3Genetic diversity and association mapping for salinity tolerance in Bangladeshi rice landraces显示文摘Breeding for salinity tolerance using Bangladeshi rice landraces and understand genetic diversity has been limited by the complex and polygenic nature of salt tolerance in rice genotypes. A genetic diversity and association mapping analysis was conducted using 96 germplasm accessions with variable response to salt stress at the seedling stage. These included86 landraces and 10 indica varieties and lines including Nona Bokra, from southern Bangladesh. A total of 220 alleles were detected at 58 Simple Sequence Repeat(SSR) marker loci randomly distributed on all 12 rice chromosomes and 8 Sequence Tagged Site(STS) markers developed for genes SKC1, DST, and SalT. The average gene diversity was 0.5075 and polymorphism information content value was 0.4426, respectively. Cluster analysis revealed that 68 and 21 accessions were clustered into 2 distinct groups, possibly corresponding to indica and japonica groups, respectively and the remaining 7 landraces were classified as an admixed group. In addition to Wn11463, the STS marker for SKC1, RM22418 on Chr. 8 was significantly associated with salinity tolerance, at the location of a QTL detected in previous studies. Our findings of favorable alleles associated with salinity tolerance in Bangladeshi rice landraces, as well as the development of STS markers for salt tolerance genes, will be helpful in future efforts to breed salinity tolerance in rice.Reza M.Emon Mirza M.Islam Jyotirmoy Halder Yeyang Fan 2015The Crop Journal2015,3,5:1

6Systematic mutational analysis of epitope-grafted ED3 immunogenicity reveals a DENV3-DENV4 bi-serospecific ED3 mutant显示文摘Objective:To identify the residue determinants of the serospecificity and sero-cross-reactivity of dengue virus(DENV)envelope protein domain 3(ED3),which contains two major putative epitopes of DENV.Methods:We constructed ED3 from DENV3(3ED3)and DENV4(4ED3),and six epitope-grafted variants,where we transferred epitope 1(L304I,K305D,V309M,and S310A)and/or epitope 2(D383N,K384S,K387T,and N389H)of 4ED3 onto 3ED3 and vice-versa.Swiss albino mice aged 3-4 weeks were immunized against wildtype and epitope-grafted ED3 variants and anti-ED3 IgG antibody responses were determined using ELISA.Results:Mouse immunization using 3ED3 and 4ED3 generated serotype-specific antisera,as expected.Similarly,most epitope-grafted ED3s produced antisera serospecific to the template ED3 with little or no cross-recognition of ED3 of the serotype from which the epitopes were taken.These indicated that a mere grafting of the epitope was not sufficient to transfer serospecificity,contrary to our expectations.However,one epitope-grafted ED3 mutant,where epitope 1 of 3ED3 was grafted onto 4ED3(4ED3^(epi1)),generated antisera that was serospecific to both 4ED3 and 3ED3.Conclusions:The 4ED3^(epi1)is a chimeric ED3 that produces antisera possessing serospecificity to both 3ED3 and 4ED3 onto a common 4ED3 scaffold.The 4ED3^(epi1),therefore,provides a unique tool for analyzing serospecificity and sero-cross-reactivity in dengue.We believe that chimeric ED3 may provide a template for future recombinant ED3 possessing serospecificity of multiple DENVs onto a single scaffold and may pave a way developing tri-and/or tetravalent anti-DENV antisera.Mamtaz Sultana Nazmul Hasan Mamunur R.Mahib Mohammad M.Islam 2022Asian Pacific Journal of Tropical Medicine2022,15,2:0

8Dynamic response in two-dimensional transversely isotropic thick plate with spatially varying heat sources and body forces显示文摘This paper deals with a two-dimensional (2D) problem for a transverselyisotropic thick plate having heat sources and body forces. The upper surface of the plate is stress free with the prescribed surface temperature, while the lower surface of the plate rests on a rigid foundation and is thermally insulated. The study is carried out in the context of the generalized thermoelasticity proposed by Green and Naghdi. The governing equations for displacement and temperature fields are obtained in the Laplace-Fourier transform domain by applying the Laplace and Fourier transforms. The inversion of the double transform is done numerically. Numerical inversion of the Laplace transform is done based on the Fourier series expansion. Numerical computations are carried out for magnesium (Mg), and the results are presented graphically. The results for an isotropic material (Cu) are obtained numerically and presented graphically to be compared with those of a transversely isotropic material (Mg). 9Predicting short-term adverse outcomes in the geriatric population presenting with syncope:a comparison of existing syncope rules and beyond显示文摘OBJECTIVES Syncope at age 65+is associated with increased mortality,irrespective of cause.Syncope rules were designed to aid in risk-stratification but were only validated in the general adult population.Our objective was to determine if they can be applied to a geriatric population in predicting short-term adverse outcomes.METHODS In this single-center retrospective study,we evaluated 350 patients aged 65+presenting with syncope.Exclusion criteria included confirmed non-syncope,active medical condition,drug or alcohol-related syncope.Patients were stratified into high or low risk based on Canadian Syncope Risk Score(CSRS),Evaluation of Guidelines in Syncope Study(EGSYS),San Francisco Syncope Rule(SFSR),and Risk Stratification of Syncope in the Emergency Department(ROSE).Composite adverse outcomes at 48-hour and 30-day included all-cause mortality,major adverse cardiac and cerebrovascular events(MACCE),return emergency department visit,hospitalization,or medical intervention.We assessed each score's ability to predict the outcomes using logistic-regression and compared performances using receiver-operator curves.Multivariate analyses were performed to study the associations between recorded parameters and outcomes.RESULTS CSRS outperformed with AUC of 0.732(95%CI:0.653-0.812)and 0.749(95%CI:0.688-0.809)for 48-h and 30-day outcomes,respectively.Sensitivities for CSRS,EGSYS,SFSR,and ROSE for 48-hour outcomes were 48%,65%,42% and 19%;and for 30-day outcomes were 72%,65%,30% and 55%,respectively.Atrial fibrillation/flutter on EKG,congestive heart failure,antiarrhythmics,systolic blood-pressure<90 at triage,and associated chest pain highly correlated with 48-h outcomes.An EKG abnormality,heart disease history,severe pulmonary hypertension,BNP>300,vasovagal predisposition,and antidepressants highly correlated with 30-day outcomes.CONCLUSIONS Performance and accuracy of four prominent syncope rules were suboptimal in identifying high-risk geriatric patients with short-term adverse outcomes.We identified some significant clinical and laboratory information that may play a role in predicting short-term adverse events in a geriatric cohort.Suud A.Kiradoh Timothy E.Craven Maria O.Rangel Lillian M.Nosow Erfan Zarrinkhoo Suma Menon Parag A.Chevli Tareq M.Islam Luqman A.Thazhatuveetil-Kunhahamed 2023Journal of Geriatric Cardiology2023,20,1:0
