by immunohistochemistry,and the inflammatory cytokines were detected by enzyme-linked immunoassay.CD4+T lymphocytes present in the animal’s spleen were separated and cultured with or without A2AR agonist and antagonist.Western blotting and real-time quantitative polymerase chain reaction were performed to determine the effect of A2AR on the cells and intestinal tissue.Cytokine production was determined.The protein and mRNA levels of A2AR associated signaling pathway molecules were also evaluated.Furthermore,A2AR agonist and antagonist were injected into the mouse model and the clinical features were observed.RESULTS The PI-IBS mouse model showed increased expression of ATP and A2AR(P<0.05),and inhibition of A2AR improved the clinical features in PI-IBS,including the abdominal withdrawal reflex and colon transportation test(P<0.05).The number of intestinal CD4+T cells and interleukin-17(IL-17)protein levels increased during PI-IBS,which was reversed by administration of the A2AR antagonist(P<0.05).CD4+T cells expressed A2AR and produced IL-17 in vitro,which was regulated by the A2AR agonist and antagonist.The A2AR antagonist increased the production of IL-17 by CD4+T cells via the Janus kinase-signal transducer and activator of transcriptionreceptor-related orphan receptorγsignaling pathway.CONCLUSION The results of the present study suggested that the upregulation of A2AR increases PI-IBS by promoting the Th17 polarization of CD4+T cells.Li-Wei Dong Zhi-Chao Ma Jiao Fu Bai-Li Huang Fu-Jin Liu Deming Sun Cheng Lan 2022World Journal of Gastroenterology2022,28,25:0
13Effects of transplantation of microencapsulated rabbit sciatic nerve on nuclear factor-kappa B expression after spinal cord injury in rats显示文摘BACKGROUND:It has been reported that nuclear factor-kappa B(NF-κB),activated after spinal cord injury in rats,plays a key role in inflammatory responses in the central nervous system. OBJECTIVE:To investigate the effects of transplantation of microencapsulated rabbit sciatic nerve on NF-κB expression and motor function after spinal cord injury in rats,and to compare the results with the transplantation of rabbit sciatic nerve alone. DESIGN,TIME AND SETTING:This completely randomized,controlled study was performed at the Department of Neurobiology,Medical College of Nanchang University between December 2007 and July 2008. MATERIALS:A rabbit anti-NF-κB P65 monoclonal antibody was made by the Santa Cruz Company,USA and a streptavidin peroxidase immunohistochemical kit was provided by the Sequoia Company,China. METHODS:Eight rabbits were used to prepare a sciatic nerve cell suspension that was divided into two parts:one stored for transplantation,and the other mixed with a 1.5%sodium alginate solution.One hundred and twenty adult Sprague Dawley rats weighing 220-250 g were randomly divided into four groups:the microencapsulated cell group(n = 36),the non-encapsulated cell group(n = 36),the saline group(n = 36) and the sham operation group(n = 12).The first three groups underwent a right hemisection injury of the spinal cord at the T_(10) level,into which was transplanted a gelatin sponge soaked with 10μL of a microencapsulated nerve tissue/cell suspension(microencapsulated cell group),a tissue/cell suspension(non-encapsulated cell group) or physiological saline(saline group).In the sham operation group the vertebrae were exposed,but the spinal cord was not injured,and no implantation was given. MAIN OUTCOME MEASURES:Pathological changes were detected using hematoxylin-eosin staining;NF-κB expression was quantified using immunohistochemical staining;motor function was assessed using the Basso,Beattie and Bresnahan(BBB) scale. RESULTS:Spinal cord injuries,such as neuronal death and inflammatory cell infiltration,were found in the microencapsulated cell group,the non-encapsulated cell group and the saline group. However,the damage in the microencapsulated cell group was milder than in the non-encapsulated cell or saline groups.NF-κB expression in the microencapsulated cell group, the non-encapsulated cell group and the saline group was increased after spinal cord injury;it reached a peak after 24 hours,gradually decreased after 3 days,and was close to normal levels after 7 days.NF-κB expression in the microencapsulated cell group was significantly lower than in the saline group and the non-encapsulated cell group(P< 0.05).With time,the motor function of the animals in each group improved to a certain extent,but did not reach normal levels.There were no significant differences in BBB scores between the different groups on post-operative day 3;however,the BBB scores for the microencapsulated cell group and the non-encapsulated cell group were significantly higher than the saline group on post-operative day 7(P < 0.05).In addition,the motor function recovered better in the microencapsulated cell group than in the non-encapsulated cell group(P< 0.05). 15Assimilation of FY-3D MWTS-Ⅱ Radiance with 3D Precipitation Detection and the Impacts on Typhoon Forecasts显示文摘Precipitation detection is an essential step in radiance assimilation because the uncertainties in precipitation would affect the radiative transfer calculation and observation errors.The traditional precipitation detection method for microwave only detects clouds and precipitation horizontally,without considering the three-dimensional distribution of clouds.Extending precipitation detection from 2D to 3D is expected to bring more useful information to the data assimilation without using the all-sky approach.In this study,the 3D precipitation detection method is adopted to assimilate Microwave Temperature Sounder-2(MWTS-Ⅱ)onboard the Fengyun-3D,which can dynamically detect the channels above precipitating clouds by considering the near-real-time cloud parameters.Cycling data assimilation and forecasting experiments for Typhoons Lekima(2019)and Mitag(2019)are carried out.Compared with the control experiment,the quantity of assimilated data with the 3D precipitation detection increases by approximately 23%.The quality of the additional MWTS-Ⅱradiance data is close to the clear-sky data.The case studies show that the average root-mean-square errors(RMSE)of prognostic variables are reduced by 1.7%in the upper troposphere,leading to an average reduction of4.53%in typhoon track forecasts.The detailed diagnoses of Typhoon Lekima(2019)further show that the additional MWTS-Ⅱradiances brought by the 3D precipitation detection facilitate portraying a more reasonable circulation situation,thus providing more precise structures.This paper preliminarily proves that 3D precipitation detection has potential added value for increasing satellite data utilization and improving typhoon forecasts.Luyao QIN Yaodeng CHEN Gang MA Fuzhong WENG Deming MENG Peng ZHANG 2023Advances in Atmospheric Sciences2023,40,5:0
