1Augmented cellular uptake and homologous targeting of exosome-based drug loaded IOL for posterior capsular opacification prevention and biosafety improvement显示文摘Posterior capsular opacification(PCO),the most common complication after cataract surgery,is caused by the proliferation,migration and differentiation of residual lens epithelial cells(LECs)on the surface of the intraocular lens(IOL).Although drug-loaded IOLs have been successfully developed,the PCO prevention efficacy is still limited due to the lack of targeting and low bioavailability.In this investigation,an exosome-functionalized drug-loaded IOL was successfully developed for effective PCO prevention utilizing the homologous targeting and high biocompatibility of exosome.The exosomes derived from LECs were collected to load the anti-proliferative drug doxorubicin(Dox)through electroporation and then immobilized on the aminated IOLs surface through electrostatic interaction.In vitro experiments showed that significantly improved cellular uptake of Dox@Exos by LECs was achieved due to the targeting ability of exosome,compared with free Dox,thus resulting in superior anti-proliferation effect.In vivo animal investigations indicated that Dox@Exos-IOLs effectively inhibited the development of PCO and showed excellent intraocular biocompatibility.We believe that this work will provide a targeting strategy for PCO prevention through exosome-functionalized IOL.Siqing Zhu Huiying Huang Dong Liu Shimin Wen Liangliang Shen Quankui Lin 2022Bioactive Materials2022,7,9:1

6Heat Insulation and Dissipation Processes in Nordic Seas in the Summer显示文摘The Nordic Seas have a significant impact on the climate system.Here 23-day air-sea heat fluxes were analyzed from an in situ air-sea coupled buoy deployed in the Lofoten Basin from 5 August 2012 to 27 August 2012.The buoy captured two stages of strong south and north winds.The observations indicate that warm and wet air transported by the south wind can lead to decreased sensible and latent heat fluxes and net longwave radiation.The total oceanic heat loss was 50-60Wm−2.Thus,this stage was called the heat insulation process.By contrast,the heat dissipation process occurred with the north wind condition dur-ing advection of the cold and dry air.During this process,sensible and latent heat fluxes and net longwave radiation notably in-creased.The total oceanic heat loss during the heat dissipation process reached 240Wm−2,which was four-fold greater than that in the heat insulation process.Given that the heat insulation process is dominant in summertime,the ocean lost minimal heat but absorbed strong solar energy.Thus,a large quantity of energy was stored in the ocean.Heat was transported to the Arctic Ocean and accelerated Arctic warming.The heat dissipation process is dominant in autumn and winter when the ocean releases consid-erably more energy.The two processes revealed in this paper can be applied to warm-water areas in high latitudes.GAO Lin ZHAO Jinping LI Shimin FAN Xiutao LIU Shixuan 2021Journal of Ocean University of China2021,20,6:1
7Inactivation of OslRX10 Leads to Decreased Xylan Content in Rice Culm Cell Walls and Improved Biomass Saccharification显示文摘Dear Editor, Xylan polysaccharides constitute the major non-cellulosic components in secondary cell walls of dicots and in both primary and secondary cell walls of grasses (Scheller and Ulvskov,2010).Xylan is composed of a linear backbone of 13(1-4)-linked xylose (Xyl).In grasses,the xylan backbone is substituted with O3-1inked and O2-1inked arabinose residues and,to a lesser extent,α(1-3)-Iinked glucuronic acid residues.A unique feature of grass xylans is the presence of hydroxycinnamate esters (ferulic and p-coumaric acid residues)attached to C5 of some of the arabinosyl residues (Carpita,1996;Vogel,2008;Scheller and Ulvskov,2010).Xylan contributes to the recalcitrance of plant cell walls to enzymatic degradation probably through direct interaction with cellulose microfibrils and also through formation of ferulate cross-links (liyama et al.,1994;Yin et al.,2011).Xuewei Chen Miguel E. 9Magnesium incorporation activates perovskite cobaltites toward efficient and stable electrocatalytic oxygen evolution显示文摘Cobalt-rich perovskite oxides play a paramount role in catalyzing oxygen evolution reaction(OER)on account of their acceptable intrinsic activity but are still challenging due to the high costs and undesired stability.In response to the defects,herein,the Mg-incorporated perovskite cobaltite SrCo_(0.6)Fe_(0.3M)g_(0.1)O_(3-δ)(SCFM-0.1)is proposed as a novel earth-abundant and durable OER electrocatalyst.A well-consolidated cubic-symmetry structure and more active oxygen intermediates are enabled upon Mg substitution.Hence,the optimized SCFM-0.1 perovskite oxide achieves prominent OER electrocatalytic performance,that is,a low overpotential of only 320 mV at 10 mA cm^(-2),a small Tafel slope of 65 mV dec^(-1),as well as an outstanding durability within 20 h,substantially outperforming that of the pristine SrCo_(0.7)Fe_(0.3)O_(3-δ)and benchmark Ba_(0.5)Sr_(0.5)Co_(0.8)Fe_(0.2)O_(3-δ)and IrO_(2) catalysts.The strong pHdependent behavior associated with lattice oxygen activation mechanism for SCFM-0.1 catalyst is also confirmed.This work paves a unique avenue to develop cost-effective and robust perovskite cobaltites for efficient OER electrocatalysis.Siyu Pan Zilin Ma Wenying Yang Biaokui Dongyang Huizhi Yang Shimin Lai Feifei Dong Xixian Yang Zhan Lin 2023Materials Reports(Energy)2023,3,3:0

13Hypoxia induces mitochondrial protein lactylation to limit oxidative phosphorylation显示文摘Oxidative phosphorylation(OXPHOS)consumes oxygen to produce ATP.However,the mechanism that balances OXPHOS activity and intracellular oxygen availability remains elusive.Here,we report that mitochondrial protein lactylation is induced by intracellular hypoxia to constrain OXPHOS.We show that mitochondrial alanyl-tRNA synthetase(AARS2)is a protein lysine lactyltransferase,whose proteasomal degradation is enhanced by proline 377 hydroxylation catalyzed by the oxygen-sensing hydroxylase PHD2.Hypoxia induces AARS2 accumulation to lactylate PDHA1 lysine 336 in the pyruvate dehydrogenase complex and carnitine palmitoyltransferase 2(CPT2)lysine 457/8,inactivating both enzymes and inhibiting OXPHOS by limiting acetyl-CoA influx from pyruvate and fatty acid oxidation,respectively.PDHA1 and CPT2 lactylation can be reversed by SIRT3 to activate OXPHOS.In mouse muscle cells,lactylation is induced by lactate oxidation-induced intracellular hypoxia during exercise to constrain high-intensity endurance running exhaustion time,which can be increased or decreased by decreasing or increasing lactylation levels,respectively.Our results reveal that mitochondrial protein lactylation integrates intracellular hypoxia and lactate signals to regulate OXPHOS.Yunzi Mao Jiaojiao Zhang Qian Zhou Xiadi He Zhifang Zheng Yun Wei Kaiqiang Zhou Yan Lin Haowen Yu Haihui Zhang Yineng Zhou Pengcheng Lin Baixing Wu Yiyuan Yuan Jianyuan Zhao Wei Xu Shimin Zhao 2024Cell Research2024,34,1:0
