1Periostin mediates epithelial-mesenchymal transition through the MAPK/ERK pathway in hepatoblastoma显示文摘Objective: The aim of the present study was to analyze the prognostic factors in patients with hepatoblastoma(HB) in our single center and to evaluate periostin(POSTN) expression in HB and its association with clinicopathological variables. In addition, the underlying mechanism of how POSTN promotes HB progression was discussed.Methods: POSTN expression was investigated in HB tumors by immunohistochemistry(IHC), immunofluorescence(IF) and Western blot(WB). The association among POSTN expression, clinicopathological features and overall survival(OS) was also evaluated. The migration and adhesion ability of HB cells were measured using chemotaxis and cell-matrix adhesion assays,respectively. Epithelial-mesenchymal transition(EMT)-associated markers and activation of the ERK pathway were detected by WB.Results: HB patients had poor prognosis which displayed lymph node metastasis, vascular invasion, POSTN and vimentin expression. POSTN expression was also associated with lymph node metastasis. Furthermore, overexpressed POSTN promoted migration and the adhesive ability of HB cells in vitro. In addition, we demonstrated that POSTN activated the MAPK/ERK pathway, upregulated the expression of Snail and decreased the expression of OVOL2. 11Microstructure and mechanical properties of a novel Zr-based bulk metallic glass composite显示文摘Zr48.5Cu46.5Al5 bulk metallic glass (BMG) com- posites with diameters of 3 and 4 mm were prepared through suction casting in an arc melting furnace by modulating the alloy composition around the monothetic BMG composition of the high glass forming ability. Microstructural charac- terization reveals that the composites contain micron-sized CuZr phase with martensite structure, as well as nano-sized Zr2Cu crystalline particles and Cu10Zr7 plate-like phase em- bedded in an amorphous matrix. 15Mutations in Exons of the CYP17-Ⅱ Gene Affect Sex Steroid Concentration in Male Japanese Flounder(Paralichthys olivaceus)显示文摘As a specific gene of fish,cytochrome P450c17-Ⅱ(CYP17-Ⅱ) gene plays a key role in the growth,development and reproduction level of fish.In this study,the single-stranded conformational polymorphism(SSCP) technique was used to characterize polymorphisms within the coding region of CYP17-Ⅱ gene in a population of 75 male Japanese flounder(Paralichthys olivaceus).Three single nucleotide polymorphisms(SNPs) were identified in CYP17-Ⅱ gene of Japanese flounder.They were c.G594A(p.G188R),c.G939A and c.G1502A(p.G490D).SNP1(c.G594A),located in exon 4 of CYP17-Ⅱ gene,was significantly associated with gonadosomatic index(GSI).Individuals with genotype GG of SNP1 had significantly lower GSI(P < 0.05) than those with geno-type AA or AG.SNP2(c.G939A) located at the CpG island of CYP17-Ⅱ gene.The mutation changed the methylation of exon 6.Indi-viduals with genotype AA of SNP2 had significantly lower serum testosterone(T) level and hepatosomatic index(HSI) compared to those with genotype GG.The results suggested that SNP2 could influence the reproductive endocrine of male Japanese flounder.How-ever,the SNP3(c.G1502A) located in exon 9 did not affect the four measured reproductive traits.This study showed that CYP17-Ⅱ gene could be a potentially useful candidate gene for the research of genetic breeding and physiological aspects of Japanese flounder.MA Ruiqin HE Feng WEN Haishen LI Jifang SHI Bao SHI Dan LIU Miao MU Weijie ZHANG Yuanqing HU Jian HAN Weiguo ZHANG Jianan WANG Qingqing YUAN Yuren LIU Qun 2012Journal of Ocean University of China2012,11,1:0
16Manipulation of Colloidal Droplets in Space and the Instability of Thermocapillary Convection in Large Prandtl Number Liquid Bridge in Microgravity显示文摘In 2016 and 2017, SJ-10 and TG-2 satellites were launched. In this short paper, we report recent progress on the studies of manipulation of colloidal droplets and instability of thermocapillary convection in large Prandtl number liquid bridge that based on the space experiments boarding SJ-10 and TG-2 satellites,separately. It was shown that the colloidal droplets can be successfully formed and manipulated in microgravity through the patterned substrates. In another aspect, the coffee-ring effect was observed at the first time in space. 17Deformation behavior during nanoindentation in Ce-based bulk metallic glasses显示文摘The deformation behavior and the ef- fect of the loading rate on the plastic deformation in Ce-based bulk metallic glasses (BMGs) were inves- tigated through nanoindentation tests. The results showed that the loading rate dependence of plastic deformation during nanoindentation measurements in the Ce-based BMGs is quite unique in contrast to that of other BMG alloys. The load-displacement (P-h) curves of Ce60Al15Cu10Ni15 BMG exhibit a homoge- neous plastic deformation at low loading rates, and a prominent serrated flow at high strain rates, whereas, the P-h curves of Ce65Al10Cu10Ni10Nb5 exhibit ho- mogenous plastic deformation at all studied loading rates. The room temperature creep behavior could clearly be observed in these two alloys.
