1利用重组反转录病毒表达马立克氏病病毒Meq基因显示文摘为研究马立克氏病病毒(MDV)Meq蛋白的生物学功能,应用磷酸钙与MDVMeq基因重组的反转录病毒转染载体质粒RCAS-Meq的DNA,转染于鸡胚成纤维细胞系DF1。经间接免疫荧光技术研究表明,转染后第3天即开始有少量DF1细胞在细胞核表达Meq蛋白,以后阳性细胞增多,到转染后第7天阳性细胞最多,之后荧光即衰减;通过酶联免疫吸附试验检测,发现细胞培养上清中游离病毒在转染后第5天即可检出,直至第7天仍维持最高水平,第9天开始下降。用细胞培养上清感染DF1细胞单层,于感染后第2天即可检出Meq蛋白阳性细胞,之后阳性细胞逐渐增多,到第4~5天达高峰,之后逐渐衰减;培养上清中的游离病毒在感染后第4天即可被检出,第5~7天达最高值。研究结果表明:(1)磷酸钙法能很好地将重组质粒RCAS-MeqDNA转染到DF1细胞中,而对细胞的损伤很小;(2)转染后细胞释放出的重组病毒能感染DF1细胞并稳定地表达Meq蛋白。表明Meq基因在构建的重组病毒基因组中的存在及其转录表达都是稳定的。这为在体内和体外研究MDVMeq基因的生物学功能提供了一种非常有效的方法和途径;(3)Meq蛋白仅分布于细胞核(包括核质和核仁)。这一结果证实了?韦平 Luck F.Lee 1999中国兽医学报1999,19,2:7
2A terrestrial observatory approach to the integrated investigation of the effects of deforestation on water,energy,and matter fluxes显示文摘Integrated observation platforms have been set up to investigate consequences of global change within a terrestrial network of observatories(TERENO)in Germany.The aim of TERENO is to foster the understanding of water,energy,and matter fluxes in terrestrial systems,as well as their biological and physical drivers.Part of the Lower Rhine Valley-Eifel observatory of TERENO is located within the Eifel National Park.Recently,the National Park forest management started to promote the natural regeneration of near-natural beech forest by removing a significant proportion of the spruce forest that was established for timber production after World War II.Within this context,the effects of such a disturbance on forest ecosystem functioning are currently investigated in a deforestation experiment in the Wüstebach catchment,which is one of the key experimental research sites within the Lower Rhine Valley-Eifel observatory.Here,we present the integrated observation system of the Wüstebach test site to exemplarily demonstrate the terrestrial observatory concept of TERENO that allows for a detailed monitoring of changes in hydrological and biogeochemical states and fluxes triggered by environmental disturbances.We present the observation platforms and the soil sampling campaign,as well as preliminary results including an analysis of data consistency.We specifically highlight the capability of integrated datasets to enable improved process understanding of the post-deforestation changes in ecosystem functioning.H. 3Carbon dioxide insufflation during colonoscopy in deeply sedated patients显示文摘AIM:To compare the impact of carbon dioxide(CO2) and air insufflation on patient tolerance/safety in deeply sedated patients undergoing colonoscopy.METHODS:Patients referred for colonoscopy were randomized to receive either CO2 or air insufflation during the procedure.Both the colonoscopist and patient were blinded to the type of gas used.During the procedure,insertion and withdrawal times,caecal intubationrates,total sedation given and capnography readings were recorded.The level of sedation and magnitude of patient discomfort during the procedure was assessed by a nurse using a visual analogue scale(VAS)(0-3).Patients then graded their level of discomfort and abdominal bloating using a similar VAS.Complications during and after the procedure were recorded.RESULTS:A total of 142 patients were randomized with 72 in the air arm and 70 in the CO2 arm.Mean age between the two study groups were similar.Insertion time to the caecum was quicker in the CO2 group at 7.3 min vs 9.9 min with air(P = 0.0083).The average withdrawal times were not significantly different between the two groups.Caecal intubation rates were 94.4% and 100% in the air and CO2 groups respectively(P = 0.012).The level of discomfort assessed by the nurse was 0.69(air) and 0.39(CO2)(P = 0.0155) and by the patient 0.82(air) and 0.46(CO2)(P = 0.0228).The level of abdominal bloating was 0.97(air) and 0.36(CO2)(P = 0.001).Capnography readings trended to be higher in the CO2 group at the commencement,caecal intubation,and conclusion of the procedure,even though this was not significantly different when compared to readings obtained during air insufflation.There were no complications in both arms.CONCLUSION:CO2 insufflation during colonoscopy is more efficacious than air,allowing quicker and better cecal intubation rates.Abdominal discomfort and bloating were significantly less with CO2 insufflation.Rajvinder Singh Eu Nice Neo Nazree Nordeen Ganesananthan Shanmuganathan Angelie Ashby Sharon Drummond Garry Nind Elizabeth Murphy Andrew Luck Graeme Tucker William Tam 2012World Journal of Gastroenterology2012,18,25:3

5实验室进行混凝土路面砖透水路面的堵塞试验研究显示文摘在全球化城市发展过程中,为促进雨水渗透、减少地表雨水径流量,改善下游承受水体的水质,正越来越多地利用连锁混凝土路面砖透水性路面系统(英文简称:PICPs)。但是,随着时间推移,地表雨水径流中的沉积物会在PICPs中造成堵塞,大大降低PICPs的渗透能力。过去曾对PICPs的堵塞问题进行过大量研究,其中很多研究都采用在实验室的PICP模型,并使用人工合成的雨水,试图重现和预测出现场铺装PICP的实际堵塞过程。试验使用了各种各样的沉积物类型和颗粒尺寸,以及不同的降雨强度,故研究所获得的结果并不相同。本项目是在实验室内,研究不同类型沉积物、不同颗粒尺寸分布、不同试验期限对PICP模型堵塞情况的影响。利用加速半合成雨水的模拟技术,在实验室分别对18个专门建造的PICP模型进行了试验。对以下三种不同类型的沉积物进行了试验:(1)专门配制的硅砂;(2)颗粒尺寸<300μm的实际雨水沉积物;(3)颗粒尺寸<1.18mm的实际雨水沉积物。本研究采用6个不同降雨速率(降雨强度)进行了试验。研究结果普遍显示,硅砂沉积物的堵塞过程与降雨强度成反比。也就是说,较低降雨强度(较长的试验期限)下的PICP的堵塞程度较弱。相反,颗粒尺寸<300μm的实际雨水沉积物,堵塞过程与降雨强度成正比,即:较低降雨强度(较长的试验期限)下的PICP的堵塞程度更严重。对于颗粒尺寸<1.18mm的沉积物,降雨强度对堵塞程度似乎没有影响。对于判断实验室PICP模型堵塞试验结果的可靠性和有效性,以及如何看待这些结果,提出了一种新的见解。Terry Lucke Carsten Dierkes 崔玉忠(译) 杜建东(校) 2017建筑砌块与砌块建筑2017,,4:3
