1PCN-Fe(Ⅲ)-PTX nanoparticles for MRI guided high efficiency chemo-photodynamic therapy in pancreatic cancer through alleviating tumor hypoxia显示文摘As nanomedicine-based clinical strategies have continued to develop,the possibility of combining chemotherapy and singlet oxygen-dependent photodynamic therapy(PDT)to treat pancreatic cancer(PaC)has emerged as a viable therapeutic modality.The efficacy of such an approach,however,is likely to be constrained by the mechanisms of drug release and tumor oxygen levels.In the present study,we developed an Fe(Ⅲ)-complexed porous coordination network(PCN)which we then used to encapsulate PTX(PCN-Fe(Ⅲ)-PTX)nanoparticles(NPs)in order to treat PaC via a combination of chemotherapy and PDT.The resultant NPs were able to release drug in response to both laser irradiation and pH changes to promote drug accumulation within tumors.Furthermore,through a Fe(Ⅲ)-based Fenton-like reaction these NPs were able to convert H2O2 in the tumor site to O2,thereby regulating local hypoxic conditions and enhancing the efficacy of PDT approaches.Also these NPs were suitable for use as a T1-MRI weighted contrast agent,making them viable for monitoring therapeutic efficacy upon treatment.Our results in both cell line and animal models of PaC suggest that these NPs represent an ideal agent for mediating effective MRI-guided chemotherapy-PDT,giving them great promise for the clinical treatment of PaC.Tao Zhang Zhenqi Jiang Libin Chen Chunshu Pan Shan Sun Chuang Liu Zihou Li Wenzhi Ren Aiguo Wu Pintong Huang 2020Nano Research2020,13,1:4
2Domesticated cynomolgus monkey embryonic stem cells allow the generation of neonatal interspecies chimeric pigs显示文摘Blastocyst complementation by pluripotent stem cell(PSC)injection is believed to be the most promising method to generate xenogeneic organs.However,ethical issues prevent the study of human chimeras in the late embryonic stage of development.Primate embryonic stem cells(ESCs),which have similar pluripotency to human ESCs,are a good model for studying interspecies chimerism and organ generation.However,whether primate ESCs can be used in xenogenous grafts remains unclear.In this study,we evaluated the chimeric ability of cynomolgus monkey(Macaca fascicularis)ESCs(cmESCs)in pigs,which are excellent hosts because of their many similarities to humans.We report an optimized culture medium that enhanced the anti-apoptotic ability of cmESCs and improved the development of chimeric embryos,in which domesticated cmESCs(D-ESCs)injected into pig blastocysts differentiated into cells of all three germ layers.In addition,we obtained two neonatal interspecies chimeras,in which we observed tissue-specific D-ESC differentiation.Taken together,the results demonstrate the capability of D-ESCs to integrate and differentiate into functional cells in a porcine model,with a chimeric ratio of 0.001-0.0001 in different neonate tissues.We believe this work will facilitate future developments in xenogeneic organogenesis,bringing us one step closer to producing tissue-specific functional cells and organs in a large animal model through interspecies blastocyst complementation.Rui Fu Dawei Yu Jilong Ren Chongyang Li Jing Wang Guihai Feng Xuepeng Wang Haifeng Wan Tianda Li Libin Wang Ying Zhang Tang Hai Wei Li Qi Zhou 2020Protein & Cell2020,11,2:4
3Construction of polysulfides defense system for greatly improving the long cycle life of metal sulfide anodes for sodium-ion batteries显示文摘Metal sulfides are promising anode materials for sodium ion batteries(SIBs)due to their high theoretical specific capacity and abundant source.Nevertheless,significant challenges,including large volume change,sluggish Na^(+)transport kinetics and polysulfides intermediates,have greatly affect their long cycle stability.Unfortunately,the majority of current studies only focus on the first two aspects,but lack of sufficient attention and insights into the effect of polysulfides intermediates.Here,a porous of CoS_(x)(P-CoS_(x))electrode material is fabricated as an example to investigate the influence of polysulfides on its cycling performance.The results show that polysulfides cause a slight loss of reversible capacity during the battery cycling,while the failure of the battery is due to its significant fluctuations in reversible capacity after extensive cycles.Detailed analyses demonstrate that the intense fluctuation in capacity originates from the faster growth of dendrites caused by the reaction of sodium polysulfides with sodium foil and/or the reaction of elemental sulfur with sodium foil to penetrate the separator,resulting in a local short circuit.To suppress these undesirable side reaction,N,S co-doped porous carbon tubes(N,S-PC)rich in C–S and C–N bonds have been added to adsorb polysulfides and alleviate their reaction with sodium foil.As a result,the capacity of the P-CoS_(x) electrode with N,S-PC(P-CoS_(x)/N,S-PC)remains stable without significant fluctuations for 1000 cycles,which is much better than that of the pure P-CoS_(x) electrode(intense fluctuation in capacity after 320 cycles).Our work offers insights into the crucial influence of polysulfides on the cycle performance of the P-CoS_(x) anode and provides a feasible strategy to prolong the cycle life of metal sulfide anode for SIBs.Xucai Yin Yang Ren Libin Wu Zhiguo Zhang Chunyu Du Jiajun Wang Gepin Yin Hua Huo 2022Journal of Energy Chemistry2022,31,8:1
4A joint call for actions to advance taxonomy in China显示文摘Taxonomy plays an important role in understanding the origin, evolution, and ecological functionality of biodiversity. There are large number of unknown species yet to be described by taxonomists, which together with their ecosystem services cannot be effectively protected prior to description. Despite this, taxonomy has been increasingly underrated insufficient funds and permanent positions to retain young talents. Further, the impact factordriven evaluation systems in China exacerbate this downward trend, so alternative evaluation metrics are urgently necessary. When the current generation of outstanding taxonomists retires,there will be too few remaining taxonomists left to train the next generation. 6Pathogenic Effects of Cloned Genomic DNA of Porcine Circovirus-like Virus P1 on Neonatal Mice via Different Inoculation Routes显示文摘[Objective] The paper was to explore the pathogenicity of cloned genomic DNA of porcine circovirus-like virus P1 to neonatal mice via different inoculation routes(brain, liver and muscle). [Method] Cloned genomic DNA of P1 was inoculated to neonatal mice via different routes of brain, liver and muscle. Tissues of heart, liver, spleen, lung, kidney and brain were taken from neonatal mice at 7, 14 and 21 d post inoculation, re-spectively. P1 in various tissues were qualitatively and quantitatively detected by using ordinary PCR and quantitative real-time PCR. Meanwhile,histopathological changes were analyzed. [Result] P1 was detected in neonatal mice inoculated through three different routes. The viral load of tis-sues at 7 d post inoculation was significantly higher than those at 14 and 21 d post inoculation. Moreover, muscle inoculation led to the highest vi-ral load in all tissues of neonatal mice. 7Processable Potassium Metal Anode for Stable Batteries显示文摘The future of high-energy density electrochemical energy storage systems relies on the advancement of rechargeable batteries that utilize reactive metals as anodes.In the alkaline metal,secondary battery systems because of abundant resource,high capacity and low redox potential,potassium(K)metal secondary battery(KMB)is expected to replace the existing lithiumion battery as a versatile platform for high-energy density,cost-effective energy storage devices.However,the difficulty in processing metal K results in nonstandard electrodes and hinders the development of KMBs.Furthermore,the mobility of the K metal anode due to its unique lowmelting point character at elevated temperatures in practical conditions leads to severe instability and risks in chemical/electrochemical processes.Herein,we fabricate a processable and moldable composite K metal anode by encapsulating K into reduced graphene oxide(rGO).The composite electrode can be engineered into various shapes discretionarily with precise sizes and stabilize the K metal anode at relatively high temperatures.Remarkably,the composite anode exhibits excellent cycling performance at high current density(8 mA cm^(-2)) with dendrite-free morphology.Paired with a Prussian blue cathode,the rGO-K composite anode shows much improved electrochemical performance and extended lifetime.Zhenghang Wei Aoxuan Wang Xuze Guan Guojie Li Zhiwei Yang Chengde Huang Jing Zhang Libin Ren Jiayan Luo Xingjiang Liu 2022Energy & Environmental Materials2022,5,4:0
8Revealing the solid electrolyte interface on calcium metal anodes显示文摘Owing to its low potential, crustal abundances and environmental friendliness, calcium metal anode(CMA) is emerging as a powerful contender in post-lithium era. However, the passivation of CMA fatally hinders its development. Recently, several feasible electrolytes have been developed. Nevertheless, as a pivotal part, the solid electrolyte interface(SEI) formed on CMA has not been paid enough attention to. In this review, based on the passivation mechanism of CMA, the favorable composition of SEI is emphasized with the corresponding electrolytes. It is considered that boron-containing and organic–inorganic hybrid SEI might be preferred. By comparing electrolytes and SEI on CMA with lithium and magnesium metal anodes, the root causes of CMA passivation are further elaborated, enlightening rational design rules of suitable SEI. Furthermore, some noteworthy details when assembling secondary calcium metal batteries(CMBs) are put forward.
