2Emergence and prevalence of naturally occurring lower virulent African swine fever viruses in domestic pigs in China in 2020显示文摘African swine fever virus(ASFV)has been circulating in China for more than two years,and it is not clear whether the biological properties of the virus have changed.Here,we report on our surveillance of ASFVs in seven provinces of China,from June to December,2020.A total of 22 viruses were isolated and characterized as genotype II ASFVs,with mutations,deletions,insertions,or short-fragment replacement occurring in all isolates compared with Pig/HLJ/2018(HLJ/18),the earliest isolate in China.Eleven isolates had four different types of natural mutations or deletion in the EP402R gene and displayed a nonhemadsorbing(non-HAD)phenotype.Four isolates were tested for virulence in pigs;two were found to be as highly lethal as HLJ/18.However,two non-HAD isolates showed lower virulence but were highly transmissible;infection with 106 TCID50 dose was partially lethal and caused acute or sub-acute disease,whereas 103 TCID50 dose caused non-lethal,sub-acute or chronic disease,and persistent infection.The emergence of lower virulent natural mutants brings greater difficulty to the early diagnosis of ASF and creates new challenges for ASFV control.Encheng Sun Zhenjiang Zhang Zilong Wang Xijun He Xianfeng Zhang Lulu Wang Wenqing Wang Lianyu Huang Fei Xi Haoyue Huangfu Ghebremedhin Tsegay Hong Huo Jianhong Sun Zhijun Tian Wei Xia Xuewu Yu Fang Li Renqiang Liu Yuntao Guan Dongming Zhao Zhigao Bu 2021Science China(Life Sciences)2021,64,5:46
3TCT标本检测高危HPV E6/E7mRNA及在宫颈病变中的应用研究显示文摘目的探讨新柏超薄液基细胞学检测(Thin-prep cytology test,TCT)标本检测高危人乳头瘤病毒(human papilloma virus,HPV)E6/E7mRNA可行性及在宫颈癌筛查中的应用价值。方法 两种标本处理方法,分别在22例经超薄液基细胞学技术标本检测HPV E6/E7mRNA及HPV E6/E7DNA;对108例不同病变程度宫颈癌标本采取超薄液基细胞学技术检测高危HPV(high-risk HPV,HR-HPV)E6/E7mRNA及高危HPV E6/E7DNA,并行统计学处理。结果 两种方法处理标本行Willcoxon配对秩和检验比较,差异无显著意义(P>0.05)。随着宫颈病变程度增加,HPV E6/E7mRNA表达率相应增加。新柏液基细胞学技术检测108例标本高危HPV E6/E7mRNA表达的相对拷贝数为320.554±779.104,高危HPV E6/E7DNA为1540.963±3575.255,两者秩相关检验,相关系数为1。结论 超薄液基细胞学检测标本可行高危HPV E6/E7mRNA及HPV E6/E7DNA检测;宫颈癌筛查检测HPV E6/E7mRNA有一定临床意义。刘桐宇 谢榕 Lulu Zhang 郑雄伟 陈刚 何诚 林玉珍 杨琳 李桑 2011中华妇幼临床医学杂志(电子版)2011,7,3:35
4Challenges in diagnosis of pancreatic cancer显示文摘Pancreatic cancer is a growing source of cancer related death, yet has poor survival rates which have not improved in the last few decades. Its high mortality rate is attributed to pancreatic cancer biology, difficulty in early diagnosis and the lack of standardised international guidelines in assessing suspicious pancreatic masses. This review aims to provide an update in the current state of play in pancreatic cancer diagnosis and to evaluate the benefits and limitations of available diagnostic technology. The main modalities discussed are imaging with computed tomography, magnetic resonance imaging, endoscopic ultrasound and positron emission tomography and tissue acquisition with fine needle aspiration. We also review the improvements in the techniques used for tissue acquisition and the opportunity for personalised cancer medicine. Screening of high risk individuals, promising biomarkers and common mimickers of pancreatic cancer are also explored, as well as suggestions for future research directions to allow for earlier detection of pancreatic cancer. 5HPV E6/E7 mRNA检测对宫颈癌筛查意义的初步评价显示文摘目的:利用超薄液基细胞学检测标本(LCT)分析高危人乳头瘤病毒(HPV)E6/E7mRNA检测在宫颈癌筛查中的意义。方法:选取231例液基细胞学检测标本,根据病理级别分为炎症/良性组(n=78)、ASCUS组(n=140)、LSIL组(n=8)和HSIL组(n=5)。利用bDNA技术检测细胞学标本中HPV E6/E7mRNA、DNA,结合病理诊断资料,进行统计学分析。结果:对不同细胞学级别中mRNA及DNA检测情况行配对χ2检验显示,炎症/正常细胞组DNA和mR-NA,阳性检出率差异无统计学意义,χ2=2.307,P>0.05;ASCUS组阳性检出率差异有统计学意义,χ2=9.082,P=0.020;LSIL组与HSIL组例数少,阳性检出率差异无统计学意义,P值分别为0.464和0.800。34例行宫颈活检的AS-CUS标本,2种检测指标在检出率差异无统计学意义,P值均>0.05;在进行活检的43例标本中,对不同病理级别HPVE6/E7mRNA及DNA检测情况行配对Fisher精确概率检验,2种检测指标检出率差异无统计学意义,P值均>0.05。分析预测效能,HPV E6/E7mRNA对LSIL/HSIL诊断敏感性为33.33%,95%CI为17.19~54.63;特异性为72.73%,95%CI为51.85~86.85;阳性预测值为53.85%,95%CI为29.14~76.79;阴性预测值为53.33%,95%CI为36.14~69.77。HPV E6/E7mRNA联合细胞学检测诊断敏感性为100%,95%CI为84.54~100.00;阴性预测值为100%,95%CI为43.85~100.00。结论:高危HPV E6/E7mRNA可以应用于宫颈癌筛查,在筛查中联合HPV E6/E7mRNA及细胞学检测有利于提高诊断准确性。黄宝英 周伦顺 富显果 余兰 卢少玲 陈惠华 缪韵仪 刘桐宇 lulu zhang 2013中华肿瘤防治杂志2013,20,14:34

7Ablation of gut microbiota alleviates obesity-induced hepatic steatosis and glucose intolerance by modulating bile acid metabolism in hamsters显示文摘Since metabolic process differs between humans and mice, studies were performed in hamsters, which are generally considered to be a more appropriate animal model for studies of obesityrelated metabolic disorders. The modulation of gut microbiota, bile acids and the farnesoid X receptor(FXR) axis is correlated with obesity-induced insulin resistance and hepatic steatosis in mice. However,the interactions among the gut microbiota, bile acids and FXR in metabolic disorders remained largely unexplored in hamsters. In the current study, hamsters fed a 60% high-fat diet(HFD) were administeredvehicle or an antibiotic cocktail by gavage twice a week for four weeks. Antibiotic treatment alleviated HFD-induced glucose intolerance, hepatic steatosis and inflammation accompanied with decreased hepatic lipogenesis and elevated thermogenesis in subcutaneous white adipose tissue(sWAT). In the livers of antibiotic-treated hamsters, cytochrome P450 family 7 subfamily B member 1(CYP7 B1) in the alternative bile acid synthesis pathway was upregulated, contributing to a more hydrophilic bile acid profile with increased tauro-β-muricholic acid(TβMCA). The intestinal FXR signaling was suppressed but remained unchanged in the liver. 8Improving bubble-particle attachment during the flotation of low rank coal by surface modification显示文摘Based on the problems caused by many oxygen-containing functional groups and poor floatability on the surface of low rank coal,the characteristics of low rank coal were analyzed systematically by means of scanning electron microscopy(SEM),X-ray diffraction(XRD)and X-Ray photoelectron spectroscopy(XPS)techniques.The bubble-particle induction time was used to determine the characterization of the bubble-particle attachment,and the bubble-particle attachment of low rank coal modified by soaking the coal samples in an acid or alkaline solution was analyzed.The floatability of the modified coal surface was verified by flotation tests.The results show that the particle size of 0.125–0.074 mm of the coal sample exhibited better bubble-particle attachment characteristics.The small bubble,the larger approach velocity of bubble and the larger bubble deformation were more helpful to enhance the bubbleparticle attachment.For an acid solution,the smaller the p H was and the longer the soaking time was,the better the floatability of the coal sample and the higher the combustible material recovery were.The combustible material recovery of low rank coal was increased to 78.79%by soaking the sample in an acid solution of pH=0 for 180 min.On the contrary there was a best concentration for the alkaline solution.Haishan Hu Ming Li Lulu Li Xiuxiang Tao 2020International Journal of Mining Science and Technology2020,30,2:27
9Single-cell RNA-seq highlights intra-tumoral heterogeneity and malignant progression in pancreatic ductal adenocarcinoma显示文摘Pancreatic ductal adenocarcinoma (PDAC) is the most common type of pancreatic cancer featured with high intra-tumoral heterogeneity and poor prognosis. To comprehensively delineate the PDAC intra-tumoral heterogeneity and the underlying mechanism for PDAC progression, we employed single-cell RNA-seq (scRNA-seq) to acquire the transcriptomic atlas of 57,530 individual pancreatic cells from primary PDAC tumors and control pancreases, and identified diverse malignant and stromal cell types, including two ductal subtypes with abnormal and malignant gene expression profiles respectively, in PDAC. We found that the heterogenous malignant subtype was composed of several subpopulations with differential proliferative and migratory potentials. Cell trajectory analysis revealed that components of multiple tumor-related pathways and transcription factors (TFs) were differentially expressed along PDAC progression. Furthermore, we found a subset of ductal cells with unique proliferative features were associated with an inactivation state in tumor-infiltrating T cells, providing novel markers for the prediction of antitumor immune response. The nanostructure of roughly 150 nm in diameter with a spatial resolution of 6 nm is purely constructed by folding DNA. The picture observed by atomic force microscopy (AFM) is almost identical with the de-signed shape. 14Neutrophil extracellular traps mediate the crosstalk between glioma progression and the tumor microenvironment via the HMGB1/RAGE/IL-8 axis显示文摘Objective:Neutrophil extracellular traps(NETs)produced by tumor-infiltrating neutrophils(TINs)are associated with poor prognosis in patients with several types of cancer.However,the mechanisms underlying the involvement of NETs in glioma progression remain largely unknown.This study aimed to elucidate the roles of NETs in biological processes that drive the crosstalk between glioma progression and the tumor microenvironment.Methods:Neutrophil infiltration and NETs formation were investigated in glioma tissue through immunohistochemistry,and their relationships with clinicopathological features and outcomes were statistically evaluated.The effects of NETs on glioma cell progression were studied in a co-culture system.In vivo and in vitro experiments validated the reactive oxygen species activity and cytokine production of TINs,as well as the ERK signaling pathway activation and the metastasis of gliomas.Results:Neutrophil infiltration and NETs formation were induced in high-grade glioma compared with low-grade glioma.NETs induced by TINs were determined to be an oncogenic marker of high-grade gliomas and to be involved in cell proliferation and invasion.NETs overproduction promoted glioma cell proliferation,migration,and invasion.Furthermore,HMGB1 was found to bind to RAGE and activate the NF-κB signaling pathway in vitro.In addition,NETs stimulated the NF-κB signaling pathway,thus promoting IL-8 secretion in glioblastoma.Subsequently,IL-8 recruited neutrophils which in turn mediated NETs formation via the PI3 K/AKT/ROS axis in TINs.Conclusions:Our results suggest that NETs produced by TINs mediate the crosstalk between glioma progression and the tumor microenvironment by regulating the HMGB1/RAGE/IL-8 axis.Targeting NETs formation or IL-8 secretion may be an effective approach to inhibit glioma progression.Caijun Zha Xiangqi Meng Lulu Li Shan Mi Da Qian Ziwei Li Pengfei Wu Shaoshan Hu Shihong Zhao Jinquan Cai Yanhong Liu 2020Cancer Biology & Medicine2020,17,1:19
15Urimem,a membrane that can store urinary proteins simply and economically,makes the large-scale storage of clinical samples possible显示文摘By nature,biomarker is the measurable change associated with a physiological or pathophysiological process.Unlike blood which has mechanisms to minimize changes and to keep the internal environment homeostatic,urine is more likely to reflect changes of the body and is a better biomarker source.Because of its potential in biomarker discovery,urinary proteins should be preserved comprehensively as the duration of the patients’corresponding medical records.Here,we propose a method to adsorb urinary proteins onto a membrane we named Urimem.This simple and inexpensive method requires minimal sample handling,uses no organic solvents,and is environmentally friendly.Urine samples were filtered through the membrane,and urinary proteins were adsorbed onto the membrane.The proteins on the membrane were dried and stored in a vacuum bag,which keeps the protein pattern faithfully preserved.The membrane may even permit storage at room temperature for weeks.Using this simple and inexpensive method,it is possible to begin preserving urine samples from all consenting people.Thus,medical research especially biomarker research can be conducted more economically.Even more objective large-scale prospective studies will be possible.This method has the potential to change the landscape of medical research and medical practice.JIA LuLu LIU XueJiao LIU Liu LI MingXi GAO YouHe 2014Science China(Life Sciences)2014,57,3:19

17Finite-time adaptive consensus of a class of multi-agent systems显示文摘Multi-agent systems(MASs) are ubiquitous in natural and artificial systems. This paper aims to establish the finite-time adaptive consensus criterion for a class of MASs with nonlinear dynamics. Traditionally, the finite-time consensus criterion is often established based on the prior information on Lipschitz constants and the eigenvalues of Laplacian matrix. However, it is difficult to acquire the above prior information for most real-world engineering systems. To overcome the above difficulty, this paper develops the finite-time consensus criteria for a class of MASs with nonlinear dynamics via adaptive technique. In detail, we design the finite-time distributed node-based and edge-based adaptive consensus protocols for a class of MASs with fixed and switching topologies. 19Study on multi-size seed-metering device for vertical plate soybean precision planter显示文摘Aiming to solve the problem of small range of the appropriately sowing seeds existing in a vertical disc seed-metering device,the planter plate series with four sizes were developed according to the variety and size distribution of all soybeans in China.The structure and working principle of the vertical disc soybean seed-metering device were detailed,and the influence of the diameter of soybean on the working performance of the seed-metering device was analyzed through the software EDEM virtual simulation,so as to achieve the goal of covering the soybean seeds with all sizes by the minimum planter plate series as well as to obtain the most appropriate operating speed of each planter plate by optimization.For the planter plates with the hole diameter of 7,9,12,16 mm,the appropriate size ranges of sowing seeds are 4.5-6.0,6.0-8.0,8.0-10.5,10.5-13.0 mm,respectively,and the appropriate operating speeds are 9,8,7,6 km/h,respectively.The results show that this planter plate series can meet the requirements of seeding with all sizes of soybeans at the range of the most appropriate operating speed.The study method can provide a reference for design and optimization of precision planters.Liu Hongxin Guo Lifeng Fu Lulu Tang Shifa 2015International Journal of Agricultural and Biological Engineering2015,8,1:13
20Experimental research on coal seam similar material proportion and its application显示文摘In this paper, the optimization design of the low strength mechanical test and orthogonal test have been analyzed in order to simulate the mechanical properties of thick and extra-thick coal seam accurately in a similar material simulation test. The results show that the specimen can reach a wider range of strength when cement has been used compared to that of gypsum, suggesting that cement is more suitable for making coal seam in similar material simulation tests. The uniaxial compressive strength is more sensitive to cement than coal or sand. The proportion of coal and sand do not play a decisive role in uniaxial compressive strength. The uniaxial compressive strength and specimen density decrease as the mass percent of coal and aggregate–binder ratio rise. There is a positive correlation between uniaxial compressive strength and density. The No. 5 proportion(cement: sand: water: activated carbon: coal = 6:6:7:1.1:79.9)was chosen to be used in the similar material simulation test of steeply dipping and extra-thick coal seam with a density of 0.913 g/cm^3 and an uniaxial compressive strength of 0.076 MPa which are in accordance with the similarity theory.
