1Pioneering function of Isl1 in the epigenetic control of cardiomyocyte cell fate显示文摘Generation of widely differing and specialized cell types from a single totipotent zygote involves large-scale transcriptional changes and chromatin reorganization. Pioneer transcription factors play key roles in programming the epigenome and facilitating recruitment of additional regulatory factors during successive cell lineage specification and differentiation steps. Here we show that Isl1 acts as a pioneer factor driving cardiomyocyte lineage commitment by shaping the chromatin landscape of cardiac progenitor cells. Using an Isl1 hypomorphic mouse line which shows congenital heart defects, genome-wide profiling of Isl1 binding together with RNA- and ATAC-sequencing of cardiac progenitor cells and their derivatives, we uncover a regulatory network downstream of Isl1 that orchestrates cardiogenesis. Mechanistically, we show that Isl1 binds to compacted chromatin and works in concert with the Brg1-Baf60c-based SWI/SNF complex to promote permissive cardiac lineage-specific alterations in the chromatin landscape not only of genes with critical functions in cardiac progenitor cells, but also of cardiomyocyte structural genes that are highly expressed when Isl1 itself is no longer present. 3Directional radiation enhancement of nanowire quantum dots based on line-array plasmonic antenna coupling显示文摘The integration of a single III-V semiconductor quantum dot with a plasmonic nanoantenna as a means toward efficient single-photon sources(SPEs)is limited due to its weak,wide-angle emission,and low emission rate.These limitations can be overcome by designing a unique linear array of plasmonic antenna structures coupled to nanowire-based quantum dot(NWQD)emitters.A linear array of a coupled device composed of multiple plasmonic antennas at an optimum distance from the quantum dot emitter can be designed to enhance the directionality and the spontaneous emission rate of an integrated single-photon emitter.Finite element modeling has been used to design these compact structures with high quantum efficiencies and directionality of single-photon emission while retaining the advantages of NWQDs.The Purcell enhancement factor of these structures approaches 66.1 and 145.8,respectively.Compared to a single NWQD of the same diameter,the fluorescence was enhanced by 1054 and 2916times.The predicted collection efficiencies approach 85%(numerical aperture,NA=0.5)and 80%(NA=0.5),respectively.Unlike single-photon emitters based on bulky conventional optics,this is a unique nanophotonic single-emission photon source based on a line-array configuration that uses a surface plasmon-enhanced design with minimum dissipation.The designs presented in this work will facilitate the development of SPEs with potential integration with semiconductor optoelectronics.PEIHANG LI PENG YU JIACHEN SUN ZHIMIN JING JIANG WU LUCAS V.BESTEIRO ROBERTO CAPUTO ARUP NEOGI HONGXING XU ZHIMING WANG 2022Photonics Research2022,10,9:0
4An integrative taxonomic study of Santolina(Asteraceae)from southern France and north-eastern Spain reveals new endemic taxa显示文摘Santolina is a clear example of a genus lying in an alpha-taxonomic status,with species accepted only based on qualitative morphological descriptions.In particular,taxonomic issues still need to be resolved for Santolina populations from southern France and north-eastern Spain,and so,we carried out an integrative taxonomic study involving morphometrics,cypsela morphometrics,niche overlap,and phylogenetic analysis based on six plastid markers(trnH-psbA,trnL-trnF,trnQ-rps16,rps15-ycf1,psbM-trnD,and trnS-trnG).Our results revealed that the current taxonomic circumscription is not adequate.In particular,the Santolina populations at the foothills of eastern Pyrenees,previously included in the variability of Santolina benthamiana,have to be considered as a distinct species,namely,Santolina intricata.In addition,despite their high phylogenetic relatedness,S.benthamiana s.str.and Santolina ericoides can still be considered as distinct species due to clear morphological and ecological differentiation.Finally,we demonstrated that three different subspecies can be recognized in Santolina decumbens,a species endemic to Provence.For one of these subspecies,due to its extremely restricted distribution range,conservation issues are pointed out.Antonio Giacò Lucia Varaldo Gabriele Casazza Daniele De Luca Paolo Caputo Marco Sarigu Gianluigi Bacchetta LlorençSáez Lorenzo Peruzzi 2023Journal of Systematics and Evolution2023,61,5:0
