| 1 | Permian–Triassic Conodonts from Dajiang(Guizhou, South China) and Their Implication for the Age of Microbialite Deposition in the Aftermath of the End-Permian Mass Extinction显示文摘The widespread microbialites deposition that followed the End-Permian mass extinction in the Tethyan realm have been intensively studied because of the evidence they provide on the nature of this crisis and its aftermath. However, the age of the microbialite event remains controversial. New conodont collection across the Permian–Triassic(P–T) transition from Dajiang(Guizhou Province, South China) in this study enable us to discriminate four conodont zones, in ascending order, they are: Hindeodus parvus zone, Isarcicella lobata zone, Isarcicella isarcica zone and Hindeodus sosioensis zone. The age of microbialite in the P–T transition at the Dajiang Section is considered to be within the Hindeodus parvus zone and thus to clearly post-date the main extinction crisis. Reviewing the age of onset of microbialites throughout the Tethyan regions reveals two different ages: a Hindeodus changxingensis zone age is dominant in south-western and westernmost Tethys, whilst most other regions show microbialite deposition began in the Hindeodus parvus zone. | 2 | Experimental small bowel preservation using Polysol: A new alternative to University of Wisconsin solution, Celsior and histidine-tryptophan-ketoglutarate solution?显示文摘AIM: To evaluate the potential of Polysol, a newly developed preservation solution, in cold storage of small bowel grafts, compared with the current standards, University of Wisconsin solution (UW), Celsior and histidine-tryptophan-ketoglutarate solution (HTK). METHODS: Male Wistar rats were used as donors. Small bowels were retrieved, flushed and then stored in the respective 4 solutions for 18 h at 4℃. Functional integrity of the grafts was evaluated by isolated reperfusion with oxygenated Krebs-Henseleit buffer at 37℃ for 30 min in all 4 groups. RESULTS: Polysol preservation exhibited the highest tissue ATP concentration and the lowest release of LDH. Malondialdehyde, an index for tissue lipid peroxidation, was also the lowest in Polysol. Tissue oxygen consumption was significantly higher in Polysol than in the others. Of interest, UW-storage promoted 10-fold higher apoptosis than in the others. Moreover, electron microscopy revealed that the mucosal villi/micro-villi formation and the cell organelles, including mitochondria, were both significantly better preserved in Polysol, while deleterious alterations were apparent in the others, most notably in UW. Although Celsior and HTK exhibited the better trend of results than UW in some parameters, but could not reach the over-all superiority to UW.CONCLUSION: Cold storage using Polysol resulted in significantly better integrity and function of small bowel grafts than UW. | 4 | The dependences of osteocyte network on bone compartment, age, and disease显示文摘Osteocytes, the most abundant bone cells, form an interconnected network in the lacunar-canalicular pore system(LCS) buried within the mineralized matrix, which allows osteocytes to obtain nutrients from the blood supply, sense external mechanical signals, and communicate among themselves and with other cells on bone surfaces. In this study, we examined key features of the LCS network including the topological parameter and the detailed structure of individual connections and their variations in cortical and cancellous compartments, at different ages, and in two disease conditions with altered mechanosensing(perlecan deficiency and diabetes).LCS network showed both topological stability, in terms of conservation of connectivity among osteocyte lacunae(similar to the ‘‘nodes' ' in a computer network), and considerable variability the pericellular annular fluid gap surrounding lacunae and canaliculi(similar to the ‘‘bandwidth'' of individual links in a computer network). Age, in the range of our study(15–32 weeks), affected only the pericellular fluid annulus in cortical bone but not in cancellous bone. Diabetes impacted the spacing of the lacunae, while the perlecan deficiency had a profound influence on the pericellular fluid annulus.
