| 1 | Engineering Escherichia coli to improve tryptophan production via genetic manipulation of precursor and cofactor pathways显示文摘Optimizing the supply of biosynthetic precursors and cofactors is usually an effective metabolic strategy to improve the production of target compounds.Here,the combination of optimizing precursor synthesis and balancing cofactor metabolism was adopted to improve tryptophan production in Escherichia coli.First,glutamine synthesis was improved by expressing heterologous glutamine synthetase from Bacillus subtilis and Bacillus megaterium in the engineered Escherichia coli strain KW001,resulting in the best candidate strain TS-1.Then icd and gdhA were overexpressed in TS-1,which led to the accumulation of 1.060 g/L tryptophan.Subsequently,one more copy of prs was introduced on the chromosome to increase the flux of 5-phospho-α-D-ribose 1-diphosphate followed by the expression of mutated serA and thrA to increase the precursor supply of serine,resulting in the accumulation of 1.380 g/L tryptophan.Finally,to maintain cofactor balance,sthA and pntAB,encoding transhydrogenase,were overexpressed.With sufficient amounts of precursors and balanced cofactors,the engineered strain could produce 1.710 g/L tryptophan after 48 h of shake-flask fermentation,which was 2.76-times higher than the titer of the parent strain.Taken together,our results demonstrate that the combination of optimizing precursor supply and regulating cofactor metabolism is an effective approach for high-level production of tryptophan.Similar strategies could be applied to the production of other amino acids or related derivatives. | Zhu Li Dongqin Ding Huiying Wang Linxia Liu Huan Fang Tao Chen Dawei Zhang | 2020 | Synthetic and Systems Biotechnology2020,5,3: | 5 |

| 13 | Word Segmentation for Chinese Judicial Documents显示文摘Word segmentation is an integral step in many knowledge discovery applications. However, existing word segmentation methods have problems when applying to Chinese judicial documents:(1) existing methods rely on large-scale labeled data which is typically unavailable in judicial documents, and (2) judicial document has its own language features and writing formats. In this paper, a word segmentation method is proposed for Chinese judicial documents. The proposed method consists of two steps:(1) automatically generating some labeled data as legal dictionaries, and (2) applying a hybrid multilayer neural networks to do word segmentation incorporating legal dictionaries. | 14 | Fungal diversity and mycotoxins identification of mouldy rapeseed in China显示文摘Investigation on fungal populations and mycotoxins of mouldy rapeseed was conducted in this paper. A total of 578 rapeseed samples were collected from 11 Chinese provinces in 2015, and 141 samples were contaminted by mycete. Mouldy rapeseed samples with exceeding specified standard were found in all areas. The highest percentages of samples were from Hunan, Jiangxi and Hubei provinces with above standard rate of 31.91%, 24.54% and 20.45% respectively. Mycete in 29 severely mouldy samples were isolatedand determined by using dilution method. Eighty-three dominant isolates were found,among which fungal strain number ranged from 1 to 8. Aspergillus flavus, Penicillium steckii and Penicillium citrinum were dominant myco-populations identified by morphological and molecular biological methods. Rapeseed from the Southern provinces including Jiangxi,Hunan and Guangxi were contaminated by more isolates. Mycotoxins content were evaluated by LC–MS/MS. AFB1, BEA and FB1 were the top 3 prevalent mycotoxins. BEA and FB1 contaminations in rapeseed were relatively low. | 15 | Amplitude-frequency-aware deep fusion network for optimal contact selection on STN-DBS electrodes显示文摘Parkinson’s disease(PD)is treated effectively by deep brain stimulation(DBS)of the subthalamic nucleus(STN),using an electrode inserted into the head of a PD patient.The electrode has multiple electrical contacts along its length,so the best may be chosen for selectively stimulating the STN.Neurosurgeons usually determine the optimal stimulated contact via the clinical experience of the neurosurgeon and the motor improvement of PD patients.This is a time-consuming and labor-intensive trial-and-error process.The selection of optimal stimulated contact highly depends on the locations of sweet spots,which are manually identified by the characteristic features of microelectrode recordings(MERs).This paper presents an amplitude-frequency-aware deep fusion network for optimal contact selection on STN-DBS electrodes.The method first obtains the amplitude-frequency fusion features by combining the MERs time sequence features and the amplitude sequence features,and then uses the convolutional neural network(CNN)with convolutional block attention module(CBAM)to identify both the border of the STN and the sweet spots to implant the electrode.The optimal stimulated contact can be selected according to the distribution of the sweet spots.Experimental results indicate that,for successful surgeries,neurosurgeons and the proposed AI solution selected the same optimal contacts.Furthermore,the proposed method outperforms the state-of-the-art methods for STN and sweet spot identification.The proposed method shows great potential for optimal contact selection to improve the efficiency of STN-DBS surgery and reduce the dependence on clinicians’experience. | Linxia XIAO Caizi LI Yanjiang WANG Weixin SI Hai LIN Doudou ZHANG Xiaodong CAI Pheng-Ann HENG | 2022 | Science China(Information Sciences)2022,65,4: | 0 |
| 16 | Preliminary security investigation and short-time follow-up of intraoperative intraperitoneal chemotherapy with lobaplatin for advanced colorectal cancer显示文摘Objective The aim of this study was to conduct a security assessment of intraoperative intraperitoneal chemotherapy using lobaplatin for advanced colorectal cancer.Methods From February 2015 to February 2016,143 patients with colorectal cancer who underwent surgery in our department were selected prospectively.All patients were randomly screened and enrolled into the intraperitoneal chemotherapy(IPC)(74 cases)and control(69 cases)groups,depending on the distribution of cases in the random table.In the trial group,patients were administered 40 mg lobaplatin by intraperitoneal implantation intraoperatively,together with intravenous chemotherapy post-operatively using a typical FOLFOX strategy with oxaliplatin,fluorouracil,and leucovorin.In the control group,only FOLFOX was administered.Bowel function recovery time,adverse reactions and complications,and preand post-chemotherapy laboratory examinations were compared.In addition,a 5-year-long follow-up was performed.Results Recovery times of bowel function were 73.5±9.7 h and 74.8±10.3 h respectively,and the difference was not significant(P>0.05).Wound fat liquefaction was observed in five cases in both groups(6.8%vs.7.2%,P>0.05).The outcomes of nausea and vomiting(57 cases,77.0%vs.50 cases,72.5%),constipation(43 cases,58.1%vs.36 cases,52.2%),and diarrhea(5 cases,6.8%vs.5 cases,7.2%)were not statistically significant(all P>0.05).Indices of white blood cell count,blood platelet count,and hepatorenal function were not significantly different(all P>0.05)neither post-operatively nor post-chemotherapy.The 5-year survival rate was not significantly different between the groups(58.1%vs.56.5%,P>0.05).Conclusion Intraoperative chemotherapy with lobaplatin for advanced colorectal cancer is safe and tolerable. | Qin Li Xianrong Li Libo Feng Xiaolong Chen Dong Xia Linxia Xu | 2023 | Oncology and Translational Medicine2023,9,2: | 0 |
| 17 | Single-cell RNA-seq of Lotus japonicus provide insights into identification and function of root cell types of legume显示文摘The roots of legume plant play a crucial role in nitrogen fixation. However, the transcriptomes of different cell types of legume root and their functions remain largely unknown. Here, we performed single-cell RNA sequencing and profiled more than 22,000 single cells from root tips of Lotus japonicus, a model species of legume.We identified seven clusters corresponding to seven major cell types, which were validated by in situ hybridization. Further analysis revealed regulatory programs including phytohormone and nodulation associated with specific cell types, and revealed conserved and diverged features for the cell types.
