1Epidemiology of Helicobacter pylori among multiracial community in Northern Peninsular,Malaysia:effect of age across race and gender显示文摘Objective:To study the epidemiology of Helicobacter pylori(H.pylori) infection according to age group.Methods:H.pylori infection data among 1 965 consecutive patients referred to the Endoscopy Unit collected at Sungai Petani Hospital for oesophagogastro-duodenoscopy(OGD). The patients were divided into 9 age groups(10-19,20-29,30-39,40-49,50-59,60-69,70-79, 80-89 and 90-99 years).In addition these groups were further divided into three minor group namely young adults(10-39),older adults(40-69) and geriatric groups(70-99).Results:Overall prevalence of infection of H.pylori was analyzed and found that the prevalence increase with age (P<0.05).When the patients divided by ethnic and gender group with age,prevalence rate among young adults and older adults significantly higher(P<0.05) compared to geriatric groups across all races and gender(P<0.05).Furthermore,significantly higher number of males were infected compared to female(P<0.05) but such trend was only observed among older adult groups.In addition,there is a significant differences in H.pylori infection prevalence rates among ethnic groups(highest in Indians adults,followed Chinese and low in Malays,P<0.05).Conclusions: The overall prevalence of H.pylori did increase with age group across ethnicity and gender,in Northern Peninsular Malaysia.Sreenivasan Sasidharan Subramanion Jo Thy Lachumy Manickam Ravichandran Lachimanan Yoga Latha Surasa Rao Surya Gegu 2011Asian Pacific Journal of Tropical Medicine2011,4,1:11
2Antimicrobial drug resistance of Staphylococcus aureus in dairy products显示文摘Objective:To evaluate the prevalence of multidrug resistant Staphylococcus aureus(S.aureus) in dairy products.Methods:Isolation and identification of S.aureus were performed in 3 dairybased food products.The isolates were tested for their susceptibility to 5 different common antimicrobial drugs.Results:Of 50 samples examined,5(10%) were contaminated with 5. aureus.Subsequently,the 5 isolates were subjected to antimicrobial resistance pattern using five antibiotic discs(methicillin,vancomycin,kanamycin,chloramphenicol and tetracycline).Sample 29 showed resistance to methicillin and vancomycin.Sample 18 showed intermediate response to tetracycline.The other samples were susceptible to all the antibiotics tested.Conclusions:The results provide preliminary data on sources of food contamination which may act as vehicles for the transmission of antimicrobial-resistant Staphylococcus.Therefore,it enables us to develop preventive strategies to avoid the emergence of new strains of resistant S.aureus.Sasidharan S Prema B Yoga Latha L 2011Asian Pacific Journal of Tropical Biomedicine2011,1,2:10
3Antioxidant activity and phytochemical screening of the methanol extracts of Euphorbia hirta L显示文摘Objective:To assess antioxidant activities of different parts of Euphorbia hirta(E.hirta),and to search for new sources of safe and inexpensive antioxidants.Methods:Samples of leaves,stems, flowers and roots from E.hirta were tested for total phenolic content,and flavonoids content and in vitro antioxidant activity by diphenyl-1-picrylhydrazyl(DPPH) assay and reducing power was measured using cyanoferrate method.Results:The leaves extract exhibited a maximum DPPH scavenging activity of(72.96±0.78)%followed by the flowers,roots and stems whose scavenging activities were(52.45±0.66)%,(48.59±0.97)%,and(44.42±0.94)%,respectively.The standard butylated hydroxytoluene(BHT) was(75.13±0.75)%.The IC50,for leaves,flowers,roots,stems and BHT were 0.803,0.972,0.989,1.358 and 0.794 mg/mL,respectively.The reducing power of the leaves extract was comparable with that of ascorbic acid and found to be dose dependent. Leaves extract had the highest total phenolic content[(206.17±1.95) mg GAE/g],followed by flowers,roots and stems extracts which were(117.08±3.10) mg GAE/g,(83.15±1.19) mg GAE/g,and (65.70±1.72) mg GAE/g,respectively.On the other hand,total flavonoids content also from leave had the highest value[(37.970±0.003) mg CEQ/g],followed by flowers,roots and stems extracts which were(35.200±0.002) mg CEQ/g,(24.350±0.006) mg CEQ/g,and(24.120±0.004) mg CEQ/ g,respectively.HPTLC bioautography analysis of phenolic and antioxidant substance revealed phenolic compounds.Phytochemical screening of E.hirta leaf extract revealed the presence of reducing sugars,terpenoids,alkaloids,steroids,tannins,flavanoids and phenolic compounds. 5Pharmacognostic studies of insect gall of Quercus infectoria Olivier(Fagaceae)显示文摘Objective:To study the detailed pharmacognostic profile of galls of Quercus infectoria Olivier(Q.infectoria olivier)(Fagaceae),an important medicinal plant used in the Indian system of medicine.Methods:Samples of galls of Q.infectoria were studied by macroscopical,microscopical,physiochemical,phytochemical.fluorescence analysis and othjer methods for standardization as recommended by WHO.Results:Macroscopically,the crude drug is globose with horny appearances on external surface(1.4-2.3 cm in length and 1-1.5 cm in diameter),with greyish-brown to brownish-black in colour externally and dark brown buff colored.Surface is smooth with numerous horny protuberances giving rough touch,and witfi unpleasant odour.Microscopically,a wide zone of radially elongated parenchyma cells between upper and lower epidermis were found.The vascular strands were present at all places and radially elongated sclerides touched the lower epidermis.In physico-chemical studies,the moisture,total ash,acid insoluble ash,alcohol soluble,water soluble,petroleum ether,chloroform extractive value and tannin content were found to be 2.790,5.020,0.110,38.780,41.210,0.402,1.590 and 49.200 percentage respectively.Prcliminary phytochemical screening showed the presence of phenols,flavonoids,steroids,triterpenes,tannins,saponins and alkaloids.Cooclusions:The results of the present study serve as a valuable source of information and provide suitable standards for identification of this medicinally important plant drug material for future investigations and applications.Savitri Shrestha Vasuki Srinivas Kaushik Ravi Shankara Birur Eshwarappa Sundara Rajan Subaramaihha Latha Muuaiah Ramanna Dhananjaya Bhadrapura Lakkappa 2014Asian Pacific Journal of Tropical Biomedicine2014,4,1:4

7Pirfenidone inhibits epithelial–mesenchymal transition in keloid keratinocytes显示文摘Background:Keloids are benign fibroproliferative skin lesions that are difficult to treat and become a lifetime predicament for patients.Several treatment modalities have been put forth,but as yet no satisfactory approach to the prevention or treatment of keloids has been identified.The process of epithelial-to-mesenchymal transition(EMT)has been implicated in keloid scarring,as keloid keratinocytes display an EMT-like phenotype.This study investigated the potential of pirfenidone,an antifibrotic agent,to counteract EMT-like alterations in keloid keratinocytes,including gene expression,cell migratory and proliferative functions.Methods:Normal and keloid keratinocytes were isolated from discarded normal skin tissues and from resected keloid tissues,respectively.Cells were quiesced for 24 h without epidermal growth factor DS-Qi1MCDigital and were exposed to transforming growth factor-beta1(TGF-β1;10 ng/mL),with or without pirfenidone(400μg/mL),for an additional 24 h.The effects of pirfenidone on cytotoxicity,cell migration,cell proliferation,and on expression of genes and proteins involved in EMT were assayed.Statistical significance was determined by two-way ANOVA using Sigma Plot.Results:We found that pirfenidone did not elicit any cytotoxic effect at concentrations up to 1000μg/mL.A statistically significant dose-dependent decrease in basal cell proliferation rate was noted in both normal and keloid keratinocytes when exposed to pirfenidone at concentrations ranging from 200 to 1000μg/mL.Pirfenidone significantly decreased basal cell migration in both normal and keloid keratinocytes,but a significant decrease in TGF-β1-induced cell migration was seen only in keloid keratinocytes.Significant inhibition of the expression of TGF-β1-induced core EMT genes,namely hyaluronan synthase 2,vimentin,cadherin-11,and wingless-type MMTV integration site family,member 5A along with fibronectin-1,was observed in both normal and keloid keratinocytes treated with pirfenidone.In addition,the protein levels of vimentin and fibronectin were significantly reduced by pirfenidone(400μg/mL)in both normal and keloid keratinocytes.Conclusions:For the first time,this study shows the efficacy of pirfenidone in inhibiting the EMTlike phenotype in keratinocytes derived from keloids,suggesting that pirfenidone may counteract a critical contributor of keloid progression and recurrence.Latha Satish Alexander Evdokiou Eleni Geletu Jennifer M.Hahn Dorothy M.Supp 2020Burns & Trauma2020,8,1:3
8Integrated lipidomics and proteomics network analysis highlights lipid and immunity pathways associated with Alzheimer's disease显示文摘Background:There is an urgent need to understand the pathways and processes underlying Alzheimer's disease(AD)for early diagnosis and development of effective treatments.This study was aimed to investigate Alzheimer's dementia using an unsupervised lipid,protein and gene multi-omics integrative approach.Methods:A lipidomics dataset comprising 185 AD patients,40 mild cognitive impairment(MCI)individuals and 185 controls,and two proteomics datasets(295 AD,159 MCI and 197 controls)were used for weighted gene CO-expression network analyses(WGCNA).Correlations of modules created within each modality with clinical AD diagnosis,brain atrophy measures and disease progression,as well as their correlations with each other,were analyzed.Gene ontology enrichment analysis was employed to examine the biological processes and molecular and cellular functions of protein modules associated with AD phenotypes.Lipid species were annotated in the lipid modules associated with AD phenotypes.The associations between established AD risk loci and the lipid/protein modules that showed high correlation with AD phenotypes were also explored.Results:Five of the 20 identified lipid modules and five of the 17 identified protein modules were correlated with clinical AD diagnosis,brain atrophy measures and disease progression.The lipid modules comprising phospholipids,triglycerides,sphingolipids and cholesterol esters were correlated with AD risk loci involved in immune response and lipid metabolism.The five protein modules involved in positive regulation of cytokine production,neutrophil-mediated immunity,and humoral immune responses were correlated with AD risk loci involved in immune and complement systems and in lipid metabolism(the APOE ε4 genotype).Conclusions:Modules of tightly regulated lipids and proteins,drivers in lipid homeostasis and innate immunity,are strongly associated with AD phenotypes.Jin Xu Giulia Bankov Min Kim Asger Wretlind Jodie Lord Rebecca Green Angela Hodges Abdul Hye Dag Aarsland Latha Velayudhan Richard J.B.Dobson Petroula Proitsi Cristina Legido-Quigley on behalf of the AddNeuroMed Consortium 2020Translational Neurodegeneration2020,9,3:3
9Evaluation of rapid molecular diagnostics for differentiating medicinal Kaempferia species from its adulterants显示文摘Accurate detection of unique herbs is crucial for herbal medicine preparation. Zingiberaceae species, which are important in Ayurvedic medicine of India, are often misidentified in Northeast (NE) Indian herbal markets. Kaempferia galanga (Zingiberaceae) is one of the major components of popular Ayurvedic drugs used for rheumatic diseases (i.e.,“Gandha Thailam” and “Rasnairandadi Kashayam”), contusions, fractures, and sprains. In NE India, herbal healers often misidentify plants from the Marantaceae family (e.g., Calathea bachemiana and Maranta leuconeura) as Kaempferia, which leads to adulteration of the medicinal herb. This misidentification of herbs occurs in NE India because Zingiberaceae plant barcoding information is inadequate. As a consequence, herbal medicine is not only therapeutically less effective but may also cause adverse reactions that range from mild to life-threatening. In this study, we used eight barcoding loci to develop “fingerprints” for four Kaempferia species and two species frequently mistaken for Kaempferia. The PCR and sequencing success of the loci matK, rbcL and trnH-psbA were found to be 100%;the combination of matK, rbcL, and trnH-psbA proved to be the ideal locus for discriminating the Kaempferia species from their adulterants because the combined loci showed greater variability than individual loci. Hook.f.& Thoms.roots(ACP) on APAP/CC1_4 induced liver toxicity in Wistar rats and in vitro free radical scavenging property显示文摘Objective:To evaluate the hepatoprotective and antioxidant properties of alkaloid extract of Cyclea peltata(C.peltata) against paracetamol/carbon tetra chloride induced liver damage in Wistar rats.Methods:In viro paracetamol/carbon tetrachloride induced liver damage in Wistar rats,in vitro free radical scavenging studies,HPTLC estimation of letrandrine and direct analysis in real timemass spectrometry of alkaloid extract of C.peltata were used for the validation.Results:The results showed that pretreatment with alkaloid extract of C.peltata caused significant reduction of serum glutamate pyruvate transaminase,serum glutamate oxaloacetate transaminase,serum alkaline phosphatase,serum cholesterol,liver malondialdehyde levels.The reduced glutathione,catalase,superoxide dismutase levels in liver were increased with alkaloid extract of C.peltata treatment.These results were almost comparable to silymarin and normal control.Histopathological studies also substantiated the biochemical findings.The in vitro hydroxyl,superoxide and DPPH scavenging study of alkaloid extract of C. peltata showed significant free radical scavenging property.Conclusions:The hepatoprotective property of alkaloid extract of C.peltata against paracetamol/carbon tetrachloride may be due the synergistic action of alkaloids especially tetrandrine,fangchinoline through free radical scavenging and thus preventing oxidative stress.Varghese Jancy Shine Panikamparambil Gopalakrishnan Latha Somasckharan Nair Rajam Suja Gangadharan Indira Anuja Gopan Raj Sreedharan Nair Rajasekharan 2014Asian Pacific Journal of Tropical Biomedicine2014,4,2:2

14Risk factors for endophthalmitis following cataract surgery-our experience at a tertiary eye care centre in India显示文摘AIM: To determine the risk factors for acute endophthalmitis after cataract extraction in a tertiary care centre in India.·METHODS: We performed a nested case control study within a retrospective cohort. The surgical records of all patients with clinically diagnosed endophthalmitis within one month after cataract surgery, performed between January 2006 and December 2009, were reviewed. These were compared with randomly selected age and gender-matched controls, from patients having routine cataract surgery within ±1wk of the endophthalmitis case.Univariable and multivariable analysis were performed to identify risk factors for endophthalmitis.· RESULTS: Of the total 33 856 cataract surgeries performed during this period, there were 57 cases of postoperative acute endophthalmitis that met our study criteria. Thus, the overall incidence of endophthalmitis in our cohort was 1.6 per 1000 cataract extractions performed. Mean age of cases was 55.9y(SD: 10.9y) and for controls was 55.6y(SD: 9.8y). Thirty-five cases(61.4%)and 133 controls(59.6%) were males. Median time of onset of endophthalmitis was 4d(IQR 2-9d; range: 1-30d).Thirty-nine cases(68.4%) presented within 7d and 27 cases(47.4%) were culture positive. Two hundred and twenty-three age and gender matched controls were selected. In multivariate analysis, endophthalmitis was associated with posterior capsular rupture(PCR) during surgery(OR 6.98, 95%CI: 2.22-21.98), phacoemulsification via scleral incision with a foldable intraocular lens(IOL)implantation(OR 3.02, 95%CI: 1.13-8.04) and ocular comorbidity(OR 2.32, 95%CI: 1.11-4.87).·CONCLUSION: PCR, presence of ocular co-morbidity,and phacoemulsification via scleral incision with foldable-IOL were found to be independent risk factors for acute endophthalmitis.Rohit C.Khanna Vanita Pathak Ray Madhavi Latha Sandra D Cassard Annie Mathai Garudadri C Sekhar 2015International Journal of Ophthalmology(English edition)2015,8,6:2
