2The RING-H2 gene LdXERICO plays a negative role in dormancy release regulated by low temperature in Lilium davidii var. unicolor显示文摘Dormancy regulation is the basis of the sustainable development of the lily industry.Therefore,basic research on lily dormancy is crucial for innovation in lily cultivation and breeding.Previous studies revealed that dormancy release largely depends on abscisic acid(ABA)degradation.However,the key genes and potential regulatory network remain unclear.We used exogenous ABA and ABA inhibitors to elucidate the effect of ABA on lily dormancy.Based on the results of weighted gene coexpression network analysis(WGCNA),the hub gene LdXERICO was identified in modules highly related to endogenous ABA,and a large number of coexpressed genes were identified.LdXERICO was induced by exogenous ABA and expressed at higher levels in tissues with vigorous physiological activity.Silencing LdXERICO increased the low-temperature sensitivity of bulblets and accelerated bulblet sprouting.LdXERICO rescued the ABA insensitivity of xerico mutants during seed germination in Arabidopsis,suggesting that it promotes seed dormancy and supporting overexpression studies on lily bulblets.The significant increase in ABA levels in transgenic Arabidopsis expressing LdXERICO indicated that LdXERICO played a role by promoting ABA synthesis.We generated three transgenic lines by overexpressing LdICE1 in Arabidopsis thaliana and showed that,in contrast to LdXERICO,LdICE1 positively regulated dormancy release.Finally,qRT-PCR confirmed that LdXERICO was epistatic to LdICE1 for dormancy release.We propose that LdXERICO,an essential gene in dormancy regulation through the ABA-related pathway,has a complex regulatory network involving temperature signals.This study provides a theoretical basis for further exploring the mechanism of bulb dormancy release.Xinyue Fan Xiaoman Zou Linlan Fu Yue Yang Min Li Chunxia Wang Hongmei Sun 2023Horticulture Research2023,10,4:0
3Photocatalytic oxidative of Keggin-type polyoxometalate ionic liquid for enhanced extractive desulfurization in binary deep eutectic solvents显示文摘A series of novel binary deep eutectic solvents(DESs)composed of choline chloride(ChCl)and formic acid(HCOOH)with different molar ratios have been successfully synthesized and applied in extractive desulfurization(EDS).Keggin-type polyoxometallate ionic liquid[TTPh]_(3)PW_(12)O_(40) was prepared and used as catalyst to enhance the EDS capacity by means of photocatalytic oxidative process.Both of the DESs and[TTPh]_(3)PW_(12)O_(40) ionic liquid catalyst were characterized in detail by Fourier transform infrared spectroscopy spectra(FT-IR),elemental analysis,and X-ray photoelectron spectroscopy(XPS).It was found that the molar ratios of Ch Cl:HCOOH had a major impact on desulfurization performance,and the optimal desulfurization capacity 96.5%was obtained by ChCl/5 HCOOH.Besides dibenzothiophene(DBT),the desulfurization efficiencies of 4-methylbenzothiophene(4-MDBT)and 4,6-dimethyldibenzothiophene(4,6-DMDBT),two kinds of DBT derivatives,were also investigated under the same experimental conditions.Moreover,the free radical scavenging experiments manifested that superoxide radical(·O_(2)^(-)) and hole(h^(+)) played important roles in the desulfurization system.After further analysis of the oxidation products by gas chromatography-mass spectrometry(GC–MS),the possible reaction mechanism was proposed.Thus,photocatalytic oxidative has been proved to be one of the efficient approaches for enhancing the extractive desulfurization performance in DES.Linlan Wu Zhengxin Jiao Suhang Xun Minqiang He Lei Fan Chao Wang Wenshu Yang Wenshuai Zhu Huaming Li 2022Chinese Journal of Chemical Engineering2022,35,4:0
4Enhanced lysosome escape mediated by 1,2-dicarboxylic-cyclohexene anhydride-modified poly-L-lysine dendrimer as a gene delivery system显示文摘Antisense oligodeoxynucleotide(ASODN)can directly interfere a series of biological events of the target RNA derived from tumor cells through Watson-Crick base pairing,in turn,plays antitumor therapeutic roles.In the study,a novel HIF-1αASODN-loaded nanocomposite was formulated to efficiently deliver gene to the target RNA.The physicochemical properties of nanocomposite were characterized using TEM,FTIR,DLS and zeta potentials.The mean diameter of resulting GEL-DGL-FA-ASODN-DCA nanocomposite was about 170–192 nm,and according to the agarose gel retardation assay,the loading amount of ASODN accounted for 166.7 mg/g.The results of cellular uptake showed that the nanocomposite could specifically target to HepG2 and Hela cells.The cytotoxicity assay demonstrated that the toxicity of vectors was greatly reduced by using DCA to reversibly block the cationic DGL.The subcellular distribution images clearly displayed the lysosomal escape ability of the DCA-modified nanocomposite.In vitro exploration of molecular mechanism indicated that the nanocomposite could inhibit m RNA expression and HIF-1αprotein translation at different levels.In vivo optical images and quantitative assay testified that the formulation accumulated preferentially in the tumor tissue.In vivo antitumor efficacy research confirmed that this nanocomposite had significant antitumor activity and the tumor inhibitory rate was 77.99%.These results manifested that the GEL-DGL-FA-ASODNDCA nanocomposite was promising in gene therapeutics for antitumor by interacting directly with target RNA.Jianmin Shen Jing Chen Jingbo Ma Linlan Fan Xiaoli Zhang Ting Yue Yaping Yan Yuhang Zhang 2020Asian Journal of Pharmaceutical Sciences2020,15,6:0
