1内毒素诱导大鼠葡萄膜炎的组织平片和切片研究显示文摘目的:探讨内毒素诱导的视网膜脉络膜改变和平片、切片结果的差异。方法:从内毒素注射的Lewis鼠制备视网膜和脉络膜-巩膜复合体平片以及眼组织冰冻切片,并进行免疫组织化学染色。结果:平片上显示,注射后4小时单核细胞浸润,随后,大量的单核巨噬细胞浸润至整个视网膜和脉络膜,脉络膜中尚有MHC-Ⅱ类抗原阳性细胞增多;切片上发现,于注射后一定时期,视网膜和脉络膜中有上述细胞浸润。结论:平片技术为视网膜脉络膜原发性严重受累提供了确凿的证据,内毒素诱导的葡萄膜炎可作为人类全葡萄膜炎的动物模型。杨培增 Alex F. de Vos Lidy Broersma Aize kijlstra 1996中华眼底病杂志1996,12,1:16
2Bone tissue engineering via growth factor delivery:from scaffolds to complex matrices显示文摘In recent years,bone tissue engineering has emerged as a promising solution to the limitations of current gold standard treatment options for bone related-disorders such as bone grafts.Bone tissue engineering provides a scaffold design that mimics the extracellular matrix,providing an architecture that guides the natural bone regeneration process.During this period,a new generation of bone tissue engineering scaffolds has been designed and characterized that explores the incorporation of signaling molecules in order to enhance cell recruitment and ingress into the scaffold,as well as osteogenic differentiation and angiogenesis,each of which is crucial to successful bone regeneration.Here,we outline and critically analyze key characteristics of successful bone tissue engineering scaffolds.We also explore candidate materials used to fabricate these scaffolds.Different growth factors involved in the highly coordinated process of bone repair are discussed,and the key requirements of a growth factor delivery system are described.Finally,we concentrate on an analysis of scaffold-based growth factor delivery strategies found in the recent literature.In particular,the incorporation of two-phase systems consisting of growth factor-loaded nanoparticles embedded into scaffolds shows great promise,both by providing sustained release over a therapeutically relevant timeframe and the potential to sequentially deliver multiple growth factors.Tinke-Marie De Witte Lidy E.Fratila-Apachitei Amir A.Zadpoor Nicholas A.Peppas 2018Regenerative Biomaterials2018,5,4:12

4Lipopolysaccharide induced intestinal epithelial injury:a novel organoids-based model for sepsis in vitro显示文摘Background: Advances in organoid culture technology have provided a greater understanding of disease pathogenesis, which has been rarely studied in sepsis before. We aim to establish a suitable organoids-based intestinal injury model for sepsis.Methods: Stable passaged organoids were constructed and pre-treated with lipopolysaccharide (LPS) to mimic sepsis-induced intestinal injury. The LPS-induced sepsis model was used as a reference. We used quantitative real-time polymerase chain reaction to evaluate the RNA levels of inflammatory factors and antimicrobial peptides. Enzyme-linked immunosorbent assay was used to evaluate the protein levels, hematoxylin and eosin staining was used to evaluate the pathology of the small intestine of mice, and immunohistochemistry and immunofluorescence were used to evaluate the intestinal epithelial barrier function. Perkin Elmer OperettaTM was used to obtain high-resolution images of three-dimensional organoids.Results: An LPS concentration >150 μg/mL after 24 h was identified to cause organoid growth restriction. The fluorescence intensity of zonula occludens-1 and occludins at LPS concentrations >100 μg/mL decreased significantly after 24 h. After LPS stimulation for 8 h, the RNA expression levels of interleukin (IL)-1α, tumor necrosis factor alpha, granulocyte-macrophage colony-stimulating factor, IL-6, and regenerating islet-derived protein 3 alpha, beta, and gamma increased. These results resembled those of intestinal epithelial layer alterations in a mouse sepsis model. For IL-10, the RNA expression level increased only when the LPS level >200 μg/mL for 24 h.Conclusions: This study provides the primary intestinalin vitro model to study the effects of LPS-induced intestinal injury resembling sepsis.
