1An Overview of Autism Spectrum Disorder, Heterogeneity and Treatment Options显示文摘Since the documented observations of Kanner in1943, there has been great debate about the diagnoses, the sub-types, and the diagnostic threshold that relates to what is now known as autism spectrum disorder(ASD).Re?ecting this complicated history, there has been continual re?nement from DSM-III with ‘Infantile Autism' to the current DSM-V diagnosis. The disorder is now widely accepted as a complex, pervasive, heterogeneous condition with multiple etiologies, sub-types, and developmental trajectories. Diagnosis remains based on observation of atypical behaviors, with criteria of persistent de?cits in social communication and restricted and repetitive patterns of behavior. This review provides a broad overview of the history, prevalence, etiology, clinical presentation, and heterogeneity of ASD. Factors contributing to heterogeneity, including genetic variability, comorbidity, and gender are reviewed. 3NETosis,complement,and coagulation:a triangular relationship显示文摘NETosis is a regulated form of neutrophil cell death that contributes to the host defense against pathogens and was linked to various diseases soon after its first description in 2004.During NETosis,neutrophils release neutrophil extracellular traps(NETs),which can capture and kill bacteria and other pathogens to prevent them from spreading.Although substantial progress has been made in our understanding of NETosis,the precise mechanism underlying NETosis is still a matter of debate.Research continues to elucidate the molecular pathways involved in NETosis.In recent years,interactions with the complement and coagulation systems have become increasingly apparent.Activated complement proteins can stimulate NET formation,and NETs,in turn,can serve as a platform for complement activation.In addition,NETs can act as a scaffold for thrombus formation during coagulation.While crosstalk between the coagulation and complement systems has been previously described,NETosis appears to be a third important player in this consortium to protect the host against pathogens.This review summarizes our current knowledge on the mutual interactions between NETosis,the complement system and the coagulation system,with an emerging description of their complex triangular relationship.Cynthia M.de Bont Wilbert C.Boelens Ger J.M.Pruijn 2019Cellular & Molecular Immunology2019,16,1:22

5Contemporary diagnostics and treatment options for female stress urinary incontinence显示文摘Stress urinary incontinence is not a deadly disease,but for the large population of women suffering from it,it is a very important issue.Especially in the continuously aging population all over the world,there is more and more need for treatment of this serious medical condition.Treatment of female stress urinary incontinence exists already for ages.In the 20th century invasive treatments like Burch colposuspension and pubovaginal slings were the mainstay of surgical treatments.The introduction of the midurethral sling made the procedure less invasive and accessible for more caregivers.Luckily there are many options available and the field is developing quickly.In recent years many new medical devices have been developed,that increase the number of treatment options available and make it possible to find a suitable solution for the individual patient based on subjective and objective results and the chances of complications.This manuscript provides an introduction to the therapeutical options that are available nowadays for female stress urinary incontinence.Allert M.de Vries John P.F.A.Heesakkers 2018Asian Journal of Urology2018,5,3:12

7Metastatic prostate cancer remains incurable, why?显示文摘Metastatic prostate cancer patients present in two waysdwith already disseminated disease at the time of presentation or with disease recurrence after definitive local therapy.Androgen deprivation therapy is given as the most effective initial treatment to patients.However,after the initial response,almost all patients will eventually progress despite the low levels of testosterone.Disease at this stage is termed castration resistant prostate cancer(CRPC).Before 2010,the taxane docetaxel was the first and only life prolonging agent for metastatic CRPC(mCRPC).The last decade has witnessed robust progress in CRPC therapeutics development.Abiraterone,enzalutamide,apalutamide and sipuleucel-T have been evaluated as first-and second-line agents in mCRPC patients,while cabazitaxel was approved as a second-line treatment.Radium-223 dichloride was approved in symptomatic patients with bone metastases and no known visceral metastases pre-and post-docetaxel.However,despite significant advances,mCRPC remains a lethal disease.Both primary and acquired resistance have been observed in CRPC patients treated by these new agents.It could be solely cell intrinsic or it is possible that the clonal heterogeneity in treated tumors may result from the adaptive responses to the selective pressures within the tumor microenvironment.The aim of this review is to list current treatment agents of CRPC and summarize recent findings in therapeutic resistance mechanisms.Liang Dong Richard C.Zieren Wei Xue Theo M.de Reijke Kenneth J.Pienta 2019Asian Journal of Urology2019,6,1:9

10Multiple responses optimization in the development of a headspace gas chromatography method for the determination of residual solvents in pharmaceuticals显示文摘An efficient generic static headspace gas chromatography(HSGC) method was developed,optimized and validated for the routine determination of several residual solvents(RS) in drug substance,using a strategy with two sets of calibration.Dimethylsulfoxide(DMSO) was selected as the sample diluent and internal standards were used to minimize signal variations due to the preparative step.A gas chromatograph from Agilent Model 6890 equipped with flame ionization detector(FID) and a DB-624(30 m × 0.53 mm i.d.,3.00 μm film thickness) column was used.The inlet split ratio was 5:1.The influencing factors in the chromatographic separation of the analytes were determined through a fractional factorial experimental design.Significant variables:the initial temperature(IT),the final temperature(FT) of the oven and the carrier gas flow rate(F) were optimized using a central composite design.Response transformation and desirability function were applied to find out the optimal combination of the chromatographic variables to achieve an adequate resolution of the analytes and short analysis time.These conditions were 30 ℃ for IT,158 ℃ for FT and 1.90 mL/min for F.The method was proven to be accurate,linear in a wide range and very sensitive for the analyzed solvents through a comprehensive validation according to the ICH guidelines.Carla M.Teglia Milagros Montemurro María M.De Zan María S.Cmara 2015Journal of Pharmaceutical Analysis2015,5,5:8

12High-speed water impacts of flat plates in different ditching configuration through a Riemann--ALE SPH model显示文摘The violent water entry of flat plates is investigated using a Riemann-arbitrary Eulerian-Lagrangian(ALE) smoothed particle hydrodynamics(SPH) model. The test conditions are of interest for problems related to aircraft and helicopter emergency landing in water. Three main parameters are considered: the horizontal velocity, the approach angle(i.e., vertical to horizontal velocity ratio) and the pitch angle, a. Regarding the latter, small angles are considered in this study. As described in the theoretical work by Zhao and Faltinsen(1993), for small a a very thin, high-speed jet of water is formed, and the time-spatial gradients of the pressure field are extremely high. These test conditions are very challenging for numerical solvers. In the present study an enhanced SPH model is firstly tested on a purely vertical impact with deadrise angle α=4°. An in-depth validation against analytical solutions and experimental results is carried out, highlighting the several critical aspects of the numerical modelling of this kind of flow, especially when pressure peaks are to be captured. A discussion on the main difficulties when comparing to model scale experiments is also provided. Then, the more realistic case of a plate with both horizontal and vertical velocity components is discussed and compared to ditching experiments recently carried out at CNR-INSEAN. 19重症肌无力致病性自身抗体的人工构建显示文摘目的构建一株具有致病性的人抗乙酰胆碱受体(AchR)自身抗体,用于诱导猴实验性自身免疫性重症肌无力(EAMG)模型。方法将分离自重症肌无力(MG)患者胸腺的抗AchR抗体的抗原结合片段(Fab637)克隆至含有免疫球蛋白(IgG1)基因的载体pIgG1,构建能在哺乳类细胞表达完整IgG1类抗AchR抗体(IgG1-637)的重组表达载体(pIgG1-637),将pIgG1-637转染CHO-k1细胞系,筛选出稳定表达IgG1-637的细胞株扩大培养。将纯化的IgG1-637注射5只恒河猴,通过检查猴肌肉收缩强度和复合肌肉运动电位(CMAP),判定猴EAMG发病情况,从而鉴定IgG1-637的致病性。结果20μmol/L MSX(L-methionine sulfoxmine)选择的CHO-k1细胞表达最高水平的抗体IgG1-637,表达产量为17.4μg/ml。3只猴(分别接受1.7、1.7和5.0mg/kg的注射剂量)在注射IgG1-637后1周内出现肌肉收缩无力临床症状,并表现出CMAP减弱现象。结论由Fab637构建的完整的IgG1-637是一株致病性抗AchR抗体,可在猴诱导出EAMG。杨康鹃 孟繁平 魏晶 李强 李红花 李英信 M.Stassen M.de Baets 2006中华微生物学和免疫学杂志2006,26,8:2
20Simulations of helicopter ditching using smoothed particle hydrodynamics显示文摘The present work has been performed in the context of the European H2020 project increased SAfety and Robust certification for ditching of Aircrafts and Helicopters(SARAH)dedicated to improving the safety during aircraft ditching,together with a better understanding of the physics involved during those crucial events.Both numerical and experimental aspects are explored during this project.The present study focuses on the application of the smoothed particle hydrodynamics(SPH)method to the simulation of helicopter ditching,as this method has proved to be particularly adapted to free surface impact cases.Simulations are performed for three different impact configurations,for which the numerical solutions are compared with the experimental results(forces and kinematics)obtained at the wave basin of Ecole Centrale Nantes on a mock-up shape provided by Airbus Helicopters.Elements of sensitivity analysis are also provided when needed,to assess the role of some parameters involved in the helicopter behavior and the fluid pressure forces exerted during the impact.G.Oger A.Vergnaud B.Bouscasse J.Ohana M.Abu Zarim M.De Leffe A.Bannier L.Chiron Y.Jus M.Gamier S.Halbout D.Le Touze 2020Journal of Hydrodynamics2020,32,4:2
