| 1 | The cartilage matrisome in adolescent idiopathic scoliosis显示文摘The human spinal column is a dynamic,segmented,bony,and cartilaginous structure that protects the neurologic system and simultaneously provides balance and flexibility.Children with developmental disorders that affect the patterning or shape of the spine can be at risk of neurologic and other physiologic dysfunctions.The most common developmental disorder of the spine is scoliosis,a lateral deformity in the shape of the spinal column.Scoliosis may be part of the clinical spectrum that is observed in many developmental disorders,but typically presents as an isolated symptom in otherwise healthy adolescent children.Adolescent idiopathic scoliosis(AIS)has defied understanding in part due to its genetic complexity.Breakthroughs have come from recent genome-wide association studies(GWAS)and next generation sequencing(NGS)of human AIS cohorts,as well as investigations of animal models.These studies have identified genetic associations with determinants of cartilage biogenesis and development of the intervertebral disc(IVD).Current evidence suggests that a fraction of AIS cases may arise from variation in factors involved in the structural integrity and homeostasis of the cartilaginous extracellular matrix(ECM).Here,we review the development of the spine and spinal cartilages,the composition of the cartilage ECM,the so-called'matrisome'and its functions,and the players involved in the genetic architecture of AIS.We also propose a molecular model by which the cartilage matrisome of the IVD contributes to AIS susceptibility. | Carol AWise Diane Sepich Aki Ushiki Anas MKhanshour Yared HKidane Nadja Makki Christina AGurnett Ryan SGray Jonathan JRios Nadav Ahituv Lila Solnica-Krezel | 2020 | Bone Research2020,8,1: | 5 |
| 2 | Antihydatic and immunomodulatory effects of Punica granatum peel aqueous extract in a murine model of echinococcosis显示文摘Objective:To investigate the effect of pomegranate peel aqueous extract(PGE) on the development of secondary experimental echinococcosis and on the viability of Echinococcux granulosus protoscoleces,and the immunomodulatory properties of PGE.Methods:Swiss mice were inoculated intraperitoneally with viable protoscoleces.Then,PGE was orally administered daily during cystic echinococcosis development.Cyst development and hepatic damage were macroscopically and histologically analyzed.The production of nitric oxide and TNF-α was assessed in plasma and the hepatic expression of iNOS.TNF-α,NF-κB and CD68 was examined.Moreover,protoscoleces were cultured and treated with different concentrations of PGE.Results:It was observed that in vitro treatment of protoscoleces caused a significant decrease in viability in a PGE-dose-dependent manner.In vivo,after treatment of cystic echinococcosis infected mice with PGE,a significant decrease in nitric oxide levels(P<0.000 1)and TNF-α levels(P<0.001) was observed.This decline was strongly related to the inhibition of cyst development(rate of hydatid cyst growth inhibition=63.08%) and a decrease in CD68 expression in both the pericystic layer of hepatic hydatid cysts and liver tissue(P<0.000 1).A significant diminution of iNOS,TNF-α and NF-κB expression was also observed in liver tissue of treated mice(P<0.000 1).Conclusions:Our results indicate an antihydatic scolicidal effect and immunomodulatory properties of PGE,suggesting its potential therapeutic role against Echinococcuss granulosus infection. | Moussa Labsi Lila Khelifi Dalila Mezioug Imene Soufli Chafia Touil-Boukoffa | 2016 | Asian Pacific Journal of Tropical Medicine2016,9,3: | 5 |
| 3 | The functional role of miRNAs in colorectal cancer: insights from a large population-based study显示文摘Identification of causal microRNAs(miRNAs) in colorectal cancer(CRC) is elusive, due to our lack of understanding of how specific miRNAs affect biological pathways and outcomes. An miRNA can regulate many mRNAs and an mRNA can be associated with many miRNAs;appreciation of these complex networks in which miRNAs operate is necessary to transition from identifying dysregulated miRNAs to identifying individual miRNAs or groups of miRNAs that are suitable for therapeutic purposes. The aim of the paper is to compile results from a population-based study(n = 1,954 cases with matched carcinoma/normal tissue) of miRNAs in CRC. The information gained allows for cohesive and comprehensive insight into miRNAs and CRC in terms of function and impact. Comparison of miRNA expression with mRNA expression from nine signaling pathways in carcinogenic processes allowed us to identify miRNA targets within a biological context. | 5 | High-density linkage map construction and identification of loci regulating fruit quality traits in blueberry显示文摘Fruit quality traits play a significant role in consumer preferences and consumption in blueberry(Vaccinium corymbosum L).The objectives of this study were to construct a high-density linkage map and to identify the underlying genetic basis of fruit quality traits in blueberry.A total of 287 F_(1) individuals derived from a cross between two southern highbush blueberry cultivars,‘Reveille’and‘Arlen’,were phenotyped over three years(2016–2018)for fruit quality-related traits,including titratable acidity,pH,total soluble solids,and fruit weight.A high-density linkage map was constructed using 17k single nucleotide polymorphisms markers.The linkage map spanned a total of 1397 cM with an average inter-loci distance of 0.08 cM.The quantitative trait loci interval mapping based on the hidden Markov model identified 18 loci for fruit quality traits,including seven loci for fruit weight,three loci for titratable acidity,five loci for pH,and three loci for total soluble solids.Ten of these loci were detected in more than one year.These loci explained phenotypic variance ranging from 7 to 28% for titratable acidity and total soluble solid,and 8-13% for pH.However,the loci identified for fruit weight did not explain more than 10%of the phenotypic variance.We also reported the association between fruit quality traits and metabolites detected by Proton nuclear magnetic resonance analysis directly responsible for these fruit quality traits.Organic acids,citric acid,and quinic acid were significantly(P<0.05)and positively correlated with titratable acidity.Sugar molecules showed a strong and positive correlation with total soluble solids.Overall,the study dissected the genetic basis of fruit quality traits and established an association between these fruit quality traits and metabolites. | Molla F.Mengist Hamed Bostan Elisheba Young Kristine L.Kay Nicholas Gillitt James Ballington Colin D.Kay Mario G.Ferruzzi Hamid Ashrafi Mary Ann Lila Massimo Iorizzo | 2021 | Horticulture Research2021,8,1: | 2 |

| 16 | Field assessment of dog as sentinel animal for plague in endemic foci of Madagascar显示文摘The epidemiology of Yersinia pestis,the causative agent of plague,involves vectors and reservoirs in its transmission cycle.The passive plague surveillance in Madagascar targets mainly rodent and fleas.However,carnivores are routinely surveyed as sentinels of local plague activity in some countries.The aim of this study is to assess the use of domestic dog(Canis familiaris)as sentinel animal for field surveillance of plague in a highly endemic area in Madagascar.Cross-sectional surveys of plague antibody prevalence in C.familiaris were conducted in endemic areas with contrasting histories of plague cases in humans,as well as a plague free area.Rodent capture was done in parallel to evaluate evidence for Y.pestis circulation in the primary reservoirs.In 2 sites,dogs were later re-sampled to examine evidence of seroconversion and antibody persistence.Biological samplings were performed between March 2008 and February 2009.Plague antibody detection was assessed using anti-F1 ELISA.Our study showed a significant difference in dog prevalence rates between plague-endemic and plague-free areas,with no seropositive dogs detected in the plague free area.No correlation was found between rodents and dog prevalence rates,with an absence of seropositive rodents in some area where plague circulation was indicated by seropositive dogs.This is consistent with high mortality rates in rodents following infection.Re-sampling dogs identified individuals seropositive on both occasions,indicating high rates of re-exposure and/or persistence of plague antibodies for at least 9 months.Seroconversion or seropositive juvenile dogs indicated recent local plague circulation.In Madagascar,dog surveillance for plague antibody could be useful to identify plague circulation in new areas or quiescent areas within endemic zones.Within active endemic areas,monitoring of dog populations for seroconversion(negative to positive)or seropositive juvenile dogs could be useful for identifying areas at greatest risk of human outbreaks. | Minoarisoa RAJERISON Voahangy ANDRIANAIVOARIMANANA Maherisoa RATSITORAHINA Soanandrasana RAHELINIRINA Suzanne CHANTEAU Sandra TELFER Lila RAHALISON | 2021 | Integrative Zoology2021,16,6: | 1 |
