1Relationship between ATPase activity and conjugated polyamines in mito-chondrial membrane from wheat seedling roots under osmotic stress显示文摘The effects of osmotic stress on the ATPase activity, the contents of —SH group and conjugated polyamines in mitochondrial membrane from wheat seedling [Triticum aestivum L. cv. Yumai No.18(drought-tolerant) and cv. Yumai No.9(drought-sensitive)] roots were investigated. The results showed that ATPase activity and —SH group content decreased with polyethylene glycol(PEG) 6000(-0.55 MPa) treatment for 7 d, in concert with the decrease of the ratio of noncovalently conjugated spermidine(NCC-Spd)/noncovalently conjugated putrescine(NCC-Put) and increase of the covalently conjugated putrescine(CC-Put). Osmotic stress injury to Yangmai No.9 seedlings was alleviated greatly with 1 mmol/L exogenous spermidine(Spd), in concert with marked increases of the ratio of NCC-Spd/NCC-Put, —SH group contents and ATPase activity in mitochondrial membrane. Under osmotic stress, the concomitant treatment of Yumai No.18 seedlings with methylglyoxyl bis(guanylhydrazone) (MGBG), an inhibitor of S-adenosyl methionine decarboxylase(SAMDC), and phenanthrolin (o-Phen), an inhibitor of transglutaminase(TGase), caused a significant decrease of the ratio of NCC-Spd / NCC-Put, CC-Put contents, respectively, in concert with the marked decreases of ATPase activity, —SH group content and its tolerance to osmotic stress. 2Effects of Variation in Activities of Starch-Sugar Metabolic Enzymes on Reducing Sugar Accumulation and Processing Quality of Potato Tubers显示文摘The experiment was designed, via storing potato tubers of cv. E-Potato1 and E-Potato3 indifferent temperatures, to explore the variation patterns of reducing sugar (RS) andtotal sugar (TS) contents and enzyme activities that are involved in the pathway ofstarch-sugar metabolism aiming at identifying the main factors that influence the chipcolor. The results showed that low temperature in storage was a main factor thataccelerated the accumulation of RS of the stored tubers and a very significant linearrelationship existed between RS content and chip color index (CCI) of the tubers. Furtheranalysis elucidated that when tubers stored at 4℃, the activities of ADP glucosepyrophosphorylase (AGPase), UDP glucose pyrophosphorylase (UGPase) and sucrose synthase(SuSy) were negatively exponential to the RS content significantly while that of acidinvertase and alkaline invertase was significantly linear to RS content. 3Advances in engineering RuO_(2) electrocatalysts towards oxygen evolution reaction显示文摘Water electrolysis technology holds the perfect promise of the hydrogen production,yet control of efficiency and rate of water electrolysis greatly relies on the availability of high-performance electrode materials for kinetic-sluggish oxygen evolution reaction(OER).Accordingly,substantial endeavors have been made to explore advanced electrode materials over the past decade.Recently,RuO_(2) and RuO_(2)-based materials have been demonstrated to be promising for OER due to their remarkable electrocatalytic activity and pH-universal application.Herein,the great achievements and progresses of this flourishing spot are comprehensively reviewed,which are started by a general description of OER to understand the reaction mechanism in detail.Subsequently,the key advantages and issues of RuO_(2) towards OER are also introduced,followed by proposing many advanced strategies for further promoting the electrocatalytic OER performance of RuO_(2).Finally,the daunting challenges and future progresses of RuO_(2) electrocatalysts toward practical water oxidation are highlighted,aiming to provide guidance for the fabrication of desirable RuO_(2)-based electrocatalysts toward OER.Cheng Wang Liujun Jin Hongyuan Shang Hui Xu Yukihide Shiraishi Yukou Du 2021Chinese Chemical Letters2021,32,7:6
4The long non-coding RNA expression profile of Coxsackievirus A16 infected RD cells identified by RNA-seq显示文摘Coxsackievirus A16(CVA16) is one of major pathogens of hand, foot and mouth disease(HFMD) in children. Long non-coding RNAs(Inc RNAs) have been implicated in various biological processes,but they have not been associated with CVA16 infection. In this study, we comprehensively characterized the landscape of Inc RNAs of normal and CVA16 infected rhabdomyosarcoma(RD)cells using RNA-Seq to investigate the functional relevance of Inc RNAs. We showed that a total of 760 Inc RNAs were upregulated and 1210 Inc RNAs were downregulated. Out of these dysregulated Inc RNAs, 43.64% were intergenic, 22.31% were sense, 15.89% were intronic, 8.67% were bidirectional, 5.59% were antisense, 3.85% were s RNA host Inc RNAs and 0.05% were enhancer. Six dysregulated Inc RNAs were validated by quantitative PCR assays and the secondary structures of these Inc RNAs were projected. Moreover, we conducted a bioinformatics analysis of an Inc RNAs(ENST00000602478) to elucidate the diversity of modification and functions of Inc RNAs. In summary, the current study compared the dysregulated Inc RNAs profile upon CVA16 challenge and illustrated the intricate relationship between coding and Inc RNAs transcripts. 5Gas phase tricholoethylene removal at low concentration using activated carbon fiber显示文摘The breakthrough adsorption behaviors of gas phase trichloroethylene in a packed bed of activated carbon fibers(ACF) were investigated. The specific surface area of the ACF was 600 m 2/g, 1400 m 2/g and 1600 m 2/g, respectively, and the concentration of trichloroethylene ranged from 270 mg/m 3 to 2700 mg/m 3. Results showed that the capacity of adsorption increased with increasing specific surface area, the relationship between the logarithms of 10% breakthrough time and concentration was approximately linear over the experimental range, the breakthrough time decreased with increasing temperature and humidity. 6Experimental Study on the Interaction of Fine Water Spray with Liquid Pool Fires显示文摘An experimental study of the interaction of a fine water spray with liquid pool fires was performed. Spatially resolved droplet size, velocity distributions, and volume flux of the fine water spray have been obtained in two types of pressure-atomized fine spray, using LDAIAPV technology. A method to correct for the presence of droplets is presented and used to determine the heat release rate using a cone calorimeter system, the effects of water spray application on the heat release rate for liquid pool fires were studied. It was found that the volume flux of the water spray is an important factor to determine the liquid pool fire behavior. For the current experimental conditions, the flame was enlarged when lower volume flux of water is used, and suppressed significantly when larger volumes of water are used. Two opposing mechanisms compete when water sprays are applied, which is more complex than the halon agents, such as 1301. 7Effect of angiotensin converting enzyme inhibitor on the calcium transients and calcium handling proteins in ventricular myocytes from rats with heart failure显示文摘Background Chronic heart failure (CHF) is associated with calcium transients and calcium handling proteins. Angiotensin converting enzyme (ACE) inhibitor has been demonstrated to have beneficial effect on CHF. Yet studies addressed to the relationship between ACE inhibitor and calcium transients in CHF are rare. The aim of this study was to investigate the influence of ACE inhibitor (perindopril) on the contractility and calcium transients and calcium handling proteins in ventricular myocytes from rats with experimental heart failure.Methods Male Wistar rats were randomized to heart failure group treated with perindopril (CHF-T, 3 mg·kg -1 ·d -1 ), heart failure group without treatment (CHF-C) and sham-operated group (PS). Heart failure was induced by abdominal aortic constriction. All groups were further followed up for 12 weeks. Left ventricular myocytes were then isolated. Single cell shortening fraction and [Ca 2+ ]i were simultaneously measured by laser scanning confocal microscope under the field stimulation (1.0 Hz). Reverse transcriptase-polymerase chain reaction (RT-PCR) and Western blot were performed to evaluate the changes of mRNA and protein of Na +-Ca 2+ exchanger (NCX1), sarcoplasmic reticulum Ca 2+ -ATPase (SERCA2) and phospholamban (PLB).Results The fraction of cell shortening (FS%) and [Ca 2+ ] imax (nmol/L) were significantly reduced in group CHF-C compared with group PS (FS%: 7.51±1.15 vs 13.21±1.49;[Ca 2+ ] imax :330.85±50.05 vs 498.16±14.07; both P <0.01), and restored at least partially in CHF-T group. In CHF-C group, the left ventricular mRNA of NCX1 and PLB were significantly upregulated in comparing with PS group (R NCX1/β-Actin : 0.51±0.12 vs 0.19±0.06, P <0.01; R PLB/β-Actin : 0.26±0.12 vs 0.20±0.08, P <0.05), while SERCA2 mRNA was downregulated (0.48±0.10 vs 0.80±0.11, P <0.01). The mRNA levels of NCX1 and SERCA2 in CHF-T group were between the CHF-C and PS group, and the differences of the latter two groups were significant (all P <0.05). In CHF-C and CHF-T groups, the protein expression of NCX1 were 1.141±0.047 and 1.074±0.081 times of that in PS group respectively (both P <0.05), and SERCA2 protein levels were 0.803±0.100 and 0.893±0.084 times of that in PS group respectively (both P <0.05). 8Application of sodium alginate microspheres in ischemic stroke modeling in miniature pigs显示文摘The miniature pig is an optimal animal model for studying nervous system disease because of its physiologic and pathologic features. However, the rete mirabile composed of arteries and veins at the skull base limits their application as a model of ischemic stroke by middle cerebral artery occlusion. The present study investigated the possibility of establishing an ischemic stroke model in the miniature pig by blocking the skull base retia with sodium alginate microspheres. Three Bama miniature pigs were used. Using the monitor of C-arm X-ray machine, sodium alginate microspheres (100-300 μm), a novel embolic material, were injected through the femoral artery, aortic arch, common carotid artery, ascending pharyngeal artery and the retia. Results were evaluated using carotid arteriography, MRI, behavior observation and histology. The unilateral rete mirabile was completely blocked, resulting in disturbance in blood supply to the basal ganglia, astasia of the right hind limb and salivation. MRI and hematoxylin-eosin staining showed an evident infarction focus in the basal ganglia. 14Lnc-RP5 Regulates the mi R-129-5p/Notch1/PFV Internal Promoter Axis to Promote the Expression of the Prototype Foamy Virus Transactivator Tas显示文摘Prototype foamy virus(PFV)is a unique retrovirus that infects animals and humans and does not cause clinical symptoms.Long noncoding RNAs(lncRNAs)are believed to exert multiple regulatory functions during viral infections.Previously,we utilized RNA sequencing(RNA-seq)to characterize and identify the lncRNA lnc-RP5-1086 D14.3.1-1:1(lnc-RP5),which is markedly decreased in PFV-infected cells.However,little is known about the function of lnc-RP5 during PFV infection.In this study,we identified lnc-RP5 as a regulator of the PFV transcriptional transactivator(Tas).Lnc-RP5 enhanced the activity of the PFV internal promoter(IP).The expression of PFV Tas was found to be promoted by lnc-RP5.Moreover,mi R-129-5 p was found to be involved in the lnc-RP5-mediated promotion of PFV IP activity,while the Notch1 protein suppressed the activity of PFV IP and the expression of Tas.Our results demonstrate that lnc-RP5 promotes the expression of PFV Tas through the miR-129-5 p/Notch1/PFV IP axis.This work provides evidence that host lnc RNAs can manipulate PFV replication by employing mi RNAs and proteins during an early viral infection.Shanshan Xu Liujun Chen Yinglian Tang Peipei Yuan Jun Yan Yingcheng Zheng Li Huang Zhi Li Yan Sun Song Han Jun Yin Qin Pan Biwen Peng Xiaohua He Wanhong Liu 2020Virologica Sinica2020,35,1:1
