1Overexpression of the tonoplast aquaporin AtTIP5;1 conferred tolerance to boron toxicity in Arabidopsis显示文摘Boron (B) toxicity to plants is responsible for low crop productivity in many regions of the world. Here we report a novel and effective means to alleviate the B toxicity to plants under high B circumstance. Functional characterization of AtTIP5;1, an aquaporin gene, re-vealed that overexpression of AtTIP5;1 (OxAtTIP5;1) in Arabidopsis significantly increased its tolerance to high B toxicity. Compared to wild-type plants, OxAtTIP5;1 plants exhibited longer hypocotyls, accelerated development, increased silique production under high B treatments. GUS staining and quantitative RT-PCR (qRT-PCR) results demonstrated that the expression of AtTIP5;1 was induced by high B concentration treatment. Subcellular localization analysis revealed that the AtTIP5;1-GFP fusion protein was localized on the tonoplast membrane, which was consistent with the prediction based on bioinformatics. 2Pinpointing Source of Mekong and Measuring Its Length Through Analysis of Satellite Imagery and Field Investigations显示文摘For centuries, explorers and scientists from different countries had made their own conclusions on the source of the Mekong. However, the geographic source of the Mekong is still arguable because of the complexity of the Mekong source water system, inaccessible environment and the varied technologies used by those explorers and scientists. The satellite remote sensing technology has been used to pinpoint the source of the Mekong, associated with the on-the-spot investigations made by the authors in June 1999 and September 2002. 3Arabidopsis PTD Is Required for Type Ⅰ Crossover Formation and Affects Recombination Frequency in Two Different Chromosomal Regions显示文摘In eukaryotes,crossovers together with sister chromatid cohesion maintain physical association between homologous chromosomes,ensuring accurate chromosome segregation during meiosis I and resulting in exchange of genetic information between homologues.The Arabidopsis PTD(Parting Dancers)gene affects the level of meiotic crossover formation,but its functional relationships with other core meiotic genes,such as AtSPO11-1,AtRAD51,and AtMSH4,are unclear;whether PTD has other functions in meiosis is also unknown.To further analyze PTD function and to test for epistatic relationships,we compared the meiotic chromosome behaviors of Atspol 1-1 ptd and AtradSl ptd double mutants with the relevant single mutants.The results suggest that PTD functions downstream of AtSPOll-1 and AtRAD51 in the meiotic recombination pathway.Furthermore,we found that meiotic defects in rck ptd and Atmsh4 ptd double mutants showed similar meiotic phenotypes to those of the relevant single mutants,providing genetic evidences for roles of PTD and RCK in the type I crossovers pathway.Moreover,we employed a pollen tetrad-based fluorescence method and found that the meiotic crossover frequencies in two genetic intervals were significantly reduced from 6.63%and 22.26%in wild-type to 1.14%and 6.36%,respectively,in the ptd-2 mutant.These results revealed new aspects of PTD function in meiotic crossover formation.Pingli Lu Asela J.Wijeratne Zhengjia Wang Gregory P.Copenhaver Hong Ma 2014Journal of Genetics and Genomics2014,41,3:5
4Attitude coordination for spacecraft formation with multiple communication delays显示文摘Communication delays are inherently present in information exchange between spacecraft and have an effect on the control performance of spacecraft formation. In this work, attitude coordination control of spacecraft formation is addressed, which is in the presence of multiple communication delays between spacecraft. Virtual system-based approach is utilized in case that a constant reference attitude is available to only a part of the spacecraft. The feedback from the virtual systems to the spacecraft formation is introduced to maintain the formation. Using backstepping control method, input torque of each spacecraft is designed such that the attitude of each spacecraft converges asymptotically to the states of its corresponding virtual system. Furthermore, the backstepping technique and the Lyapunov–Krasovskii method contribute to the control law design when the reference attitude is time-varying and can be obtained by each spacecraft. 5Identification of key gene networks controlling organic acid and sugar metabolism during watermelon fruit development by integrating metabolic phenotypes and gene expression profiles显示文摘The organoleptic qualities of watermelon fruit are defined by the sugar and organic acid contents,which undergo considerable variations during development and maturation.The molecular mechanisms underlying these variations remain unclear.In this study,we used transcriptome profiles to investigate the coexpression patterns of gene networks associated with sugar and organic acid metabolism.We identified 3 gene networks/modules containing 2443 genes highly correlated with sugars and organic acids.Within these modules,based on intramodular significance and Reverse Transcription Quantitative polymerase chain reaction(RT-qPCR),we identified 7 genes involved in the metabolism of sugars and organic acids.Among these genes,Cla97C01G000640,Cla97C05G087120 and Cla97C01G018840(r^(2)=0.83 with glucose content)were identified as sugar transporters(SWEET,EDR6 and STP)and Cla97C03G064990(r^(2)=0.92 with sucrose content)was identified as a sucrose synthase from information available for other crops.Similarly,Cla97C07G128420,Cla97C03G068240 and Cla97C01G008870,having strong correlations with malic(r^(2)=0.75)and citric acid(r^(2)=0.85),were annotated as malate and citrate transporters(ALMT7,CS,and ICDH).The expression profiles of these 7 genes in diverse watermelon genotypes revealed consistent patterns of expression variation in various types of watermelon.These findings add significantly to our existing knowledge of sugar and organic acid metabolism in watermelon.Muhammad Jawad Umer Luqman Bin Safdar Haileslassie Gebremeskel Shengjie Zhao Pingli Yuan Hongju Zhu M.O.Kaseb Muhammad Anees Xuqiang Lu Nan He Chengsheng Gong Wenge Liu 2020Horticulture Research2020,7,1:4
6Fast self-adapting high-order sliding mode control for a class of uncertain nonlinear systems显示文摘A fast self-adapting high-order sliding mode(FSHOSM)controller is designed for a class of nonlinear systems with unknown uncertainties.As for uncertainty-free nonlinear system,a new switching condition is introduced into the standard geometric homogeneity.Different from the existing geometric homogeneity method,both state variables and their derivatives are considered to bring a reasonable effective switching condition.As a result,a faster convergence rate of state variables is achieved.Furthermore,based on the integral sliding mode(ISM)and above geometric homogeneity,a self-adapting high-order sliding mode(HOSM)control law is proposed for a class of nonlinear systems with uncertainties.The resulting controller allows the closed-loop system to conduct with the expected properties of strong robustness and fast convergence.Stable analysis of the nonlinear system is also proved based on the Lyapunov approach.The effectiveness of the resulting controller is verified by several simulation results.GUO Fuhui LU Pingli 2021Journal of Systems Engineering and Electronics2021,32,3:1
7Tumor extracellular vesicles mediate anti-PD-L1 therapyresistance by decoying anti-PD-L1显示文摘PD-L1+tumor-derived extracellular vesicles(TEVs)cause systemic immunosuppression and possibly resistance to anti-PD-L1 antibody(αPD-L1)blockade.However,whether and how PD-L1+TEVs mediateαPD-L1 therapy resistance is unknown.Here,we show that PD-L1+TEVs substantially decoyαPD-L1 and that TEV-boundαPD-L1 is more rapidly cleared by macrophages,causing insufficient blockade of tumor PD-L1 and subsequentαPD-L1 therapy resistance.Inhibition of endogenous production of TEVs by Rab27a or Coro1a knockout reversesαPD-L1 therapy resistance.Either an increasedαPD-L1 dose or macrophage depletion mediated by the clinical drug pexidartinib abolishesαPD-L1 therapy resistance.Moreover,in the treatment cycle with the same total treatment dose ofαPD-L1,high-dose and low-frequency treatment had better antitumor effects than low-dose and highfrequency treatment,induced stronger antitumor immune memory,and eliminatedαPD-L1 therapy resistance.Notably,in humanized immune system mice with human xenograft tumors,both increasedαPD-L1 dose and high-dose and low-frequency treatment enhanced the antitumor effects ofαPD-L1.Furthermore,increased doses ofαPD-L1 andαPD-1 had comparable antitumor effects,butαPD-L1 amplified fewer PD-1+Treg cells,which are responsible for tumor hyperprogression.Altogether,our results reveal a TEV-mediated mechanism ofαPD-L1-specific therapy resistance,thus providing promising strategies to improveαPD-L1 efficacy.Jiming Chen Jie Yang Wenhui Wang Danfeng Guo Chengyan Zhang Shibo Wang Xinliang Lu Xiaofang Huang Pingli Wang Gensheng Zhang Jing Zhang Jianli Wang Zhijian Cai 2022Cellular & Molecular Immunology2022,19,11:1

11Post-transcriptional regulation of grain weight and shape by the RBP-A-J-K complex in rice显示文摘RNA-binding proteins(RBPs)are components of the post-transcriptional regulatory system,but their regulatory effects on complex traits remain unknown.Using an integrated strategy involving map-based cloning,functional characterizations,and transcriptomic and population genomic analyses,we revealed that RBP-K(LOC_Os08g23120),RBP-A(LOC_Os11g41890),and RBP-J(LOC_Os10g33230)encode proteins that form an RBP-A-J-K complex that negatively regulates rice yield-related traits.Examinations of the RBP-A-J-K complex indicated RBP-K functions as a relatively non-specific RBP chaperone that enables RBP-A and RBP-J to function normally.Additionally,RBP-J most likely affects GA pathways,resulting in considerable increases in grain and panicle lengths,but decreases in grain width and thickness.In contrast,RBP-A negatively regulates the expression of genes most likely involved in auxin-regulated pathways controlling cell wall elongation and carbohydrate transport,with substantial effects on the rice grain filling process as well as grain length and weight.Evolutionarily,RBP-K is relatively ancient and highly conserved,whereas RBP-J and RBP-A are more diverse.Thus,the RBP-A-J-K complex may represent a typical functional model for many RBPs and protein complexes that function at transcriptional and post-transcriptional levels in plants and animals for increased functional consistency,efficiency,and versatility,as well as increased evolutionary potential.Our results clearly demonstrate the importance of RBP-mediated post-transcriptional regulation for the diversity of complex traits.Furthermore,rice grain yield and quality may be enhanced by introducing various complete or partial loss-of-function mutations to specific RBP genes using clustered regularly interspaced palindromic repeats(CRISPR)/CRISPR-associated protein 9 technology and by exploiting desirable natural tri-genic allelic combinations at the loci encoding the components of the RBP-A-J-K complex through marker-assisted selection.Ding Ren Hui Liu Xuejun Sun Fan Zhang Ling Jiang Ying Wang Ning Jiang Peiwen Yan Jinhao Cui Jinshui Yang Zhikang Li Pingli Lu Xiaojin Luo 2024Journal of Integrative Plant Biology2024,66,1:0

13Watermelon domestication was shaped by stepwise selection and regulation of the metabolome显示文摘Although crop domestication has greatly aided human civilization,the sequential domestication and regulation of most quality traits remain poorly understood.Here,we report the stepwise selection and regulation of major fruit quality traits that occurred during watermelon evolution.The levels of fruit cucurbitacins and flavonoids were negatively selected during speciation,whereas sugar and carotenoid contents were positively selected during domestication.Interestingly,fruit malic acid and citric acid showed the opposite selection trends during the improvement.We identified a novel gene cluster(CGC1,cucurbitacin gene cluster on chromosome 1)containing both regulatory and structural genes involved in cucurbitacin biosynthesis,which revealed a cascade of transcriptional regulation operating mechanisms.In the CGC1,an allele caused a single nucleotide change in Cl ERF1 binding sites(GCC-box)in the promoter of Cl Bh1,which resulted in reduced expression of Cl Bh1 and inhibition of cucurbitacin synthesis in cultivated watermelon.Functional analysis revealed that a rare insertion of 244 amino acids,which arose in C.amarus and became fixed in sweet watermelon,in Cl OSC(oxidosqualene cyclase)was critical for the negative selection of cucurbitacins during watermelon evolution.This research provides an important resource for metabolomics-assisted breeding in watermelon and for exploring metabolic pathway regulation mechanisms.Pingli Yuan Congping Xu Nan He Xuqiang Lu Xingping Zhang Jianli Shang Hongju Zhu Chengsheng Gong Hanhui Kuang Tang Tang Yong Xu Shuangwu Ma Dexi Sun Weiqin Zhang Muhammad J.Umer Jian Shi Alisdair R.Fernie Wenge Liu Jie Luo 2023Science China(Life Sciences)2023,66,3:0
